12018 Chinese Pediatric Cardiology Society(CPCS) guideline for diagnosis and treatment of syncope in children and adolescents显示文摘Syncope belongs to the transient loss of consciousness(TLOC), characterized by a rapid onset, short duration, and spontaneous complete recovery. It is common in children and adolescents, accounting for 1% to 2% of emergency department visits.Recurrent syncope can seriously affect children's physical and mental health, learning ability and quality of life and sometimes cardiac syncope even poses a risk of sudden death. The present guideline for the diagnosis and treatment of syncope in children and adolescents was developed for guiding a better clinical management of pediatric syncope. Based on the globally recent development and the evidence-based data in China, 2018 Chinese Pediatric Cardiology Society(CPCS) guideline for diagnosis and treatment of syncope in children and adolescents was jointly prepared by the Pediatric Cardiology Society, Chinese Pediatric Society, Chinese Medical Association(CMA)/Committee on Pediatric Syncope, Pediatricians Branch, Chinese Medical Doctor Association(CMDA)/Committee on Pediatric Cardiology, Chinese College of Cardiovascular Physicians, Chinese Medical Doctor Association(CMDA)/Pediatric Cardiology Society, Beijing Pediatric Society, Beijing Medical Association(BMA). 3Thermal-responsive genetic and epigenetic regulation of DAM cluster controlling dormancy and chilling requirement in peach floral buds显示文摘The Dormancy-associated MADS-box(DAM)gene cluster in peach serves as a key regulatory hub on which the seasonal temperatures act and orchestrate dormancy onset and exit,chilling response and floral bud developmental pace.Yet,how different temperature regimes interact with and regulate the six linked DAM genes remains unclear.Here,we demonstrate that chilling downregulates DAM1 and DAM3–6 in dormant floral buds with distinct patterns and identify DAM4 as the most abundantly expressed one.We reveal multiple epigenetic events,with tri-methyl histone H3 lysine 27(H3K27me3)induced by chilling specifically in DAM1 and DAM5,a 21-nt sRNA in DAM3 and a ncRNA induced in DAM4.Such induction is inversely correlated with downregulation of their cognate DAMs.We also show that the six DAMs were hypermethylated,associating with the production of 24-nt sRNAs.Hence,the chilling-responsive dynamic of the different epigenetic elements and their interactions likely define distinct expression abundance and downregulation pattern of each DAM.We further show that the expression of the five DAMs remains steadily unchanged or continuously downregulated at the ensuing warm temperature after chilling,and this state of regulation correlates with robust increase of sRNA expression,H3K27me3 and CHH methylation,which is particularly pronounced in DAM4.Such robust increase of repressive epigenetic marks may irreversibly reinforce the chillingimposed repression of DAMs to ensure flower-developmental programming free from any residual DAM inhibition.Taken together,we reveal novel information about genetic and epigenetic regulation of the DAM cluster in peach,which will be of fundamental significance in understanding of the regulatory mechanisms underlying chilling requirement and dormancy release,and of practical application for improvement of plasticity of flower time and bud break in fruit trees to adapt changing climates.Hong Zhu Pao-Yang Chen Silin Zhong Chris Dardick Ann Callahan Yong-Qiang An Steve van Knocker Yingzhen Yang Gan-Yuan Zhong Albert Abbott Zongrang Liu 2020Horticulture Research2020,7,1:9

5Diversity and redundancy of the ripening regulatory networks revealed by the fruitENCODE and the new CRISPR/Cas9 CNR and NOR mutants显示文摘Tomato is considered as the genetic model for climacteric fruits,in which three major players control the fruit ripening process:ethylene,ripening transcription factors,and DNA methylation.The fruitENCODE project has now shown that there are multiple transcriptional circuits regulating fruit ripening in different species,and H3K27me3,instead of DNA methylation,plays a conserved role in restricting these ripening pathways.In addition,the function of the core tomato ripening transcription factors is now being questioned.We have employed CRISPR/Cas9 genome editing to mutate the SBP-CNR and NAC-NOR transcription factors,both of which are considered as master regulators in the current tomato ripening model.These plants only displayed delayed or partial non-ripening phenotypes,distinct from the original mutant plants,which categorically failed to ripen,suggesting that they might be gain-of-function mutants.Besides increased DNA methylation genome-wide,the original mutants also have hyper-H3K27me3 in ripening gene loci such as ACS2,RIN,and TDR4.It is most likely that multiple genetic and epigenetic factors have contributed to their strong non-ripening phenotypes.Hence,we propose that the field should move beyond these linear and twodimensional models and embrace the fact that important biological processes such as ripening are often regulated by highly redundant network with inputs from multiple levels.Ying Gao Ning Zhu Xiaofang Zhu Meng Wu Cai-Zhong Jiang Donald Grierson Yunbo Luo Wei Shen Silin Zhong Da-Qi Fu Guiqin Qu 2019Horticulture Research2019,6,1:7

8Recent advances in enzyme-mediated peptide ligation显示文摘Artificial synthesis and site-specific modification of peptides and proteins has evolved into an indispensable tool for protein engineers and chemical biologists. Chemical and enzymatic approaches to peptide ligation are important alternatives of recombinant DNA technology for protein synthesis and modification. Although as old as that of chemical procedures, enzyme-mediated peptide ligation is far less developed than that of chemical counterpart due to the difficult availability of peptide ligase.Fortunately, this situation has been changed slowly with the fast development of biological techniques. In the past decades, several natural peptide ligases have been discovered. Protein engineering to improve the ligation efficiencies of the natural peptide ligase and to reverse the functionality of protease provide more powerful peptide ligases. 10Comparative WGBS identifies genes that influence non-ripe phenotype in tomato epimutant Colourless non-ripening显示文摘Whole-genome bisulfite sequencing(WGBS)allows single-base resolution and genome-wide profiling of DNA methylation in plants and animals.This technology provides a powerful tool to identify genes that are potentially controlled by dynamic changes of DNA methylation and demethylation.However,naturally occurring epimutants are rare and genes under epigenetic regulation as well as their biological relevances are often difficult to define.In tomato,fruit development and ripening are a complex process that involves epigenetic control.We have taken the advantage of the tomato epimutant Colourless non-ripening(Cnr)and performed comparative mining of the WGBS datasets for the Cnr and Sl CMT3-silenced Cnr fruits.We compared DNA methylation profiles for the promoter sequences of approximately 5,000 bp immediately upstream of the coding region of a list of20 genes.Differentially methylated regions were found for some of these genes.Virus-induced gene silencing(VIGS)of differentially methylated gene Sl DET1 or Sl PDS resulted in unusual brown pigmentation in Cnr fruits.These results suggest that comparative WGBS coupled with VIGS can be used to identify genes that may contribute to the colourless unripe phenotype of fruit in the Cnr epimutant.Weiwei Chen Zhiming Yu Junhua Kong Hui Wang Yichen Li Mei Zhao Xiaohong Wang Qianqian Zheng Nongnong Shi Pengcheng Zhang Silin Zhong Paul Hunter Mahmut Tor Yiguo Hong 2018Science China(Life Sciences)2018,61,2:3

12The silk road economic belt: balance of interests显示文摘The article makes an analogy between the Great Silk Road and the modern Belt and Road Initiative(BRI,also known as‘One Belt,One Road’[OBOR])of China in the context of their significance for the development of international trade,as well as the Central Asian region.The historical role of the Silk Road is considered from the viewpoint of mutual benefit of states along the trade route.The authors consider the concept as the co-development of states,suggested by China within the framework of the BRI,which involves international cooperation on the basis of mutual benefits for all participants,promotion of economic growth and welfare,development of modern infrastructure through joint investments.An attempt is made to assess the interests of Central Asian countries in the Silk Road Economic Belt(SREB)projects.Common interests include construction of modern road infrastructure,exploration and extraction of mineral deposits,transportation of energy resources,establishment of logistics and industrial zones.There is an increase in the transit role of the Central Asian countries within the framework of the BRI,as well an increase in investments from China.The countries along SREB are concerned about potential threats of the inflow of Chinese labour,the use of technologies that can cause environmental problems,the loss of control over strategic assets,and general growth of financial dependence on China.The article analyses Russia’s perception of the Chinese initiative.It is concluded that the main mutual interest of the two countries is opposing the US’s attempts to establish a dominating role in the region and thus ensuring the security of the transport system through the institutions of the Eurasian Economic Union(EAEU)and the Shanghai Cooperation Organisation(SCO).The authors reveal a trend of changing the role of Russia as the main transit corridor for Chinese goods exported to Europe and European ones imported to China.The key long-term economic interests of China implementing the BRI have been identified:expansion of foreign markets,access to critical resources in the countries of Central Asia,export of capital,transfer of production abroad,increased exports of high-tech products and services,expansion of the RMB currency area and initiation of structural changes in the global management system.The assessment of China’s initiative by the US and Europe is given as a claim to economic leadership and the struggle to balance the country’s geopolitical position with its economic strength.The authors identify the areas of mutual and opposing interests of some European countries and China.It is concluded that,despite high risks and existing threats,the projects of the BRI can be implemented successfully that achieve the balance of interests and long-term mutual benefits of the countries involved.Yakov Silin Larisa Kapustina Italo Trevisan Andrei Drevalev 2018Economic and Political Studies2018,6,3:2
