1Predicting oral disintegrating tablet formulations by neural network techniques显示文摘Oral disintegrating tablets(ODTs) are a novel dosage form that can be dissolved on thetongue within 3 min or less especially for geriatric and pediatric patients. Current ODT for-mulation studies usually rely on the personal experience of pharmaceutical experts andtrial-and-error in the laboratory, which is inefficient and time-consuming. The aim of cur-rent research was to establish the prediction model of ODT formulations with direct com-pression process by artificial neural network(ANN) and deep neural network(DNN) tech-niques. 145 formulation data were extracted from Web of Science. All datasets were dividedinto three parts: training set(105 data), validation set(20) and testing set(20). ANN andDNN were compared for the prediction of the disintegrating time. The accuracy of the ANNmodel have reached 85.60%, 80.00% and 75.00% on the training set, validation set and testingset respectively, whereas that of the DNN model were 85.60%, 85.00% and 80.00%, respec-tively. Compared with the ANN, DNN showed the better prediction for ODT formulations.It is the first time that deep neural network with the improved dataset selection algorithmis applied to formulation prediction on small data. The proposed predictive approach couldevaluate the critical parameters about quality control of formulation, and guide researchand process development. 3Establishment of a Sandwich ELISA Method for Detection of Vascular Endothelial Growth Factor in Serum Samples of Hepatocellular Carcinoma Patients显示文摘Objective To establish a sandwich ELISA method for detecting vascular endothelial growth factor (VEGF) in sera of population and the patients with hepatocellular carcinoma (HCC). Methods Full length and two truncated human VEGF cDNA sequences were amplified from a commercial plasmid pBLAST49-hVEGF by PCR and inserted into the prokaryotic-expression plasmid pET-32a or pGEX-2T. Various VEGF proteins were expressed and purified from E. coli in His-Trx or GST fusion forms. The specific VEGF antibodies were elicited in experimental rabbits and mice by immunization of the full length VEGF fusion protein His-Trx-VEGF1-165. After purification of antibodies with chromatograph of Protein G, a sandwich ELISA technique was established. Serum VEGF levels were evaluated in 229 adults and 291 HCC patients. Results SDS-PAGE displayed that the molecular weights of the expressed full length (His-Trx-VEGF1-165), N-terminal (His-Trx-VEGF1-100) and C-terminal (GST-VEGF100-165) human VEGF fusion proteins were about 38KD, 31KD, and 33KD, respectively. Western blots confirmed that the prepared antisera were able to recognize both prokaryoticly and eukaryoticly expressed recombinant VEGF proteins. Assays of serially diluted His-Trx-VEGF1-100 by the established sandwich ELISA method showed that the linear range of the standard curve was 0.625-320 ng/mL, with the squared correlation coefficient R2=0.991. Screening of a serum panel containing 291 serum samples of HCC patients and 229 health adults revealed that the average VEGF level in HCC patients was higher than that in healthy controls, with a statically significant difference. 5PHYTOCHROME-INTERACTING FACTORS Interact with the ABA Receptors PYL8 and PYL9 to Orchestrate ABA Signaling in Darkness显示文摘PHYTOCHROME-INTERACTING FACTORS(PIFs)are a group of basic helix-loop-helix transcription factors that can physically interact with photoreceptors,including phytochromes and cryptochromes.It was previously demonstrated that PIFs accumulated in darkness and repressed seedling photomorphogenesis,and that PIFs linked different photosensory and hormonal pathways to control plant growth and development.In this study,we show that PIFs positively regulate the ABA signaling pathway during the seedling stage specifically in darkness.We found that PIFs positively regulate ABI5 transcript and protein levels in darkness in response to exogenous ABA treatment by binding directly to the G-box motifs in the ABI5 promoter.Consistently,PIFs and the G-box motifs in the ABI5 promoter determine ABI5 expression in darkness,and overexpression of ABI5 could rescue the ABA-insensitive phenotypes of pifq mutants in the dark.Moreover,we discovered that PIFs can physically interact with the ABA receptors PYL8 and PYL9,and that this interaction is not regulated by ABA.Further analyses showed that PYL8 and PYL9 promote PIF4 protein accumulation in the dark and enhance PIF4 binding to the ABI5 promoter,but negatively regulate PIF4-mediated ABI5 activation.Taken together,our data demonstrate that PIFs interact with ABA receptors to orchestrate ABA signaling in darkness by controlling ABI5 expression,providing new insights into the pivotal roles of PIFs as signal integrators in regulating plant growth and development.Lijuan Qi Shan Liu Cong Li Jingying Fu Yanjun Jing Jinkui Cheng Hong Li Dun Zhang Xiaoji Wang Xiaojing Dong Run Han Bosheng Li Yu Zhang Zhen Li William Terzaghi Chun-Peng Song Rongcheng Lin Zhizhong Gong Jigang Li 2020Molecular Plant2020,13,3:3

8A LexA-based yeast two-hybrid system for studying light- switchable interactions of phytochromes with their interacting partners显示文摘Phytochromes are a family of photoreceptors in plants that perceive the red(R)and far-red(FR)components of their light environment.Phytochromes exist in vivo in two forms,the inactive Pr form and the active Pfr form,that are interconvertible by treatments with R or FR light.It is believed that phytochromes transduce light signals by interacting with their signaling partners.A GAL4-based lightswitchable yeast two-hybrid(Y2H)system was developed two decades ago and has been successfully employed in many studies to determine phytochrome interactions with their signaling components.However,several pairs of interactions between phytochromes and their interactors,such as the phyACOP1 and phyA-TZP interactions,were demonstrated by other assay systems but were not detected by this GAL4 Y2H system.Here,we report a modified LexA Y2H system,in which the LexA DNA-binding domain is fused to the C-terminus of a phytochrome protein.The conformational changes of phytochromes in response to R and FR light are achieved in yeast cells by exogenously supplying phycocyanobilin(PCB)extracted from Spirulina.The well-defined interaction pairs,including phyA-FHY1 and phyB-PIFs,are well reproducible in this system.Moreover,we show that our system is successful in detecting the phyA-COP1 and phyA-TZP interactions.Together,our study provides an alternative Y2H system that is highly sensitive and reproducible for detecting light-switchable interactions of phytochromes with their interacting partners.Hong Li Xinyan Qin Pengyu Song Run Han Jigang Li 2021aBIOTECH2021,2,2:2

12SARS Patients-derived Human Recombinant Antibodies to S and M Proteins Efficiently Neutralize SARS-Coronavirus Infectivity显示文摘Objective To develop a specific SARS virus-targeted antibody preparation for emergent prophylaxis and treatment of SARS virus infection. Methods By using phage display technology, we constructed a naive antibody library from convalescent SARS patient lymphocytes. To obtain the neutralizing antibody to SARS virus surface proteins, the library panning procedure was performed on purified SARS virions and the specific Fab antibody clones were enriched by four rounds of repeated panning procedure and screened by highthroughput selection. The selected Fab antibodies expressed in the periplasma of E. Coli were soluble and further purified and tested for their binding properties and antiviral function to SARS virus. The functional Fab antibodies were converted to full human IgG antibodies with recombinant baculovirus/insect cell systems and their neutralizing activities were further determined. Results After four rounds of the panning, a number of SARS-CoV virus-targeted human recombinant Fab antibodies were isolated from the SARS patient antibody library. Most of these were identified to recognize both natural and recombinant SARS spike (S) proteins, two Fab antibodies were specific for the virus membrane (M) protein, only one bound to SARS-CoV nucleocapsid protein. The SARS-CoV S and M protein-targeted Fab or IgG antibodies showed significant neutralizing activities in cytopathic effect (CPE) inhibition neutralization test, these antibodies were able to completely neutralize the SARS virus and protect the Vero cells from CPE after virus infection. However, the N protein-targeted Fab or IgG antibodies failed to neutralize the virus. In addition, the SARS N protein-targeted human Fab antibody reacted with the denatured N proteins, whereas none of the S and M protein specific neutralizing antibodies did. These results suggested that the S and M protein-specific neutralizing antibodies could recognize conformational epitopes which might be involved in the binding of virions to cellular receptors and the fusion activity of the virus.Conclusion The SARS-CoV spike protein and membrane proteins are able to elicite efficient neutralizing antibodies in SARS patients. 14Coexistence of meningioma and other intracranial benign tumors in non-neurofibromatosis type 2 patients:A case report and review of literature显示文摘BACKGROUND The coexistence of meningioma and other intracranial primary benign tumors is rare,especially in non-neurofibromatosis type 2,and there is limited guidance for the management of such patients.Here,we report a series of 5 patients with concomitant meningioma and other intracranial benign tumors,including subependymoma and pituitary adenoma.CASE SUMMARY Five non-neurofibromatosis type 2 patients with simultaneous occurrence of meningioma and other intracranial benign tumors were retrospectively reviewed.The patients had no history of previous irradiation.The clinical features,pre-and postoperative imaging,surgical procedure and pathological findings were extracted from electronic medical records.There were 4 female patients(80%)and 1 male patient(20%).The mean age was 42.8 years(range:29-52 years).The coexisting tumors included subependymoma in 1 case(20%)and pituitary adenoma in 4 cases(80%).The most common clinical symptom was headache(3/5,60%).Four patients(80%)underwent craniotomy.One patient(20%)underwent transsphenoidal surgery followed by transcranial operation.All tumor diagnoses were confirmed by histopathological examination.The mean follow-up was 38.8 mo(range:23-96 mo),and all 5 patients were in a stable condition at the last follow-up.CONCLUSION The simultaneous occurrence of meningioma and other intracranial benign tumors is a rare clinical event.Histological examination is necessary for the accurate diagnosis.Neurosurgeons should select the appropriate surgical strategy according to the clinical features of each patient,which may provide a more favorable prognosis for individual patients.Tian-Hao Hu Run Wang Hai-Yun Wang Yi-Fu Song Juan-Han Yu Zi-Xun Wang Yu-Zhou Duan Ting Liu Sheng Han 2022World Journal of Clinical Cases2022,10,13:1

16Nanograins on Ti-25Nb-3Mo-2Sn-3Zr alloy facilitate fabricating biological surface through dual-ion implantation to concurrently modulate the osteogenic functions of mesenchymal stem cells and kill bacteria显示文摘Surface mechanical attrition treatment(SMAT)method is an effective way to generate nanograined(NG)surface on Ti-25 Nb-3 Mo-2 Sn-3 Zr(wt.%)(named as TLM),a kind ofβ-type titanium alloy,and the achieved nanocrystalline surface was proved to promote positive functions of osteoblastic cells.In this work,to further endow the NG TLM alloy with both good osteogenic and antibacterial properties,magnesium(Mg),silver(Ag)ion or both were introduced onto the NG TLM surface by ion implantation process,as a comparison,the Mg and Ag ions were also co-implanted onto coarsegrained(CG)TLM surface.The obtained results show that subsequent ion implantation does not remarkably induce the surface roughness and topography alteration of the SMAT-treated layers,and it also has little impact on the microstructure of the SMAT-derivedβ-Ti nanograins.In addition,the implanted Mg and Ag ions are observed to exist as MgO and metallic Ag na noparticles(NPs)embedding tightly in theβ-Ti matrix with grain size of about 15 and 7 nm,respectively.Initial cell adhesion and functions(including proliferation,osteo-differentiation and extracellular matrix mineralization)of rabbit bone marrow mesenchymal stem cells(rBMMSCs)and the bacterial colonization of Staphylococcus aureus(S.aureus)on the different surfaces were investigated.The in-vitro experimental results reveal that the Mg and Ag single-ion implanted NG surface either significantly promotes the rBMMSCs response or inhibits the growth ofS.aureus,whereas the Mg/Ag coimplanted NG surface could concurrently enhance the rBMMSCs functions as well as inhibit the bacterial growth compared to the NG surface,and this efficacy is more pronounced as compared to the Mg/Ag co-implantation in the CG surface.The SMAT-achieved nanograins in the TLM surface layer are identified to not only play a leading role in determining the fate of rBMMSCs but also facilitate fabricating dualfunctio nal surface with both good osteogenic and antibacterial activities through co-implantation of Mg and Ag ions.Our investigation provides a new strategy to develop high-performance Ti-based implants for clinical application.Run Huang Lei Liu Bo Li Liang Qin Lei Huang Kelvin W.K.Yeung Yong Han 2021Journal of Materials Science & Technology2021,,14:1
17Sand source and formation mechanism of riverine sand dunes: a case study in Xiangshui River, China显示文摘Riverine sand dunes develop as a result of fluvial-aeolian interactions. The primarily barchan dune chains along the Xiangshui River(a branch of the Xar Moron River in the western part of the Horqin Sandy Land of China) form a typical riverine dune field. We collected a series of samples from the riverine sand dunes parallel to the direction of the prevailing wind and investigated the sand sources and formation mechanisms of these dunes by determining the grain size, heavy mineral content and optically stimulated luminescence(OSL) of the samples. The sand of the near-river dunes was coarser than the sand of the dunes distant from the river, indicating that coarse sand of the valley mainly deposited on near-river dunes. The heavy mineral analysis suggested that wind-sand activity levels were intense on the upwind dunes, but relatively weak on the downwind dunes. This indicated that the sand sources for the near-river dunes were more abundant than those of the distant dunes. Our OSL analysis of samples suggested that the deposition rates on dunes near the river were greater than the deposition rates on dunes distant from the river. The development of dunes along the river indicated that the river played an important role in dune formation and development. In addition, airflow fluctuation and the formation of the waveform dunes had a type of feedback relationship. Grain size, heavy mineral and OSL analyses are widely used methods in wind-sand research. Sand dune grain size characteristics reflect the effects of airflow on the transport and separation of sand materials, as well as the physical characteristics of the sand sources. Heavy mineral characteristics are often used to investigate the relationships between sediments and sand sources. OSL indicates dune age, revealing formation of dunes. Therefore, it is useful to explore dune sand sources, as well as the mechanisms underlying dune formation, by determining grain size, heavy mineral content and OSL.
