| 1 | Psychometrics of the chronic liver disease questionnaire for Southern Chinese patients with chronic hepatitis B virus infection显示文摘AIM:To test the psychometric properties of a Chinese [(Hong Kong) HK] translation of the chronic liver disease questionnaire (CLDQ).METHODS:A Chinese (HK) translation of the CLDQ was developed by iterative translation and cognitive debriefing.It was then administered to 72 uncomplicated and 78 complicated chronic hepatitis B (CHB) patients in Hong Kong together with a structured questionnaire on service utilization,and the Chinese (HK) SF-36 Health Survey Version 2 (SF-36v2).RESULTS:Scaling success was ≥ 80% for all but three items.A new factor assessing sleep was found and items of two (Fatigue and Systemic Symptoms) subscales tended to load on the same factor.Internal consistency and test-retest reliabilities ranged from 0.58-0.90 for different subscales.Construct validity was confirmed by the expected correlations between the SF-36v2 Health Survey and CLDQ scores.Mean scores of CLDQ were significantly lower in complicated compared with uncomplicated CHB,supporting sensitivity in detecting differences between groups.CONCLUSION:The Chinese (HK) CLDQ is valid,reliable and sensitive for patients with CHB.Some modif ications to the scaling structure might further improve its psychometric properties. | Elegance Ting Pui Lam Cindy Lo Kuen Lam Ching Lung Lai Man Fung Yuen Daniel Yee Tak Fong | 2009 | World Journal of Gastroenterology2009，15，26： | 16 |
| 2 | Metamorphism of the northern Liaoning Complex:Implications for the tectonic evolution of Neoarchean basement of the Eastern Block,North China Craton显示文摘As one of the areas where typical late Archean crust is exposed in the Eastern Block of the North China Craton,the northern Laioning Complex consists principally of tonalitic-trondhjemitic-granodioritic(TTG) gneisses,massive granitoids and supracrustal rocks.The supracrustal rocks,named the Qingyuan Group, consist of interbedded amphibolite,hornblende granulite,biotite granulite and B1F.Petrological evidence indicates that the amphibolites experienced the early prograde(M1),peak(M2) and post-peak(M3) metamorphism.The early prograde assemblage(M1) is preserved as mineral inclusions,represented by actinotite + hornblende + plagioclase + epidote + quartz + sphene,within garnet porphyroblasts.The peak assemblage(M2) is indicated by garnet + clinopyroxene + hornblende + plagioclase + quartz + ilmenite, which occur as major mineral phases in the rock.The post-peak assemblage(M3) is characterized by the garnet + quartz symplectite.The P—T pseudosections in the NCFMASHTO system constructed by using THERMOCALC define the P-T conditions of Ml,M2 and M3 at 490-550℃/<4.5 kbar,780-810℃/7.65- 8.40 kbar and 630-670℃/8.15-9.40 kbar,respectively.As a result,an anticlockwise P-T path involving isobaric cooling is inferred for the metamorphic evolution of the amphibolites.Such a P-T path suggests that the late Archean metamorphism of the northern Liaoning Complex was related to the intrusion and underplating of mantle-derived magmas.The underplating of voluminous mantle-derived magmas leading to metamorphism with an anticlockwise P-T path involving isobaric cooling may have occurred in continental magmatic arc regions,above hot spots driven by mantle plumes,or in continental rift environments. | 3 | Precision medicine in acute lymphoblastic leukemia显示文摘The cure rate of childhood acute lymphoblastic leukemia(ALL)has exceeded 90%in some contemporary clinical trials.However,the dose intensity of conventional chemotherapy has been pushed to its limit.Further improvement in outcome will need to rely more heavily on molecular therapeutic as well as immuno-and cellular-therapy approaches together with precise risk stratification.Children with ETV6-RUNX1 or hyperdiploid>50 ALL who achieve negative minimal residual disease during early remission induction are suitable candidates for reduction in treatment.Patients with Philadelphia chromosome(Ph)-positive or Ph-like ALL with ABL-class fusion should be treated with dasatinib.BH3 profiling and other preclinical methods have identified several high-risk subtypes,such as hypodiplod,early T-cell precursor,immature T-cell,KMT2A-rearranged,Ph-positive and TCF-HLF-positive ALL,that may respond to BCL-2 inhibitor venetoclax.There are other fusions or mutations that may serve as putative targets,but effective targeted therapy has yet to be established.For other high-risk patients or poor early treatment responders who do not have targetable genetic lesions,current approaches that offer hope include blinatumomab,inotuzumab and CAR-T cell therapy for B-ALL,and daratumumab and nelarabine for T-ALL.With the expanding therapeutic armamentarium,we should start focus on rational combinations of targeted therapy with non-overlapping toxicities. | Ching-Hon Pui | 2020 | Frontiers of Medicine2020，14，6： | 13 |

| 5 | Bioengineered miR-27b-3p and miR-328-3p modulate drug metabolism and disposition via the regulation of target ADME gene expression显示文摘Drug-metabolizing enzymes, transporters, and nuclear receptors are essential for the absorption, distribution, metabolism, and excretion(ADME) of drugs and xenobiotics. MicroRNAs participate in the regulation of ADME gene expression via imperfect complementary Watson–Crick base pairings with target transcripts. We have previously reported that Cytochrome P450 3A4(CYP3A4) and ATP-binding cassette sub-family G member 2(ABCG2) are regulated by miR-27b-3p and miR-328-3p,respectively. Here we employed our newly established RNA bioengineering technology to produce bioengineered RNA agents(BERA), namely BERA/miR-27b-3p and BERA/miR-328-3p, via fermentation. When introduced into human cells, BERA/miR-27b-3p and BERA/miR-328-3p were selectively processed to target miRNAs and thus knock down CYP3A4 and ABCG2 mRNA and their protein levels,respectively, as compared to cells treated with vehicle or control RNA. Consequently, BERA/miR-27b-3p led to a lower midazolam 10-hydroxylase activity, indicating the reduction of CYP3A4 activity. Likewise,BERA/miR-328-3p treatment elevated the intracellular accumulation of anticancer drug mitoxantrone, a classic substrate of ABCG2, hence sensitized the cells to chemotherapy. | 12 | Effects of different acupuncture manipulations on protein expression in the parietal cortex of spontaneously hypertensive rats显示文摘Objective: To analyze the effects of twirling reinforcing and reducing manipulations on protein expression in parietal cortex of spontaneously hypertensive rats(SHRs), and elucidate the main mechanisms and differences between two manipulations in hypertension treatment.Methods: Rats were randomly divided into the control, model, twirling reinforcing manipulation(TRFM),and twirling reducing manipulation(TRDM) groups. The control and model groups received catch and fixation stimulations once a day for 14 days. The TRFM and TRDM groups were intervened once a day for 20 min for 14 days. On days 0, 2, 6, 10, and 14 after acupuncture, rat systolic blood pressures(SBPs) were measured. Differential protein(DP) expression in the rat parietal cortices was detected. Thereafter, GO functional significance and KEGG pathway enrichment analyses were performed.Results: Compared with the model group, SBP of rats in the TRDM and TRFM groups decreased on days 6 and 10 of acupuncture, respectively(P=.009;P <.001). Moreover, SBP of the TRDM group was significantly lower than that of the TRFM group on days 10 and 14 of acupuncture(P=.015;P=.013).Compared with control group, 601 and 1040 DPs were up-and downregulated, respectively, in the model group. Compared with model group, 44 and 28 up-and downregulated DPs were expressed, respectively,in the TRFM group. Compared with model group, expression of 616 and 427 up-and downregulated DPs,respectively, was found in the TRDM group. | 16 | Differentiation potential of human adipose tissue derived stem cells into photoreceptors through explants culture and enzyme methods显示文摘● AIM: To investigate the retinal photoreceptor differentiation potential of human orbital adipose tissue-derived stem cells(ADSCs) generated by enzyme(EN) and explant(EX) culture methods.● METHODS: We investigated potentials of human orbital ADSCs to differentiate into photoreceptors through EN and EX culture methods. EN and EX orbital ADSCs were obtained from the same donor during rehabilitative orbital decompression, and then were subject to a 3-step induction using Noggin, DKK-1, IGF-1 and b-FGF at different time points for 38 d. Stem cell, eye-field and photoreceptor-related gene and protein markers were measured by reverse transcription-polymerase chain reaction(RT-PCR) and immunofluorescent(IMF) staining. ● RESULTS: Both EX and EN orbital ADSCs expressed CD133, a marker of cell differentiation. Moreover, PAX6 and rhodopsin, markers of the retinal progenitor cells, were detected from EX and EN orbital ADSCs. In EX orbital ADSCs, PAX6 m RNA was detected on the 17^(th) day and then the rhodopsin m RNA was detected on the 24^(th) day. In contrast, the EN orbital ADSCs expressed PAX6 and rhodopsin m RNA on the 31^(st) day. | 17 | Molecular Evidence for the Hybrid Origin of Bauhinia blakeana(Caesalpinioideae)显示文摘Bauhinia blakeana Dunn is the Hong Kong Special Administrative Region emblem and a popular horticultural species in many Asian countries. It was first described as a new species from Hong Kong almost a century ago. This plant is sterile and has long been considered a hybrid,possibly from two related species,B. purpurea and B. variegata. However,not much evidence based on molecular methods was available to support this hypothesis. In this study,sequences of internal transcribed spacer 1(ITS1) ,rbcL and atpB-rbcL intergenic spacer for five Bauhinia species and two varieties of one of the species were determined and compared. There were two types of ITS1 sequences in B. blakeana,one indistinguishable from that of B. purpurea and the other one identical to that of B. variegata. This confirmed that B. blakeana was a hybrid of these two species. Chloroplast atpB-rbcL intergenic spacer sequence of B. blakeana was identical to that of B. purpurea,indicating that B. purpurea was the female parent. The hybridization event seemed to occur only recently and was a rare incident. Its occurrence was likely facilitated by interspecific pollen competition. | 18 | Screening high-quality fetal bovine serum for porcine oocyte maturation in vitro显示文摘Fetal bovine serum(FBS) is widely used in cell cultures due to its high stability and easy access. It was also used as a substitute for porcine follicular fluid(PFF) in previous studies. However, FBS components are unclear, and the presence of FBS in culture media may introduce a variation from batch to batch. This study aimed to establish an effective method to screen FBS in place of PFF in the culture media for porcine oocytes in vitro. We screened FBS from different sources by using porcine fetal fibroblast cells. The effects of six FBS samples on porcine fetal fibroblast cell growth were tested via frozen cell survival assay, cell clone formation assay, cell growth curve, and cell passage activity assay. The best serum that we called GFBS(heat-inactivated FBS, cat. no. 10500-64;Gibco) showed a similar effect on the maturation and development of porcine oocytes to that of PFF and can be used as a good substitute for PFF.
