| 3 | Modeling of elastic wave propagation on a curved free surface using an improved finite-difference algorithm显示文摘Based on existing direct and imaging methods of a staggered finite-difference scheme, an improved algorithm for staggered finite-difference is proposed to implement rugged topographic free boundary conditions. This method assumes that the free surface can be implemented with horizontal and vertical free surface segments and their corners; the free surface passes through the grid points of shear stress components, instead of the normal stress components. Imaging is carried out for stress components in both horizontal and vertical directions, thus increasing the accuracy. To update particle- velocities, imaging and updating are first performed in the horizontal direction, and then in the vertical direction. The numerical results for elastic flat horizontal free surface with the imaging method and those for flat free surfaces of various slope angles with the proposed method are compared, and are shown to be in good agreement. | 4 | Cloning of cytochrome P-450 2C9 cDNA from human liver and its expression in CHL cells显示文摘AIM: Using bacterial, yeast, or mammalian cell expressing a human drug metabolism enzyme would seem good way to study drug metabolism-related problems. Human cytochrome P-450 2C9 ( CYP2 C9) is a polymorphic enzyme responsible for the metabolism of a large number of clinically important drugs. It ranks among the most important drug metabolizing enzymes in humans. In order to provide a sufficient amount of the enzyme for drug metabolic research, the CYP2 C9 eDNA was cloned and expressed stably in CHL cellsMETHODS: After extraction of total RNA from human livertissue, the human CYP2C9 eDNA was amplified withreverse transcription-polymerase chain reaction (RT-PCR),and cloned into cloning vector pGEM-T. The cDNA fragmentwas identified by DNA sequencing and subcloned into amammalian expression vector pREP9. A transgenic cell linewas established by transfecting the recombinant vector ofpREP9-CYP2C9 into CHL cells. The enzyme activity ofCYP2C9 catalyzing oxidation of tolbutamide to hydroxytolbutamide in S9 fraction of the cell was determined by highperformance liquid chromatography(HPLC).RESULTS: The amino acid sequence predicted from theeDNA segment was identical to that of CYP2 C9 * 1, the wildtype CYP2 C9. However, there were two base differences, i.e. 21T > C, 1146C > T, but the encoding amino acidsequence was the same, L7, P382. | 5 | EXPERIMENTAL STUDY OF BEDLOAD TRANSPORT IN UNSTEADY OPEN-CHANNEL FLOW显示文摘Experiments are performed to study bedload transport in both steady uniform and unsteady open-channel flows. Data of Steady uniform flow fit well with the existing formula in the literature. If the formula for sediment transport in a uniform flow is used to predict the bedload transport rate in unsteady flow, the calculated and the measured bedload transport rates are different. For the runs with predominately accelerating flow, the calculated sediment transport rate is smaller than the measured one, while for the runs with predominately decelerating flow, the calculated sediment transport rate is much larger than the measured one. | 7 | High Expression of NKG2A/CD94 and Low Expression of Granzyme B Are Associated with Reduced Cord Blood NK Cell Activity显示文摘Human umbilical cord blood (CB) has recently been used as a source of stem cells in transplantation. NK cells derived from CB are the key effector cells involved in graft-versus-host disease (GVHD) and graft-versus-leukemia (GVL). It was reported that the activity of CB NK cells was lower than that of adult peripheral blood (PB) NK cells. In this study,we analyzed the expression of some NK cell receptors and cytotoxicity-related molecules in CB and PB NK cells. The expressions of activating NK receptors,CD16,NKG2D and NKp46,did not show significant difference between CB and PB NK cells. But the expression of inhibitory receptor NKG2A/CD94 was significantly higher on CB NK cells. As to the effector function molecules,granzyme B was expressed significantly lower in CB NK cells,but the expressions of intracellular perforin,IFN-γ,TNF-α and cell surface FasL and TRAIL did not show difference between CB and PB NK cells. | 8 | Surgical treatment for Nevin stage IV and V gallbladder carcinoma: report of 70 cases显示文摘BACKGROUND: The role of aggressive surgery for end-stage gallbladder carcinoma is controversial. This retrospective study was designed to evaluate the outcome of surgical treatment for Nevin stage Ⅳ and Ⅴ gallbladder carcinoma at a single institution. METHODS: A retrospective analysis was made on 70 patients with Nevin stage Ⅳ and Ⅴ gallbladder carcinoma undergoing surgical treatment from January 1993 to June 2004. RESULTS: There were 22 cases of stage Ⅳ and 48 of stage V. Cholecystectomy was performed in 37 cases with a resection rate of 53%,9 cases received radical resection, 13 extended radical resection, and 15 palliative resection. The curative resection rate was 31% and the morbidity rate was 36%. Postoperative 1-, 3-, 5-year survival rates of curative and palliative resection were 69%, 33%, 8% and 27%, 13%, 0, respectⅣely (P <0.01). The 1- and 3-year survival rates of patients undergoing exploratory laparotomy only were 3% and 0, respectively. CONCLUSIONS: Nevin stage Ⅳ and Ⅴ gallbladder carcinoma should be treated by aggressive surgery. | 11 | Low temperature induces oocytes p34^(cdc2) synthesis and accumulation-the acquisition of competence to resume meiosis in toad oocytes显示文摘Full grown oocytes derived from Bufo Bufo gargarizans rearing at high temperature environment (24℃),never underwent GVBD after progesterone treatment. No p34cdc2, H1 kinase activity was detected in the oocytes after progesterone stimulation or OA microinjection; Western blotting analysis showed that the level of p3cdc2, and p33 in the oocytes are significantly lower than those in the oocytes derived from the hibernating toads (below 10 ℃). 25S-Met incorporation analysis showed that when the oocytes were incubated at 6℃, synthesis of about thirty defferent polypeptides was promoted or induced, including p34cdc2 and some other p13suc1-binding proteins. All these results indicated that a low temperature environment is essential for the oocytes of Bufo Bufo gargarizans to express and store some cell cycle drivers and its regulators, and to gain the maturation competence. The results showed that the inhibjtoryIX)tency of thirteen varieties of PAs differed greatly, which may be related to theirchemical structures; the blocking rate (BR) of different killds of PAs were as follows:caf feic acid, 92.5%; tannic acid, 90.0%; gallic acid, 86.8%; sinaPinic acid, 86.2%; ferulicacid, 8l.l%; chlorogenic acid, 69.4%; gentisic acid, 69.2%; syringic acid, 62.1%; protocatechuic acid, 56.0%; p-coumaric acid, 52.5%; vannilic acid, 35.4%; moreover,p-hydroxybenzoic acid and m-coumaric acid had the least blocking effect or even slightcatalyzing effect.
