1Epidemiology, causes, clinical manifestation and diagnosis, prevention and control of coronavirus disease (COVID-19) during the early outbreak period: a scoping review显示文摘Background:The coronavirus disease(COVID-19)has been identified as the cause of an outbreak of respiratory illness in Wuhan,Hubei Province,China beginning in December 2019.As of 31 January 2020,this epidemic had spread to 19 countries with 11791 confirmed cases,including 213 deaths.The World Health Organization has declared it a Public Health Emergency of International Concern.Methods:A scoping review was conducted following the methodological framework suggested by Arksey and O'Malley.In this scoping review,65 research articles published before 31 January 2020 were analyzed and discussed to better understand the epidemiology,causes,clinical diagnosis,prevention and control of this virus.The research domains,dates of publication,journal language,authors'affiliations,and methodological characteristics were included in the analysis.All the findings and statements in this review regarding the outbreak are based on published information as listed in the references.Results:Most of the publications were written using the English language(89.2%).The largest proportion of published articles were related to causes(38.5%)and a majority(67.7%)were published by Chinese scholars.Research articles initially focused on causes,but over time there was an increase of the articles related to prevention and control.Studies thus far have shown that the virus7 origination is in connection to a seafood market in Wuhan,but specific animal associations have not been confirmed.Reported symptoms include fever,cough,fatigue,pneumonia,headache,diarrhea,hemoptysis,and dyspnea.Preventive measures such as masks,hand hygiene practices,avoidance of public contact,case detection,contact tracing,and quarantines have been discussed as ways to reduce transmission.To date,no specific antiviral treatment has proven effective;hence,infected people primarily rely on symptomatic treatment and supportive care.Conclusions:There has been a rapid surge in research in response to the outbreak of COVID-19.During this early period,published research primarily explored the epidemiology,causes,clinical manifestation and diagnosis,as well as prevention and control of the novel coronavirus.Although these studies are relevant to control the current public emergency,more high-quality research is needed to provide valid and reliable ways to manage this kind of public health emergency in both the short-and long-term.Sasmita Poudel Adhikari Sha Meng Yu-Ju Wu Yu-Ping Mao Rui-Xue Ye Qing-Zhi Wang Chang Sun Sean Sylvia Scott Rozelle Hein Raat Huan Zhou 2020Infectious Diseases of Poverty2020,9,2:28

3Soil respiration in tropical seasonal rain forest in Xishuangbanna, SW China显示文摘With the static opaque chamber and gas chromatography technique, from January 2003 to January 2004 soil respiration was investigated in a tropical seasonal rain forest in Xishuangbanna, SW China. In this study three treatments were applied, each with three replicates: A (bare soil), B (soil+litter), and C (soil+litter+seedling). The results showed that soil respiration varied seasonally, low from December 2003 to February 2004, and high from June to July 2004. The annual average values of CO2 efflux from soil respiration differed among the treatments at 1% level, with the rank of C (14642 mgCO2· m-2. h-1)>B (12807 mgCO2· m-2. h-1)>A (9532 mgCO2· m-2. h-1). Diurnal variation in soil respiration was not apparent due to little diurnal temperate change in Xishuangbanna. There was a parabola relationship between soil respiration and soil moisture at 1% level. Soil respiration rates were higher when soil moisture ranged from 35% to 45%. There was an exponential relationship between soil respiration and soil temperature (at a depth of 5cm in mineral soil) at 1% level. The calculated Q1o values in this study,ranging from 2.03 to 2.36, were very near to those of tropical soil reported. 4Active learning for accelerated design of layered materials显示文摘Hetero-structures made from vertically stacked monolayers of transition metal dichalcogenides hold great potential for optoelectronic and thermoelectric devices.Discovery of the optimal layered material for specific applications necessitates the estimation of key material properties,such as electronic band structure and thermal transport coefficients.However,screening of material properties via brute force ab initio calculations of the entire material structure space exceeds the limits of current computing resources.Moreover,the functional dependence of material properties on the structures is often complicated,making simplistic statistical procedures for prediction difficult to employ without large amounts of data collection.Here,we present a Gaussian process regression model,which predicts material properties of an input hetero-structure,as well as an active learning model based on Bayesian optimization,which can efficiently discover the optimal hetero-structure using a minimal number of ab initio calculations.The electronic band gap,conduction/valence band dispersions,and thermoelectric performance are used as representative material properties for prediction and optimization.The Materials Project platform is used for electronic structure computation,while the BoltzTraP code is used to compute thermoelectric properties.Bayesian optimization is shown to significantly reduce the computational cost of discovering the optimal structure when compared with finding an optimal structure by building a regression model to predict material properties.The models can be used for predictions with respect to any material property and our software,including data preparation code based on the Python Materials Genomics(PyMatGen)library as well as python-based machine learning code,is available open source.Lindsay Bassman Pankaj Rajak Rajiv K.Kalia Aiichiro Nakano Fei Sha Jifeng Sun David J.Singh Muratahan Aykol Patrick Huck Kristin Persson Priya Vashishta 2018npj Computational Materials2018,,1:10

7Three-dimensional dynamic contrast-enhanced MR angiography for evaluating recipient vessels in orthotopic liver transplantation显示文摘BACKGROUND:With the improvement of MR technology, three-dimensional dynamic contrast-enhanced MR angiography(3D-DCE-MRA)may be the optimal vascular imaging method for preoperative evaluation of liver transplantation candidates.This study was undertaken to determine the value of 3D-DCE-MRA in the assessment of recipient vessels in orthotopic liver transplantation(OLT). METHODS:The surgical and pathological records were taken as the'gold standards'.Eighteen cases of OLT were retrospectively analyzed to assess the image quality of MRA,including the signal-to-noise ratio(SNR)in arteries and veins,depiction of vascular variation and vessel disease, and the accuracy of vascular diameter measurement. RESULTS:3D-DCE-MRA of 34 cases was carried out before OLT.The rates(excellent and good)showing hepatic arteries and portal vein for 3D-DCE-MRA were 94.1%(32/34)and 88.2%(30/34),respectively.The SNRs of the celiac axis and portal vein measurements from 3D-DCE-MRA were 20.58±3.74 and 13.43±4.12, and the mean diameters were 3.4±0.3 mm and 13.1±3.2 mm,respectively.There were 5 cases of vessel variation according to the Michel's classification.Of the 34 patients, 18 were compared radiologically and pathologically.The accuracy of depiction of the hepatic artery and portal vein with 3D-DCE-MRA was 100%for both;3D-DCE-MRA precisely assessed 4 cases of more than moderate stenosis in hepatic arteries,2 cases of small-caliber hepatic artery 3 cases of venous stenosis at the second porta hepatis, 6 cases of collateral vasculature,1 case of portal vein thrombosis and 1 case of portal vein aneurysm;all were confirmed pathologically.CONCLUSION:3D-DCE-MRA may be the first choice for recipient vascular assessment before OLT.Bian, Jie Sha, Lin Yang, Chao Sun, Chuan-Shu 2008Hepatobiliary & Pancreatic Diseases International2008,7,5:8
8Urodynamic assessment of bladder storage function after radical hysterectomy for cervical cancer显示文摘Background:After radical hysterectomy for cervical cancer,the most common complication is lower urinary tract symptoms.Post-operatively,bladder capacity can alter bladder function for a prolonged period.This study aimed to identify factors affecting bladder storage function.Methods:A multicenter,retrospective cohort study was conducted.Information of patients with stages IA2 to IIB cervical cancer with urodynamic study results were retrospectively collected from nine hospitals between June 2013 and June 2018 according to the inclusion criteria.Demographic,surgical,and oncological data were collected.The univariate and multivariate logistic regression was used to identify clinical factors associated with bladder storage function.Results:Two hundred and three patients with cervical cancer had urodynamic testing post-operatively.Ninety-five(46.8%)patients were diagnosed with stress urinary incontinence(SUI).The incidence of low bladder compliance(LBC)was 23.2%.Twenty-seven(13.3%)patients showed detrusor overactivity(DO).Fifty-seven patients(28.1%)presented with a decreased maximum cystometric capacity(DMCC).The probability of composite bladder storage dysfunction was 68.0%.Multivariate analysis confirmed that laparoscopy represents a protective factor for SUI with an odds ratio of 0.498(P=0.034).Patients who underwent a nerve-sparing procedure were less odds to experience SUI(P=0.014).A significant positive correlation between LBC and DO was observed(P<0.001).A greater length of the resected vagina and chemoradiotherapy were common risk factors for LBC and DO,while radiotherapy exerted a stronger effect than chemotherapy.Additionally,patients who received chemoradiotherapy frequently developed a DMCC.The follow-up time was not correlated with bladder storage function.Conclusion:A nerve-sparing procedure without longer resected vagina is recommended for protecting the bladder storage function.Ting-Ting Cao Hong-Wu Wen Yu-Nong Gao Qiu-Bo Lyu Hui-Xin Liu Sha Wang Shi-Yan Wang Hua-Xin Sun Na Yu Hai-Bo Wang Yi Li Zhi-Qi Wang Olivia H.Chang Xiu-Li Sun Jian-Liu Wang 2020Chinese Medical Journal2020,,19:7
9Lipoprotein(a) as a Marker for Predicting the Presence and Severity of Coronary Artery Disease in Untreated Chinese Patients Undergoing Coronary Angiography显示文摘Objective Low-density lipoprotein cholesterol(LDL-C) has been well known as the risk factor of coronary artery disease(CAD). However, the role of lipoprotein(a) [Lp(a)] in the development of CAD is of great interest but still controversial. Thus, we aim to explore the effect of Lp(a) on predicting the presence and severity of CAD in Chinese untreated patients, especially in combination with LDL-C. Methods We consecutively recruited 1,980 non-treated patients undergoing coronary angiography, among which 1,162 patients were diagnosed with CAD. Gensini score(GS) was used to assess the severity of CAD. Lp(a) was measured by immunoturbidimetric method. Results Patients with CAD had higher level of LDL-C and Lp(a) compared with non-CAD(P < 0.05). Multivariable logistic regression revealed that Lp(a) > 205 mg/L(highest tertile) predicted 1.437-fold risk for CAD(95% CI: 1.108-1.865, P = 0.006) and 1.480-fold risk for high GS(95% CI: 1.090-2.009, P = 0.012) respectively. Interestingly, concomitant elevated level of Lp(a) and LDL-C conferred the highest risk for both presence [OR = 1.845, 95% CI: 1.339-2.541, P < 0.001] and severity [OR = 1.736, 95% CI: 1.188-2.538, P = 0.004] of CAD. 10Circulating MicroRNAs as Novel Diagnostic Biomarkers for Very Early-onset(≤40 years) Coronary Artery Disease显示文摘Objective Very early-onset coronary artery disease(CAD) is a great challenge in cardiovascular medicine throughout the world,especially regarding its early diagnosis.This study explored whether circulating micro RNAs(mi RNAs) could be used as potential biomarkers for patients with very early-onset CAD.Methods We performed an initial screening of mi RNA expression using RNA isolated from 20 patients with angiographically documented very early-onset CAD and 20 age-and sex-matched normal controls.For further confirmation,we prospectively examined the mi RNAs selected from 40 patients with very early-onset CAD and 40 angiography-normal controls.Results A total of 22 overexpressed mi RNAs and 22 underexpressed mi RNAs were detected in the initial screening.RT-q PCR analysis of the mi RNAs obtained from the initial screening revealed that four mi RNAs including mi R-196-5p,mi R-3163-3p,mi R-145-3p,and mi R-190a-5p exhibited significantly decreased expression in patients compared with that in controls(P<0.05).The areas under the receiver operating characteristic curve for these mi RNAs were 0.824(95% CI,0.731-0.917;P<0.001),0.758(95% CI,0.651-0.864;P<0.001),0.753(95% CI,0.643-0.863;P<0.001),and 0.782(95% CI,0.680-0.884;P<0.001),respectively,in the validation set.Conclusion To our knowledge,this is an advanced study to report about four serum mi RNAs(mi R-196-5p,mi R-3163-3p,mi R-145-3p,and mi R-190a-5p) that could be used as novel biomarkers for the diagnosis of very early-onset CAD.DU Ying YANG Sheng Hua LI Sha CUI Chuan Jue ZHANG Yan ZHU Cheng Gang GUO Yuan Lin WU Na Qiong GAO Ying SUN Jing DONG Qian LIU Geng LI Jian Jun 2016Biomedical and Environmental Sciences2016,29,8:5
11Effects of land-use types on the vertical distribution of fractions of oxidizable organic carbon on the Loess Plateau, China显示文摘The oxidizability of soil organic carbon(SOC_) influences soil quality and carbon sequestration. Four fractions of oxidizable organic carbon(very labile(C_1), labile(C_2), less labile(C_3) and non-labile(C_4)) reflect the status and composition of SOC_ and have implications for the change and retention of SOC_. Studies of the fractions of oxidizable organic carbon(OC_) have been limited to shallow soil depths and agroecosystems. How these fractions respond at deep soil depths and in other types of land-use is not clear. In this study, we evaluated the vertical distributions of the fractions of oxidizable organic carbon to a soil depth of 5.0 m in 10 land-use types in the Zhifanggou Watershed on the Loess Plateau, C_hina. Along the soil profile, C_1 contents were highly variable in the natural grassland and shrubland I(C_aragana microphylla), C_2 and C_4 contents were highly variable in the natural grassland and two terraced croplands, respectively, and C_3 contents varied little. Among the land-use types, natural grassland had the highest C__1 and C_2 contents in the 0–0.4 m layers, followed by shrubland I in the 0–0.1 m layer. Natural grassland had the highest C_4 contents in the 1.0–4.5 m layers. 12Integrating full-length transcriptomics and metabolomics reveals the regulatory mechanisms underlying yellow pigmentation in tree peony(Paeonia suffruticosa Andr.)flowers显示文摘Tree peony(Paeonia suffruticosa Andr.)is a popular ornamental plant in China due to its showy and colorful flowers.However,yellow-colored flowers are rare in both wild species and domesticated cultivars.The molecular mechanisms underlying yellow pigmentation remain poorly understood.Here,petal tissues of two tree peony cultivars,“High Noon”(yellow flowers)and“Roufurong”(purple–red flowers),were sampled at five developmental stages(S1–S5)from early flower buds to full blooms.Five petal color indices(brightness,redness,yellowness,chroma,and hue angle)and the contents of ten different flavonoids were determined.Compared to“Roufurong,”which accumulated abundant anthocyanins at S3–S5,the yellow-colored“High Noon”displayed relatively higher contents of tetrahydroxychalcone(THC),flavones,and flavonols but no anthocyanin production.The contents of THC,flavones,and flavonols in“High Noon”peaked at S3 and dropped gradually as the flower bloomed,consistent with the color index patterns.Furthermore,RNA-seq analyses at S3 showed that structural genes such as PsC4Hs,PsDFRs,and PsUFGTs in the flavonoid biosynthesis pathway were downregulated in“High Noon,”whereas most PsFLSs,PsF3Hs,and PsF3’Hs were upregulated.Five transcription factor(TF)genes related to flavonoid biosynthesis were also upregulated in“High Noon.”One of these TFs,PsMYB111,was overexpressed in tobacco,which led to increased flavonols but decreased anthocyanins.Dual-luciferase assays further confirmed that PsMYB111 upregulated PsFLS.These results improve our understanding of yellow pigmentation in tree peony and provide a guide for future molecular-assisted breeding experiments in tree peony with novel flower colors.Xiaoning Luo Daoyang Sun Shu Wang Sha Luo Yaqi Fu Lixin Niu Qianqian Shi Yanlong Zhang 2021Horticulture Research2021,8,1:5
13Effects of Sc Addition and Annealing Treatment on the Microstructure and Mechanical Properties of the As-rolled Mg-3Li alloy显示文摘The microstructures and mechanical properties of the as-rolled Mg-3Li and Mg-3Li-1Sc (wt%) alloys before and after annealing treatment have been investigated. Results show that the grains are refined evidently and the recrystallization temperature is improved for more than 100°C by adding 1 wt% Sc into the Mg-3Li alloy. After complete recrystallization, both the strength and ductility of the Mg-3Li alloy are improved evidently with the addition of minor Sc. 15Consensus on the digestive endoscopic tunnel technique显示文摘With the digestive endoscopic tunnel technique(DETT), many diseases that previously would have been treated by surgery are now endoscopically curable by establishing a submucosal tunnel between the mucosa and muscularis propria(MP). Through the tunnel, endoscopic diagnosis or treatment is performed for lesions in the mucosa, in the MP, and even outside the gastrointestinal(GI) tract.At present, the tunnel technique application range covers the following:(1)Treatment of lesions originating from the mucosal layer, e.g., endoscopic submucosal tunnel dissection for oesophageal large or circular early-stage cancer or precancerosis;(2) treatment of lesions from the MP layer, per-oral endoscopic myotomy, submucosal tunnelling endoscopic resection, etc.; and(3) diagnosis and treatment of lesions outside the GI tract, such as resection of lymph nodes and benign tumour excision in the mediastinum or abdominal cavity. With the increasing number of DETTs performed worldwide, endoscopic tunnel therapeutics, which is based on DETT, has been gradually developed and optimized. However, there is not yet an expert consensus on DETT to regulate its indications, contraindications, surgical procedure, and postoperative treatment.The International DETT Alliance signed up this consensus to standardize the procedures of DETT. 16Warburg Effects in Cancer and Normal Proliferating Cells:Two Tales of the Same Name显示文摘It has been observed that both cancer tissue cells and normal proliferating cells(NPCs)have the Warburg effect.Our goal here is to demonstrate that they do this for different reasons.To accomplish this,we have analyzed the transcriptomic data of over 7000 cancer and control tissues of 14 cancer types in TCGA and data of five NPC types in GEO.Our analyses reveal that NPCs accumulate large quantities of ATPs produced by the respiration process before starting the Warburg effect,to raise the intracellular pH from 6.8 to 7.2 and to prepare for cell division energetically.Once cell cycle starts,the cells start to rely on glycolysis for ATP generation followed by ATP hydrolysis and lactic acid release,to maintain the elevated intracellular pH as needed by cell division since together the three processes are pH neutral.The cells go back to the normal respirationbased ATP production once the cell division phase ends.In comparison,cancer cells have reached their intracellular pH at 7.4 from top down as multiple acid-loading transporters are up-regulated and most acid-extruding ones except for lactic acid exporters are repressed.Cancer cells use continuous glycolysis for ATP production as way to acidify the intracellular space since the lactic acid secretion is decoupled from glycolysis-based ATP generation and is pH balanced by increased expressions of acid-loading transporters.Co-expression analyses suggest that lactic acid secretion is regulated by external,non-pH related signals.Overall,our data strongly suggest that the two cell types have the Warburg effect for very different reasons.Huiyan Sun Liang Chen Sha Cao Yanchun Liang Ying Xu 2019Genomics, Proteomics & Bioinformatics2019,17,3:4
17The role of pineal microRNA-325 in regulating circadian rhythms after neonatal hypoxic-ischemic brain damage显示文摘Circadian rhythm disorder is a common,but often neglected,consequence of neonatal hypoxic-ischemic brain damage(HIBD).However,the underlying molecular mechanisms remain largely unknown.We previously showed that,in a rat model of HIBD,up-regulation of microRNA-325(miR-325)in the pineal gland is responsible for the suppression of Aanat,a key enzyme involved in melatonin synthesis and circadian rhythm regulation.To better understand the mechanism by which miR-325 affects circadian rhythms in neonates with HIBD,we compared clinical samples from neonates with HIBD and samples from healthy neonates recruited from the First Affiliated Hospital of Soochow University(Dushuhu Branch)in 2019.We found that circulating miR-325 levels correlated positively with the severity of sleep and circadian rhythm disorders in neonates with HIBD.Furthermore,a luciferase reporter gene assay revealed that LIM homeobox 3(LHX3)is a novel downstream target of miR-325.In addition,in miR-325 knock-down mice,the transcription factor LHX3 exhibited an miR-325-dependent circadian pattern of expression in the pineal gland.We established a neonatal mouse model of HIBD by performing doublelayer ligation of the left common carotid artery and exposing the pups to a low-oxygen environment for 2 hours.Lhx3 mRNA expression was significantly down-regulated in these mice and partially rescued in miR-325 knockout mice subjected to the same conditions.Finally,we showed that improvement in circadian rhythm-related behaviors in animals with HIBD was dependent on both miR-325 and LHX3.Taken together,our findings suggest that the miR-325-LHX3 axis is responsible for regulating circadian rhythms and provide novel insights into the identification of potential therapeutic targets for circadian rhythm disorders in patients with neonatal HIBD.The clinical trial was approved by Institutional Review Board of Children’s Hospital of Soochow University(approval No.2015028)on July 20,2015.Animal experiments were approved by Animal Care and Use Committee,School of Medicine,Soochow University,China(approval No.XD-2016-1)on January 15,2016.Ning Sha Hua-Wei Wang Bin Sun Min Gong Po Miao Xiao-Lu Jiang Xiao-Feng Yang Mei Li Li-Xiao Xu Chen-Xi Feng Yuan-Yuan Yang Jie Zhang Wen-Jing Zhu Yuan-Yuan Gao Xing Feng Xin Ding 2021Neural Regeneration Research2021,16,10:4
18Long-term adenosine A1 receptor activation-induced sortilin expression promotes α-synuclein upregulation in dopaminergic neurons显示文摘Prolonged activation of adenosine A1 receptor likely leads to damage of dopaminergic neurons and subsequent development of neurodegenerative diseases.However,the pathogenesis underlying long-term adenosine A1 receptor activation-induced neurodegeneration remains unclear.In this study,rats were intraperitoneally injected with 5 mg/kg of the adenosine A1 receptor agonist N6-cyclopentyladenosine(CPA)for five weeks.The mobility of rats was evaluated by forced swimming test,while their cognitive capabilities were evaluated by Y-maze test.Expression of sortilin,α-synuclein,p-JUN,and c-JUN proteins in the substantia nigra were detected by western blot analysis.In addition,immunofluorescence staining of sortilin andα-synuclein was performed to detect expression in the substantia nigra.The results showed that,compared with adenosine A1 receptor antagonist 8-cyclopentyl-1,3-dipropylxanthine(5 mg/kg)+CPA co-treated rats,motor and memory abilities were reduced,surface expression of sortin andα-synuclein in dopaminergic neurons was reduced,and total sortilin and totalα-synuclein were increased in CPA-treated rats.MN9D cells were incubated with 500 nM CPA alone or in combination with 10μM SP600125(JNK inhibitor)for 48 hours.Quantitative real-time polymerase chain reaction analysis of sortilin andα-synuclein mRNA levels in MN9D cells revealed upregulated sortilin expression in MN9D cells cultured with CPA alone,but the combination of CPA and SP600125 could inhibit this expression.Predictions made using Jasper,PROMO,and Alibaba online databases identified a highly conserved sequence in the sortilin promoter that was predicted to bind JUN in both humans and rodents.A luciferase reporter assay of sortilin promoter plasmid-transfected HEK293T cells confirmed this prediction.After sortilin expression was inhibited by sh-SORT1,expression of p-JUN and c-JUN was detected by western blot analysis.Long-term adenosine A1 receptor activation levels upregulatedα-synuclein expression at the post-transcriptional level by affecting sortilin expression.The online tool Raptor-X-Binding and Discovery Studio 4.5 prediction software predicted that sortilin can bind toα-synuclein.Co-immunoprecipitation revealed an interaction between sortilin andα-synuclein in MN9D cells.Our findings indicate that suppression of prolonged adenosine A1 receptor activation potently inhibited sortilin expression andα-synuclein accumulation,and dramatically improved host cognition and kineticism.This study was approved by the University Committee of Animal Care and Supply at the University of Saskatchewan(approval No.AUP#20070090)in March 2007 and the Animals Ethics Committee of University of South China(approval No.LL0387-USC)in June 2017.Yun-Cheng Lv An-Bo Gao Jing Yang Li-Yuan Zhong Bo Jia Shu-Hui Ouyang Le Gui Tian-Hong Peng Sha Sun Francisco S.Cayabyab 2020Neural Regeneration Research2020,15,4:4
19Sortilin promotes macrophage cholesterol accumulation and aortic atherosclerosis through lysosomal degradation of ATP-binding cassette transporter A1 protein显示文摘Sortilin is closely associated with hyperlipidemia and the risk of atherosclerosis(AS).The role of sortilin and the underlying mechanism in peripheral macrophage are not fully understood.In this study,we investigated the effect of macrophage sortilin on ATP-binding cassette transporter A1(ABCA1)expression,ABCA1-mediated cholesterol efflux,and aortic AS.Macrophage sortilin expression was upregulated by oxidized low-density lipoproteins(ox-LDLs)in both concentration-and time-dependent manners.Its expression reached the peak level when cells were incubated with 50μg/ml ox-LDL for 24 h.Overexpression of sortilin in macrophage reduced cholesterol efflux,leading to an increase in intracellular total cholesterol,free cholesterol,and cholesterol ester.Sortilin was found to bind with ABCA1 protein and suppress macrophage ABCA1 expression,resulting in a decrease in cholesterol efflux from macrophages.The inhibitory effect of sortilin in cholesterol efflux was partially reversed by treatment with chloroquine,a lysosomal inhibitor.On the contrary,the ABCA1 protein level and ABCA1-mediated cholesterol efflux is increased by sortilin short hairpin RNA transfection.The fecal and biliary cholesterol 3H-sterol from cholesterol-laden mouse peritoneal macrophage was reduced by sortilin overexpression through lentivirus vector(LV)-sortilin in low-density lipoprotein receptor knockout mice,which was prevented by co-treatment with chloroquine.Treatment with LV-sortilin reduced plasma high-density lipoprotein and increased plasma ox-LDL levels.Accordingly,aortic lipid deposition and plaque area were exacerbated,and ABCA1 expression was reduced in mice in response to infection with LV-sortilin alone.These effects of LV-sortilin were partially reversed by chloroquine.Sortilin enhances lysosomal degradation of ABCA1 protein and suppresses ABCA1-mediated cholesterol efflux from macrophages,leading to foam cell formation and AS development.Yuncheng Lv Jing Yang Anbo Gao Sha Sun Xilong Zheng Xi Chen Wei Wan Chaoke Tang Wei Xie Suyun Li Dongming Guo Tianhong Peng Guojun Zhao Liyuan Zhong 2019Acta Biochimica et Biophysica Sinica2019,51,5:4
