| 1 | Emergence of antibiotic resistance Pseudomonas aeruginosa in intensive care unit;a critical review显示文摘The emergence of antibiotic resistant bacteria in the healthcare is a serious concern.In the Healthcare premises precisely intensive care unit are major sources of microbial diversity.Recent findings have demonstrated not only microbial diversity but also drug resistant microbes largely habitat in ICU.Pseudomonas aeruginosa found as a part of normal intestinal flora and a significant pathogen responsible for wide range of ICU acquired infection in critically ill patients.Nosocomial infection associated with this organism including gastrointestinal infection,urinary tract infections and blood stream infection.Infection caused by this organism are difficult to treat because of the presence of its innate resistance to many antibiotics(b-lactam and penem group of antibiotics),and its ability to acquire further resistance mechanism to multiple class of antibiotics,including Beta-lactams,aminoglycosides and fluoroquinolones.In the molecular evolution microbes adopted several mechanism to maintain genomic plasticity.The tool microbe use for its survival is mainly biofilm formation,quorum sensing,and horizontal gene transfer and enzyme promiscuity.Such genomic plasticity provide an ideal habitat to grow and survive in hearse environment mainly antibiotics pressure.This review focus on infection caused by Pseudomonas aeruginosa,its mechanisms of resistance and available treatment options.The present study provides a systemic review on major source of Pseudomonas aeruginosa in ICU.Further,study also emphasizes virulence gene/s associated with Pseudomonas aeruginosa genome for extended drug resistance.Study gives detailed overview of antibiotic drug resistance mechanism. | Preeti Pachori Ragini Gothalwal Puneet Gandhi | 2019 | Genes & Diseases2019,6,2: | 20 |

| 3 | Interactive Effects of Elevated CO_2 and Growth Temperature on the Tolerance of Photosynthesis to Acute Heat Stress in C_3 and C_4 Species显示文摘Determining effects of elevated CO2 on the tolerance of photosynthesis to acute heat-stress (heat wave) is necessary for predicting plant responses to global warming,as photosynthesis is thermolabile and acute heat-stress and atmospheric CO2 will increase in the future.Few studies have examined this,and past results are variable,which may be due to methodological variation.To address this,we grew two C3 and two C4 species at current or elevated CO2 and three different growth temperatures (GT).We assessed photosynthetic thermotolerance in both unacclimated (basal tolerance) and preheat-stressed (preHS=acclimated) plants.In C3 species,basal thermotolerance of net photosynthesis (Pn) was increased in high CO2,but in C4 species,Pn thermotlerance was decreased by high CO2 (except Zea mays at low GT); CO2 effects in preHS plants were mostly small or absent,though high CO2 was detrimental in one C3 and one C4 species at warmer GT.Though high CO2 generally decreased stomatal conductance,decreases in Pn during heat stress were mostly due to non-stomatal effects.Photosystem Ⅱ (PSⅡ efficiency was often decreased by high CO2 during heat stress,especially at high GT; CO2 effects on post-PSII electron transport were variable.Thus,high CO2 often affected photosynthetic theromotolerance,and the effects varied with photosynthetic pathway,growth temperature,and acclimation state.Most importantly,in heat-stressed plants at normal or warmer growth temperatures,high CO2 may often decrease,or not benefit as expected,tolerance of photosynthesis to acute heat stress.Therefore,interactive effects of elevated CO2 and warmer growth temperatures on acute heat tolerance may contribute to future changes in plant productivity,distribution,and diversity. | E. | 5 | Neuroprotective potential of Quercetin in combination with piperine against 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine-induced neurotoxicity显示文摘1-Methy-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP)is a neurotoxin that selectively damages dopaminergic neurons in the substantia nigra pars compacta and induces Parkinson’s like symptoms in rodents.Quercetin(QC)is a natural polyphenolic bioflavonoid with potent antioxidant and anti-inflammatory properties but lacks of clinical attraction due to low oral bioavailability.Piperine is a well established bioavailability enhancer used pre-clinically to improve the bioavailability of antioxidants(e.g.,Quercetin).Therefore,the present study was designed to evaluate the neuroprotective potential of QC together with piperine against MPTP-induced neurotoxicity in rats.MPTP(100μg/μL/rat,bilaterally)was injected intranigrally on days 1,4 and 7 using a digital stereotaxic apparatus.QC(25 and 50 mg/kg,intragastrically)and QC(25 mg/kg,intragastrically)in combination with piperine(2.5 mg/kg,intragastrically)were administered daily for 14 days starting from day 8 after the 3^(rd) injection of MPTP.On day 22,animals were sacrificed and the striatum was isolated for oxidative stress parameter(thiobarbituric acid reactive substances,nitrite and glutathione),neuroinflammatory cytokine(interleukin-1β,interleukin-6,and tumor necrosis factor-α)and neurotransmitter(dopamine,norepinephrine,serotonin,gamma-aminobutyric acid,glutamate,3,4-dihydroxyphenylacetic acid,homovanillic acid,and 5-hydroxyindoleacetic acid)evaluations.Bilateral infusion of MPTP into substantia nigra pars compacta led to significant motor deficits as evidenced by impairments in locomotor activity and rotarod performance in open field test and grip strength and narrow beam walk performance.Both QC(25 and 50 mg/kg)and QC(25 mg/kg)in combination with piperine(2.5 mg/kg),in particular the combination therapy,significantly improved MPTP-induced behavioral abnormalities in rats,reversed the abnormal alterations of neurotransmitters in the striatum,and alleviated oxidative stress and inflammatory response in the striatum.These findings indicate that piperine can enhance the antioxidant and anti-inflammatory properties of QC,and QC in combination with piperine exhibits strong neuroprotective effects against MPTP-induced neurotoxicity. | Shamsher Singh Sumit Jamwal Puneet Kumar | 2017 | Neural Regeneration Research2017,12,7: | 6 |

| 7 | Vitamin D,calcium homeostasis and aging显示文摘Osteoporosis is characterized by low bone mass and microarchitecture deterioration of bone tissue, leading to enhanced bone fragility and consequent increase in fracture risk. Evidence is accumulating for an important role of calcium deficiency as the process of aging is associated with disturbed calcium balance. Vitamin D is the principal factor that maintains calcium homeostasis. Increasing evidence indicates that the reason for disturbed calcium balance with age is inadequate vitamin D levels in the elderly. In this article, an overview of our current understanding of vitamin D, its metabolism, and mechanisms involved in vitamin D-mediated maintenance of calcium homeostasis is presented. In addition, mechanisms involved in age-related dysregulation of 1,25(OH)_2D_3action, recommended daily doses of vitamin D and calcium, and the use of vitamin D analogs for the treatment of osteoporosis(which remains controversial) are reviewed. | 8 | Effect of Elevated CO_2 and Drought on Soil Microbial Communities Associated with Andropogon gerardii显示文摘Our understanding of the effects of elevated atmospheric CO2,singly and in combination with other environmental changes,on plant-soil interactions is incomplete.Elevated CO2 effects on C4 plants,though smaller than on C3 species,are mediated mostly via decreased stomatal conductance and thus water loss.Therefore,we characterized the interactive effect of elevated CO2 and drought on soil microbial communities associated with a dominant C4 prairie grass,Andropogon gerardii Vitman.Elevated CO2 and drought both affected resources available to the soil microbial community.For example,elevated CO2 increased the soil C:N ratio and water content during drought,whereas drought alone decreased both.Drought significantly decreased soil microbial biomass.In contrast,elevated CO2 increased biomass while ameliorating biomass decreases that were induced under drought.Total and active direct bacterial counts and carbon substrate use (overall use and number of used sources) increased significantly under elevated CO2.Denaturing gradient gel electrophoresis analysis revealed that drought and elevated CO2,singly and combined,did not affect the soil bacteria community structure.We conclude that elevated CO2 alone increased bacterial abundance and microbial activity and carbon use,probably in response to increased root exudation.Elevated CO2 also limited drought-related impacts on microbial activity and biomass,which likely resulted from decreased plant water use under elevated CO2.These are among the first results showing that elevated CO2 and drought work in opposition to modulate plant-associated soil-bacteria responses,which should then influence soil resources and plant and ecosystem function. | Issmat I. | 9 | Magnesium acetyltaurate prevents retinal damage and visual impairment in rats through suppression of NMDA-induced upregulation of NF-κB,p53 and AP-1(c-Jun/c-Fos)显示文摘Magnesium acetyltaurate(MgAT)has been shown to have a protective effect against N-methyl-D-aspartate(NMDA)-induced retinal cell apoptosis.The current study investigated the involvement of nuclear factor kappa-B(NF-κB),p53 and AP-1 family members(c-Jun/c-Fos)in neuroprotection by MgAT against NMDA-induced retinal damage.In this study,Sprague-Dawley rats were randomized to undergo intravitreal injection of vehicle,NMDA or MgAT as pre-treatment to NMDA.Seven days after injections,retinal ganglion cells survival was detected using retrograde labelling with fluorogold and BRN3A immunostaining.Functional outcome of retinal damage was assessed using electroretinography,and the mechanisms underlying antiapoptotic effect of MgAT were investigated through assessment of retinal gene expression of NF-κB,p53 and AP-1 family members(c-Jun/c-Fos)using reverse transcription-polymerase chain reaction.Retinal phospho-NF-κB,phospho-p53 and AP-1 levels were evaluated using western blot assay.Rat visual functions were evaluated using visual object recognition tests.Both retrograde labelling and BRN3A immunostaining revealed a significant increase in the number of retinal ganglion cells in rats receiving intravitreal injection of MgAT compared with the rats receiving intravitreal injection of NMDA.Electroretinography indicated that pre-treatment with MgAT partially preserved the functional activity of NMDA-exposed retinas.MgAT abolished NMDA-induced increase of retinal phospho-NF-κB,phospho-p53 and AP-1 expression and suppressed NMDA-induced transcriptional activity of NF-κB,p53 and AP-1 family members(c-Jun/c-Fos).Visual object recognition tests showed that MgAT reduced difficulties in recognizing the visual cues(i.e.objects with different shapes)after NMDA exposure,suggesting that visual functions of rats were relatively preserved by pre-treatment with MgAT.In conclusion,pre-treatment with MgAT prevents NMDA induced retinal injury by inhibiting NMDA-induced neuronal apoptosis via downregulation of transcriptional activity of NF-κB,p53 and AP-1-mediated c-Jun/c-Fos.The experiments were approved by the Animal Ethics Committee of Universiti Teknologi MARA(UiTM),Malaysia,UiTM CARE No 118/2015 on December 4,2015 and UiTM CARE No 220/7/2017 on December 8,2017 and Ethics Committee of Belgorod State National Research University,Russia,No 02/20 on January 10,2020. | Lidawani Lambuk Igor Iezhitsa Renu Agarwal Puneet Agarwal Anna Peresypkina Anna Pobeda Nafeeza Mohd Ismail | 2021 | Neural Regeneration Research2021,16,11: | 5 |

| 11 | Taurine protects against retinal and optic nerve damage induced by endothelin-1 in rats via antioxidant effects显示文摘Endothelin-1(ET-1), a potent vasoconstrictor, is involved in retinal vascular dysregulation and oxidative stress in glaucomatous eyes. Taurine(TAU), a naturally occurring free amino acid, is known for its neuroprotective and antioxidant properties. Hence, we evaluated its neuroprotective properties against ET-1 induced retinal and optic nerve damage. ET-1 was administered intravitreally to Sprague-Dawley rats and TAU was injected as pre-, co-or post-treatment. Animals were euthanized seven days post TAU injection. Retinae and optic nerve were examined for morphology, and were also processed for caspase-3 immunostaining. Retinal redox status was estimated by measuring retinal superoxide dismutase, catalase, glutathione, and malondialdehyde levels using enzyme-linked immuosorbent assay. Histopathological examination showed significantly improved retinal and optic nerve morphology in TAU-treated groups. Morphometric examination showed that TAU pre-treatment provided marked protection against ET-1 induced damage to retina and optic nerve. In accordance with the morphological observations, immunostaining for caspase showed a significantly lesser number of apoptotic retinal cells in the TAU pre-treatment group. The retinal oxidative stress was reduced in all TAU-treated groups, and particularly in the pre-treatment group. The findings suggest that treatment with TAU, particularly pre-treatment, prevents apoptosis of retinal cells induced by ET-1 and hence prevents the changes in the morphology of retina and optic nerve. | 13 | Immunophenotypic signature of primary glioblastoma multiforme: A case of extended progression free survival显示文摘Glioblastoma-multiforme(GBM), the most aggressive glial tumor, has a worldwide age-adjusted incidence ranging from 0.59-3.69/100000 persons. Despite current multimodal-treatment approach, median-survival time and progression-free survival(PFS) remains short. Glioblastomas display a variety of molecular alterations, which necessitates determining which of these have a prognostic significance. This is a case of a 45-yearold patient who presented with progressive slurring of speech and features of raised intracranial pressure. Computed tomography(CT) scan revealed a large heterogeneously enhancing lesion in the left front-temporalperisylvian region with solid, cystic areas, suggestive of malignant glioma. Partial tumor-excision was followed by concurrent chemo-radiotherapy. Histopathologically, the tumor was astrocytoma grade-IV. Patient had an extended PFS of 12 mo, with an overall survival of 26 mo. Primary-GBM was confirmed using molecular markers and the immunophenotypic signature was defined by evaluating systemic expression of human telomerase reverse transcriptase, interleukin-6, neutrophil-lymphocyte ratio, tissue inhibitor of metalloproteinases-1, human chitinase-3-like-protein-1(YKL-40) and high mobility group-A1. | 15 | Unique case of oligoastrocytoma with recurrence and grade progression:Exhibiting differential expression of high mobility group-A1 and human telomerase reverse transcriptase显示文摘Mixed gliomas, primarily oligoastrocytomas, account for about 5%-10% of all gliomas. Distinguishing oligoastrocytoma based on histological features alone has limitations in predicting the exact biological behavior, necessitating ancillary markers for greater specificity. In this case report, human telomerase reverse transcriptase(hT ERT) and high mobility group-A1(HMGA1); markers of proliferation and stemness, have been quantitatively analyzed in formalin-fixed paraffin-embedded tissue samples of a 34 years old patient with oligoastrocytoma. Customized florescence-based immunohistochemistry protocol with enhanced sensitivity and specificity is used in the study. The patient presented with a history of generalized seizures and his magnetic resonance imaging scans revealed infiltrative ill-defined mass lesion with calcified foci within the left frontal white matter, suggestive of glioma. He was surgically treated at our center for four consecutive clinical events. Histopathologically, the tumor was identified as oligoastrocytoma-grade Ⅱ followed by two recurrence events and final progression to grade Ⅲ. Overall survival of the patient without adjuvant therapy was more than 9 years. Glial fibrillary acidic protein, p53, Ki-67, nuclear atypia index, pre-operative neutrophillymphocyte ratio, are the other parameters assessed. Findings suggest that hT ERT and HMGA1 are linked to tumor recurrence and progression. | 16 | Novel molecular panel for evaluating systemic inflammation and survival in therapy naïve glioma patients显示文摘BACKGROUND Inflammation is crucial to tumor progression.A traumatic event at a specific site in the brain activates the signaling molecules,which triggers inflammation as the initial response within the tumor and its surroundings.The educated immune cells and secreted proteins then initiate the inflammatory cascade leading to persistent chronic inflammation.Therefore,estimation of the circulating inflammatory indicators kynurenine(KYN),interleukin-6(IL-6),tissue-inhibitor of matrix-metalloproteinase-1 and human telomerase reverse transcriptase(hTERT)along with neutrophil-lymphocyte ratio(NLR)has prognostic value.AIM To assess the utility of chosen inflammatory marker panel in estimating systemic inflammation.METHODS The chosen markers were quantitatively evaluated in 90 naive,molecularly subtyped plasma samples of glioma.A correlation between the markers and confounders was assessed to establish their prognostication power.Follow-up on the levels of the indicators was done 3-mo post-surgery.To establish the validity of circulating KYN,it was also screened qualitatively by dot-immune-assay and by immunofluorescence-immunohistochemistry in tumor tissues.RESULTS Median values of circulating KYN,IL-6,hTERT,tissue-inhibitor of matrixmetalloproteinase-1 and NLR in isocitrate-dehydrogenase-mutant/wildtype and within the astrocytic sub-groups were estimated,which differed from controls,reaching statistical significance(P<0.0001).All markers negatively correlated with mortality(P<0.0001).Applying combination-statistics,the panel of KYN,IL-6,hTERT and NLR achieved higher sensitivity and specificity(>90%)than standalone markers,to define survival.The inflammatory panel could discriminate between WHO grades,and isocitrate-dehydrogenase-mutant/wildtype and define differential survival between astrocytic isocitrate-dehydrogenase-mutant/wildtype.Therefore,its assessment for precise disease prognosis is indicated.Association of KYN with NLR,IL-6 and hTERT was significant.Cox-regression described KYN,IL-6,NLR,and hTERT as good prognostic markers,independent of confounders.Multivariate linear-regression analysis confirmed the association of KYN and hTERT with inflammation marker IL-6.There was a concomitant significant decrease in their levels in a 3-mo follow-up.CONCLUSION The first evidence-based study of circulating-KYN in molecularly defined gliomas,wherein the tissue expression was found to be concomitant with plasma levels.A non-invasive model for assessing indicators of chronic systemic inflammation is proposed. | Puneet Gandhi Richa Shrivastava Nitin Garg Sandeep K Sorte | 2021 | World Journal of Clinical Oncology2021,12,10: | 2 |
