1Endosonography-guided cholangiopancreatography as a salvage drainage procedure for obstructed biliary and pancreatic ducts显示文摘Endoscopic ultrasound allows transmural access to the bile or pancreatic ducts and subsequent contrast injection to provide ductal drainage under fluoroscopy using endoscopic retrograde cholangiopancreatography (ERCP)-based techniques.Differing patient specifics and operator techniques result in six possible variant approaches to this procedure,known as endosonography-guided cholangiopancreatography(ESCP). 2Diffuse intestinal ganglioneuromatosis an uncommon manifestation of Cowden syndrome显示文摘Diffuse intestinal ganglioneuromatosis is a hamartomatous polyposis characterized by a disseminated, intramural or transmural proliferation of neural elements involving the enteric plexuses. It has been associated with MEN Ⅱ, neurofibromatosis type 1 and hamartomatous polyposis associated with phosphatase and tensin homolog mutation. We report the case of a female patient with a history of a breast and endometrial tumor who presented in a colonoscopy performed for rectal bleeding diffuse ganglioneuromatosis, which oriented the search for other characteristic findings of Cowden syndrome given the personal history of the patient. The presence of an esophagogastric polyposis was also noted. Cowden syndrome is characterized by skin lesions, but it is rarely diagnosed by these lesions, because they are usually overlooked. Intestinal polyposis is not a major diagnostic criterion but it is very useful for early diagnosis. 3Nonlinear optical properties of integrated GeSbS chalcogenide waveguides显示文摘In this paper, we report the experimental characterization of highly nonlinear Ge Sb S chalcogenide glass waveguides.We used a single-beam characterization protocol that accounts for the magnitude and sign of the real and imaginary parts of the third-order nonlinear susceptibility of integrated Ge23 Sb7 S70(GeSbS) chalcogenide glass waveguides in the near-infrared wavelength range at λ =1580 nm. We measured a waveguide nonlinear parameter of 7.0±0.7 W^(-1)· m(-1), which corresponds to a nonlinear refractive index of n_2=0.93±0.08 × 10^(-18) m^2∕W,comparable to that of silicon, but with an 80 times lower two-photon absorption coefficient βTPA=0.010± 0.003 cm∕GW, accompanied with linear propagation losses as low as 0.5 dB/cm. 6Controlling self-assembling and tumor cell-targeting of protein-only nanoparticles through modular protein engineering显示文摘Modular protein engineering is suited to recruit complex and multiple functionalities in single-chain polypeptides. Although still unexplored in a systematic way, it is anticipated that the positioning of functional domains would impact and refine these activities, including the ability to organize as supramolecular entities and to generate multifunctional protein materials. To explore this concept, we have repositioned functional segments in the modular protein T22-GFP-H6 and characterized the resulting alternative fusions. In T22-GFP-H6, the combination of T22 and H6 promotes selfassembling as regular nanoparticles and selective binding and internalization of this material in CXCR4-overexpressing tumor cells, making them appealing as vehicles for selective drug delivery. The results show that the pleiotropic activities are dramatically affected in module-swapped constructs, proving the need of a carboxy terminal positioning of H6 for protein self-assembling, and the accommodation of T22 at the amino terminus as a requisite for CXCR4^+ cell binding and internalization. 7Hybrid silicon slotted photonic crystal waveguides:how does third order nonlinear performance scale with slow light?显示文摘We investigate in this paper the influence of slow light on the balance between the Kerr and two-photon absorption(TPA) processes in silicon slotted hybrid nonlinear waveguides. Three typical silicon photonic waveguide geometries are studied to estimate the influence of the light slow-down factor on the mode field overlap with the silicon region, as well as on the complex effective nonlinear susceptibility. It is found that slotted photonic crystal modes tend to focalize in their hollow core with increasing group index(n_G) values. Considering a hybrid integration of nonlinear polymers in such slotted waveguides, a relative decrease of the TPA process by more factor of 2 is predicted from n_G=10 to n_G=50. 9Bioengineering bacterial encapsulin nanocompartments as targeted drug delivery system显示文摘The development of Drug Delivery Systems(DDS)has led to increasingly efficient therapies for the treatment and detection of various diseases.DDS use a range of nanoscale delivery platforms produced from polymeric of inorganic materials,such as micelles,and metal and polymeric nanoparticles,but their variant chemical composition make alterations to their size,shape,or structures inherently complex.Genetically encoded protein nanocages are highly promising DDS candidates because of their modular composition,ease of recombinant production in a range of hosts,control over assembly and loading of cargo molecules and biodegradability.One example of naturally occurring nanocompartments are encapsulins,recently discovered bacterial organelles that have been shown to be reprogrammable as nanobioreactors and vaccine candidates.Here we report the design and application of a targeted DDS platform based on the Thermotoga maritima encapsulin reprogrammed to display an antibody mimic protein called Designed Ankyrin repeat protein(DARPin)on the outer surface and to encapsulate a cytotoxic payload.The DARPin9.29 chosen in this study specifically binds to human epidermal growth factor receptor 2(HER2)on breast cancer cells,as demonstrated in an in vitro cell culture model.The encapsulin-based DDS is assembled in one step in vivo by co-expressing the encapsulin-DARPin9.29 fusion protein with an engineered flavin-binding protein mini-singlet oxygen generator(MiniSOG),from a single plasmid in Escherichia coli.Purified encapsulin-DARPin_miniSOG nanocompartments bind specifically to HER2 positive breast cancer cells and trigger apoptosis,indicating that the system is functional and specific.The DDS is modular and has the potential to form the basis of a multi-receptor targeted system by utilising the DARPin screening libraries,allowing use of new DARPins of known specificities,and through the proven flexibility of the encapsulin cargo loading mechanism,allowing selection of cargo proteins of choice.Alexander Van de Steen Rana Khalife Noelle Colant Hasan Mustafa Khan Matas Deveikis Saverio Charalambous Clare M.Robinson Rupali Dabas Sofia Esteban Serna Diana A.Catana Konstantin Pildish Vladimir Kalinovskiy Kenth Gustafsson Stefanie Frank 2021Synthetic and Systems Biotechnology2021,6,3:2
