| 1 | Prehospital system delay in patients with ST-segment elevation myocardial infarction in Singapore显示文摘BACKGROUND: Timely reperfusion in ST-segment elevation myocardial infarction(STEMI)improves outcomes. System delay is that between first medical contact and reperfusion therapy,comprising prehospital and hospital components. This study aimed to characterize prehospital system delay in Singapore.METHODS: A retrospective chart review was performed for 462 consecutive STEMI patients presenting to a tertiary hospital from December 2006 to April 2008. Patients with cardiac arrest secondarily presented were excluded. For those who received emergency medical services(EMS),ambulance records were reviewed. Time intervals in the hospital were collected prospectively. The patients were divided into two equal groups of high/low prehospital system delay using visual binning technique.RESULTS: Of 462 patients, 76 received EMS and 52 of the 76 patients were analyzed. The median system delay was 125.5 minutes and the median prehospital system delay was 33.5minutes(interquartile range [IQR]=27.0, 42.0). Delay between call-received-by-ambulance and ambulance-dispatched was 2.48 minutes(IQR=1.47, 16.55); between ambulance-dispatch and arrival-at-patient-location was 8.07 minutes(IQR=1.30, 22.13); between arrival-at- and departurefrom-patient-location was 13.12 minutes(IQR=3.12, 32.2); and between leaving-patient-location to ED-registration was 9.90 minutes(IQR=1.62, 32.92). Comparing patients with prehospital system delay of less than 35.5 minutes versus more showed that the median delay between ambulancedispatch and arrival-at-patient-location was shorter(5.75 vs. 9.37 minutes, P<0.01). The median delay between arrival-at-patient-location and leaving-patient-location was also shorter(10.78 vs.14.37 minutes, P<0.01).CONCLUSION: Prehospital system delay in our patients was suboptimal. | 2 | Multiple haploids, triploids, and tetraploids found in modern-day “living fossil” Ginkgo biloba显示文摘Ginkgo biloba,the last extant representative of a lineage of Mesozoic gymnosperms,is one of the few seed plants with an exceptionally long(~300 Myr)evolutionary history free of genome-wide duplications(polyploidy).Despite this genome conservatism,we have recently found a viable spontaneous tetraploid Ginkgo sapling during routine screening of several plants,demonstrating that natural polyploidy is possible in Ginkgo.Here we provide a much wider flow cytometry survey of ploidy in some European Ginkgo collections,and own seedlings(>2200 individuals and~200 cultivars).We found a surprisingly high level of ploidy variation in modern-day Ginkgo and documented altogether 13 haploid,3 triploid,and 10 tetraploid Ginkgo plants or cultivars,most of them being morphologically distinct from common diploids.Haploids frequently produced polyploid(dihaploid)buds or branches.Tetraploids showed some genome size variation.The surveyed plants provide a unique resource for future Ginkgo research and breeding,and they might be used to accelerate the modern diversification of this nearly extinct plant lineage. | PetrŠmarda Lucie Horová Ondřej Knápek Heidi Dieck Martin Dieck Katarína Ražná Pavel Hrubík Laszlo Orlóci Laszlo Papp Kristýna Veselá Pavel Veselý Petr Bureš | 2018 | Horticulture Research2018,5,1: | 4 |
| 3 | In-memory computing with emerging nonvolatile memory devices显示文摘The von Neumann bottleneck and memory wall have posed fundamental limitations in latency and energy consumption of modern computers based on von Neumann architecture. In-memory computing represents a radical shift in the computer architecture that can address such problems by merging computing functions within the memory itself. In this article, we review the emerging nonvolatile memory devices,such as resistance-based and charge-based memory devices, that are explored for in-memory computing applications. We will provide an overview of the materials, mechanisms, and integration of these devices,and discuss the optimizations at the device and array levels that are required to better support in-memory computing. Recent progress in the application of in-memory computing in artificial neural networks, spiking neural networks, digital logic in memory as well as hardware security will also be discussed. | 4 | Humanized mice are susceptible to Salmonella typhi infection显示文摘Salmonella enterica serovar Typhi is a pathogen that only infects humans.Currently,there is no animal model for studying this pathogen.Recently,alymphoid RAG2^(-/-)/γc^(-/-) mice engrafted with human leukocytes,known as humanized mice,have been successfully utilized to develop experimental models for several human-specific viral infections,including HIV,human-like dengue fever and hepatitis C virus.Little is known about the usefulness and feasibility of the humanized mouse model for the study of human-specific bacterial pathogens,such as S.typhi.The aim of this study was to determine if Salmonella enterica serovar Typhi could establish productive infection in humanized mice.Here we report that intravenous inoculation of S.typhi into humanized mice,but not controls,established S.typhi infections.High bacterial loads were found in the liver,spleen,blood and bone marrow of mice reconstituted with human leukocytes,but not in the unreconstituted control mice.Importantly,S.typhi-infected humanized mice lost significant body weight,and some of the infected mice displayed neurological symptoms.Our data suggest,for the first time,that humanized mice are susceptible to S.typhi challenge and that this model can be utilized to study the pathogenesis of S.typhito develop novel therapeutic strategies. | M Firoz Mian Elisabeth A Pek Meghan J Chenoweth Ali A Ashkar | 2011 | Cellular & Molecular Immunology2011,8,1: | 2 |

| 15 | Heat shock protein 20 promotes sirtuin 1-dependent cell proliferation in induced pluripotent stem cells显示文摘BACKGROUND Heat shock proteins(HSPs)are molecular chaperones that protect cells against cellular stresses or injury.However,it has been increasingly recognized that they also play crucial roles in regulating fundamental cellular processes.HSP20 has been implicated in cell proliferation,but conflicting studies have shown that it can either promote or suppress proliferation.The underlying mechanisms by which HSP20 regulates cell proliferation and pluripotency remain unexplored.While the effect of HSP20 on cell proliferation has been recognized,its role in inducing pluripotency in human-induced pluripotent stem cells(iPSCs)has not been addressed.AIM To evaluate the efficacy of HSP20 overexpression in human iPSCs and evaluate the ability to promote cell proliferation.The purpose of this study was to investigate whether overexpression of HSP20 in iPSCs can increase pluripotency and regeneration.METHODS We used iPSCs,which retain their potential for cell proliferation.HSP20 overexpression effectively enhanced cell proliferation and pluripotency.Overexpression of HSP20 in iPSCs was characterized by immunocytochemistry staining and realtime polymerase chain reaction.We also used cell culture,cell counting,western blotting,and flow cytometry analyses to validate HSP20 overexpression and its mechanism.RESULTS This study demonstrated that overexpression of HSP20 can increase the pluripotency in iPSCs.Furthermore,by overexpressing HSP20 in iPSCs,we showed that HSP20 upregulated proliferation markers,induced pluripotent genes,and drove cell proliferation in a sirtuin 1(SIRT1)-dependent manner.These data have practical applications in the field of stem cell-based therapies where the mass expansion of cells is needed to generate large quantities of stem cell-derived cells for transplantation purposes.CONCLUSION We found that the overexpression of HSP20 enhanced the proliferation of iPSCs in a SIRT1-dependent manner.Herein,we established the distinct crosstalk between HSP20 and SIRT1 in regulating cell proliferation and pluripotency.Our study provides novel insights into the mechanisms controlling cell proliferation that can potentially be exploited to improve the expansion and pluripotency of human iPSCs for cell transplantation therapies.These results suggest that iPSCs overexpressing HSP20 exert regenerative and proliferative effects and may have the potential to improve clinical outcomes. | Mujib Ullah Nicole Pek Min Qian Gustavo Yannarelli Asma Akbar | 2021 | World Journal of Stem Cells2021,13,6: | 1 |
