1Study on real working performance and overload safety factor of high arch dam显示文摘Considering the fact that high arch dams have problems such as complicated stress,high cost,and hazards after being damaged,this paper intends to study the effects of load,material strength,and safety analysis method on dam safety and working performance of arch dams.In this article,the effects of temperature,self weight exaction way and water loading on structure response are first discussed,and a more reasonable way of considering is then put forward.By taking into consideration the mechanical property of materials and comparing the effects of different yield criteria on overloading safety of high arch dams,this paper concludes that brittle characteristics of concrete should be fully considered when conducting safety assessment for high arch dams to avoid overestimating the bearing capacity of the dams.By comparing several typical projects,this paper works out a safety assessment system of multiple safety and relevant engineering analogical analysis methods,which is closer to the actual situation,and thus is able to assess the response of high arch dam structure in a more comprehensive way,elicit the safety coefficients in different situations,and provide a new way of considering the safety assessment of high arch dams.ZHANG GuoXin,LIU Yi & ZHOU QiuJing Department of Structures and Materials,China Institute of Water Resources and Hydropower Research,Beijing 100038,China 2008Science China(Technological Sciences)2008,51,S2:25
2Reproductive management through integration of PGD and MPS-based noninvasive prenatal screening/diagnosis for a family with GJB2-associated hearing impairment显示文摘A couple with a proband child of GJB2(encoding the gap junction protein connexin 26)-associated hearing impairment and a previous pregnancy miscarriage sought for a reproductive solution to bear a healthy child. Our study aimed to develop a customized preconception-to-neonate care trajectory to fulfill this clinical demand by integrating preimplantation genetic diagnosis(PGD), noninvasive prenatal testing(NIPT), and noninvasive prenatal diagnosis(NIPD) into the strategy. Auditory and genetic diagnosis of the proband child was carried out to identify the disease causative mutations. The couple then received in-vitro-fertilization treatment, and eight embryos were obtained for day 5 biopsy. PGD was performed by short-tandem-repeat linkage analysis and Sanger sequencing of GJB2 gene. Transfer of a GJB2 c.235del C heterozygous embryo resulted in a singleton pregnancy. At the 13 th week of gestation, genomic DNA(g DNA) from the trio family and cell-free DNA(cf DNA) from maternal plasma were obtained for assessment of fetal chromosomal aneuploidy and GJB2 mutations. NIPT and NIPD showed the absence of chromosomal aneuploidy and GJB2-associated disease in the fetus, which was later confirmed by invasive procedures and postnatal genetic/auditory diagnosis. 3DNA vaccine encoding Der p 2 allergen generates immunologic protection in recombinant Der p 2 allergen-induced allergic airway inflammation mice model显示文摘Background DNA immunization is a promising novel type of immunotherapy against allergy. An estimated 79.2% patients with asthma, wheezing and/or rhinitis suffer from Dermatophagoides pteronyssinus group 2 (Der p 2) allegen. The aim of the present study was to determine whether DNA vaccine encoding Der p 2 could generate immunologic protection in recombinant Der p 2 (rDer p 2) allergen-induced allergic airway inflammation mice model and to understand the role of DNA vaccination in specific-allergen immunotherapy for asthma. Methods After DNA vaccination, BALB/c mice were sensitized by intraperitoneal injection (i.p) and challenged by intranasal instillation of rDer p 2. The lung tissues were assessed using hematoxylin and eosin. Mucus-producing goblet cells were identifed using periodic acid-Schiff(PAS)/alcian blue. The total cell number and composition of bronchoalveolar lavage samples were determined. The levels of the cytokines IL-4 and IFN-γ, as well as IgE and IgG2ain the serum were determined by enzyme-linked immunosorbent assay. Allergen-specific IL-4 and IFN-γ production by spleen cells were also measured by enzyme-linked immunosorbent assay. Expression of signal transducer and activator of transcription 6 (STAT6) in splenocytes were determined by Western blot. Results DNA vaccine encoding Der p 2 allergen inhibited extensive infiltration of inflammatory cells and production of mucin induced by allergen. The influx of eosinophils into the lung interstitium was significantly reduced after administration of DNA vaccine. Significant reductions of IL-4 and increase in levels of IFN-γ in bronchoalveolar lavage fluid were observed. The allergen-specific IgE was markedly decreased in mice receiving DNA vaccination. Allergen could induce higher IFN-γ, weaker IL-4 in cultured spleen cells from mice receiving DNA vaccine. DNA vaccination inhibited STAT6 expression of spleen cells induced by allergen. 4Serine metabolism orchestrates macrophage polarization by regulating the IGF1–p38 axis显示文摘Serine metabolism is reportedly involved in immune cell functions, but whether and how serine metabolism regulates macrophage polarization remain largely unknown. Here, we show that suppressing serine metabolism, either by inhibiting the activity of the key enzyme phosphoglycerate dehydrogenase in the serine biosynthesis pathway or by exogenous serine and glycine restriction, robustly enhances the polarization of interferon-γ-activated macrophages (M(IFN-γ)) but suppresses that of interleukin-4-activated macrophages (M(IL-4)) both in vitro and in vivo. Mechanistically, serine metabolism deficiency increases the expression of IGF1 by reducing the promoter abundance of S-adenosyl methionine-dependent histone H3 lysine 27 trimethylation. IGF1 then activates the p38-dependent JAK–STAT1 axis to promote M(IFN-γ) polarization and suppress STAT6-mediated M(IL-4) activation. 5On the critical particle size of soil with clogging potential in shield tunneling显示文摘Shield tunneling is easily obstructed by clogging in clayey strata with small soil particles.However,soil clogging rarely occurs in strata with coarse-grained soils.Theoretically,a critical particle size of soils should exist,below which there is a high risk of soil clogging in shield tunneling.To determine the critical particle size,a series of laboratory tests was carried out with a large-scale rotary shear apparatus to measure the tangential adhesion strength of soils with different particle sizes and water contents.It was found that the tangential adhesion strength at the soilesteel interface gradually increased linearly with applied normal pressure.When the particle size of the soil specimen was less than 0.15 mm,the interfacial adhesion force first increased and then decreased as the water content gradually increased;otherwise,the soil specimens did not manifest any interfacial adhesion force.The amount of soil mass adhering to the steel disc was positively correlated with the interfacial adhesion force,thus the interfacial adhesion force was adopted to characterize the soil clogging risk in shield tunneling.The critical particle size of soils causing clogging was determined to be 0.15 mm.Finally,the generation mechanism of interfacial adhesion force was explored for soils with different particle sizes to explain the critical particle size of soil with clogging risk in shield tunneling.Shuying Wang Zihao Zhou Pengfei Liu Zhao Yang Qiujing Pan Weizhong Chen 2023Journal of Rock Mechanics and Geotechnical Engineering2023,15,2:3
6Self-thinning lines and allometric relation in Chinese fir(Cunninghamia lanceolata) stands显示文摘We calculated a self-thinning exponent of 1.05 for tree mass using the 3/2 power equation in 93 Cunninghamia lanceolata plots.According to Weller's allometric model,the self-thinning exponent for tree mass was calculated as 1.28 from the allometric exponents h and d.The both self-thinning exponents were significantly lower than 3/2.The self-thinning exponent of organs was estimated to be 1.42 for stems,0.93 for branches,0.96 for leaves,1.35 for roots and 1.28 for shoots,respectively.The self-thinning exponent of stem mass was not significantly different from 3/2,whereas thinning exponents of trees,branches,leaves and roots were significantly lower than 3/2.The stand leaf mass and stand branch mass were constant regardless of the stand density.The scaling relations among branch,leaf,stem,root and shoot mass(MB,ML,MS,MR and MA,respectively) showed that MB and ML scaled as the3/4 power of MS,whereas MS or MA scaled isometrically with respect to MR.Li Xue Xiaoli Hou Qiujing Li Yunting Hao 2015Journal of Forestry Research2015,26,2:3
