| 2 | The fabrication of biomimetic biphasic CAN-PAC hydrogel with a seamless interfacial layer applied in osteochondral defect repair显示文摘Cartilage tissue engineering based on biomimetic scaffolds has become a rapidly developing strategy for repairing cartilage defects. In this study, a biphasic CAN-PAC hydrogel for osteochondral defect(OCD)regeneration was fabricated based on the density difference between the two layers via a thermally reactive,rapid cross-linking method. The upper hydrogel was cross-linked by CSMA and NIPAm, and the lower hydrogel was composed of PECDA, AAm and PEGDA. The interface between the two layers was first grafted by the physical cross-linking of calcium gluconate and alginate, followed by the chemical cross-linking of the carbon-carbon double bonds in the other components. The pore sizes of the upper and lower hydrogels were ~ 187.4 and ~ 112.6 μm, respectively. The moduli of the upper and lower hydrogels were ~ 0.065 and~ 0.261 MPa. This prepared bilayer hydrogel exhibited the characteristics of mimetic composition, mimetic structure and mimetic stiffness, which provided a microenvironment for sustaining cell attachment and viability. Meanwhile, the biodegradability and biocompatibility of the CAN-PAC hydrogel were examined in vivo. Furthermore, an osteochondral defect model was developed in rabbits, and the bilayer hydrogels were implanted into the defect. | 3 | FHY3 and FAR1 Integrate Light Signals with the miR156-SPL Module-Mediated Aging Pathway to Regulate Arabidopsis Flowering显示文摘In response to competition for light from their neighbors,shade-intolerant plants flower precociously to ensure reproductive success and survival.However,the molecular mechanisms underlying this key developmental switch are not well understood.Here,we show that a pair of Arabidopsis transcription factors essential for phytochrome A signaling,FAR-RED ELONGATED HYPOCOTYL3(FHY3)and FAR-RED IMPAIRED RESPONSE1(FAR1),regulate flowering time by integrating environmental light signals with the miR156-SPL module-mediated aging pathway.We found that FHY3 and FAR1 directly interact with three flowering-promoting SQUAMOSA-PROMOTER BINDING PROTEIN-LIKE(SPL)transcription factors,SPL3,SPL4,and SPL5,and inhibit their binding to the promoters of several key flowering regulatory genes,including FRUITFUL(FUL),LEAFY(LFY),APETALA1(AP1),and MIR172C,thus downregulating their transcript levels and delaying flowering.Under simulated shade conditions,levels of SPL3/4/5 proteins increase,whereas levels of FHY3 and FAR1 proteins decline,thus releasing SPL3/4/5 from FHY3/FAR1 inhibition to allow activation of FUL,LFY,AP1,and MIR172C and,consequently,early flowering.Taken together,these results unravel a novel mechanism whereby plants regulate flowering time by integrating environmental cues(such as light conditions)and an internal developmental program(the miR156-SPL module-mediated aging pathway). | Yurong Xie Qin Zhou Yongping Zhao Quanquan Li Yang Liu Mengdi Ma Baobao Wang Rongxin Shen Zhigang Zheng Haiyang Wang | 2020 | Molecular Plant2020,13,3: | 8 |
| 4 | Longitudinal Alterations of Local Spontaneous Brain Activity in Parkinson's Disease显示文摘We used resting-state fMRI to evaluate longitudinal alterations in local spontaneous brain activity in Parkinson's disease(PD) over a 2-year period. Data were acquired from 23 PD patients at baseline and follow-up,and 27 age-and sex-matched normal controls. Regional homogeneity(ReHo) and voxel-based-morphometry(VBM) were used to identify differences in local spontaneous brain activity and grey matter volume. With disease progression, we observed a progressive decrease in ReHo in the sensorimotor cortex, default-mode network, and left cerebellum, but increased ReHo in the supplementary motor area, bilateral temporal gyrus, and hippocampus.Moreover, there was a significant positive correlation between the rates of ReHo change in the left cerebellum and the rates of change in the Unified Parkinson's Disease Rating Scale-III scores. VBM revealed no significant differences in the grey matter volume among the three sets of acquisitions. | 6 | Experimental and mechanistic understanding of photo-oxidation of methanol catalyzed by CuO/TiO2-spindle nanocomposite:Oxygen vacancy engineering显示文摘We report experimental and mechanistic understanding of methanol oxidation to produce methyl formate using CuO/Ti02-spindle composite as a promising photocatalyst under mild conditions with over 97%conversion and 83%selectivity.The catalysts are obtained via precise depositing of CuO nanoclusters(size:~3.5 nm)at the{101}facet of the TiO2 to optimally tune exciton recombination through oxygen vacancies generation,evidenced by photoluminescence and Raman spectroscopy measurements.The turnover frequency(TOF)and the apparent quantum efficiency(AQE)of the 7%CuO/TiO2-spindle composites reach up to 23.8 molmethanol·gcat^-1·h^-1 and 55.2%at 25℃,respectively,which are substantially higher than these previously reported photocatalysts.Further,the in-situ attenuated total reflection infrared spectroscopy analysis reveals that the methanol oxidation most likely takes place through the conversion of adsorbed methoxy(CH30^*)to formaldehyde(CHO^*)intermediate,a subject of major debate for a long time.The adsorbed formaldehyde(CHO^*)thus produced reacts with another CH30^*species in its close proximity to form the final product of methyl formate.Results of this study provide insights into the reaction mechanism,and offer guidelines to systematically develop and apply photocatalysts for methanol conversion and related reactions via surface engineering. | Quanquan Shi Zhaoxian Qin Changlin Yu Ammara Waheed Hui Xu Yong Gao Hadi Abroshan Gao Li | 2020 | Nano Research2020,13,4: | 3 |

| 8 | Stress concentration in a finite functionally graded material plate显示文摘This paper is to study the two-dimensional stress distribution of a finite functionally graded material (FGM) plate with a circular hole under arbitrary constant loads. Using the method of piece-wise homogeneous layers, the stress analysis of the finite FGM plate having radial arbitrary elastic properties is made based on the complex variable method combined with the least square boundary collocation technique. Numerical results of stress distribution around the hole are then presented for different loading conditions, different material properties and different plate sizes, respectively. | 9 | Novel derivative of Danshensu acts as anti-thrombotic agent for modulation of protein disulfide isomerase family member ERp57显示文摘OBJECTIVE To identify the specific targets of a novel derivative of Danshensu ADTM as the protein disulfide isomerase(PDI)family proteins including ERp57.To further investigate the underlying mechanism of ADTM to modulate ERp57 to regulate platelet function with direct interaction withαⅡbβ3 integrin.METHODS To isolate the protein targets that bound to ADTM,a biotin-conjugated ADTM analogue(BAA)was designed and synthesized.BAA(300μmol·L-1)was incubated with rat blood platelet lysates and the BAA-protein complexes were pulled down with NeutrAvidin-agarose followed by protein profiling using LC-MS/MS.To determine platelet aggregation in vitro,rabbit platelets were incubated with the indicated concentrations of compounds and aggregation was induced by ADP(10μmol·L-1)or AA(200μmol·L-1)and measured using a platelet aggregometer.To determine platelet aggregation-induced by ADP in rat in vivo,ADTM(5-20mg·kg-1)in comparison with DSS(10mg·kg-1)and clopidogrel(18mg·kg-1)were administered daily by i.v.injection for 5d,respectively.To determine the action of ADTM on the ERp57/αⅡbβ3 interaction,it was examined by immunoprecipitation with anti-αⅡbβ3antibody,followed by detection of ERp57 immunoreactivity using immunoblotting.RESULTS BAA could bind to various proteins involved in platelet function.In particular,platelet aggregation-associated proteins were identified with>95% protein identification probability including ERp72,ERp57ERp5 and PDI,which are members of the protein disulfide isomerase(PDI)family related to platelet function and redox homeostasis.ADTM exhibited potent inhibition on the redox activity of ERp57 in a concentration-dependent manner(IC50=100 300μmol·L-1).In in vitro studies,ADTM exhibited concentration-dependent inhibition on ADP-induced and AA-induced platelet aggregation with comparable effects to aspirin and clopidogrel.In vivo study showed that ADP-induced platelet aggregation was significantly compromised(>40%reduction)in rats treated with ADTM(20mg·kg-1).Similarly,ADTM also exhibited significant anti-thrombotic effect in vivo as shown in the ferric chloride(FeCl3)-induced venous thrombosis.Immunoprecipitation with anti-αⅡbβ3antibody,followed by detection of ERp57 immunoreactivity using immunoblotting showed that ADTM disrupted the interaction of ERp57 with αⅡ bβ3.CONCLUSION These results demonstrated that ADTM exhibited broad-spectrum anti-platelet activities and ERp57 is a potential therapeutic target for anti-platelet therapy. | Guo-zhenCUI Lu-chenSHAN LinGUO IvanKeungCHU Guo-huiLI QuanQUAN YunZHAO Chenong-mengCHONG ZaijunZHANG PeiYU Pui-manHOI Ye-weiSUN Yu-qiangWANG Simonming-yuenLEE | 2015 | 中国药理学与毒理学杂志2015,29,S1: | 3 |
| 10 | Serum ficolin-2 concentrations are significantly changed in patients with hepatitis B virus infection and liver diseases显示文摘Human ficolin-2 is an important lectin complement pathway activator that is secreted from liver cells and has been implicated as an anti-infection innate immune molecule. However, the role of ficolin-2 protein and its dynamic changes over the course of and in the prognosis of chronic hepatitis B(CHB) and hepatocellular carcinoma(HCC) remain unclear. In this study, we analyzed ficolin-2 protein expression in a cohort of individuals with CHB infection, HCC and cirrhosis. A sandwich enzyme-linked immunosorbent assay(ELISA) method was used to measure serum ficolin-2 concentrations. Ficolin-2 expression in liver tissues was detected by immunohistochemical staining. Serum ficolin-2 concentrations in CHB patients were significantly higher than in healthy controls and HBV carriers. After 48 weeks of routine amelioration liver function treatment, serum ficolin-2 concentrations decreased and were positively correlated with favorable alanine aminotransferase(ALT), HBV DNA and HBe Ag-seroconversion outcomes. Interestingly, we observed much lower expression of serum and intrahepatic ficolin-2 in HCC and cirrhosis compared with healthy controls. | 11 | Linking key intervention timings to rapid declining effective reproduction number to quantify lessons against COVID-19显示文摘Coronavirus disease 2019(COVID-19)is currently under a global pandemic trend.The efficiency of containment measures and epidemic tendency of typical countries should be assessed.In this study,the efficiency of prevention and control measures in China,Italy,Iran,South Korea,and Japan was assessed,and the COVID-19 epidemic tendency among these countries was compared.Results showed that the effective reproduction number(Re)in Wuhan,China increased almost exponentially,reaching a maximum of 3.98 before a lockdown and rapidly decreased to below 1 due to containment and mitigation strategies of the Chinese government.The Re in Italy declined at a slower pace than that in China after the implementation of prevention and control measures.The Re in Iran showed a certain decline after the establishment of a national epidemic control command,and an evident stationary phase occurred because the best window period for the prevention and control of the epidemic was missed.The epidemic in Japan and South Korea reoccurred several times with the Re fluctuating greatly.The epidemic has hardly rebounded in China due to the implementation of prevention and control strategies and the effective enforcement of policies.Other countries suffering from the epidemic could learn from the Chinese experience in containing COVID-19. | Zhihang Peng Wenyu Song Zhongxing Ding Quanquan Guan Xu Yang Qiaoqiao Xu Xu Wang Yankai Xia | 2020 | Frontiers of Medicine2020,14,5: | 2 |

| 15 | A measuring method of effects of herbs based on classification used for intelligent calculation of efficacies of prescriptions显示文摘It is important to translate dosage of each medical herb in a prescription into intensity of its effects measured by mathematical models. In this paper, we present intelligent calculational algorithms on efficacies of prescriptions of Chinese Medicine (CM), study 'how curative effects of any herb depend on dosage' when constructing CM intelligent systems, and discuss relevant issues and definitions. In order to measure intensities of effects of all herbs reasonably, the natural classification method is proposed to classify herbs into 38 natural classes according to their dosage ranges using the knowledge of CM dosage and distribution rules of herbs. Then the 38 natural classes are clustered to 6 basic classes reflecting characteristic attributes of various herbs. Based on the results of classified herbs, a set of nonlinear measuring functions of intensities of effects is designed for various basic classes. These functions have been embedded in our intelligent calculation system of efficacies of prescriptions. The effects of 560 herbs and curative efficacies of more than 1000 prescriptions derived from 'ShangHan Lun', 'JinGui YaoLue', 'Ming Jia Yi An', etc., have been calculated and discovered successfully.
