1Expression changes and bioinformatic analysis of Wallerian degeneration after sciatic nerve injury in rat显示文摘Wallerian degeneration (WD) remains an important research topic. Many genes are differentially expressed during the process of WD, but the precise mechanisms responsible for these differentiations are not completely understood. In this study, we used microarrays to analyze the expression changes of the distal nerve stump at 0, 1, 4, 7, 14, 21 and 28 days after sciatic nerve injury in rats. The data revealed 6 076 differentially-expressed genes, with 23 types of expression, specifically enriched in genes associated with nerve development and axonogenesis, cytokine biosynthesis, cell differentiation, cytokine/chemokine production, neuron differentiation, cytokinesis, phosphorylation and axon regeneration. Kyoto Encyclopedia of Genes and Genomes pathway analysis gave findings related mainly to the MAPK signaling pathway, the Jak-STAT signaling pathway, the cell cycle, cytokine-cytokine receptor interaction, the p53 signaling pathway and the Wnt signaling pathway. Some key factors were NGF, MAG, CNTF, CTNNA2, p53, JAK2, PLCB1, STAT3, BDNF, PRKC, collagen II, FGF, THBS4, TNC and c-Src, which were further validated by real-time quantitative PCR, Western blot, and immunohistochemistry. 2Gene expression profiling of the rat sciatic nerve in early Wallerian degeneration after injury显示文摘Wallerian degeneration is an important area of research in modern neuroscience.A large number of genes are differentially regulated in the various stages of Wallerian degeneration,especially during the early response.In this study,we analyzed gene expression in early Wallerian degeneration of the distal nerve stump at 0,0.5,1,6,12 and 24 hours after rat sciatic nerve injury using gene chip microarrays.We screened for differentially-expressed genes and gene expression patterns.We examined the data for Gene Ontology,and explored the Kyoto Encyclopedia of Genes and Genomes Pathway.This allowed us to identify key regulatory factors and recurrent network motifs.We identified 1 546 differentially-expressed genes and 21 distinct patterns of gene expression in early Wallerian degeneration,and an enrichment of genes associated with the immune response,acute inflammation,apoptosis,cell adhesion,ion transport and the extracellular matrix.Kyoto Encyclopedia of Genes and Genomes pathway analysis revealed components involved in the Jak-STAT,ErbB,transforming growth factor-β,T cell receptor and calcium signaling pathways.Key factors included interleukin-6,interleukin-1,integrin,c-sarcoma,carcinoembryonic antigen-related cell adhesion molecules,chemokine(C-C motif) ligand,matrix metalloproteinase,BH3 interacting domain death agonist,baculoviral IAP repeat-containing 3 and Rac.The data were validated with real-time quantitative PCR.This study provides a global view of gene expression profiles in early Wallerian degeneration of the rat sciatic nerve.Our findings provide insight into the molecular mechanisms underlying early Wallerian degeneration,and the regulation of nerve degeneration and regeneration.Dengbing Yao Meiyuan Li Dingding Shen Fei Ding Shibi Lu Qin Zhao Xiaosong Gu 2012Neural Regeneration Research2012,7,17:5
3Scar-modulating treatments for central nervous system injury显示文摘Traumatic injury to the adult mammalian central nervous system(CNS) leads to complex cellular responses. Among them, the scar tissue formed is generally recognized as a major obstacle to CNS repair, both by the production of inhibitory molecules and by the physical impedance of axon regrowth. Therefore, scar-modulating treatments have become a leading therapeutic intervention for CNS injury. To date, a variety of biological and pharmaceutical treatments, targeting scar modulation, have been tested in animal models of CNS injury, and a few are likely to enter clinical trials. In this review, we summarize current knowledge of the scar-modulating treatments according to their specific aims:(1) inhibition of glial and fibrotic scar formation, and(2) blockade of the production of scar-associated inhibitory molecules. The removal of existing scar tissue is also discussed as a treatment of choice. 5In-situ imaging the electrochemical reactions of Li-CO_(2) nanobatteries at high temperatures in an aberration corrected environmental transmission electron microscope显示文摘Rechargeable lithium-carbon dioxide(Li-CO_(2))batteries have attracted much attention due to their high theoretical energy densities and capture of C0_(2).However,the electrochemical reaction mechanisms of rechargeable Lo-CO_(2) batteries,particularly the decomposition mechanisms of the discharge product Li_(2)CO_(3) are still unclear,impeding their practical applications.Exploring electrochemistry of Li_(2)CO_(3) is critical for improving the performance of Li-C0_(2) batteries.Herein,in-situ environmental transmission electron microscopy(ETEM)technique was used to study electrochemistry of Li_(2)CO_(3) in Li-C0_(2) batteries during discharge and charge processes.During discharge,Li_(2)CO_(3) was nucleated and accumulated on the surface of the cathode media such as carbon nanotubes(CNTs)and Ag nanowires(Ag NWs),but it was hard to decompose during charging at room temperature.To promote the decomposition of Li2C03,the charge reactions were conducted at high temperatures,during which Li_(2)CO_(3) was decomposed to lithium with release of gases.Density functional theory(DFT)calculations revealed that the synergistic effect of temperature and biasing facilitates the decomposition of Li_(2)CO_(3).This study not only provides a fundamental understanding to the high temperature Li-C0_(2) nanobatteries,but also offers a valid technique,i.e.,discharging/charging at high temperatures,to improve the cyclability of Li-CO_(2) batteries for energy storage applications.Peng Jia Meiqi Yu Xuedong Zhang Tingting Yang Dingding Zhu Tongde Shen Liqiang Zhang Yongfu Tang Jianyu Huang 2022Nano Research2022,15,1:0
6Circulating proteomic biomarkers for diagnosing sporadic amyotrophic lateral sclerosis:a cross-sectional study显示文摘Biomarke rs are required for the early detection,prognosis prediction,and monitoring of amyotrophic lateral sclerosis,a progressive disease.Proteomics is an unbiased and quantitative method that can be used to detect neurochemical signatures to aid in the identification of candidate biomarke rs.In this study,we used a label-free quantitative proteomics approach to screen for substantially differentially regulated proteins in ten patients with sporadic amyotrophic lateral scle rosis compared with five healthy controls.Su bstantial upregulation of serum proteins related to multiple functional clusters was observed in patients with spo radic amyotrophic lateral sclerosis.Potential biomarke rs were selected based on functionality and expression specificity.To validate the proteomics profiles,blood samples from an additional cohort comprising 100 patients with sporadic amyotrophic lateral sclerosis and 100 healthy controls were subjected to enzyme-linked immunosorbent assay.Eight substantially upregulated serum proteins in patients with spora dic amyotrophic lateral sclerosis were selected,of which the cathelicidin-related antimicrobial peptide demonstrated the best discriminative ability between patients with sporadic amyotrophic lateral sclerosis and healthy controls(area under the curve[AUC]=0.713,P<0.0001).To further enhance diagnostic accuracy,a multi-protein combined discriminant algorithm was developed incorporating five proteins(hemoglobin beta,cathelicidin-related antimicrobial peptide,talin-1,zyxin,and translationally-controlled tumor protein).The algo rithm achieved an AUC of 0.811 and a P-value of<0.0001,resulting in 79%sensitivity and 71%specificity for the diagnosis of sporadic amyotrophic lateral scle rosis.Subsequently,the ability of candidate biomarkers to discriminate between early-stage amyotrophic lateral sclerosis patients and controls,as well as patients with different disease severities,was examined.A two-protein panel comprising talin-1 and translationally-controlled tumor protein effectively distinguished early-stage amyotrophic lateral sclerosis patients from controls(AUC=0.766,P<0.0001).Moreove r,the expression of three proteins(FK506 binding protein 1A,cathelicidin-related antimicrobial peptide,and hemoglobin beta-1)was found to increase with disease progression.The proteomic signatures developed in this study may help facilitate early diagnosis and monitor the progression of sporadic amyotrophic lateral sclerosis when used in co mbination with curre nt clinical-based parameters.Lu He Qinming Zhou Chaoyang Xiu Yaping Shao Dingding Shen Huanyu Meng Weidong Le Sheng Chen 2024Neural Regeneration Research2024,19,8:0
7A perspective on therapies for amyotrophic lateral sclerosis:can disease progression be curbed?显示文摘Amyotrophic lateral sclerosis(ALS)is a progressive neurodegenerative disease involving both upper and lower motor neurons,leading to paralysis and eventually death.Symptomatic treatments such as inhibition of salivation,alleviation of muscle cramps,and relief of spasticity and pain still play an important role in enhancing the quality of life.To date,riluzole and edaravone are the only two drugs approved by the Food and Drug Administration for the treatment of ALS in a few countries.While there is adequate consensus on the modest efficacy of riluzole,there are still open questions concerning the efficacy of edaravone in slowing the disease progression.Therefore,identification of novel therapeutic strategies is urgently needed.Impaired autophagic process plays a critical role in ALS pathogenesis.In this review,we focus on therapies modulating autophagy in the context of ALS.Furthermore,stem cell therapies,gene therapies,and newly-developed biomaterials have great potentials in alleviating neurodegeneration,which might halt the disease progression.In this review,we will summarize the current and prospective therapies for ALS.Xiaojiao Xu Dingding Shen Yining Gao Qinming Zhou You Ni Huanyu Meng Hongqin Shi Weidong Le Shengdi Chen Sheng Chen 2021Translational Neurodegeneration2021,10,3:0
8Improving the electroconductivity and mechanical properties of cellulosic paper with multi-walled carbon nanotube/polyaniline nanocomposites显示文摘Cellulose is the most abundant renewable polymer in the nature,and cellulosic paper is widely used in our daily life.Conferring electroconductivity to cellulosic paper would allow this conventional material to hold great promise for a wide range of energy-related applications.In the present work,multi-walled carbon nanotube(MWCNT)/polyaniline(PANI)nanocomposites were synthesized via in situ oxidation polymerization process and characterized by FT-IR and TEM.Subsequently,the application of the synthesized MWCNT/PANI nanocomposites as a wet-end filler for the production of electro-conductive paper was demonstrated/developed.Results showed that the cellulosic paper was imparted with an electro-conductivity of up to 0.14 S·m^(-1) while exhibiting a pronounced improvement in mechanical properties as a function of the added MWCNT/PANI nanocomposites.Xiaochuang Shen Yanjun Tang Dingding Zhou Junhua Zhang Daliang Guo Gustavo Friederichs 2016Journal of Bioresources and Bioproducts2016,1,1:0
9Increased expression of coronin-1a in amyotrophic lateral sclerosis: a potential diagnostic biomarker and therapeutic target显示文摘Amyotrophic lateral sclerosis(ALS)is the most common motor neuron disease.At present,no definite ALS biomarkers are available.In this study,exosomes from the plasma of patients with ALS and healthy controls were extracted,and differentially expressed exosomal proteins were compared.Among them,the expression of exosomal coronin-1a(CORO1A)was 5.3-fold higher than that in the controls.CORO1A increased with disease progression at a certain proportion in the plasma of patients with ALS and in the spinal cord of ALS mice.CORO1A was also overexpressed in NSC-34 motor neuron-like cells,and apoptosis,oxidative stress,and autophagic protein expression were evaluated.CORO1A overexpression resulted in increased apoptosis and oxidative stress,overactivated autophagy,and hindered the formation of autolysosomes.Moreover,CORO1A activated Ca^(2+)-dependent phosphatase calcineurin,thereby blocking the fusion of autophagosomes and lysosomes.The inhibition of calcineurin activation by cyclosporin A reversed the damaged autolysosomes.In conclusion,the role of CORO1A in ALS pathogenesis was discovered,potentially affecting the disease onset and progression by blocking autophagic flux.Therefore,CORO1A might be a potential biomarker and therapeutic target for ALS.Qinming Zhou Lu He Jin Hu Yining Gao Dingding Shen You Ni Yuening Qin Huafeng Liang Jun Liu Weidong Le Sheng Chen 2022Frontiers of Medicine2022,16,5:0
10利用激光离子源产生质子束显示文摘In 2018,the technology of the proton beam production with a laser ion source was developed.To produce proton beams with the laser ion source,a solidified hydrogen target system had been developed by the end of 2017.The experiment on the production of proton beams with the laser ion source was carried out at the beginning of 2018.The 8 ns laser pulse with the 0.5 J energy was focused with a lens of f=500 mm onto the hydrogen ice deposited on a copper sheet attached at the second stage of a 4.2 K cryocooler.The laser power density on the ice was estimated to 2.5109 Wcm−2.As shown in Fig.1(a)and(b),the condition of the hydrogen ice formation can be observed through the laser window before each laser shot.The waveform of the proton beam,shown in Fig.1(c),was measured with a Faraday Cup located 2.5 m away from the target.As shown in Fig.1(c),the current intensity of the proton beam was much lower than other ion species.And the repeatability of the proton beam pulses was also poor,the main reason of which is that the conditions of the deposited hydrogen ice were quite different from shot to shot.Zhao Huanyu Zhang Junjie Shen Dingding Wang Guicai Zhang Xuezhen Sun Liangting Zhao Hongwei 2018IMP & HIRFL Annual Report2018,,1:0

13Slot-die coated large-area flexible all-polymer solar cells by nonhalogenated solvent显示文摘The slot-die coating is recognized as the most compatible method for the roll-to-roll(R2R)processing of large-area flexible organic solar cells(OSCs).However,the photovoltaic performance of the large-area flexible all-polymer solar cells was significantly lagging behind that of polymer donors with small molecule non-fullerene acceptors devices.In this work,the 1 cm^(2) flexible device of an all-polymer system,PTQ10:PYF-T-o,fabricated by slot-die coating,achieves an excellent efficiency of 11.24%via controlling the coating temperatures.It is found that,compared with the donor,the crystallinity of PYF-T-o plays a crucial role in device performance.The all-polymer flexible devices show superior mechanical bending stability,maintaining an efficiency of over 95%of the initial value during a 1000-cycle bending test.Yi-Fan Shen Jianqi Zhang Chenyang Tian Dingding Qiu Zhixiang Wei 2023Nano Research2023,16,12:0
