1Screening high-quality fetal bovine serum for porcine oocyte maturation in vitro显示文摘Fetal bovine serum(FBS) is widely used in cell cultures due to its high stability and easy access. It was also used as a substitute for porcine follicular fluid(PFF) in previous studies. However, FBS components are unclear, and the presence of FBS in culture media may introduce a variation from batch to batch. This study aimed to establish an effective method to screen FBS in place of PFF in the culture media for porcine oocytes in vitro. We screened FBS from different sources by using porcine fetal fibroblast cells. The effects of six FBS samples on porcine fetal fibroblast cell growth were tested via frozen cell survival assay, cell clone formation assay, cell growth curve, and cell passage activity assay. The best serum that we called GFBS(heat-inactivated FBS, cat. no. 10500-64;Gibco) showed a similar effect on the maturation and development of porcine oocytes to that of PFF and can be used as a good substitute for PFF. 2Quasi-4-dimension ionospheric modeling and its application in PPP显示文摘Ionospheric delay modeling is not only important for Global Navigation Satellite System(GNSS)based space weather study and monitoring,but also an efficient tool to speed up the convergence time of Precise Point Positioning(PPP).In this study,a novel model,denoted as Quasi-4-Dimension Ionospheric Modeling(Q4DIM)is proposed for wide-area high precision ionospheric delay correction.In Q4DIM,the Line Of Sight(LOS)ionospheric delays from a GNSS station network are divided into different clusters according to not only the location of latitude and longitude,but also satellite elevation and azimuth.Both Global Ionosphere Map(GIM)and Slant Ionospheric Delay(SID)models that are traditionally used for wide-area and regional ionospheric delay modeling,respectively,can be regarded as the special cases of Q4DIM by defining proper grids in latitude,longitude,elevation,and azimuth.Thus,Q4DIM presents a resilient model that is capable for both wide-area coverage and high precision.Four different sets of clusters are defined to illustrate the properties of Q4DIM based on 200 EUREF Permanent Network(EPN)stations.The results indicate that Q4DIM is compatible with the GIM products.Moreover,it is proved that by inducting the elevation and azimuth angle dependent residuals,the precision of the 2-dimensional GIM-like model,i.e.,Q4DIM 2-Dimensional(Q4DIM-2D),is improved from around 1.5 Total Electron Content Units(TECU)to better than 0.5 TECU.In addition,treating Q4DIM as a 4-dimensional matrix in latitude,longitude,elevation,and azimuth,whose sparsity is less than 5%,can result in its feasibility in a bandwidth-sensitive applications,e.g.,satellite-based Precising Point Positioning Real-Time Kinematic(PPP-RTK)service.Finally,the advantages of Q4DIM in PPP over the 2-dimensional models are demonstrated with the one month's data from 30 EPN stations in both high solar activity year 2014 and low solar activity year 2020.Shengfeng Gu Chengkun Gan Chengpeng He Haixia Lyu Manuel Hernandez-Pajares Yidong Lou Jianghui Geng Qile Zhao 2022Satellite Navigation2022,3,3:1
3Insulin exerts direct, IGF-1 independent actions in growth plate chondrocytes显示文摘Insufficient insulin production or action in diabetic states is associated with growth retardation and impaired bone healing, while the underling mechanisms are unknown. In this study, we sought to define the role of insulin signaling in the growth plate. Insulin treatment of embryonic metatarsal bones from wild-type mice increased chondrocyte proliferation. Mice lacking insulin receptor(IR) selectively in chondrocytes(CartIR2/2)had no discernable differences in total femoral length compared to control littermates. However, CartIR2/2mice exhibited an increase in chondrocyte numbers in the growth plate than that of the controls. Chondrocytes lacking IR had elevated insulin-like growth factor(IGF)-1R mRNA and protein levels. Subsequently, IGF-1induced phosphorylation of Akt and ERK was enhanced, while this action was eliminated when the cells were treated with IGF-1R inhibitor Picropodophyllin. Deletion of the IR impaired chondrogenic differentiation, and the effect could not be restored by treatment of insulin, but partially rescued by IGF-1 treatment. Intriguingly,the size of hypertrophic chondrocytes was smaller in CartIR2/2mice when compared with that of the control littermates, which was associated with upregulation of tuberous sclerosis complex 2(TSC2). 4Illumination and regolith temperature at China's next candidate lunar landing site Shackleton crater显示文摘This study focuses on the illumination and temperature at China’s next lunar candidate landing site Shackleton crater.We used the NASA’s SPICE system to evaluate the terrain obscuration effect on real-time illumination;the resulting illumination map resembles previous studies,validating the methodologies used in our study.In addition,we estimated an accumulated illumination map for the period of likely rover movement.The map indicates the illuminated inner wall of the Shackleton crater is close to 27%of the whole,meaning that the rover will likely receive solar radiation during its movement.Using the real-time illumination and the distributed 1-D thermal diffusion model,we continuously evaluated the regolith temperature for more than 20 years to stabilize the temperature,and selected the temperature of the end time as the initial value used in a thermal study set for July 20,2023 and May 8,2027.Our results indicate the temperature in the permanent shadow region remains nearly constant,thus validating the stability of our estimated initial temperature.Our results also indicate that the surface temperature is more sensitive to transient illumination,but the subsurface temperature is more likely to be associated with the accumulated illumination.This difference indirectly implies that the conductivity of the lunar regolith is inefficient.The locations receiving more solar radiation show a temperature larger than the threshold(~112 K)of ice stability.The permanently shadowed regions can be as cold as 25 K,and such extreme coldness is a hazard to the rover.There are suitable temperature locations which have a warm surface but cold subsurface to preserve water ice.To further ensure normal rover movement,we provided a map of suitable temperature sites and found that these locations exist not only in the Shackleton crater’s inner wall,but also outside the crater.We suggested four trade-off sampling sites with suitable temperatures and gradual slopes.Zhen ZHONG Jianguo YAN Huaiyu HE Qiling WEN Deyun LIU Jean-Pierre BARRIOT 2023Science China Earth Sciences2023,66,2:0
5Identification and functional characterization of female antennae-biased odorant receptor 23 involved in acetophenone detection of the Indian meal moth Plodia interpunctella显示文摘The Indian meal moth,Plodia interpunctella(Lepidoptera:Pyralidae),a globally distributed storage pest,relies on odors that are emitted from stored foods to select a suitable substrate for oviposition.However,the molecular mechanism underlying the chemical communication between P.interpunctella and its host remains elusive.In this study,130 chemosensory genes were identified from the transcriptomes of 7 P.interpunctella tissues,and the quantitative expression levels of all 56 P.interpunctella odorant receptor genes(PintORs)were validated using real-time quantitative polymerase chain reaction.The functional characteristics of 5 PintORs with female antennae-biased expression were investigated using 2-electrode voltage clamp recordings in Xenopus laevis oocytes.PintOR23 was found to be specifically tuned to acetophenone.Acetophenone could elicit a significant electrophysiological response and only attracted mated females when compared with males and virgin females.In addition,molecular docking predicted that the hydrogen bonding sites,TRP-335 and ALA-167,might play key roles in the binding of PintOR23 to acetophenone.Our study provides valuable insights into the olfactory mechanism of oviposition substrate detection and localization in P.interpunctella and points toward the possibility of developing eco-friendly odorant agents to control pests of stored products.Qi Chen Xiaoyan Zhu Guoqing Kang Qiling Yu Qingxin Liu Lin Du Yi Yang Xinyu He Ying Zhao Junjie Zhang Ying Hu Bingzhong Ren 2024Insect Science2024,31,1:0

7Resilience assessment model for urban public transportation systems based on structure and function显示文摘Urbanization and the increasing frequency of extreme climates affect the sustainability of urban public transportation systems,and improving resilience is one of the primary directions for sustainable development.To scientifically assess the resilience of urban public transportation systems,a resilience assessment model based on structure and function is established in this study.This model mathematically quantifies and simulates the structural and functional changes in public transportation systems under disruption scenarios and provides a comprehensive assessment of six abilities:1)structural resistance,2)structural recoverability,3)functional resistance,4)functional recoverability,5)passenger adaptability,and 6)management adaptability.Depending on the initial failure stations,this model can simulate the resilience of a public transportation system under various scenarios.This model is applied to assess the resilience of public transportation systems in a provincial capital city under an equipment failure scenario.The results show that the impact of equipment failure on resilience varies according to the metro lines,and improvement strategies for functional recoverability and management adaptability are proposed.The weaknesses in the resilience of urban public transportation systems can be identified using the proposed model,which helps provide strategies for improving the capacity to face perturbations.Changkun Chen Fan He Rongfu Yu Siqi Wang Qile Dai 2023Journal of Safety Science and Resilience2023,4,4:0
