2Cloning the Structure Genes and Expression the N Gene of Porcine Epidemic Diarrhea Virus DX显示文摘The structure genes spike(S) ,nucleocapsid(N) ,membrane(M) ,small membrane(sM) of a porcine epidemic diarrhea virus(PEDV) strain DX isolated in Gansu province,North-west of China,were cloned,sequenced and compared with published sequences of PEDV strains. The nucleotide sequences encoding the entire S,sM,M and N genes open reading frame(ORF) of DX were 4 152,231,681 and 1 326 bases long respectively. There were transcription regulatory sequences(TRSs) upstream of the initiator ATG of the S,N and M genes. The amino acids sequences of S,M and N contained 30,3 and 7 potential asparagine(N) -linked glycosylation sites. Homologous analysis and phylogenetic trees showed that DX had the closest relationship with strains LJB/06,JS-2004-2Z and CH/HLJH/06 that were also isolated from China and indicated the prevalence of some PEDV isolates in China were widespread since the JS-2004-2Z strain originated from the south of the China,and LJB/06 and CH/HLJH/06 were isolated from northeast China. The N gene was cloned using two primers which contained Nco I and BamH I restriction enzyme sites and subcloned into expression vector pET30a. The recombinant plasmid was then transformed into E.coli Rossta. 3PHYTOCHROME-INTERACTING FACTORS Interact with the ABA Receptors PYL8 and PYL9 to Orchestrate ABA Signaling in Darkness显示文摘PHYTOCHROME-INTERACTING FACTORS(PIFs)are a group of basic helix-loop-helix transcription factors that can physically interact with photoreceptors,including phytochromes and cryptochromes.It was previously demonstrated that PIFs accumulated in darkness and repressed seedling photomorphogenesis,and that PIFs linked different photosensory and hormonal pathways to control plant growth and development.In this study,we show that PIFs positively regulate the ABA signaling pathway during the seedling stage specifically in darkness.We found that PIFs positively regulate ABI5 transcript and protein levels in darkness in response to exogenous ABA treatment by binding directly to the G-box motifs in the ABI5 promoter.Consistently,PIFs and the G-box motifs in the ABI5 promoter determine ABI5 expression in darkness,and overexpression of ABI5 could rescue the ABA-insensitive phenotypes of pifq mutants in the dark.Moreover,we discovered that PIFs can physically interact with the ABA receptors PYL8 and PYL9,and that this interaction is not regulated by ABA.Further analyses showed that PYL8 and PYL9 promote PIF4 protein accumulation in the dark and enhance PIF4 binding to the ABI5 promoter,but negatively regulate PIF4-mediated ABI5 activation.Taken together,our data demonstrate that PIFs interact with ABA receptors to orchestrate ABA signaling in darkness by controlling ABI5 expression,providing new insights into the pivotal roles of PIFs as signal integrators in regulating plant growth and development.Lijuan Qi Shan Liu Cong Li Jingying Fu Yanjun Jing Jinkui Cheng Hong Li Dun Zhang Xiaoji Wang Xiaojing Dong Run Han Bosheng Li Yu Zhang Zhen Li William Terzaghi Chun-Peng Song Rongcheng Lin Zhizhong Gong Jigang Li 2020Molecular Plant2020,13,3:3

11Structure and Properties of Sol-Gel Derived CuNiMnZrO2 Catalysts for CO Hydrogenation显示文摘Two CuNiMnZrO2 catalysts (crystallized or non-crystallized) were prepared by co-hydrolyzing zirconium n-propoxide with Cu(NO3)2, Mn(NO3)2 and Ni(NO3)2 in an ethanol solution in the presence of diglycol. The physical and chemical properties of the catalysts were characterized using BET, XRD,TEM, XPS and CO-DRIFT techniques. 12Research on the Critical Buckling Pressure of Bucket Foundations with Inner Compartments for Offshore Wind Turbines显示文摘In the process of suction penetration of bucket foundations with inner compartments for offshore wind turbines,most researches focus on soil seepage failure and soil plugs,while the buckling of foundations is rarely investigated.Therefore,theoretical calculation methods for critical buckling pressures of the skirt and bulkheads of the bucket foundation are first presented according to the stability theory of a cylindrical shell and the small deflection theory of a thin plate,respectively.Furthermore,two types of models with and without considering the skirt-soil interaction are developed for the calculation of critical buckling pressure of the bucket foundation.Taking a practical project as an example,theoretical and numerical methods are used to obtain the critical buckling pressures of a bucket foundation.In this work,the theoretical method and the finite element model considering the skirt-soil interaction for calculating the critical buckling pressure of bucket foundations are firstly proposed.The results can help to optimize the design process of offshore wind turbine foundations and improve the safety of offshore wind power systems.SUN Guodong LIU Run LI Qingxin HUANG Guoyu ZHOU Zhenbo ZHANG Cheng 2023Journal of Ocean University of China2023,22,2:0
