| 1 | Essential role of monocytes and macrophages in the progression of acute pancreatitis显示文摘Acute pancreatitis(AP) is an inflammatory condition of the pancreas caused by an imbalance in factors involved in maintaining cellular homeostasis.Earliest events in AP occur within acinar cells accompanied by other principal contributors to the inflammatory response i.e.the endothelial cells,immunocytes(granulocytes,monocytes/macrophages,lymphocytes) and neutrophils.Monocytes/macrophages are important inflammatory mediators,involved in the pathophysiology of AP,known to reside in the peritoneal cavity(in the vicinity of the pancreas) and in peripancreatic tissue.Recent studies suggested that impaired clearance of injured acini by macrophages is associated with an altered cytokine reaction which may constitute a basis for progression of AP.This review focuses on the role of monocytes/macrophages in progression of AP and discusses f indings on the inflammatory process involved. | Pratima Shrivastava Madhav Bhatia | 2010 | World Journal of Gastroenterology2010,16,32: | 33 |
| 2 | CAN catalyzed one-pot synthesis and docking study of some novel substituted imidazole coupled 1,2,4-triazole-5-carboxylic acids as antifungal agents显示文摘The present work describes a facile,one-pot three component synthesis of a series of 3-[(4,5-diphenyl-2-substituted aryl/heteryl)-1H-imidazol-1-yl]-1H-1,2,4-triazole-5-carboxylic acid derivatives M(1-15).Benzil,aromatic aldehydes and 3-amino-l,2,4-triazole-5-carboxylic acid was refluxed in ethanol using cerric ammonium nitrate(CAN) as a catalyst to give the title compounds in good yields.The compounds were evaluated for their in vitro antifungal and antibacterial activity.Compounds M1,M9,and M15 were found to be equipotent against Candida albicans when compared with fluconazole.Compounds M2.M5,and M14 showed higher activity against Streptococcus pneumoniae.Escherichia coli and Streptococcus pyogenes,respectively,compared with ampicillin.Docking study of the newly synthesized compounds was performed,and the results showed good binding mode in the active sites of C albicans enzyme cytochrome P450 lanosterol 14α-demethylase.The results of in vitro antifungal activity and docking study showed that synthesized compounds had potential antifungal activity and can be further optimized and developed as a lead compound. | Anna Pratima G.Nikalje Mangesh S.Ghodke Firoz A.Kalam Khan Jaiprakash N.Sangshetti | 2015 | Chinese Chemical Letters2015,26,1: | 3 |
| 3 | Leukemic stromal hematopoietic microenvironment negatively regulates the normal hematopoiesis in mouse model of leukemia显示文摘Background and Objective: Leukemic microenvironment has a major role in the progression of leukemia. Leukemic cells can induce reversible changes in microenvironmental components, especially the stromal function which results in improved growth conditions for maintaining the malignant leukemic cells. This study aimed to investigate the survival advantage of leukemic cells over normal hematopoietic cells in stromal microenvironment in long term. Methods: The mice were injected intraperitoneally with N-N’ ethylnitrosourea (ENU) to induce leukemia; the mice received injection of normal saline were used as control. At 180 days after ENU induction, the mice were killed and the bone marrows were cultured for 19 days. Colony-forming assays were used to analyze the formation of various cell colonies. The expression of Sca-1, CD146, VEGFR2, CD95, pStat3, pStat5, and Bcl-xL in marrow cells were detected by flow cytometry. Results: Long-term leukemic bone marrow culture showed abnormal elongated stromal fibroblasts with almost absence of normal hematopoietic cells. Adherent cell colonies were increased, but CFU-F and other hematopoietic cell colonies were significantly decreased in leukemia group (P< 0.001). Primitive progenitor-specific Sca-1 receptor expression was decreased with subsequent increased expression of CD146 and VEGFR-2 in leukemic bone marrow cells. Decreased Fas antigen expression with increased intracellular pStat3, pStat5 and Bcl-xL proteins were observed in leukemic bone marrow cells. Conclusions: Stromal microenvironment shows altered morphology and decreased maturation in leukemia. Effective progenitor cells are decreased in leukemia with increased leukemia-specific cell population. | 4 | Gut microbiome profiling and colorectal cancer in African Americans and Caucasian Americans显示文摘AIM To determine whether and to what extent the gut microbiome is involved in regulating racial disparity in colorectal cancer(CRC). METHODS All patients were recruited and experiments were performed in accordance with the relevant guidelines and regulations by the Institutional Review Boards (IRB), committees of the John D. Dingell VAMC and Wayne State University guidelines. African American (AA) and Caucasian American (CA) patients were scheduled for an outpatient screening for colonoscopy, and no active malignancy volunteer patients were doubly consented, initially by the gastroenterologist and later by the study coordinator, for participation in the study. The gut microbial communities in colonic effluents from AAs and CAs were examined using 16 sRNA profiling, and bacterial identifications were validated by performing SYBR-based Real Time PCR. For metagenomic analysis to characterize the microbial communities, multiple software/tools were used, including Metastats and R statistical software.RESULTS It is generally accepted that the incidence and mortality of CRC is higher in AAs than in CAs. However, the reason for this disparity is not well understood. We hypothesize that the gut microbiome plays a role in regulating this disparity. Indeed, we found significant differences in species richness and diversity between AAs and CAs. Bacteroidetes was more abundant in AAs than in CAs. In particular, the pro-inflammatory bacteria Fusobacterium nucleatum and Enterobacter species were significantly higher in AAs, whereas probiotic Akkermansia muciniphila and Bifidobacterium were higher in CAs. The polyphyletic Clostridia class showed a divergent pattern, with Clostridium XI elevated in AAs, and Clostridium IV, known for its beneficial function, higher in CAs. Lastly, the AA group had decreased microbial diversity overall in comparison to the CA group. | 5 | Glacier-Glacial Lake Interactions and Glacial Lake Development in the Central Himalaya,India(1994–2017)显示文摘Despite several regional glacier and glacier lake inventories, the relationship between receding glacier, glacial lake evolution(glacial-lake interactions) and their sensitivity to different forcing factors have not been properly understood yet. To better understand these processes, we used satellite images collected in 1994, 2015 and 2017 to monitor the spatially-explicit evolution of glacial lakes and glacier changes. The results show a total of 1 353 glacial lakes covering an area of 7.96 km;in the year 2015. Out of these, a total of 137 glacial lakes having an area of >0.01 km;and located within 2 km periphery of mother glacier have been selected for the monitoring of spatial development between 1994 and 2017. We found an increase in the total lake area from ~4.9 to ~7.73 km;between 1994 and 2017,corresponding to an overall expansion of ~57%. The total area covered by the glaciers associated with these lakes reduced from ~365 km;in 1994 to ~358 km^(2) in 2017, accounting for a glacier loss of ~7 km^(2) and corresponding to ~1.92% reduction. Our study results are in agreement with global glacier behavior, revealing a rapid glacier recession and accelerated glacial lake expansion under an unprecedented climate change scenario. | 7 | Controlled release of FK506 from micropatterned PLGA films:potential for application in peripheral nerve repair显示文摘After decades of research,peripheral nerve injury and repair still frequently results in paralysis,chronic pain and neuropathies leading to severe disability in patients.Current clinically available nerve conduits only provide crude guidance of regenerating axons across nerve gap without additional functionality.FK506(Tacrolimus),an FDA approved immunosuppressant,has been shown to enhance peripheral nerve regeneration but carries harsh side-effects when delivered systemically.The objective of this study was to develop and evaluate a bioresorbable drug delivery system capable of local extended delivery of FK506 that also provides topological guidance cues to guide axon growth via microgrooves.Photolithography was used to create micropatterned poly(lactide-co-glycolic acid)(PLGA) films embedded with FK506.Non-patterned,10/10 μm(ridge/groove width),and 30/30 μm patterned films loaded with 0,1,and 3 μg/cm2 FK506 were manufactured and characterized.In vitro FK506 rate of release testing indicated that the films are capable of an extended(at least 56 days),controlled,and scalable release of FK506.Neurite extension bioactivity assay indicated that FK506 released from the films(concentration of samples tested ranged between 8.46–19.7 ng/m L) maintained its neural bioactivity and promoted neurite extension similar to control FK506 dosages(10 ng/m L FK506).The multi-functional FK506 embedded,micropatterned poly(lactide-co-glycolic acid) films developed in this study have potential to be used in the construction of peripheral nerve repair devices. | Brett Davis Susan Wojtalewicz Pratima Labroo Jill Shea Himanshu Sant Bruce Gale Jayant Agarwal | 2018 | Neural Regeneration Research2018,13,7: | 2 |
