| 1 | Osteopontin increases hepatocellular carcinoma cell growth in a CD44 dependant manner显示文摘AIM:To investigate the role of osteopontin(OPN) and its splice variants in the proliferation of hepatocellular carcinoma(HCC).METHODS:The expression of OPN variants in HCC cell lines as well as HCC tissue samples and nontumour tissue was studied using polymerase chain reaction.OPN variant cDNAs were cloned into a mammalian expression vector allowing both transient expression and the production of stable OPN expressing cell lines.OPN expression was studied in these cells using Western blotting,immunofluoresnce and enzyme linked immunosorbent assay.A CD44 blocking antibody and siRNA targeting of CD44 were used to examine the role of this receptor in the OPN stimulated cell growth observed in culture.Huh-7 cells stably expressing either OPN-A,-B or-C were injected subcutaneously into the flanks of nude mice to observe in vivo tumour growth.Expression of OPN mRNA and protein in these tumours was examined using reverse transcriptionpolymerase chain reaction and immunohistochemistry.RESULTS:OPN is expressed in HCC in 3 forms,the full length OPN-A and 2 splice variants OPN-B and-C.OPN variant expression was noted in HCC tissue as well as cognate surrounding cirrhotic liver tissue.Expression of these OPN variants in the HCC derived cell line Huh-7 resulted in secretion of OPN into the culture medium.Transfer of OPN conditioned media to na ve Huh-7 and HepG2 cells resulted in significant cell growth suggesting that all OPN variants can modulate cell proliferation in a paracrine manner.Furthermore the OPN mediated increase in cellular proliferation was dependent on CD44 as only CD44 positive cell lines responded to OPN conditioned media while siRNA knockdown of CD44 blocked the proliferative effect.OPN expression also increased the proliferation of Huh-7 cells in a subcutaneous nude mouse tumour model,with Huh-7 cells expressing OPN-A showing the greatest proliferative effect.CONCLUSION:This study demonstrates that OPN plays a significant role in the proliferation of HCC through interaction with the cell surface receptor CD44.Modulation of this interaction could represent a novel strategy for the control of HCC. | Renee J Phillips Karla J Helbig Kylie H Van der Hoek Devanshi Seth Michael R Beard | 2012 | World Journal of Gastroenterology2012，18，26： | 12 |
| 2 | Peripheral primitive neuroectodermal tumor of the kidney presenting with pulmonary tumor embolism: A case report显示文摘Peripheral primitive neuroectodermal tumor(PNET) of the kidney is a rare, aggressive tumor known for its recurrence and metastatic potential. Despite the frequency of venous extension to the renal veins and inferior vena cava, pulmonary tumor embolism at the initial presentation is not common. We report a case of 22-year-old female with PNET of the kidney who presented with tumor embolism in the inferior vena cava(IVC) and bilateral pulmonary artery. The patient underwent surgical resection and histopathological analysis confirmed the presence of tumor within the IVC and pulmonary arteries. | 3 | Dyslipidemia management in primary prevention of cardiovascular disease:Current guidelines and strategies显示文摘Cardiovascular disease is the leading cause of death in the United States. In 2010, the Centers for Disease Control and Prevention estimated that $444 billion was spent on cardiovascular diseases alone, about $1 of every $6 spent on health care. As life expectancy continues to increase, this annual cost will also increase, making costeffective primary prevention of cardiovascular disease highly desirable. Because of its role in development of atherosclerosis and clinical events, dyslipidemia management is a high priority in cardiovascular prevention. Multiple major dyslipidemia guidelines have been published around the world recently, four of them by independent organizations in the United States alone. They share the goal of providing clinical guidance on optimal dyslipidemia management, but guidelines differ in their emphasis on pharmacotherapy, stratification of groups, emphasis on lifestyle modification, and use of a fixed target or percentage reduction in low density lipoprotein cholesterol. This review summarizes eight major guidelines for dyslipidemia management and considers the basis for their recommendations. | 7 | Landscape features and weather influence nest survival of a ground-nesting bird of conservation concern,the greater sage-grouse,in humanaltered environments显示文摘Introduction:Ground-nesting birds experience high levels of nest predation.However,birds can make selection decisions related to nest site location and characteristics that may result in physical,visual,and olfactory impediments to predators.Methods:We studied daily survival rate[DSR]of greater sage-grouse(Centrocercus urophasianus)from 2008 to 2010 in an area in Wyoming experiencing large-scale alterations to the landscape.We used generalized linear mixed models to model fixed and random effects,and a correlation within nesting attempts,individual birds,and years.Results:Predation of the nest was the most common source of nest failure(84.7%)followed by direct predation of the female(13.6%).Generally,landscape variables at the nest site(≤30 m)were more influential on DSR of nests than features at larger spatial scales.Percentage of shrub canopy cover at the nest site(15-m scale)and distances to natural gas wells and mesic areas had a positive relationship with DSR of nests,whereas distance to roads had a negative relationship with DSR of nests.When added to the vegetation model,maximum wind speed on the day of nest failure and a 1-day lag in precipitation(i.e.,precipitation the day before failure)improved model fit whereby both variables negatively influenced DSR of nests.Conclusions:Nest site characteristics that reduce visibility(i.e.,shrub canopy cover)have the potential to reduce depredation,whereas anthropogenic(i.e.,distance to wells)and mesic landscape features appear to facilitate depredation.Last,predators may be more efficient at locating nests under certain weather conditions(i.e.,high winds and moisture). | Stephen L Webb Chad V Olson Matthew R Dzialak Seth M Harju Jeffrey B Winstead Dusty Lockman | 2012 | Ecological Processes2012，1，1： | 2 |
| 8 | Clinical outcomes for Conduits and Scaffolds in peripheral nerve repair显示文摘The gold standard of peripheral nerve repair is nerve autograft when tensionless repair is not possible. Use of nerve autograft has several shortcomings, however.These include limited availability of donor tissue,sacrifice of a functional nerve, and possible neuroma formation. In order to address these deficiencies,researchers have developed a variety of biomaterials available for repair of peripheral nerve gaps. We review the clinical studies published in the English literature detailing outcomes and reconstructive options.Regardless of the material used or the type of nerve repaired, outcomes are generally similar to nerve autograft in gaps less than 3 cm.
