The effect of dissolved oxygen concentration on long-term stability of partial nitrification process显示文摘Dissolved oxygen(DO)concentration is regarded as one of the crucial factors to influence partial nitrification process. However, achieving and keeping stable partial nitrification under different DO concentrations were widely reported. The mechanism of DO concentration influencing partial nitrification is still unclear. Therefore,in this study two same sequencing batch reactors(SBRs)cultivated same seeding sludge were built up with realtime control strategy.Different DO concentrations were controlled in SBRs to explore the effect of DO concentration on the long-term stability of partial nitrification process at room temperature. It was discovered that ammonium oxidation rate(AOR)was inhibited when DO concentration decreased from 2.5 to 0.5 mg/L.The abundance of Nitrospira increased from 1011.5 to 1013.7 copies/g DNA,and its relative percentage increased from 0.056%to 3.2%during 190 operational cycles, causing partial nitrification gradually turning into complete nitrification process. However, when DO was 2.5 mg/L the abundance of Nitrospira was stable and AOB was always kept at 1010.7 copies/g DNA.High AOR was maintained, and stable partial nitrification process was kept.Ammonia oxidizing bacteria(AOB)activity was significantly higher than nitrite oxidizing bacteria(NOB)activity at DO of 2.5 mg/L,which was crucial to maintain excellent nitrite accumulation performance.Bin Cui Qing Yang Xiuhong Liu Siting Huang Yubing Yang Zhibin Liu 2020Journal of Environmental Sciences2020,32,4:12
Cooperative Spectrum Allocation with QoS Support in Cognitive Cooperative Vehicular Ad Hoc Networks显示文摘To solve the contradiction between the increasing demand of diverse vehicular wireless applications and the shortage of spectrum resource,a novel cognitive cooperative vehicular ad-hoc network(CCVANET) framework is proposed in this paper.Firstly,we develop an adaptive cognitive spectrum sensing(ACSS) mechanism which can help to trigger and adjust the spectrum sensing window according to network traffic load status and communication quality. And then,Generalized Nash Bargaining Solution(GNBS),which can achieve a good tradeoff between efficiency and weighted fairness,is proposed to formulate the asymmetric intercell resource allocation.Finally,GNBSSafety(GNBS-S) scheme is developed to enhance the Quality of Service(QoS) of safety applications, especially in the heavy load status, where the bandwidth demanded and supplied cannot be matched well.Furthermore, the primary user activity(PUA)which can cause rate loss to secondary users,is also considered to alleviate its influence to fairness.Simulation results indicate that the proposed CC-VANET scheme can greatly improve the spectrum efficiency and reduce the transmission delay and packet loss rate on the heavy contention status. And GNBS spectrum allocation scheme outperforms both the Max-min and Max-rate schemes, and can enhance the communication reliability of safety service considerably in CC-VANET.ZHANG Lei LUO Tao LIU Wei ZHU Siting LI Jianfeng 2014China Communications2014,11,10:5
Genesis of the Changba–Lijiagou Giant Pb-Zn Deposit, West Qinling, Central China: Constraints from S-Pb-C-O isotopes显示文摘The extensive Changba-Lijiagou Pb-Zn deposit is located in the north of the Xihe–Chengxian ore cluster in West Qinling. The ore bodies are mainly hosted in the marble, dolomitic marble and biotite-calcite-quartz schist of the Middle Devonian Anjiacha Formation, and are structurally controlled by the fault and anticline. The ore-forming process can be divided into three main stages, based on field geological features and mineral assemblages. The mineral assemblages of hydrothermal stage I are pale-yellow coarse grain, low Fe sphalerite, pyrite with pits, barite and biotite. The mineral assemblages of hydrothermal stage II are black-brown cryptocrystalline, high Fe shalerite, pyrite without pits, marcasite or arsenopyrite replace the pyrite with pits, K-feldspar. The features of hydrothermal stage III are calcite-quartz-sulfide vein cutting the laminated, banded ore body. Forty-two sulfur isotope analyses, twenty-five lead isotope analyses and nineteen carbon and oxygen isotope analyses were determined on sphalerite, pyrite, galena and calcite. The δ34 S values of stage I(20.3 to 29.0‰) are consistent with the δ34 S of sulfate(barite) in the stratum. Combined with geological feature, inclusion characteristics and EPMA data, we propose that TSR has played a key role in the formation of the sulfides in stage I. The δ34 S values of stage II sphalerite and pyrite(15.1 to 23.0‰) are between sulfides in the host rock, magmatic sulfur and the sulfate(barite) in the stratum. This result suggests that multiple S reservoirs were the sources for S2-in stage II. The δ34 S values of stage III(13.1 to 22‰) combined with the structure of the geological and mineral features suggest a magmatic hydrothermal origin of the mineralization. The lead isotope compositions of the sulfides have 206 Pb/204 Pb ranging from 17.9480 to 17.9782, 207 Pb/204 Pb ranging from 15.611 to 15.622, and 208 Pb/204 Pb ranging from 38.1368 to 38.1691 in the three ore-forming stages. The narrow and symmetric distributions of the lead isotope values reflect homogenization of granite and mantle sources before the Pb-Zn mineralization. The δ13 CPDB and δ18 OSMOW values of stage I range from-0.1 to 2.4‰ and from 18.8 to 21.7‰. The values and inclusion data indicate that the source of fluids in stage I was the dissolution of marine carbonate. The δ13 CPDB and δ18 OSMOW values of stage II range from-4 to 1‰ and from 12.3 to 20.3‰, suggesting multiple C-O reservoirs in the Changba deposit and the addition of mantle-source fluid to the system. The values in stage III are-3.1‰ and 19.7‰, respectively. We infer that the process of mineralization involved evaporitic salt and sedimentary organic-bearing units interacting through thermochemical sulfate reduction through the isotopic, mineralogy and inclusion evidences. Subsequently, the geology feature, mineral assemblages, EPMA data and isotopic values support the conclusion that the ore-forming hydrothermal fluids were mixed with magmatic hydrothermal fluids and forming the massive dark sphalerite, then yielding the calcite-quartz-sulfide vein ore type at the last stage. The genesis of this ore deposit was epigenetic rather than the previously-proposed sedimentary-exhalative(SEDEX) type.WEI Ran WANG Yitian MAO Jingwen HU Qiaoqing QIN Siting LIU Shengyou YE Dejin YUAN Qunhu DOU Ping 2020Acta Geologica Sinica(English Edition)2020,94,4:4
Equilibrium and kinetic Si isotope fractionation factors and their implications for Si isotope distributions in the Earth's surface environments显示文摘Several important equilibrium Si isotope fractionation factors among minerals, organic molecules and the H_4SiO_4 solution are complemented to facilitate the explanation of the distributions of Si isotopes in Earth's surface environments. The results reveal that,in comparison to aqueous H_4SiO_4,heavy Si isotopes will be significantly enriched in secondary silicate minerals. On the contrary,quadra-coordinated organosilicon complexes are enriched in light silicon isotope relative to the solution. The extent of ^(28)Si-enrichment in hyper-coordinated organosilicon complexes was found to be the largest. In addition, the large kinetic isotope effect associated with the polymerization of monosilicic acid and dimer was calculated, and the results support the previous statement that highly ^(28)Sienrichment in the formation of amorphous quartz precursor contributes to the discrepancy between theoretical calculations and field observations. With the equilibrium Si isotope fractionation factors provided here,Si isotope distributions in many of Earth's surface systems can be explained. For example, the change of bulk soil δ^(30)Si can be predicted as a concave pattern with respect to the weathering degree, with the minimum value where allophane completely dissolves and the total amount of sesquioxides and poorly crystalline minerals reaches their maximum. When, under equilibrium conditions, the well-crystallized clays start to precipitate from the pore solutions, the bulk soil δ^(30)Si will increase again and reach a constant value.Similarly, the precipitation of crystalline smectite and the dissolution of poorly crystalline kaolinite may explain the δ^(30)Si variations in the ground water profile. The equilibrium Si isotope fractionations among the quadracoordinated organosilicon complexes and the H_4SiO_4solution may also shed light on the Si isotope distributions in the Si-accumulating plants.Hong-tao He Siting Zhang Chen Zhu Yun Liu 2016Acta Geochimica2016,35,1:2
Truncated glycoprotein E of varicella-zoster virus is an ideal immunogen for Escherichia coli-based vaccine design显示文摘Varicella-zoster virus(VZV)is a highly infectious agent responsible for both varicella and herpes zoster disease.Despite high efficacy, there remain safety and accessibility concerns with the licensed vaccines. Here,we sought to produce a VZV g E immunogen using an E.coli expression system. We found that the soluble expression and yield of g E protein could be enhanced via C-terminal truncations to the protein, thereby facilitating a robust and scalable purification process for the purpose of vaccine manufacturing. The lead truncated g E(aa 31–358),hereafter referred to as tg E,was a homogenous monomer in solution and showed excellent antigenicity. Finally,we assessed and compared the immunogenicity of tg E with commercial v Oka LAV and Shingrix vaccine. We found that aluminum-adjuvanted tg E was immunogenic as compared with v Oka LAV.When adjuvanted with AS01B,a two-dose immunization of tg E showed comparable or better potency in antibody responses and cell-mediated immunity with those of the Shingrix vaccine at the same dosage, especially in terms of the proportion of IFN-γ-expressing CD4^(+)T cells. In conclusion, this method of E.coli-mediate tg E expression offers a cost-effective and scalable strategy to generate an ideal VZV g E immunogen for the development of both varicella and zoster vaccines.Tingting Chen Jie Sun Sibo Zhang Tingting Li Liqin Liu Wenhui Xue Lizhi Zhou Siting Liang Zhili Yu Qingbing Zheng Hai Yu Tong Cheng Jun Zhang Ying Gu Shaowei Li Ningshao Xia 2023Science China(Life Sciences)2023,66,4:1

Grafting Propagation Techniques of Red Sandalwood(Pterocarpus santalinus),a Precious Hongmu Tree Species显示文摘[Objectives] This article aimed to study the grafting propagation techniques of red sandalwood ( Pterocarpus santalinus ), so as to provide technical support for the effective propagation of seedlings of this precious Hongmu species.[Methods] Based on the analysis and comparison of the growth performance of the Hongmu tree species introduced to China, taking 1 to 2-year-old Pterocarpus indicus seedlings as rootstocks and single-bud stems of P. santalinus as scions, grafting was carried out from January to April using three grafting propagation methods, cut grafting propagation, skin grafting propagation and oblique grafting propagation.[Results] The results of several years of experiments show that in early spring, P. santalinus could be propagated well using the grafting propagation methods of skin grafting propagation and oblique grafting propagation, with survival rate over 80%. For rootstocks with young age or thin stems, the joint unions of the oblique grafting propagation healed well, without apparent protrusions, unlike those of skin grafting propagation. The survival rate of oblique grafting propagation that is fast and reliable was higher than that of cut grafting propagation.[Conclusions] It is feasible to use the grafting propagation methods to propagate P. santalinus seedlings. This is a traditional and innovative technology path in the propagation of high-quality seedlings of precious Hongmu trees, and can be promoted and applied in industrial practice.Xiaonan HUANG Yanming LI Yiqing LI Tingting LIU Siting CHEN Chengxiang XU Yingshan QIU Yesheng LIANG 2019Asian Agricultural Research2019,11,7:1
Design and Analysis of Wide Speed Regulation of Variable Leakage Flux Reverse Salient-pole Motor显示文摘In this article, a new variable leakage flux reverse salient-pole motor(VLF-RSPM) is raised to widen the speed range. The innovation is to realize both reverse salient-pole characteristics and variable leakage flux characteristics by using a method of adding magnetic bridges and magnetic barriers. Firstly, the evolution of the topological structure and working principle of the motor are introduced. Secondly, based on 2D Finite Element Analysis(FEA), the electromagnetic properties and noise of the motor are analyzed in detail, and the electromagnetic properties are contrasted with that of the conventional V-type synchronous motor(CVTSM). The results show that VLF-RSPM has the advantages of small torque ripple, strong magnetic weakening ability, low noise, high efficiency, and low risk of permanent magnet demagnetization under different conditions. In addition, it is verified that the proposed motor extends the speed range.Xiping Liu Siting Zhu Dabin Liu Jianwei Liang 2023CES Transactions on Electrical Machines and Systems2023,7,3:0
RIP1/3-dependent programmed necrosis induces intestinal injuryin septic rats显示文摘The regulation of various types of cell death may help to restore the normal physiological function of cells and play a protective role in sepsis. In the current study,we explore the role of programmed cell necrosis in sepsis and the underlying mechanisms. The septic rat model is established by Cecal-ligation and perforation(CLP),and the in vitro model is established by LPS in IEC-6 cells. Our results demonstrate that receptor-interacting protein 1(RIP1)is significantly upregulated in the ileum of septic rats and LPS-treated IEC-6 cells at both the mRNA and protein levels.Nec-1,an inhibitor of RIP1,reduces the protein levels of RIP1,p-RIP3,and phosphorylated mixed-lineage kinase domain-like(MLKL)(serine 358)and relieves intestinal injury in CLP-induced septic rats with decreased IL-6 and TNF-αlevels.The in vitro experiments further reveal that LPS induces the colocalization of RIP1 and RIP3,resulting in the phosphorylation and translocation of MLKL to the plasma membrane in IEC-6 cells.LPS also facilitates ROS production in IEC-6 cells, but this effect is further reversed by Nec-1,si-RIP1 and si-RIP3.Furthermore,LPS-induced necrosis in IEC-6 cells is counteracted by NAC.Thus,we conclude that RIP1/RIP3-dependent programmed cell necrosis participates in intestinal injury in sepsis and may be associated with RIP1/RIP3-mediated ROS.Siting Ye Xinxin Qi Yuxiao Liu Liangming Zhuang Zhongmin Gu 2024Acta Biochimica et Biophysica Sinica2024,56,1:0
Deep learning for fast channel estimation in millimeter-wave MIMO systems显示文摘Channel estimation has been considered as a key issue in the millimeter-wave(mmWave)massive multi-input multioutput(MIMO)communication systems, which becomes more challenging with a large number of antennas. In this paper,we propose a deep learning(DL)-based fast channel estimation method for mmWave massive MIMO systems. The proposed method can directly and effectively estimate channel state information(CSI)from received data without performing pilot signals estimate in advance, which simplifies the estimation process.Specifically,we develop a convolutional neural network(CNN)-based channel estimation network for the case of dimensional mismatch of input and output data, subsequently denoted as channel(H)neural network(HNN).It can quickly estimate the channel information by learning the inherent characteristics of the received data and the relationship between the received data and the channel, while the dimension of the received data is much smaller than the channel matrix.Simulation results show that the proposed HNN can gain better channel estimation accuracy compared with existing schemes.LYU Siting LI Xiaohui FAN Tao LIU Jiawen SHI Mingli 2022Journal of Systems Engineering and Electronics2022,33,6:0
Supercontinuum fiber laser-based coherent anti-Stokes Raman scattering microscopy for label-free chemical imaging显示文摘Coherent anti-Stokes Raman scattering(CA RS)microscopy can resolve the chemical compo-nents and distribution of living biological systems in a label-firee manner and is favored in several disciplines.Current CA RS microscopes typically use bulky,high-performance solid-state lasers, which are expensive and sensitive to environmental changes. With their relatively low cost and environmental sensitivity, supercontinum fiber(SF)lasers with a small footprint have found increasing use in biomedical applications.Upon these features,in this paper,we homebuilt a low-cost CARS microscope based on a SF laser module(scCA RS microscope). This SF laser module is specially customized by adding a time synchronized seed source channel to the SF laser to form a dual-channel output laser. The performance of the scCARS microscope is evaluated with dimethyl sulfoxide, whose results confirm a spatial resolution of better than 500nm and a detection sensitivity of millimolar concentrations. The dual-color imaging capability is further demonstrated by imaging different species of mixed microspheres. We finally explore the potential of our scCARS microscope by mapping lipid droplets in different cancer cells and corneal stromal lenses.Nan Wang Jiaojiao Zhang Wangting Zhou Siting Liu Jing Li Xinyi Xu Qi Zeng Yong Li Shouping Zhu Xueli Chen 2022Journal of Innovative Optical Health Sciences2022,15,4:0
Investigation of an Intensifying-flux Variable Flux-leakage Interior Permanent Magnet Machine for Wide Speed Range显示文摘In this paper,a novel intensifying-flux variable flux-leakage interior permanent magnet(IFVF-IPM)machine is proposed,in which flux barriers were designed deliberately between the adjacent poles to obtain intensifying-flux effect and variable flux-leakage property. The rotor topology and design principles of the proposed machine are also introduced. Then,a multi-objective optimization method is adopted based on the sensitivity analysis, and some design variables of IFVF-IPM machine with strong sensitivity are selected to optimization progress by using the non-dominated sorting genetic algorithm-Ⅱ(NSGA-Ⅱ).Moreover,the electromagnetic characteristics of conventional IPM machine, conventional IFVF-IPM machine(CIFVF-IPM)and the novel IFVF-IPM machine are compared based on the finite element analysis(FEA)method which includes flux linkage, inductances characteristic,torque-speed envelops and power characteristic,as well as evaluation of the risk of irreversible demagnetization. Finally, the experiment results show that the IFVF-IPM machine has a better performance in flux weakening capability for wide speed range and a lower risk of irreversible demagnetization, which indicates the validity and feasibility of the proposed machine.Xiping Liu Dabin Liu Siting Zhu Jianwei Liang 2022CES Transactions on Electrical Machines and Systems2022,6,2:0
Transcriptomics reveals substance biosynthesis and transport on membranes of Listeria monocytogenes affected by antimicrobial lipopeptide brevilaterin B显示文摘Listeria monocytogenes is a worrisome food-borne pathogen threatening global food safety. Our previous study proved that lipopeptide brevilaterin B showed efficient antibacterial activity against L.monocytogenes by interacting with the cell membrane. This research further explored the antibacterial mechanism of brevilaterin B against L.monocytogenes at the sub-minimum inhibition concentration via transcriptomic analysis.Brevilaterin B induced growth inhibition rather than direct membrane lysis in L.monocytogenes at the minimum inhibitory concentration.Transcriptomic analysis showed 1779 difference expressed genes, including 895 up-regulated and 884 down-regulated genes.Gene Ontology and Kyoto Encyclopedia of Genes and Genomes analysis indicated that brevilaterin B influenced multiple pathways of L.monocytogenes, including peptidoglycan biosynthesis, membrane transport(ATP-binding cassette transports, ion transport),cellular metabolism(amino acid and lipid metabolism),ATP synthesis, and activation of the stress response(quorum sensing and bacterial chemotaxis). In conclusion, brevilaterin B affects gene expression related to biosynthesis, transport and stress response pathways on the membrane of L.monocytogenes.The present work provides the first transcriptomic assessment of the antibacterial mechanism of lipopeptide brevilaterin B at the gene level.Yangliu Liu Yawei Ning Zhou Chen Panpan Han Tongxin Zhi Siting Li Aijin Ma Yingmin Jia 2023Food Science and Human Wellness2023,12,4:0

Selective deletion of zinc transporter 3 in amacrine cells promotes retinal ganglion cell survival and optic nerve regeneration after injury显示文摘Vision depends on accurate signal conduction from the retina to the brain through the optic nerve,an important part of the central nervous system that consists of bundles of axons originating from retinal ganglion cells. The mammalian optic nerve,an important part of the central nervous system, cannot regenerate once it is injured, leading to permanent vision loss.To date, there is no clinical treatment that can regenerate the optic nerve and restore vision. Our previous study found that the mobile zinc(Zn^(2+))level increased rapidly after optic nerve injury in the retina, specifically in the vesicles of the inner plexiform layer.Furthermore, chelating Zn^(2+)significantly promoted axonal regeneration with a long-term effect. In this study,we conditionally knocked out zinc transporter 3(ZnT3)in amacrine cells or retinal ganglion cells to construct two transgenic mouse lines(VGAT^(Cre)ZnT3^(fl/fl)and VGLUT2^(Cre)ZnT3^(fl/fl),respectively).We obtained direct evidence that the rapidly increased mobile Zn^(2+)in response to injury was from amacrine cells. We also found that selective deletion of ZnT3 in amacrine cells promoted retinal ganglion cell survival and axonal regeneration after optic nerve crush injury, improved retinal ganglion cell function, and promoted vision recovery.Sequencing analysis of reginal ganglion cells revealed that inhibiting the release of presynaptic Zn^(2+)affected the transcription of key genes related to the survival of retinal ganglion cells in postsynaptic neurons, regulated the synaptic connection between amacrine cells and retinal ganglion cells, and affected the fate of retinal ganglion cells. These results suggest that amacrine cells release Zn^(2+)to trigger transcriptomic changes related to neuronal growth and survival in reginal ganglion cells, thereby influencing the synaptic plasticity of retinal networks. These results make the theory of zinc-dependent retinal ganglion cell death more accurate and complete and provide new insights into the complex interactions between retinal cell networks.Zhe Liu Jingfei Xue Canying Liu Jiahui Tang Siting Wu Jicheng Lin Jiaxu Han Qi Zhang Caiqing Wu Haishun Huang Ling Zhao Yehong Zhuo Yiqing Li 2023Neural Regeneration Research2023,18,12:0
Size Effects of Atomically Precise Gold Nanoclusters in Catalysis显示文摘The emergence of ligand-protected,atomically precise gold nanoclusters(NCs)in recent years has attracted broad interest in catalysis due to their well-defined atomic structures and intriguing properties.Especially, the precise formulas of NCs provide an opportunity to study the size effects at the atomic level without complications by the polydispersity in conventional nanoparticles that obscures the relationship between the size/structure and properties.Herein,we summarize the catalytic size effects of atomically precise,thioate-protected gold NCs in the range of tens to hundreds of metal atoms. The catalytic reactions include electrochemical catalysis, photocatalysis, and thermocatalysis. With the precise sizes and structures, the fundamentals underlying the size effects are analyzed, such as the surface area, electronic properties, and active sites. In the catalytic reactions, one or more factors may exert catalytic effects simultaneously, hence leading to different catalytic-activity trends with the size change of NCs.The summary of literature work disentangles the underlying fundamental mechanisms and provides insights into the size effects.Future studies will lead to further understanding of the size effects and shed light on the catalytic active sites and ultimately promote catalyst design at the atomic level.Site Li Xiangsha Du Zhongyu Liu Yingwei Li Yucai Shao Rongchao Jin 2023Precision Chemistry2023,1,1:0
Intratumoral Bacteria Dysbiosis Is Associated with Human Papillary Thyroid Cancer and Correlated with Oncogenic Signaling Pathways显示文摘Emerging evidence suggests that microbial dysbiosis plays vital roles in many human cancers. However, knowledge of whether the microbial community in thyroid tumor is related to tumorigenesis remains elusive. In this study,we aimed to explore the microbial community in thyroid tissues and its contribution to papillary thyroid cancer(PTC).In parallel,we performed microbial profiling and transcriptome sequencing in the tumor and adjacent normal tissues of a large cohort of 340 PTC and benign thyroid nodule(BTN)patients.Distinct microbial signatures were identified in PTC,BTN,and their adjacent nontumor tissues.Intra-thyroid tissue bacteria were verified by means of bacteria staining, fluorescence in situ hybridization, and immunoelectron microscopy. We found that 17 bacterial taxa were differentially abundant in PTC compared with BTN,which included enrichment in PTC of the pathobionts Rhodococcus,Neisseria,Streptococcus,Halomonas, and Devosia, and depletion of the beneficial bacteria Amycolatopsis. These differentially abundant bacteria could differentiate PTC tumor tissues(PTC-T)from BTN tissues(BTN-T)with an area under the curve(AUC)of 81.66%.Microbial network analysis showed increased correlation strengths among the bacterial taxa in PTC-T in comparison with BTN-T.Immunefunction-corresponding bacteria(i.e.,Erwinia,Bacillus,and Acinetobacter)were found to be enriched in PTC with Hashimoto’s thyroiditis. Moreover, our integrative analysis revealed that the PTC-enriched bacteria had a positive association with key PTC-oncogenic pathway-related genes, including BRAF,KRAS,IRAK4,CTNNB1,PIK3CA,MAP3K7,and EGFR.In conclusion, our results suggest that intratumor bacteria dysbiosis is associated with the thyroid tumorigenesis and oncogenic signaling pathways of PTC.Shuang Yu Yanqiang Ding Xuejie Wang Siu Kin Ng Siting Cao Weixin Liu Zhuming Guo Yubin Xie Shubin Hong Lixia Xu Xiaoxing Li Jie Li Weiming Lv Sui Peng Yanbing Li Joseph J.Y.Sung Jun Yu Haipeng Xiao 2023Engineering2023,,9:0

Dynamic Balance of pSTAT1 and pSTAT3 in C57BL/6 Mice Infected with Lethal or Nonlethal Plasmodium yoelii显示文摘Signal transducer and activator of transcription (STAT) proteins play an important role in cytokine signaling pathways and regulation of immune responses. The balance of the phosphorylated (activated) STAT1 (pSTAT1) and STAT3 (pSTAT3) has been documented in cancer immunology. In this study, we investigated the dynamic balance of pSTAT1 and pSTAT3 in C57BL/6 mice infected with either a nonlethal (Py17XNL) or lethal (Py17XL) strain of Plasmodium yoelii. Both Py17XNL and Py17XL infections induced a maximum activation of STAT1 and STAT3 on the first day after parasite inoculation. Additionally, the Py17XNL infection induced a pSTAT1-dominant response in mice during the early stage of infection, with the resolution of parasitemia. In contrast, Py17XL infection induced a pSTAT3-dominant response during the early phase of infection, with the death of the animals. Our results indicated that maximum activation of STAT1 and STAT3 occurred much earlier than the peak levels of cytokines induced by Plasmodium yoelii infection based on previous reports and that infection with Py17XNL and Py17XL induced different dynamic patterns of pSTAT1 and pSTAT3 balance.Xibao Shi Li Qin Guangjie Liu Siting Zhao Nanzheng Peng Xiaoping Chen 2008Cellular & Molecular Immunology2008,5,5:0
Chemical screening reveals Ronidazole is a superior prodrug to Metronidazole for nitroreductase-induced cell ablation system in zebrafish larvae显示文摘The Metronidazole(MTZ)/nitroreductase(NTR)-mediated cell ablation system is the most commonly used chemical-genetic cell ablation method in zebrafish. This system can specifically ablate target cells under spatial and temporal control. The MTZ/NTR system has become a widely used cell ablation system in biological, developmental, and functional studies. However, the inadequate cell-ablation ability of some cell types and the side effects of high concentration MTZ impede extensive applications of the MTZ/NTR system. In the present study, the US drug collection library was searched to extend the NTR system. Six MTZ analogs were found, and the cell-ablation ability of these analogs was tested in zebrafish larvae. The results revealed that two of the NTR substrates, Furazolidone and Ronidazole, ablated target cells more efficiently than MTZ at lower concentrations. Furthermore, the working concentration of Ronidazole, but not Furazolidone and MTZ, did not affect axonal bridge formation during spinal cord regeneration. Our results, taken together, indicate that Ronidazole is a superior prodrug to MTZ for the NTR system, especially for the study of neuron regeneration in zebrafish larvae.Siting Lai Ankita Kumari Jixiang Liu Yiyue Zhang Wenqing Zhang Kuangyu Yen Jin Xu 2021Journal of Genetics and Genomics2021,48,12:0
