1Expression changes and bioinformatic analysis of Wallerian degeneration after sciatic nerve injury in rat显示文摘Wallerian degeneration (WD) remains an important research topic. Many genes are differentially expressed during the process of WD, but the precise mechanisms responsible for these differentiations are not completely understood. In this study, we used microarrays to analyze the expression changes of the distal nerve stump at 0, 1, 4, 7, 14, 21 and 28 days after sciatic nerve injury in rats. The data revealed 6 076 differentially-expressed genes, with 23 types of expression, specifically enriched in genes associated with nerve development and axonogenesis, cytokine biosynthesis, cell differentiation, cytokine/chemokine production, neuron differentiation, cytokinesis, phosphorylation and axon regeneration. Kyoto Encyclopedia of Genes and Genomes pathway analysis gave findings related mainly to the MAPK signaling pathway, the Jak-STAT signaling pathway, the cell cycle, cytokine-cytokine receptor interaction, the p53 signaling pathway and the Wnt signaling pathway. Some key factors were NGF, MAG, CNTF, CTNNA2, p53, JAK2, PLCB1, STAT3, BDNF, PRKC, collagen II, FGF, THBS4, TNC and c-Src, which were further validated by real-time quantitative PCR, Western blot, and immunohistochemistry. 2Gene expression profiling of the rat sciatic nerve in early Wallerian degeneration after injury显示文摘Wallerian degeneration is an important area of research in modern neuroscience.A large number of genes are differentially regulated in the various stages of Wallerian degeneration,especially during the early response.In this study,we analyzed gene expression in early Wallerian degeneration of the distal nerve stump at 0,0.5,1,6,12 and 24 hours after rat sciatic nerve injury using gene chip microarrays.We screened for differentially-expressed genes and gene expression patterns.We examined the data for Gene Ontology,and explored the Kyoto Encyclopedia of Genes and Genomes Pathway.This allowed us to identify key regulatory factors and recurrent network motifs.We identified 1 546 differentially-expressed genes and 21 distinct patterns of gene expression in early Wallerian degeneration,and an enrichment of genes associated with the immune response,acute inflammation,apoptosis,cell adhesion,ion transport and the extracellular matrix.Kyoto Encyclopedia of Genes and Genomes pathway analysis revealed components involved in the Jak-STAT,ErbB,transforming growth factor-β,T cell receptor and calcium signaling pathways.Key factors included interleukin-6,interleukin-1,integrin,c-sarcoma,carcinoembryonic antigen-related cell adhesion molecules,chemokine(C-C motif) ligand,matrix metalloproteinase,BH3 interacting domain death agonist,baculoviral IAP repeat-containing 3 and Rac.The data were validated with real-time quantitative PCR.This study provides a global view of gene expression profiles in early Wallerian degeneration of the rat sciatic nerve.Our findings provide insight into the molecular mechanisms underlying early Wallerian degeneration,and the regulation of nerve degeneration and regeneration.Dengbing Yao Meiyuan Li Dingding Shen Fei Ding Shibi Lu Qin Zhao Xiaosong Gu 2012Neural Regeneration Research2012,7,17:5
3Novel directional comparison protection for distribution lines显示文摘For quickly clearing up a fault of distribution lines, which concerned with complex structure and operating modes, a directional comparison protection is necessary. The paper evaluated the traditional directional relay through modeling a typical distribution system and presented a novel negative sequence directional relay and a new directional comparison protection scheme specially designed for distribution systems. In the relay and the protection scheme, a particular negative sequence component has been constructed to solve the problem that there is no negative sequence component in a symmetrical fault case so that they could operate correctly in both asymmetrical fault and symmetrical fault. Extensive EMTP simulation studies proved that the protection schemes are able to provide fast and reliable responses for all fault conditions. 7Effect of grain structure on fatigue crack propagation behavior of Al-Cu-Li alloys显示文摘Recrystallization behavior during optimized heat treatments provides a potential to obtain desirable grain structure,which significantly improves the mechanical properties of aluminum alloys.The influence of grain structures on fatigue crack propagation(FCP)behaviors of Al-Cu-Li alloy with hot-rolled(HR)and cold-rolled(CR)was investigated.Subgrain boundaries have a significant impact on small crack growth rates,which is reflected in the pronounced fluctuation of fatigue crack growth of HR specimens after solution treatment.Moreover,the specific cellular structure within grains can improve the deformation capacity of alloys due to their accommodation of plastic deformation,which contributes to the lower fatigue crack growth rates and higher threshold values in HR specimens.The intragranular deflection also decelerates the FCP rate and occurs in these regions of large grain without subgrain boundaries.Recrystallization occurs in the CR specimens,resulting in small anisotropy on the fatigue resistance for the different orientations in the Paris stage due to the recrystallization texture.Fatigue cracks can be deflected and tend to propagate along the grain boundaries when it goes into the grain with a relatively low Schmidt factor value.Dingding Lu Ben Lin Tianle Liu Sanxi Deng Youjie Guo Jinfeng Li Danyang Liu 2023Journal of Materials Science & Technology2023,,17:1
8Single-bond-linked oligomeric donors for high performance organic solar cells显示文摘The effective design and synthesis of novel small-molecule donors(SMDs) is extremely essential for the in-depth study of the scientific problems of bulk heterojunction morphology and the improvement of photovoltaic performance in organic solar cells(OSCs). Importantly, developing a series of donors with different conjugated central donor(D) units is a remarkable strategy to obtain high-performance donors. Herein, two acceptor-donor-donor-acceptor(A-D-D-A) type oligomeric donors 2DTBDT and2DTBDT-2T with two dithieno[2,3-d:2,3-d’]benzo[1,2-b:4,5-b’]dithiophene(DTBDT) as D units, without and with bithiophene as the π bridge respectively are designed and synthesized successfully. The central linked-DTBDT unit can provide a larger conjugated plane and promote π electron delocalization, which can effectively improve π-π interactions between donors and regulate the crystallinity. And we found that the π bridge provided 2DTBDT-2T with 12.31% efficiency that is already a high efficiency in OSCs,whereas 2DTBDT with merely 3.63% efficiency, both with 2,2-((2Z,2 Z)-((12,13-bis(2-ethylhexyl)-3,9-diundecyl-12,13-dihydro-[1,2,5]thiadiazolo[3,4-e]thieno[2,'3 ’:4,5 ]thieno[2,3 :4,5]pyrrolo[3,2-g]thieno-[2,3 :4,5]thieno[3,2-b]indole,10-diyl)bis(methanylylidene))bis(5,6-difluoro-3-oxo-2,3-dihydro-1H-indene-2,1-diylidene)) dimalononitrile(Y6) as the acceptor. We conjecture that the main reason for the different device performance may be ascribed to the different molecular stacking orientation of the oligomeric donors and morphology features of the donors:Y6 blend films. Compared to the predominant face-on orientation of the 2DTBDT neat film, the 2DTBDT-2T neat film performed a preferential edge-on orientation, which obtained a smoother surface, stronger crystallinity and more uniform phase separation in the 2DTBDT-2T:Y6 blend films with nanofiber structure, which delivered higher and more balanced carrier mobilities, the more efficient exciton dissociation and reduced biomolecule recombination, therefore obtaining better power conversion efficiencies(PCEs). We speculate that the transformation of molecular stacking orientation of oligomeric donors is possibly due to that π bridge extended and twisted the molecular structure of 2DTDBT-2T, resulting in an edge-on orientation relative to the substrate. 9Conjugation expansion strategy enables highly stable all-polymer solar cells显示文摘The stability issue is one of the key factors hindering the commercial application of organic solar cells.All-polymer organic solar cell is one of the effective ways to solve the stability problem.In this work,we designed and synthesized two polymer donor materials PBDT and PDTBDT with different conjugation ranges,and demonstrated for the first time that extending the conjugation range of donor materials in all polymer solar cells can significantly improve device efficiency and stability.The experimental results of materials and devices show that PDTBDT with a larger conjugation range has stronger crystallinity and a more planar structure,which endows the active layer in its corresponding device with higher exciton dissociation probability,lower carrier recombination probability,more balanced charge transport properties and more favorable film morphology.As a result,the PDTBDT:PYF-T-o devices display an outstanding PCE of 13.38%,which is much higher than PBDT with smaller conjugation range based devices.Moreover,the PDTBDT:PYF-T-o device retains 0.86 of the initial PCE after over 500 h in the air atmosphere,exhibiting significantly improved stability.The improved stability is attributed to the enhanced moisture and air tolerance of active layer film thanks to the strong crystallinity of the donor material.These results demonstrate that the conjugation expansion strategy is one of the effective ways to obtain efficient and stable all-polymer organic solar cells.Dingding Qiu Yanan Shi Yi Li Jianqi Zhang Kun Lu Zhixiang Wei 2023Chinese Chemical Letters2023,34,8:0
10Circulating proteomic biomarkers for diagnosing sporadic amyotrophic lateral sclerosis:a cross-sectional study显示文摘Biomarke rs are required for the early detection,prognosis prediction,and monitoring of amyotrophic lateral sclerosis,a progressive disease.Proteomics is an unbiased and quantitative method that can be used to detect neurochemical signatures to aid in the identification of candidate biomarke rs.In this study,we used a label-free quantitative proteomics approach to screen for substantially differentially regulated proteins in ten patients with sporadic amyotrophic lateral scle rosis compared with five healthy controls.Su bstantial upregulation of serum proteins related to multiple functional clusters was observed in patients with spo radic amyotrophic lateral sclerosis.Potential biomarke rs were selected based on functionality and expression specificity.To validate the proteomics profiles,blood samples from an additional cohort comprising 100 patients with sporadic amyotrophic lateral sclerosis and 100 healthy controls were subjected to enzyme-linked immunosorbent assay.Eight substantially upregulated serum proteins in patients with spora dic amyotrophic lateral sclerosis were selected,of which the cathelicidin-related antimicrobial peptide demonstrated the best discriminative ability between patients with sporadic amyotrophic lateral sclerosis and healthy controls(area under the curve[AUC]=0.713,P<0.0001).To further enhance diagnostic accuracy,a multi-protein combined discriminant algorithm was developed incorporating five proteins(hemoglobin beta,cathelicidin-related antimicrobial peptide,talin-1,zyxin,and translationally-controlled tumor protein).The algo rithm achieved an AUC of 0.811 and a P-value of<0.0001,resulting in 79%sensitivity and 71%specificity for the diagnosis of sporadic amyotrophic lateral scle rosis.Subsequently,the ability of candidate biomarkers to discriminate between early-stage amyotrophic lateral sclerosis patients and controls,as well as patients with different disease severities,was examined.A two-protein panel comprising talin-1 and translationally-controlled tumor protein effectively distinguished early-stage amyotrophic lateral sclerosis patients from controls(AUC=0.766,P<0.0001).Moreove r,the expression of three proteins(FK506 binding protein 1A,cathelicidin-related antimicrobial peptide,and hemoglobin beta-1)was found to increase with disease progression.The proteomic signatures developed in this study may help facilitate early diagnosis and monitor the progression of sporadic amyotrophic lateral sclerosis when used in co mbination with curre nt clinical-based parameters.Lu He Qinming Zhou Chaoyang Xiu Yaping Shao Dingding Shen Huanyu Meng Weidong Le Sheng Chen 2024Neural Regeneration Research2024,19,8:0
11Halogenated thiophene substitutions on quinoxaline unit to achieve morphology optimization in efficient organic solar cells显示文摘Halogenated thiophenes are generally used units for constructing organic semiconductor materials for photovoltaic applications.Here,we introduced thiophene,2-bromothiophene,and 2-chlorothiophene units to the central core of quinoxaline-based acceptors and obtained three acceptors,Qx-H,Qx-Br,and Qx-Cl,respectively.Compared with Qx-H,Qx-Br and Qx-Cl showed enhanced absorption,down-shifted energy levels,improved crystallinity,and reduced energy disorder.The improved crystallinity significantly optimized the blend morphology,leading to efficient charge generation and transport and,therefore,less bimolecular recombination.Eventually,PM6:Qx-Br-based devices exhibited an outstanding power conversion efficiency of 17.42%with a high open-circuit voltage(VOC)of 0.915 V.Furthermore,Y6 was introduced into the PM6:Qx-Br binary system to improve the light utilization,and the resulting ternary devices delivered a high PCE of 18.36%.This study demonstrated the great potential of halogenated thiophene substitution in quinoxaline-based acceptors for building high-performance organic solar cell acceptor materials.Dingding Qiu Jianqi Zhang Kun Lu Zhixiang Wei 2023Nano Research2023,16,9:0
12Increased expression of coronin-1a in amyotrophic lateral sclerosis: a potential diagnostic biomarker and therapeutic target显示文摘Amyotrophic lateral sclerosis(ALS)is the most common motor neuron disease.At present,no definite ALS biomarkers are available.In this study,exosomes from the plasma of patients with ALS and healthy controls were extracted,and differentially expressed exosomal proteins were compared.Among them,the expression of exosomal coronin-1a(CORO1A)was 5.3-fold higher than that in the controls.CORO1A increased with disease progression at a certain proportion in the plasma of patients with ALS and in the spinal cord of ALS mice.CORO1A was also overexpressed in NSC-34 motor neuron-like cells,and apoptosis,oxidative stress,and autophagic protein expression were evaluated.CORO1A overexpression resulted in increased apoptosis and oxidative stress,overactivated autophagy,and hindered the formation of autolysosomes.Moreover,CORO1A activated Ca^(2+)-dependent phosphatase calcineurin,thereby blocking the fusion of autophagosomes and lysosomes.The inhibition of calcineurin activation by cyclosporin A reversed the damaged autolysosomes.In conclusion,the role of CORO1A in ALS pathogenesis was discovered,potentially affecting the disease onset and progression by blocking autophagic flux.Therefore,CORO1A might be a potential biomarker and therapeutic target for ALS.Qinming Zhou Lu He Jin Hu Yining Gao Dingding Shen You Ni Yuening Qin Huafeng Liang Jun Liu Weidong Le Sheng Chen 2022Frontiers of Medicine2022,16,5:0
