| 1 | Ron receptor-dependent gene regulation of Kupffer cells during endotoxemia显示文摘BACKGROUND: Ron receptor tyrosine kinase signaling in macrophages, including Kupffer cells and alveolar macrophages,suppresses endotoxin-induced proinflammatory cytokine/chemokine production. Further, we have also identified genes from Ron replete and Ron deplete livers that were differentially expressed during the progression of liver inflammation associated with acute liver failure in mice by microarray analyses.While important genes and signaling pathways have been identified downstream of Ron signaling during progression of inflammation by this approach, the precise role that Ron receptor plays in regulating the transcriptional landscape in macrophages, and particular in isolated Kupffer cells, has still not been investigated.METHODS: Kupffer cells were isolated from wild-type(TK+/+)and Ron tyrosine kinase deficient(TK-/-) mice. Ex vivo, the cells were treated with lipopolysaccharide(LPS) in the presence or absence of the Ron ligand, hepatocyte growth factor-like protein(HGFL). Microarray and qRT-PCR analyses were utilized to identify alterations in gene expression between genotypes.RESULTS: Microarray analyses identified genes expressed differentially in TK+/+ and TK-/- Kupffer cells basally as well as after HGFL and LPS treatment. Interestingly, our studies identified Mefv, a gene that codes for the anti-inflammatory protein pyrin, as an HGFL-stimulated Ron-dependent gene.Moreover, lipocalin 2, a proinflammatory gene, which is induced by LPS, was significantly suppressed by HGFL treatment.Microarray results were validated by qRT-PCR studies on Kupffer cells treated with LPS and HGFL.CONCLUSION: The studies herein suggest a novel mechanism whereby HGFL-induced Ron receptor activation promotes the expression of anti-inflammatory genes while inhibiting genes involved in inflammation with a net effect of diminished inflammation in macrophages. | Rishikesh M Kulkarni William D Stuart Susan E Waltz | 2014 | Hepatobiliary & Pancreatic Diseases International2014,13,3: | 6 |
| 2 | Soil Carbon Dynamics Under Changing Climate A Research Transition from Absolute to Relative Roles of Inorganic Nitrogen Pools and Associated Microbial Processes:A Review显示文摘It is globally accepted that soil carbon(C) dynamics are at the core of interlinked environmental problems, deteriorating soil quality and changing climate. Its management remains a complex enigma for the scientific community due to its intricate relationship with soil nitrogen(N) availability and moisture-temperature interactions. This article reviews the management aspects of soil C dynamics in light of recent advances, particularly in relation to the availability of inorganic N pools and associated microbial processes under changing climate. Globally, drastic alterations in soil C dynamics under changing land use and management practices have been primarily attributed to the variation in soil N availability, resulting in a higher decomposition rate and a considerable decline in soil organic C(SOC) levels due to increased soil CO_2 emissions, degraded soil quality, and increased atmospheric CO_2 concentrations,leading to climate warming. Predicted climate warming is proposed to enhance SOC decomposition, which may further increase soil N availability, leading to higher soil CO_2 efflux. However, a literature survey revealed that soil may also act as a potential C sink, if we could manage soil inorganic N pools and link microbial processes properly. Studies also indicated that the relative, rather than the absolute, availability of inorganic N pools might be of key importance under changing climate, as these N pools are variably affected by moisture-temperature interactions, and they have variable impacts on SOC turnover. | 3 | Impact of sole and combined application of biochar,organic and chemical fertilizers on wheat crop yield and water productivity in a dry tropical agro-ecosystem显示文摘Agriculture under changing climate scenario is facing major challenges of water scarcity and resource imbalances.Crop water productivity(WP)may act as an indicator of crop responses to water limitation.Organic amendments such as biochar and manure application to soil are suggested for improving soil quality and reducing water requirements from agricultural sector.However,studies exploring the impact of biochar as sole or in combination with organic and/or chemical fertilizers on WP in dry tropical agro-ecosystems are limited.In this study,we observed the effect of rice-husk ash(RHA,biochar)along with farm-yard manure(FYM)and chemical fertilizers(CF)under varying water conditions on soil hydro-physical properties,yield and WP of wheat crop.Water-filled pore space(WFPS),grain and straw yield,irrigation and total water productivity varied significantly(at P<0.001)at treatment level.Grain and straw yield were found higher under sole and combined CF applied treatments.Sole and combined RHA and FYM amendment improved water holding capacity(WHC)and WFPS,whereas a decrease in crop yield was observed as compared to the control.Irrigation and total water productivity were found higher under combined RHA+FYM and sole CF treatments with reduced water supply(except sole CF)as compared to control and sole RHA treatments with full water irrigation.Crop water productivity was found positively correlated with grain and straw yields,however,significant correlations were not observed with WHC and WFPS.Results indicate that increasing soil hydro-physical properties in silty-loam soil may hinder crop yield and WP under sole biochar applied soils.Overall,the implications of the study would help in devising agro-management practices based on combined application of RHA and FYM with reduced chemical fertilizer and water inputs to mitigate the impacts of climate change without compromising crop yield in the highly vulnerable dry tropical agro-ecosystem of India.Moreover,long-term studies are needed in these ecosystems to identify the appropriate agricultural package for mitigating the forthcoming water scarcity conditions. | Rishikesh Singh Pardeep Singh Hema Singh A.S.Raghubanshi | 2019 | Biochar2019,1,2: | 5 |
| 4 | Ron receptor-dependent gene regulation in a mouse model of endotoxin-induced acute liver failure显示文摘BACKGROUND:Prior experimentation has shown that loss of the tyrosine kinase(TK) signaling domain of the Ron receptor leads to marked hepatocyte protection in a model of lipopolysaccharideinduced acute liver failure(ALF) in D-galactosamine(GalN)sensitized mice.The aim of this study was to identify the role of Ron in the regulation of hepatic gene expression.METHODS:Microarray analyses were performed on liver RNA isolated sequentially from wild-type(WT) and TK-/mice during the progression of ALF.Gene array data were validated using Western and immunohistochemistry analyses as well as with ex vivo culture systems.RESULTS:At baseline,101 genes were differentially expressed between WT and TK-/-livers,which regulate processes involved in hypoxia,proliferation,apoptosis and metabolism.One hour after ALF induction,WT livers exhibited increased cytokine expression compared to TK-/-livers,and after 4 hours,an induction of suppressor of cytokine signaling(SOCS) genes as well as JAK-STAT pathway activation were prominent in TK-/livers compared to controls.CONCLUSION:Our studies suggest a novel hepato-protective mechanism in Ron TK-/-mice wherein increased and sustained SOCS production and JAK-STAT activation in the hepatocyte may inhibit the destructive proinflammatory milieu and promote survival factors which blunt hepatic death and the ensuing development of ALF. | Rishikesh M Kulkarni Louis W Kutcher William D Stuart Daniel J Carson Mike A Leonis Susan E Waltz | 2012 | Hepatobiliary & Pancreatic Diseases International2012,11,4: | 3 |
| 5 | Formulation and evaluation of novel stomach specific floating microspheres bearing famotidine for treatment of gastric ulcer and their radiographic study显示文摘Objective:To develop and characterize multiple-unit-type oral floating microsphere of famotidine to prolong gastric residence time and to target stomach ulcer.Methods:The floating microspheres were prepared by modified solvent evaporation method,Eudragil S-100 was used as polymer.Microspheres were characterized for the micromeritic properties,floating behavior,entrapment efficiency and scanning electron microscopy.The invitro release studies and floating behavior were studied in simulated gastric fluid at pH 1.2.Different drug release kinetics models were also applied for all the batches.Selected formulations were also subjected for X-ray radiographic study.Results:Floating microspheres were successfully prepared by modified solvent evaporation technique.Microspheres showed passable flow properties.The maximum yield of microspheres was up to(95.11±0.35)%.On the basis of optical microscopy particle size range was found to be ranging from(52.18±182.00) to(91.64±5.16) μm.Scanning electron microscopy showed their spherical size,perforated smooth surface and a cavity inside microspheres.Microspheres were capable to float up to 20 h in simulated gastric fluid.X-ray radiographic studies also proved its better retention in the stomach.Conclusions:On the basis of the results,such dosage forms may be a good candidate for stomach targeting and may be dispensed in hard gelatin capsules. | Rishikesh Gupta Sunil Kumar Prajapati Snigdha Pattnaik Peeyush Bhardwaj | 2014 | Asian Pacific Journal of Tropical Biomedicine2014,4,9: | 2 |
| 6 | Crystal twins:self-supervised learning for crystalline material property prediction显示文摘Machine learning(ML)models have been widely successful in the prediction of material properties.However,large labeled datasets required for training accurate ML models are elusive and computationally expensive to generate.Recent advances in Self-Supervised Learning(SSL)frameworks capable of training ML models on unlabeled data mitigate this problem and demonstrate superior performance in computer vision and natural language processing.Drawing inspiration from the developments in SSL,we introduce Crystal Twins(CT):a generic SSL method for crystalline materials property prediction that can leverage large unlabeled datasets.CT adapts a twin Graph Neural Network(GNN)and learns representations by forcing graph latent embeddings of augmented instances obtained from the same crystalline system to be similar.We implement Barlow Twins and SimSiam frameworks in CT.By sharing the pre-trained weights when fine-tuning the GNN for downstream tasks,we significantly improve the performance of GNN on 14 challenging material property prediction benchmarks. | Rishikesh Magar Yuyang Wang Amir Barati Farimani | 2022 | npj Computational Materials2022,,1: | 1 |

| 20 | Lensless polarimetric coded ptychography for high-resolution,high-throughput gigapixel birefringence imaging on a chip显示文摘Polarimetric imaging provides valuable insights into the polarization state of light interacting with a sample.It can infer crucial birefringence properties of specimens without using labels,thereby facilitating the diagnosis of diseases such as cancer and osteoarthritis.In this study,we present a novel polarimetric coded ptychography(pol-CP)approach that enables high-resolution,high-throughput gigapixel birefringence imaging on a chip.Our platform deviates from traditional lens-based systems by employing an integrated polarimetric coded sensor for lensless coherent diffraction imaging.Utilizing Jones calculus,we quantitatively determine the birefringence retardance and orientation information of biospecimens from the recovered images.Our portable pol-CP prototype can resolve the 435 nm linewidth on the resolution target,and the imaging field of view for a single acquisition is limited only by the detector size of 41 mm2.The prototype allows for the acquisition of gigapixel birefringence images with a 180 mm^(2) field of view in~3.5 min,a performance that rivals high-end whole slide scanner but at a small fraction of the cost.To demonstrate its biomedical applications,we perform high-throughput imaging of malaria-infected blood smears,locating parasites using birefringence contrast.We also generate birefringence maps of label-free thyroid smears to identify thyroid follicles.Notably,the recovered birefringence maps emphasize the same regions as autofluorescence images,underscoring the potential for rapid on-site evaluation of label-free biopsies.Our approach provides a turnkey and portable solution for lensless polarimetric analysis on a chip,with promising applications in disease diagnosis,crystal screening,and label-free chemical imaging,particularly in resource-constrained environments. | LIMING YANG RUIHAI WANG QIANHAO ZHAO PENGMING SONG SHAOWEI JIANG TIANBO WANG XIAOPENG SHAO CHENGFEI GUO RISHIKESH PANDEY GUOAN ZHENG | 2023 | Photonics Research2023,11,12: | 0 |
