3Cancer-associated adipocyte-derived G-CSF promotes breast cancer malignancy via Stat3 signaling显示文摘Adipocyte is the most predominant cell type in the tumor microenvironment of breast cancer and plays a pivotal role in cancer progression,yet the underlying mechanisms and functional mediators remain elusive.We isolated primary preadipocytes from mammary fat pads of human breast cancer patients and generated mature adipocytes and cancer-associated adipocytes(CAAs)in vitro.The CAAs exhibited significantly different gene expression profiles as assessed by transcriptome sequencing.One of the highly expressed genes in CAAs is granulocyte colony-stimulating factor(G-CSF).Treatment with recombinant human G-CSF protein or stable expression of human G-CSF in triple-negative breast cancer(TNBC)cell lines enhanced epithelial–mesenchymal transition,migration,and invasion of cancer cells,by activating Stat3.Accordantly,targeting G-CSF/Stat3 signaling with G-CSF-neutralizing antibody,a chemical inhibitor,or siRNAs for Stat3 could all abrogate CAA-or G-CSF-induced migration and invasion of breast cancer cells.The pro-invasive genes MMP2 and MMP9 were identified as target genes of G-CSF in TNBC cells.Furthermore,in human breast cancer tissues,elevated G-CSF expression in adipocytes is well correlated with activated Stat3 signal in cancer cells.Together,our results suggest a novel strategy to intervene with invasive breast cancers by targeting CAA-derived G-CSF.Li Liu Yudong Wu Cheng Zhang Chong Zhou Yining Li Yi Zeng Chunbo Zhang Rong Li Daya Luo Lieliang Wang Long Zhang Shuo Tu Huan Deng Shiwen Luo Ye-Guang Chen Xiangyang Xiong Xiaohua Yan 2020Journal of Molecular Cell Biology2020,12,9:4
4Comparison of Murraya exotica and Murraya paniculata by fingerprint analysis coupled with chemometrics and network pharmacology methods显示文摘There are two source plants for the traditional Chinese medicine Murrayae Folium et Cacumen(MFC)in Chinese Pharmacopoeia,i.e.Murraya exotica L.and M.paniculata(L.)Jack.Herein,a chemical comparison of M.exotica and M.paniculata by high performance liquid chromatography(HPLC)fingerprint analysis coupled with chemometrics and network pharmacology was performed.The main peaks in the fingerprints were identified by liquid chromatography coupled with ion trap/time-of-flight mass spectrometry(LC-IT-TOF-MS)and authenticated by references.The chemometrics results showed that the HPLC fingerprints of these two species were clearly divided into two categories using hierarchical cluster analysis(HCA)and principal component analysis(PCA),and a total of 13 significantly differentiated markers were screened out by orthogonal partial least squares-discriminant analysis(OPLS-DA).However,the following network pharmacology analysis showed that these discriminated markers were found to act via many common targets and metabolic pathways,indicating the possibly similar pharmacological effects and mechanisms for M.exotica and M.paniculata.The above results provide valuable evidence for the equivalent use of these two plants in clinical settings.Moreover,the chromatographic fingerprint analysis coupled with chemometrics and network pharmacology supplies an efficient approach for the comparative analysis of multi-source TCMs like MFC.LIANG Hai-Zhen DU Zhi-Yong YUAN Shuo LU Meng-Qiu XING Jian-Yong MA Qing HAN Zheng-Zhou TU Peng-Fei JIANG Yong 2021Chinese Journal of Natural Medicines2021,19,9:3
5KLF2 inhibits TGF-β-mediated cancer cell motility in hepatocellular carcinoma显示文摘Feedback regulation plays a pivotal role in determining the intensity and duration of TGF-p signaling and subsequently affecting the pathophysiological roles of TGF-p,including those in liver malignancy.KLF2,a member of the Kruppel-like factor(KLF)fam ily transcription factors,has been implicated in impeding hepatocellular carcinoma(HCC)development.However,the underlying molecular mechanisms are not fully understood.In the present study,we found that TGF-p stimulates the expression of KLF2 gene in several HCC cell lines.KLF2 protein is able to inhibit TGF-p/Smad signaling in HCC cells as assessed by luciferase reporter assay.Further studies indicated that KLF2 inhibits the transcriptional activity of Smad2/3 and Smad4 and ameliorates TGF-p-induced target gene expression,therefore creating a novel negative feedback loop in TGFp signaling.Functionally,stably expression of KLF2 in HCCLM3 cells attenuated TGF-卜induced cancer cell motility in wound-healing and transwell assays by interfering with TGF-p-mediated upregulation of MMP2.Together,our results revealed that KLF2 protein has a tumor-suppressive function in HCC through a negative feedback loop over TGF-p signaling.Yining Li Shuo Tu Yi Zeng Cheng Zhang Tian Deng Weicheng Luo Lingyan Lian Ling Chen Xiangyang Xiong Xiaohua Yan 2020Acta Biochimica et Biophysica Sinica2020,52,5:2
6Link between miR-19b and the mTOR signaling pathway in cancer prognosis显示文摘Objective Previous studies have reported differing conclusions regarding the prognostic value of miR-19b in cancers.Moreover,miR-19b may affect tumor growth by different pathways,mainly targeting PTENPI3K-AKT,which activates the downstream mTOR pathway.Therefore,we performed data mining to explore the possible correlation between miR-19b and mTOR in cancer prognosis.Methods We conducted online search and collected a total of 943 articles.According to different authors cross check and our study including/excluding criteria we at end retained 21 articles with 25 studies in this meta-analysis.Then TCGA data containing miR-19b level with cancer progression were obtained using OncomiR.Furthermore,Trial Sequential Analysis(TSA)was performed to determine whether the results of our meta-analysis could be used in clinical applications.After that,articles regarding the mechanism of miR-19b in various cancers were analyzed and KEGG pathway database was used to find the main regulatory function of miR-19b in human cancers.Results Overall hazard ratio(HR)results showed that higher levels of miR-19b expression were correlated with shorter overall survival time[HR=1.54,95%confidence interval(CI)=1.20-1.98]by promoting distant metastasis,but had no correlation with disease-free survival(DFS)/progression-free survival(PFS;HR=0.61,95%CI=0.31–1.19).Data from The Cancer Genome Atlas also revealed the role of miR-19b in tumorigenesis.According to trial sequential analysis results,more evidence is required to confirm that miR-19b is not correlated with DFS/PFS.Exploration of the mechanism revealed a possible link between miR-19b and the mTOR pathway.Conclusion miR-19b may have a pro-carcinogenic role through the mTOR pathway and thus,it is likely to be a therapeutic target for cancers.Yanlin Feng Souraka Tapara Dramani Maman Shuo Li Dingdong He Jiancheng Tu 2020Oncology and Translational Medicine2020,6,4:1

9Prognostic factors for pN2 non-small cell lung cancer:a comprehensive evidence from 73 studies involving 23,772 patients显示文摘Obojective Non-small-cell lung cancer(NSCLC)is a common malignancy.pN2 NSCLC,with pathologically confirmed ipsilateral mediastinal/subcarinal nodes metastasis,has been known as a very heterogeneous subgroup in terms of its anatomical,biological and patient characteristics.Prognostic factors based on patient characteristics were not well determined yet in this subgroup,and there is currently no standard treatment recommendation for these heterogeneous pN2 subjects.Apparent disagreements and inconsistency exist in study reports concerning the prognostic significance of certain factors in pN2 NSCLC,especially regarding to the issue about whether skip N2 metastasis benefit from surgery.Methods We therefore performed this comprehensive summary of the published literatures to draw a more precise and less uncertain conclusion.After a comprehensive literature search,a total of 73 studies involving 23,773 subjects were included according to eligibility criteria.Results As expected,most of the investigated factors,such as old age,male,advanced pathological T stage,advanced clinical N stage,multiple N2 stations,extended surgical resection(pneumonectomy),and incomplete resection,but not post-operation treatment(eg.chemotherapy and radiotherapy)were significantly associated with poor survival.However,skip N2 metastasis was favourable prognostic factors in operable pN2 NSCLC subjects.Other factors(histological type and primary tumour side)were neutral in terms of association with overall survival.We highlighted a number of important prognostic factors for pN2 NSCLC patients.Particularly,patients with skip N2 disease benefit from surgery.Conclusion Our findings could be used as reference information for decision-making in clinical practice and future study design.Shuo Li Yanlin Feng Chunzi Liang Jiancheng Tu 2020Oncology and Translational Medicine2020,6,2:1
10Effects of dietary amylose to amylopectin ratio on growth performance,carcass quality characteristics and meat fatty acids in Chinese Qinchuan cattle显示文摘This study evaluated the effects of the dietary starch amylose/amylopectin ratio on growth performance,rumen fermentation and blood parameters,carcass characteristics and marbling score,and meat fatty acid profiles and transcriptional changes in the genes involved in the gluconeogenesis pathway in Chinese Qinchuan cattle.Forty-five cattle were randomly divided into three groups.The bulls were fed a control diet(middle amylose/amylopectin ratio=0.47)or diets with either waxy corn starch(WS)inclusion(low amylose/amylopectin ratio=0.23)or pea starch(PS)inclusion(high amylose/amylopectin ratio=0.60)for 90 days.The bulls were individually allowed to receive the diets at the daily amount of at least 2.1%of their individual BW twice daily.The bulls were weighed at 45-day intervals,and blood and rumen fluid samples were also collected at 45-day intervals.The bulls were slaughtered and longissimus thoracis(LT)samples were collected for meat quality measurements.Compared with PS,the average daily gain(ADG)and feed efficiency in control and WS were increased(P<0.05)during d 0 to 45.However,the feed efficiency in PS was increased(P<0.05)compared with the control and WS during d 46 to 90.During the whole feeding trial,the ADG in control and WS showed trends that were higher(0.050.05)among the three groups.Compared with PS,the serum insulin concentration in WS was increased(P<0.05)at d 45,but not at d 90.Compared with control and WS,the serum SOD and T-AOC concentrations in PS were increased(P<0.05)at d 45,but not at d 90.Compared with control,the rumen microbial crude protein contents in WS and PS were increased(P<0.05)at d 0,while those of control and WS were higher(P<0.05)than that of PS at d 45,but not at d 90.Compared with PS,the backfat thickness in control and WS were increased(P<0.05),however,the marbling score and expression of genes related to the gluconeogenesis pathway in liver of the three groups did not differ(P>0.05).Chemical and physio-chemical compositions of LT did not differ(P>0.05)among the three groups.In conclusion,diets with low or middle amylose/amylopectin ratios increased the backfat thickness,and tended to improve ADG,but had no effect on other carcass characteristics,fat content in the LT,or the expression of hepatic genes related to the gluconeogenesis pathway in Chinese Qinchuan cattle.PIAO Min-yu HU Feng-ming KONG Fan-lin LIU Yun-long WANG Shuo CUI Kai SUN Tao DIAO Qi-yu TU Yan 2021Journal of Integrative Agriculture2021,20,12:1
11Multi-scale pressure-balanced structures in the solar wind observed by WIND显示文摘This work detects multi-scale, from hour to seconds, pressure-balanced structures (PBSs) in the solar wind based on the anti- correlation between the plasma thermal pressure and the magnetic pressure measured by WIND at 1 AU on April 5th, 2001. In our former research based on Cluster measurements, we showed the anti-correlation between the electron density and the magnetic field strength in multi-scales, and we supposed these structures may be pressure-balanced structures. Thus, in this work we aim to prove our speculation by the direct evidence on pressure measurements. Different from our previous work, we apply the WIND measurements this time, for they have both the magnetic pressure and the plasma pressure which Cluster could not offer. We use the wavelet cross-coherence method to analyze the correlation between the plasma pressure (P th ) and the magnetic pressure (P B ), and also the electron density (N e ) and the magnetic field strength (B) on various scales. We observe the anti-correlation between P th and P B distributed at different temporal scales ranging from 1000 s down to 10 s. This result directly indicates the existence of pressure- balanced structures (PBSs) with different sizes in the solar wind. Further, We compare the wavelet cross correlation spectrum of P th -P B and N e -B. We notice that the two spectra are similar in general. Thus this result confirms that the relation between P th -P B and N e -B are consistent with each other in the PBSs we study. Moreover, we compare the power spectrum density (PSD) of relative N e fluctuation with our previous work based on Cluster measurements. The two spectra show similar trend with Komolgorov's -5/3 as their slopes. This may imply the similarity of the structures observed by both WIND and Cluster spacecrafts. Finally, we discuss the possible formation mechanisms for these multi-scale pressure-balanced structures. 12Tea domain transcription factor TEAD4 mitigatesTGF-β signaling and hepatocellular carcinomaprogression independently of YAP显示文摘Tea domain transcription factor 4 (TEAD4) plays a pivotal role in tissue development and homeostasis by interacting with Yesassociated protein (YAP) in response to Hippo signaling inactivation. TEAD4 and YAP can also cooperate with transforminggrowth factor-β (TGF-β)-activated Smad proteins to regulate gene transcription. Yet, it remains unclear whether TEAD4 playsa YAP-independent role in TGF-β signaling. Here, we unveil a novel tumor suppressive function of TEAD4 in liver cancer viamitigating TGF-β signaling. Ectopic TEAD4 inhibited TGF-β-induced signal transduction, Smad transcriptional activity, and targetgene transcription, consequently suppressing hepatocellular carcinoma cell proliferation and migration in vitro and xenografttumorgrowth in mice. Consistently, depletion of endogenous TEAD4 by siRNAs enhanced TGF-β signaling in cancer cells. Mechanistically,TEAD4 associates with receptor-regulated Smads (Smad2/3) and Smad4 in the nucleus, thereby impairing the binding of Smad2/3to the histone acetyltransferase p300. 13Optimization method of urban rail train operational plan based on O-D time-varying demand显示文摘Purpose–Under the constraints of given passenger service level and coupling travel demand with train departure time,this study optimizes the train operational plan in an urban rail corridor to minimize the numbers of train trips and rolling stocks considering the time-varying demand of urban rail passenger flow.Design/methodology/approach–The authors optimize the train operational plan in a special network layout,i.e.an urban rail corridor with dead-end terminal yard,by decomposing it into two sub-problems:train timetable optimization and rolling stock circulation optimization.As for train timetable optimization,the authors propose a schedule-based passenger flow assignment method,construct the corresponding timetabling optimization model and design the bi-directional coordinated sequential optimization algorithm.For the optimization of rolling stock circulation,the authors construct the corresponding optimization assignment model and adopt the Hungary algorithm for solving the model.Findings–The case study shows that the train operational plan developed by the study’s approach meets requirements on the passenger service quality and reduces the operational cost to the maximum by minimizing the numbers of train trips and rolling stocks.Originality/value–The example verifies the efficiency of the model and algorithm.Feng Shi Xian Tu Shuo Zhao 2022Railway Sciences2022,1,1:0
14Accelerated design for magnetocaloric performance in Mn-Fe-P-Si compounds using machine learning显示文摘Magnetocaloric performance is of vital importance for Mn-Fe-P-Si alloys.However,when processes and compositions are considered,designing alloys with large magnetic entropy changes(ΔS_(m)),low thermal hysteresis(ΔT_(hys)),and Curie temperatures(T_(C))around room temperature become relatively complicated.In this study,we adopt machine learning methods to predict the magnetocaloric performance of Mn-Fe-P-Si compounds for the first time.To achieve this goal,503,465,and 660 data points for datasets with T_(C),ΔT_(hys),andΔS_(m)are collected,respectively.The collected datasets contain parameters of compositions,preparations,heat treatment,and magnetic field changes.We search for the optimal configuration using various methods and also compare their mean squared errors(MSE)and allowable errors.Evaluation results show that the performance of neural networks(NNs)is better than other methods.Therefore,we select NN to explore the T_(C),ΔT_(hys),andΔS_(m)values as a function of Mn,Si,metal/non-metal ratios,and B(Boron).We also propose to use the composition window with excellent magnetocaloric performance.These results not only help us gain deep insights into Mn-Fe-P-Si alloys but also accelerate the design process of alloys suitable for magnetocaloric materials.This work has the potential to solve the challenges and boost the research of Mn-Fe-P-Si alloys.Defang Tu Jianqi Yan Yunbo Xie Jun Li Shuo Feng Mingxu Xia Jianguo Li Alex Po Leung 2022Journal of Materials Science & Technology2022,,1:0
15NOGEA: A Network-oriented Gene Entropy Approach for Dissecting Disease Comorbidity and Drug Repositioning显示文摘Rapid development of high-throughput technologies has permitted the identification of an increasing number of disease-associated genes(DAGs),which are important for understanding disease initiation and developing precision therapeutics.However,DAGs often contain large amounts of redundant or false positive information,leading to difficulties in quantifying and prioritizing potential relationships between these DAGs and human diseases.In this study,a networkoriented gene entropy approach(NOGEA)is proposed for accurately inferring master genes that contribute to specific diseases by quantitatively calculating their perturbation abilities on directed disease-specific gene networks.In addition,we confirmed that the master genes identified by NOGEA have a high reliability for predicting disease-specific initiation events and progression risk.Master genes may also be used to extract the underlying information of different diseases,thus revealing mechanisms of disease comorbidity.More importantly,approved therapeutic targets are topologically localized in a small neighborhood of master genes in the interactome network,which provides a new way for predicting drug-disease associations.Through this method,11 old drugs were newly identified and predicted to be effective for treating pancreatic cancer and then validated by in vitro experiments.Collectively,the NOGEA was useful for identifying master genes that control disease initiation and co-occurrence,thus providing a valuable strategy for drug efficacy screening and repositioning.NOGEA codes are publicly available at http://gffzz188fe103f8f1460as5kqbww6nufc06fwx.ffgz.tsg.suse.edu.cn/guozihuaa/NOGEA.Zihu Guo Yingxue Fu Chao Huang Chunli Zheng Ziyin Wu Xuetong Chen Shuo Gao Yaohua Ma Mohamed Shahen Yan Li Pengfei Tu Jingbo Zhu Zhenzhong Wang Wei Xiao Yonghua Wang 2021Genomics, Proteomics & Bioinformatics2021,19,4:0
