1Preparation,characterization of Mo,Ag-loaded BiVO_4 and comparison of their degradation of methylene blue显示文摘Two types of metal-loaded visible-light-driven photocatalysts,Mo-BiVO4and Ag-BiVO4,were synthesized by wet impregnation method.Material poperties were characterized by UV-vis diffuse reflectance spectroscopy,X-ray diffraction,field emission scanning electron microscopy,X-ray photoelectron spectroscopy and low temperature nitrogen adsorption-desorption.Photocatalytic activity of the obtained materials was investigated through degrading methylene blue(MB) solution under visible-light irradiation.The results reveal that both metal loaded-BiVO4catalysts have monoclinic scheelite structure.Mo and Ag exist as oxides on the surface of the particles.The changes of absorption in visible-light region,band gap(E g) and specific surface area(A BET) caused by loading Ag are more obvious than those caused by loading Mo.But the isoelectric point of Ag-BiVO4decreases less than that of Mo-BiVO4does.Both catalysts show higher photocatalytic activity than pure BiVO4,resulting in the significantly improved efficiency of degradation of MB.And the degradation efficiency of these two metal-loaded BiVO4photocatalysts is similar to each other.However,mechanisms of such enhancement are different.The decrease of isoelectric point helps Mo-BiVO4improve the degradation efficiency.As for Ag-BiVO4,the augmentation of absorption in visible-light region as well as the abatement of E g plays more important roles.LIU KunJie CHANG ZhiDong LI WenJun CHE Ping ZHOU HuaLei 2012Science China Chemistry2012,55,9:10
2MdATG18a overexpression improves basal thermotolerance in transgenic apple by decreasing damage to chloroplasts显示文摘High temperature is an abiotic stress factor that threatens plant growth and development.Autophagy in response to heat stress involves the selective removal of heat-induced protein complexes.Previously,we showed that a crucial autophagy protein from apple,MdATG18a,has a positive effect on drought tolerance.In the present study,we treated transgenic apple(Malus domestica)plants overexpressing MdATG18a with high temperature and found that autophagy protected them from heat stress.Overexpression of MdATG18a in apple enhanced antioxidase activity and contributed to the production of increased beneficial antioxidants under heat stress.Transgenic apple plants exhibited higher photosynthetic capacity,as shown by the rate of CO_(2) assimilation,the maximum photochemical efficiency of photosystem II(PSII),the effective quantum yield,and the electron transport rates in photosystems I and II(PSI and PSII,respectively).We also detected elevated autophagic activity and reduced damage to chloroplasts in transgenic plants compared to WT plants.In addition,the transcriptional activities of several HSP genes were increased in transgenic apple plants.In summary,we propose that autophagy plays a critical role in basal thermotolerance in apple,primarily through a combination of enhanced antioxidant activity and reduced chloroplast damage.Liuqing Huo Xun Sun Zijian Guo Xin Jia Runmin Che Yiming Sun Yanfei Zhu Ping Wang Xiaoqing Gong Fengwang Ma 2020Horticulture Research2020,7,1:9
3Overexpression of MdATG18a in apple improves resistance to Diplocarpon mali infection by enhancing antioxidant activity and salicylic acid levels显示文摘Marssonina apple blotch,caused by Diplocarpon mali,is one of the most serious diseases of apple.Autophagy plays a key role in pathogen resistance.We previously showed that MdATG18a has a positive influence on drought tolerance.Herein,we describe how overexpression(OE)of MdATG18a enhances resistance to D.mali infection,probably because less H2O2 but more salicylic acid(SA)is accumulated in the leaves of OE apple plants.Expression of chitinase,β-1,3-glucanase,and SA-related marker genes was induced more strongly by D.mali in OE lines.Transcript levels of other important MdATG genes were also drastically increased by D.mali in OE plants,which indicated increased autophagy activities.Taken together,these results demonstrate that OE of MdATG18a enhances resistance to infection by D.mali and plays positive roles in H2O2-scavenging and SA accumulations.Our findings provide important information for designing strategies which could induce autophagy to minimize the impact of this disease on apple production.Xun Sun Liuqing Huo Xin Jia Runmin Che Xiaoqing Gong Ping Wang Fengwang Ma 2018Horticulture Research2018,5,1:8
4Effects of Yiqi Huoxue Recipe (益气活血方) and Coxsackie Virus B Type 3 on the Expression of Ribosomal Protein S20 in Rat Cardiac Myocytes显示文摘<正>Objective:To study the effects of Yiqi Huoxue Recipe(益气活血方) and Coxsackie B virus type 3(CVB3) on the expression of ribosomal protein S20 in rat cardiac myocytes,to explore the pathogenesis of myocarditis induced by CVB3 and the mechanism of Yiqi Huoxue Recipe on gene level,and to further investigate whether Yiqi Huoxue Recipe is an effective prescription for CVB3 myocarditis.Methods:A modified suppression subtractive hybridization(SSH) was used to isolate differentially expressed genes between the CVB3 infection group and the treatment group with Yiqi Huoxue Recipe.The results were further verified by fluorescence RT-PCR.Results:The results of SSH showed that the gene expression of ribosomal protein S20 in the treatment group was higher than that in the CVB3 infection group(P<0.05),which agreed with the results of fluorescent RT-PCR.Conclusion:Down-regulation of ribosomal protein S20 mRNA expression might be one of the mechanisms in CVB3 myocarditis,and Yiqi Huoxue Recipe could achieve the treatment of viral myocarditis by regulating the expression of ribosomal protein S20.ZHANG Ming-xue ZHANG Yu-mei Esther Jun HE Wei CHE Hong-hua GU Ping 2011Chinese Journal of Integrative Medicine2011,17,5:8
5Matrilineal History of the Rana longicrus Species Group(Rana,Ranidae,Anura) and the Description of a New Species from Hunan,southern China显示文摘Fieldwork in Jiemuxi National Nature Reserve,Hunan,China,discovered morphologically indistinguishable groups of brown frogs that bred at different,exclusive times of the year.A principal components analysis of morphometric data,molecular analyses,and the exclusive breeding season suggested the occurrence of two species.The population that breeds during the winter was found to be an undescribed species and was subsequently described as Rana jiemuxiensis sp.nov.The new cryptic species can be identified from its congeners at the same locality by having a different breeding season and a divergent DNA barcode.Six major lineages of brown frogs were resolved with high nodal support.Rana japonica,R.chaochiaoensis,R.omeimontis,and R.jiemuxiensis have independent maternal lineages.Rana hanlunica and R.maoershanensis have essentially identical maternal lineages and they appear to represent the same species.The validity of the species R.longicrus,R.zhenhaiensis,and R.culaiensis and their genealogical relationships are not resolved and deserve further study.The genealogy indicates that sympatric cryptic lineages probably widely exist in the R.longicrus group.This highlights the importance of future more fine-scaled samplings and the inclusion of nuclear genes.Fang YAN Ke JIANG Hongman CHEN Ping FANG Jieqiong JIN Yi LI Shaoneng WANG Robert W.MURPHY Jing CHE Yaping ZHANG 2011Asian Herpetological Research2011,2,2:8
6Anorectal malignant melanomas: Retrospective experience with surgical management显示文摘AIM: To present the experience and outcomes of the surgical treatment for the patients with anorectal melanoma from the Cancer Hospital, Chinese Academy of Medical Sciences. METHODS: Medical records of the diagnosis, surgery, and follow-up of 56 patients with anorectal melanoma who underwent surgery between 1975 and 2008 were retrospectively reviewed. The factors predictive for the survival rate of these patients were identified using multivariate analysis. RESULTS: The 5-year survival rate of the 56 patients with anorectal melanoma was 20%, 36 patients underwent abdominoperineal resection (APR) and 20 patients underwent wide local excision (WLE). The rates of local recurrence of the APR and WLE groups were 16.13% (5/36) and 68.75% (13/20), (P = 0.001), and the median survival time was 22 mo and 21 mo, respectively (P = 0.481). Univariate survival analysis demonstrated that the number of tumor and the depth of invasion had significant effects on the survival (P < 0.05). Multivariate analysis showed that the number of tumor [P = 0.017, 95% confidence interval (CI) = 1.273-11.075] and the depth of invasion (P = 0.015, 95% CI = 1.249-7.591) were independent prognostic factors influencing the survival rate. 8Clinicopathological analysis of metaplastic meningioma: report of 15 cases in Huashan Hospital显示文摘Objective: Metaplastic meningioma is a rare subtype of benign meningiomas, classified as WHO grade I with well prognosis. Here we presented our experiences on 15 cases of metaplastic meningioma, to investigate the clinicopathological features, therapies and prognosis of these cases. Methods: 15 patients underwent surgical treatment for intracranial metaplastic meningioma between 2001 and 2010 at Neurosurgery Department of Huashan Hospital, Shanghai, China. The clinical data, radiological manifestation, treatment strategy, pathological findings and prognosis of all patients were analyzed retrospectively. Results: Among the 15 cases (10 males and 5 females), the age ranged from 22 to 74 years old (the mean age was 50.67-year old). The clinical manifestations include headache, dizziness, seizure attack, vision decrease, and weakness of bilateral lower limbs. All the patients received surgical treatment, combined with radiotherapy in some cases. In the follow-up period, recurrence occurred in 2 cases, of which 1 patient died of other system complications. Conclusions: Metaplastic meningiomas are characterized by focal or widespread mesenchymal differentiation with formation of bone, cartilage, fat, and xanthomatous tissue elements. Surgical removal is the optimal therapy, and the overall prognosis is well. 11Efficient LEC2 activation of OLEOSIN expression requires two neighboring RY elements on its promoter显示文摘As the main structural protein of oil body,OLEOSIN is highly expressed only during seed development. OLEOSIN promoter is a very useful tool for seed-specific gene engineering and seed bioreactor designing. The B3 domain transcription factor leafy cotyledon2 (LEC2) plays an important role in regulating seed development and seed-specific gene expression. Here,we first report how seed-specific B3 domain transcription factor leafy cotyledon2 (LEC2) efficiently activates OLEOSIN expression. The central promoter region of OLEOSIN,responsible for seed specificity and LEC2 activation,was determined by 5'-deletion analysis. Binding experiments in yeast cells and electrophoretic mobility shift assays showed that LEC2 specifically bound to two conserved RY elements in this region. In transient expression assays,mutation in either RY element dramatically reduced LEC2 activation of OLEOSIN promoter activity,while double mutation abolished it. Analysis of the distribution of RY elements in seed-specific genes activated by LEC2 also supported the idea that genes containing neighboring RY elements responded strongly to LEC2 activation. Therefore,we conclude that two neighboring RY elements are essential for efficient LEC2 activation of OLEOSIN expression. 13Diagnostic Value of Cerebrospinal Fluid T-SPOT.TB for Tuberculousis Meningitis in China显示文摘The aim of this study was to evaluate the diagnostic value of the cerebrospinal fluid(CSF) T‐SPOT.TB test for the diagnosis of TB meningitis(TBM). A retrospective analysis of 96 patients with manifested meningitis was conducted; T‐SPOT.TB test was performed for diagnosing TBM to determine the diagnostic sensitivity, specificity, positive predictive value(PPV), and negative predictive value(NPV). A receiver operating characteristic(ROC) curve was also drawn to assess the diagnostic accuracy. The sensitivity, specificity, PPV, and NPV of CSF T‐SPOT.TB test were 97.8%, 78.0%, 80.3%, and 97.5%, respectively, for 52 patients(54.2%) of the 96 enrolled patients. The area under the curve(AUC) was 0.910, and the sensitivities of CSF T‐SPOT.TB for patients with stages I, II, and III of TBM were 96.7%, 97.2%, and 98.9%, respectively. 14Glucose oxidase as a blocking agent-based signal amplification strategy for the fabrication of label-free amperometric immunosensors显示文摘An effective electrochemical signal amplification strategy based on enzyme membrane modification and redox probe immobilization was proposed to construct an amperometric immunosensor.L-cysteine@ferrocene functionalized chitosan,which possessed not only efficient redox-activity but also excellent film-forming ability,was coated on the bare glass carbon electrode. Moreover,the thiol groups(SH)in the ferrocenyl compound were used for gold nanoparticles immobilization via the strong bonding interaction,which could further be utilized for the immobilization of antibody biomolecules with well-retained bioactivities.Finally,glucose oxidase(GOD)as the enzyme membrane was employed to block the possible remaining active sites and avoid the nonspecific adsorption.With the excellent electrocatalytic properties of GOD towards glucose,the amplification of antigen-antibody interaction and the enhanced sensitivity could be achieved.Under the optimal conditions,the linear range of the proposed immunosensor for the determination of carcinoembryonic antigen(CEA)was from 0.05 to 100 ng/mL with a detection limit of 0.02 ng/mL(S/N=3).Moreover,the immunosensor exhibited good selectivity,stability and reproducibility, which provided a promising potential for clinical immunoassay.SONG ZhongJu YUAN Ruo CHAI YaQin CHE Xin LV Ping 2011Science China Chemistry2011,54,3:3

16In-situ transformation of Co(OH)_(2) into NH_(4)CoPO_(4)·H_(2)O on Co foil: 3D self-supported electrocatalyst with asymmetric local atomic and electronic structure for enhanced oxygen evolution reaction显示文摘Development of high efficient and stable water oxidation catalysts is essential for the realization of industrial water-splitting systems. Herein, a novel approach involving an in-situ transformation of Co(OH)2 nanosheets into NH4 CoPO4·H2 O nanoplates on Co foil is reported. As a 3 D self-supported oxygen revolution reaction(OER) electrocatalyst, the as-prepared NH4 CoPO4·H2 O/Co exhibits remarkable catalytic activity and exceptional stability. Specifically, it can deliver a current density of 10 m A cm^(-2) at a quite low overpotential of 254 m V with a small Tafel slope of 64.4 m V dec-1 in alkaline electrolyte. Through experimental study and theoretical analysis, the excellent OER performance can be attributed to enriched exposed active sites, favorable electron/proton transfer and mass transport, and its unique asymmetric local atomic and electronic structure. 18Discovery of a wild,genetically pure Chinese giant salamander creates new conservation opportunities显示文摘Effective conservation of threatened biota relies on accurate assessments and scientific guidance.As an unfortunate example,Chinese giant salamanders(Andrias,CGS)remain critically endangered in nature.Misguided conservation efforts,e.g.,commercial propagation and releasing of millions of likely non-indigenous or interspecific hybrids,have further compromised conservation initiatives.Limited information on wild populations of CGS poses a significant conservation challenge.Following 18-month long field monitoring,we now report the discovery of a wild population of CGS in a closed nature reserve in Jiangxi Province,China.Genomic assessments reveal its genetic distinctiveness and do not detect genetic admixture with other species.Based on morphological and molecular evidences,we describe this CGS as a new species Andrias jiangxiensis sp.nov.This is the only known species of CGS today with a genetically pure,reproducing,in situ population.This discovery emphasizes the important role that closed nature reserves play in protecting species,and the necessity of integrating long-term field monitoring and genetic assessments.It sets a new pathway for discovering and conserving endangered species,especially for those biotas that are similarly being extirpated by anthropogenic translocations and overexploitation.Jing Chai Chen-Qi Lu Mu-Rong Yi Nian-Hua Dai Xiao-Dong Weng Ming-Xiao Di Yong Peng Yong Tang Qing-Hua Shan Kai Wang Huan-Zhang Liu Hai-Peng Zhao Jie-Qiong Jin Ru-Jun Cao Ping Lu Lai-Chun Luo Robert W.Murphy Ya-Ping Zhang Jing Che 2022Zoological Research2022,43,3:2

20Overexpression of MdATG8i improves water use efficiency in transgenic apple by modulating photosynthesis, osmotic balance, and autophagic activity under moderate water deficit显示文摘Water deficit is one of the major limiting factors for apple(Malus domestica)production on the Loess Plateau,a major apple cultivation area in China.The identification of genes related to the regulation of water use efficiency(WUE)is a crucial aspect of crop breeding programs.As a conserved degradation and recycling mechanism in eukaryotes,autophagy has been reported to participate in various stress responses.However,the relationship between autophagy and WUE regulation has not been explored.We have shown that a crucial autophagy protein in apple,MdATG8i,plays a role in improving salt tolerance.Here,we explored its biological function in response to long-term moderate drought stress.The results showed that MdATG8i-overexpressing(MdATG8i-OE)apple plants exhibited higher WUE than wild-type(WT)plants under long-term moderate drought conditions.Plant WUE can be increased by improving photosynthetic efficiency.Osmoregulation plays a critical role in plant stress resistance and adaptation.Under long-term drought conditions,the photosynthetic capacity and accumulation of sugar and amino acids were higher in MdATG8i-OE plants than in WT plants.The increased photosynthetic capacity in the OE plants could be attributed to their ability to maintain optimal stomatal aperture,organized chloroplasts,and strong antioxidant activity.MdATG8i overexpression also promoted autophagic activity,which was likely related to the changes described above.In summary,our results demonstrate that MdATG8i-OE apple lines exhibited higher WUE than WT under long-term moderate drought conditions because they maintained robust photosynthesis,effective osmotic adjustment processes,and strong autophagic activity.Xin Jia Ke Mao Ping Wang Yu Wang Xumei Jia Liuqing Huo Xun Sun Runmin Che Xiaoqing Gong Fengwang Ma 2021Horticulture Research2021,8,1:1
