2Genomic and genetic alterations influence the progression of gastric cancer显示文摘Gastric cancer is one of the leading causes of cancerrelated deaths worldwide, although the incidence has gradually decreased in many Western countries. Two main gastric cancer histotypes, intestinal and diffuse, are recognised. Although most of the described genetic alterations have been observed in both types, different genetic pathways have been hypothesized. 3State of the Art on Micromilling of Materials,a Review显示文摘The trend towards miniaturization has increased dramatically over the last decade, especially within the fields concerned with bioengineering, microelectronics, and aerospace. Micromilling is among the principal manufacturing processes which have allowed the development of components possessing micrometric dimensions, being used to the manufacture of both forming tools and the final product. The aim of this work is to present the principal aspects related to this technology, with emphasis on the work material requirements, tool materials and geometry, cutting forces and temperature, quality of the finished product, process modelling and monitoring and machine tool requirements. It can be noticed that size effect possesses a relevant role with regard to the selection of both work material (grain size) and tooling (edge radius). Low forces and temperature are recorded during micromilling, however, the specific cutting force may reach high values because of the ploughing effect observed as the uncut chip thickness is reduced. 8The Study of Carbamoyl Phosphate Synthetase 1 Deficiency Sheds Light on the Mechanism for Switching On/Off the Urea Cycle显示文摘Carbamoyl phosphate synthetase 1(CPS1) deficiency(CPS1D) is an inborn error of the urea cycle having autosomal(2q34) recessive inheritance that can cause hyperammonemia and neonatal death or mental retardation. We analyzed the effects on CPS1 activity, kinetic parameters and enzyme stability of missense mutations reported in patients with CPS1 deficiency that map in the 20-k Da C-terminal domain of the enzyme. This domain turns on or off the enzyme depending on whether the essential allosteric activator of CPS1, N-acetylL-glutamate(NAG), is bound or is not bound to it. To carry out the present studies, we exploited a novel system that allows the expression in vitro and the purification of human CPS1, thus permitting site-directed mutagenesis. These studies have clarified disease causation by individual mutations, identifying functionally important residues, and revealing that a number of mutations decrease the affinity of the enzyme for NAG. Patients with NAG affinity-decreasing mutations might benefit from NAG site saturation therapy with N-carbamyl-Lglutamate(a registered drug, the analog of NAG). Our results, together with additional present and prior site-directed mutagenesis data for other residues mapping in this domain, suggest an NAG-triggered conformational change in the b4-a4 loop of the C-terminal domain of this enzyme. This change might be an early event in the NAG activation process. 9Effect of alcoholic extract of Lepidium meyenii (Maca) on testicular function in male rats显示文摘Aim: To evaluate the effect of the alcoholic extract of Lepidium meyenii (Maca) on the spermatogenesisin male rats. Methods: In Holtzman rats, Maca alcoholic extract (5 %) was given by oral route at doses of 48 mg/day or 96 mg/day for 7 days, 14 days and 21 days. Testicular function was assessed by measurements of lengths of different stages of seminiferous epithelia and by epididymal sperm count. Results: Ethanolic extract of Maca increased the length of stages IX-XI of seminiferous epithelium at treatment day 7, day 14 and day 21. Progression of spermatogenesis was evident only after day 21 when lengths of stages XII-XIV of seminiferous epithelium were increased; at day 7 and day 14, no important change in spermatogenesis was observed. Epididymal sperm count was increased with 48 mg/day at all times. With 96 mg/day an increase in sperm count was observed at day 7, but it was reduced at day 14 and day 21 of treatment. Serum testosterone levels were not affected. Conclusion: The alcoholic extract of Maca activates onset ant progression of spermatogenesis at 48 mg/day or 96 mg/day in rats.Gustavo F. 10Rheostatic Control of ABA Signaling through HOS15-Mediated OST1 Degradation显示文摘Dehydrating stresses trigger the accumulation of abscisic acid(ABA),a key plant stress-signaling hormone that activates Snf1-Related Kinases(SnRK2s)to mount adaptive responses.However,the regulatory circuits that terminate the SnRK2s signal relay after acclimation or post-stress conditions remain to be defined.Here,we show that the desensitization of the ABA signal is achieved by the regulation of OST1(SnRK2.6)protein stability via the E3-ubiquitin ligase HOS15.Upon ABA signal,HOS15-induced degradation of OST1 is inhibited and stabilized OST1 promotes the stress response.When the ABA signal terminates,protein phosphatases ABI1/2 promote rapid degradation of OST1 via HOS15.Notably,we found that even in the presence of ABA,OST1 levels are also depleted within hours of ABA signal onset.The unexpected dynamics of OST1 abundance are then resolved by systematic mathematical modeling,demonstrating a desensitizing feedback loop by which OST1-induced upregulation of ABI1/2 leads to the degradation of OST1.This model illustrates the complex rheostat dynamics underlying the ABA-induced stress response and desensitization.Akhtar Ali Jae Kyoung Kim Masood Jan Haris Ali Khan Irfan Ullah Khan Mingzhe Shen Junghoon Park Chae Jin Lim Shah Hussain Dongwon Baek Kai Wang Woo Sik Chung Vicente Rubio Sang Yeol Lee Zhizhong Gong Woe Yeon Kim Ray ABressan Jose MPardo Dae-Jin Yun 2019Molecular Plant2019,12,11:5
11Fecal lactoferrin accurately reflects mucosal inflammation in inflammatory bowel disease显示文摘BACKGROUND Studies have demonstrated a potential role for fecal biomarkers such as fecal calprotectin(FC)and fecal lactoferrin(FL)in monitoring inflammatory bowel diseases(IBD)-Crohn's disease(CD)and ulcerative colitis(UC).However,their correlation to endoscopic scores,disease severity and affected intestinal surface has not been extensively investigated.AIM To correlate FL,and for comparison white blood cell(WBC)and C-reactive protein(CRP),with endoscopic scores,disease extent and location in CD and UC.METHODS Retrospective analysis in 188 patients who had FL,CRP and WBC determined within 30 d of endoscopy.Disease location,disease extent(number of intestinal segments involved),disease severity(determined by endoscopic scores),timing of FL testing in relation to colonoscopy,as well as the use of effective fast acting medications(steroids and biologics)between colonoscopy and FL measurement,were recorded.RESULTS In 131 CD and 57 UC patients,both CRP and FL-but not WBC-distinguished disease severity(inactive,mild,moderate,severe).In patients receiving fastacting(steroids or biologics)treatment in between FL and colonoscopy,FL showed a higher correlation to endoscopic scores when tested before vs after the procedure(r=0.596,P<0.001,vs r=0.285,P=0.15 for the Simple Endoscopic Score for CD;and r=0.402,P=0.01 vs r=0.054 P=0.84 for Disease Activity Index).Finally,FL was significantly correlated with the diseased mucosal surface(colon-ileocolon>small bowel)and the number of inflamed colon segments.CONCLUSION FL and CRP separated disease severity categories with FL showing lower discriminating P-values.FL showed a close correlation with the involved mucosal surface and with disease extent and was more closely correlated to endoscopy when determined before the procedure–this indicating that inflammatory activity changes associated with therapy might be rapidly reflected by FL levels.FL can accurately and timely characterize intestinal inflammation in IBD.Marrieth G Rubio Kofi Amo-Mensah James M Gray Vu Q Nguyen Sam Nakat Douglas Grider Kim Love James H Boone Dario Sorrentino 2019World Journal of Gastrointestinal Pathophysiology2019,10,5:4

15Knockdown of OLA1,a regulator of oxidative stress response,inhibits motility and invasion of breast cancer cells显示文摘To explore the role of a novel Obg-like ATPase 1 (OLA1) in cancer metastasis,small interference RNA (siRNA) was used to knockdown the protein,and the cells were subjected to in vitro cell migration and invasion assays. Knockdown of OLA1 significantly inhibited cell migration and invasion in breast cancer cell line MDA-MB-231. The knockdown caused no changes in cell growth but affected ROS production. In wound-healing assays,decreased ROS in OLA1-knockdown cells were in situ associated with the cells’ decreased motile morphology. Further,treatment of N-acetylcysteine,a general ROS scavenger,blunted the motility and invasiveness of MDA-MB-231 cells,similar to the effect of OLA1-knockdown.
