1Famitinib in combination with concurrent chemoradiotherapy in patients with locoregionally advanced nasopharyngeal carcinoma: a phase 1, open-label, dose-escalation Study显示文摘Background:Famitinib is a tyrosine kinase inhibitor against multiple targets,including vascular endothelial growth factor receptor 2/3,platelet-derived growth factor receptor,and stem cell factor receptor(c-kit).Previous studies have demonstrated anti-tumour activities of famitinib against a wide variety of advanced-stage solid cancers.We aimed to determine the safety and efficacy of famitinib with concurrent chemoradiotherapy(CCRT)in patients with locoregionally advanced nasopharyngeal carcinoma(NPC).We also evaluated the feasibility of contrast-enhanced ultrasound(D-CEUS)as a predictor of early tumour response to famitinib and to correlate functional parameters with clinical efficacy.Methods:The trial was conducted in subjects with stage III or IVa-b NPC using a 3+3 design of escalating fami-tinib doses.Briefly,subjects received 2 weeks of famitinib monotherapy followed by 7 weeks of famitinib plus CCRT.D-CEUS of the neck lymph nodes was performed at day 0,8 and 15 after famitinib was administered before starting concurrent chemoradiotherapy.End points included safety,tolerability and anti-tumour activity.Results:Twenty patients were enrolled(six each for 12.5,16.5 and 20 mg and two for 25 mg).Two patients in the 25 mg cohort developed dose-limiting toxicities,including grade 4 thrombocytopenia and grade 3 hypertension.The most common grade 3/4 adverse events were leukopenia,neutropenia and radiation mucositis.D-CEUS tests showed that more than 60%of patients achieved a perfusion parameter response after 2 weeks taking famitinib alone,and the parameter response was associated with disease improvement.In the famitinib monotherapy stage,three patients(15%)showed partial responses.The complete response rate was 65%at the completion of treatment and 95%3 months after the treatment ended.After a median follow-up of 44 months,the 3-year progression-free survival(PFS)and distant metastasis-free survival were 70%and 75%,respectively.Subjects with a decrease of perfusion parameter response,such as peak intensity decreased at least 30%after 1 week of famitinib treatment,had higher 3-year PFS(90.9%vs.44.4%,95%CI 73.7%-100%vs.11.9%-76.9%,P<0.001)than those with an increase or a reduction of less than 30%.Conclusions:The recommended famitinib dose for phase II trial is 20 mg with CCRT for patients with local advanced NPC.D-CEUS is a reliable and early measure of efficacy for famitinib therapies.Further investigation is required to confirm the effects of famitinib plus chemoradiotherapy.Qiuyan Chen Linquan Tang Na Liu Feng Han Ling Guo Shanshan Guo Jianwei Wang Huai Liu Yanfang Ye Lu Zhang Liting Liu Pan Wang Yingqin Li Qingmei He Xiaoqun Yang Qingnan Tang Yang Li YuJing Liang XueSong Sun Chuanmiao Xie Yunxian Mo Ying Guo Rui Sun Haoyuan Mo Kajia Cao Xiang Guo Musheng Zeng Haiqiang Mai Jun Ma 2018Cancer Communications2018,38,1:6

3Tunable Electronic and Magnetic Properties from Structure Phase Transition of Layered Vanadium Diselenide显示文摘The atomic geometry, structure stability, electronic and magnetic properties of VSe_2 were systematically investigated based on the density functional theory(DFT). Varying from 3D to 2D four VSe_2 structures, bulk 2H-VSe_2 and 1T-VSe_2, monolayer H-VSe_2 and T-VSe_2 are all demonstrated as thermodynamically stable by lattice dynamic calculations. More interestingly, the phase transition temperature is dramatically different due to the lattice size. Finally, owing to different crystal structures, H-VSe_2 is semimetallic whereas T-VSe_2 is totally metallic and also they have different magnetic moments. Our main argument is that being exfoliated from bulk to monolayer, 2H-VSe_2 transforms to T-VSe_2, accompanied by both semimetallic-metallic transition and dramatic magnetic moment variation. 7Vasorelaxant effect of osthole on isolated thoracic aortic rings in rats显示文摘OBJECTIVE: To investigate the effect of osthole on isolated thoracic aortic rings, and to determine the potential mechanism of action.METHODS: Thoracic aortas were isolated from Wistar rats, and were suspended in tissue organ chambers for vascular tension measurement. The effect of cumulative osthole(10-9, 10-8, 10-7, 10-6, and 10-5 mol/L) on endothelium-intact and endothelium-denuded thoracic aortic rings pre-contracted with phenylephrine(PE, 10-6 mol/L) or KCl(6 × 10-2 mol/L) was recorded. Histomorphological changes of thoracic aorta were analyzed by hematoxylin-eosin. The effects of different osthole concentrations on endothelium-intact aortic rings, which were pre-inhibited with the non-selective nitric oxide synthase inhibitor L-Arg(NO2)-OMe·HCl(3 × 10-4 mol/L), endothelium-derived nitric oxide synthase inhibitor Nω-nitro-L-arginine(3 × 10-4 mol/L), guanylate cyclase inhibitor 1 H-[1,2,4] oxadiazolo [4,3-α]quinoxaline-1-one(10-5 mol/L), cyclooxygenase inhibitor indometacin(10-5 mol/L), and the Ca2+-activated potassium channel inhibitor tetraethylammonium nitrate(10-5 mol/L), and then contracted with PE, were examined. Aortic rings incubated with osthole(10-5 mol/L), phentolamine(10-5 mol/L), or verapamil(10-5 mol/L) in Ca2+-free Krebs-Henseleit solution(KHS) were stimulated with PE or KCl.RESULTS: There was a dose-dependent increase in vasorelaxation of isolated thoracic aortic rings(both with and without endothelium) with increasing osthole concentration. Hematoxylin-eosin staining showed that osthole significantly improved thoracic aorta ring morphology. Compared with the control group, there were also significant differences after incubation with L-Arg(NO2)-OMe ω-nitro-L-arginine, and 1 H-[1,2,4] oxadiazo·lo HCl,N [4,3-α] quinoxaline-1-one(P < 0.05 for all). The relaxation rate of the rings in the osthole group incubated with indometacin and tetraethylammonium nitrate were similar to controls. In Ca2+-free KHS, the PE-induced contraction was similar between the osthole(4.37% ± 0.41%) and control(4.21% ± 1.33%)groups. However, after cumulative CaCl2(0.5, 1, 1.5,2, 2.5, and 3 mmol/L), the Ca2+-induced contraction was significantly inhibited in the osthole and phentolamine groups compared with controls(P < 0.05).After cumulative CaCl2 was added to Ca2+-free KHS(high K+ concentration), the contraction rate was significantly higher than both of the control and the osthole groups(P < 0.05). 15First detection of cutavirus DNA in stools of patients with rheumatic diseases in Guangzhou,China显示文摘Cutavirus(CuV)is a novel protoparvovirus possibly associated with diarrhea and cutaneous T-cell lymphomas.Patients with rheumatic disease are immunosuppressed and may be more vulnerable to pathogenic viruses.A descriptive study was conducted among hospitalized patients with rheumatic diseases and individuals undergoing medical health check-ups between June 2019 and June 2022 in Guangzhou,China.Stool samples of subjects were tested for CuV DNA.Demographic and fecal examination data of patients were obtained from electronic medical records.A total of 505 patients with rheumatic diseases and 244 individuals who underwent medical health check-ups were included in the study.Of the patients with rheumatic disease,5.74%[95%confidence interval(CI):4.03%–8.12%]were positive for CuV DNA,while no individual in the medical health check-up group was positive,indicating a close correlation between CuV and rheumatic disease.Men and patients with rheumatoid arthritis or ankylosing spondylitis,according to the disease classification,were more susceptible to being infected with CuV(P<0.01).After adjustments,being male remained the only significant factor,with an adjusted odd ratio(OR)of 4.4(95%CI:1.7–11.4,P?0.002).Phylogenetic analysis of the CuV VP2 sequences showed three diverse clades,one of which was segregated to be a single branching independent of previously known sequences,which is possible a new genotype.Yongzhi Li Liting Zheng Huan He Husheng Xiong Jiaqi Chen Hengbiao Sun Caiyun Chen Qiushuang Li Jiaqi Fu Fei Wu Yuhan Gao Juxian Xian Minyi Liang Gang Xiao Qing Chen 2023Virologica Sinica2023,38,6:0
16Behaviours of Multi-Stakeholders under China’s Renewable Portfolio Standards:A Game Theory-Based Analysis显示文摘China has implemented both quantitative and policy incentives for renewable energy development since 2019 and is currently in the policy transition stage.The implementation of renewable portfolio standards(RPSs)is difficult due to the interests of multiple stakeholders,including power generation enterprises,power grid companies,power users,local governments,and the central government.Based on China’s RPS policy and power system reform documents,this research sorted out the core game decision problems of China’s renewable energy industry and established a conceptual game decision model of the renewable energy industry from the perspective of local governments,power generation enterprises and power grid companies.The results reveal that for local governments,the probability of meeting the earnings quota or punishments for not reaching quota completion are the major determinants for active participation in quota supervision.For power grid firms,the willingness to accept renewable electricity quotas depends on the additional cost of receiving renewable electricity and governmental incentives.It is reasonable,from the theoretical perspective,to implement the RPS policy on the power generation side.Electricity reform will help clarify the electricity price system and increase the transparency of the quota implementation process.Policy implications are suggested to achieve sustainable development of the renewable energy industry from price incentives and quantity delivery.Bing Wang Kailei Deng Liting He Zhenming Sun 2021Energy Engineering2021,118,5:0
17Establishment of a prognostic scoring model for regional recurrent nasopharyngeal carcinoma after neck dissection显示文摘Objective:The main aim of this study was to establish a scoring model to predict risk of progression and survival in patients with regionally recurrent nasopharyngeal carcinoma(NPC).Methods:Three hundred and forty-eight patients subjected to neck dissection from 2003 to 2017 were included for study.Clinicopathologic information for each patient was analyzed.Independent prognostic factors were selected using the Cox proportional hazards model and incorporated into the scoring model.Concordance index(C-index)and calibration curves were used to verify discrimination and calibration,respectively and the results validated using bootstrap resampling.Results:Microscopic positive lymph node>2[hazard ratio(HR),2.19;95%confidence interval(CI),1.30–3.68;P=0.003],extranodal extension(HR,2.75;95%CI,1.69–4.47;P<0.001),and lower neck involvement(HR,1.78;95%CI,1.04–3.04;P=0.034)were identified from multivariate analysis as independent factors for overall survival(OS).A qualitative 4-point scale was generated to stratify patients into 4 risk groups for predicting OS and progression-free survival(PFS).The novel scoring model demonstrated enhanced discrimination(C-index=0.69;95%CI,0.62–0.76)relative to the original recurrent tumor-node-metastasis(rTNM)staging system(C-index=0.56;95%CI,0.50–0.62),and was internally validated with a bootstrap-adjusted C-index of 0.70.The calibration curve showed good agreement between predicted probabilities and actual observations.Conclusions:The scoring system established in this study based on a large regionally recurrent NPC cohort fills a gap regarding assessment of risk and prediction of survival outcomes after neck dissection in this population and could be further applied to identify high-risk patients who may benefit from more aggressive intervention.Xiaoyun Li Chao Lin Jinjie Yan Qiuyan Chen Xuesong Sun Sailan Liu Shanshan Guo Liting Liu Haojun Xie Qingnan Tang Yujing Liang Ling Guo Hao Li Xuekui Liu Xiang Guo Linquan Tang Haiqiang Mai 2020Cancer Biology & Medicine2020,17,1:0

19Assembling ionic liquid into porous molecular filler of mixed matrix membrane to trigger high gas permeability,selectivity,and stability for CO_(2)/CH_(4) separation显示文摘As an emerging zero-dimensional nano crystalline porous material,porous organic cages(POCs)with soluble properties in organic solvents,are promising candidates as molecular fillers in mixed matrix membranes(MMMs).The pore structure of POCs should be adjusted to trigger efficient gas separation performance,and the interaction between filler and matrix should be optimized.In this work,ionic liquid(IL)was introduced into the molecular fillers of CC3,to construct the IL@CC3/PIM-1 membrane to effectively separate CO_(2) from CH_(4).The advantages of doping IL include:(1)narrowing the cavity size of POCs from 4.4 to 3.9Åto enhance the diffusion selectivity,(2)strengthening the CO_(2) solubility to heighten the gas permeability,and(3)improving the compatibility between filler and matrix to upgrade membrane stability.After the optimization of the membrane composite,the IL@CC3/PIM-1-10%membrane possesses the CO_(2) permeability of 7868 Barrer and the CO_(2)/CH_(4) selectivity of 73.4,which compared to the CC3/PIM-1-10%membrane,improved by 15.9%and 106.2%,respectively.Furthermore,the membrane has maintained a stable separation performance at varied temperatures and pressures during the long-term test.The proposed method offers an efficient way to improve the performance of POCs-based MMMs in gas separation.Liting Yu Liqin Hao Yang Feng Jia Pang Mengwei Guo Liangjun Li Weidong Fan Lili Fan Rongming Wang Zixi Kang Daofeng Sun 2024Nano Research2024,17,5:0
20Intrinsic spin shielding effect in platinum-rare-earth alloy boosts oxygen reduction activity显示文摘Oxygen reduction reactions(ORRs)involve a multistep proton-coupled electron process accompanied by the conversion of the apodictic spin configuration.Understanding the role of spin configurations of metals in the adsorption and desorption of oxygen intermediates during ORRs is critical for the design of efficient ORR catalysts.Herein,a platinum-rare-earth-metal-based alloy catalyst,Pt_(2)Gd,is introduced to reveal the role of spin configurations in the catalytic activity of materials.The catalyst exhibits a unique intrinsic spin reconfiguration because of interactions between the Gd-4f and Pt-5d orbitals.The adsorption and desorption of the oxygen species are optimized by modifying the spin symmetry and electronic structures of the material for increased ORR efficiency.The Pt_(2)Gd alloy exhibits a half-wave potential of 0.95 V and a superior mass activity of 1.5 A·mg Pt^(−1)in a 0.1 M HClO_(4)electrolyte,as well as higher durability than conventional Pt/C catalysts.Theoretical calculations have proven that the spin shielding effect of Gd pairs increases the spin symmetry of Pt-5d orbitals and adsorption preferences toward spin-polarized intermediates to facilitate ORR.This work clarifies the impact of modulating the intrinsic spin state of Pt through the interaction with the local high spin 4f orbital electrons in rare-earth metals,with the aim of boosting the spin-related oxygen reduction reaction,thus fundamentally contributing to the understanding of new descriptors that control ORR activity.Siyuan Zhu Mingzi Sun Bingbao Mei Liting Yang Yuyi Chu Zhaoping Shi Jingsen Bai Xian Wang Zheng Jiang Changpeng Liu Bolong Huang Junjie Ge Wei Xing 2023National Science Review2023,10,9:0
