1An all-silk-derived functional nanosphere matrix for sequential biomolecule delivery and in situ osteochondral regeneration显示文摘Endogenous repair of osteochondral defect is usually limited by the insufficient number of cells in the early stage and incomplete cell differentiation in the later stage.The development of drug delivery systems for sequential release of pro-migratory and pro-chondrogenic molecules to induce endogenous bone marrow-derived mesenchymal stem cells(BMSCs)recruitment and chondrogenic differentiation is highly desirable for in situ osteochondral regeneration.In this study,a novel,all-silk-derived sequential delivery system was fabricated by incorporating the tunable drug-loaded silk fibroin(SF)nanospheres into a SF porous matrix.The loading efficiency and release kinetics of biomolecules depended on the initial SF/polyvinyl alcohol(PVA)concentrations(0.2%,1%and 5%)of the nanospheres,as well as the hydrophobicity of the loaded molecules,resulting in controllable and programmed delivery profiles.Our findings indicated that the 5%nanosphere-incorporated matrix showed a rapid release of E7 peptide during the first 120 h,whereas the 0.2%nanosphere-incorporated matrix provided a slow and sustained release of Kartogenin(KGN)longer than 30 days.During in vitro culture of BMSCs,this functional SF matrix incorporated with E7/KGN nanospheres showed good biocompatibility,as well as enhanced BMSCs migration and chondrogenic differentiation through the release of E7 and KGN.Furthermore,when implanted into rabbit osteochondral defect,the SF nanosphere matrix with sequential E7/KGN release promoted the regeneration of both cartilage and subchondral bone.This work not only provided a novel all-silk-derived drug delivery system for sequential release of molecules,but also a functional tissueengineered scaffold for osteochondral regeneration.Wei Zhang Chen Ling Aini Zhang Haoyang Liu Yujie Jiang ,Xiaolong Li Renwang Sheng Qingqiang Yao Jialin Chen 2020Bioactive Materials2020,5,4:5
2Synthesis of the superfine high-entropy zirconate nanopowders by polymerized complex method显示文摘The high-purity and superfine high-entropy zirconate nanopowders,namely(Y_(0.25)La_(0.25)Sm_(0.25)Eu_(0.25))_(2)Zr_(2)O_(7)nanopowders,without agglomeration,were successfully synthesized via polymerized complex method at low temperatures for the first time.The results showed that the crystallinity degree,lattice strain,and particle size of the as-synthesized powders were gradually enhanced with the increase of the synthesis temperature from 800 to 1300℃.The as-synthesized powders involved fluorite phase in the range of 800-1200℃while they underwent the phase evolution from fluorite to pyrochlore at 1300℃.It is worth mentioning that the as-synthesized powders at 900℃are of the highest quality among all the as-synthesized powders,which is due to the fact that they not only possess the particle size of 11 nm without agglomeration,but also show high purity and good compositional uniformity.Yangjie HAN Renwang YU Honghua LIU Yanhui CHU 2022Journal of Advanced Ceramics2022,11,1:2

18Crystal structures, absolute configurations and molecular docking studies of naftopidil enantiomers as α1D-adrenoceptor antagonists显示文摘Chiral drug naftopidil(NAF), a specific α1D-adrenoceptor(AR) antagonist for the treatment of benign prostatic hyperplasia, was used in racemic form for several decades. Our recent work declared that NAF enantiomers showed the same antagonistic effects on the α1D-AR, but the binding mechanism of these two stereochemical NAF isomers to the α1D receptor remained unclear. Herein, we reported the crystallographic structures of optically pure NAF stereoisomers for the first time and unambiguously determined their absolute configurations. The crystal data of R and S enantiomers matched satisfactorily the pharmacophore model for α1D-selective antagonists. Based on the constructed α1D homology model,molecular docking studies shed light on the molecular mechanism of NAF enantiomers binding to α1D-AR. 20Chemical composition-based characterization of the anti-allergic effect of Guominkang Formula on IgE-mediated mast cells activation and passive cutaneous anaphylaxis显示文摘Guominkang(GMK),a Chinese medicine formula,has been used to treat allergic diseases in clinical settings for many years.To evaluate the antiallergic effect and molecular mechanism of action of GMK extract,RBL-2H3 cell models and passive cutaneous anaphylaxis(PCA)mouse models were established.High performance liquid chromatography(HPLC)and ultra-high performance liquid chromatography-mass spectrometry(UHPLC-MS)analyses were performed to characterize the chemical composition of GMK.A total of 94 compounds were identified or tentatively identified from GMK.Three of them,emodin,ursolic acid,and hamaudol,were identified for the first time as potential active compounds in GMK,since they inhibited the degranulation of mast cells.The anti-allergic effect of hamaudol was the first to be discovered.GMK could markedly mitigate the shade of Evans Blue extravasation and ear incrassation in PCA mouse models.Additionally,GMK significantly inhibited the degranulation of mast cells,suppressed mast cell degranulation by reducing Ca2+influx and the levels of TNF-α,IL-4,and histamine,and markedly inhibited the phosphorylation of Lyn,Syk,PLCγ1,IκBα,and NF-κB p65.Molecular docking results indicated that hamaudol and emodin had strong interaction with FcεRI and NF-κB related proteins,while ursolic acid only interacted with NF-κB associated proteins.These results suggest GMK suppresses the activation of MCs both in vivo and in vitro.The underlying mechanism of its anti-allergic activity is associated with the inhibition of FcεRI and NF-κB activation.TANG Ding WANG Chen GAN Qianying WANG Zhixin JIANG Renwang 2022Chinese Journal of Natural Medicines2022,20,12:0
