5Efficacy and safety of Xinglouchengqi decoction for acute ischemic stroke with constipation:study protocol for a randomized con-trolled trial显示文摘OBJECTIVE: To investigate the efficacy and safety of Xinglouchengqi(XLCQ) decoction in treatment of acute ischemic stroke with constipation.METHODS: In this prospective, multicenter, assessor-blinded, randomized controlled trial, 360 eligible participants will be randomized to the XLCQ group or the control group. Participants in the XLCQ group will receive XLCQ decoction, while those in the control group will undergo clysis therapy using glycerin enemas or oral administration of lactulose solution. Both groups will undergo a treatment period of(5 ± 2) d and a 1-month follow-up.The primary outcome measure will be the Constipation Scale score. The secondary outcome measures will include scores on the National Institutes of Health Stroke Scale, the Traditional Chinese Medicine(TCM) Stroke Scale, the Diagnostic Scale for TCM Syndromes of Ischemic Stroke and TCM Scale for Syndrome of Phlegm-heat and Fu-organ Excess.Therapeutic mechanism outcomes and safety outcomes will also be assessed. Assessments will be conducted at baseline, at the end of the treatment period, and at the follow-up. 6Receptor tyrosine kinases positively regulate BACE activity and Amyloid-β production through enhancing BACE internalization显示文摘Amyloid-β (AP) peptide, the primary constituent of senile plaques in Alzheimer's disease (AD), is generated byβ-secretase- and γ-secretase-mediated sequential proteolysis of the amyloid precursor protein (APP). The aspartic pro-tease, β -site APP cleavage enzyme (BACE), has been identified as the main P-secretase in brain but the regulation of itsactivity is largely unclear. Here, we demonstrate that both BACE activity and subsequent Aβ production are enhancedafter stimulation of receptor tyrosine kinases (RTKs), such as the receptors for epidermal growth factor (EGF) and nervegrowth factor (NGF), in cultured cells as well as in mouse hippocampus. Furthermore, stimulation of RTKs also inducesBACE internalization into endosomes and Golgi apparatus. This enhancement of BACE activity and Aβ production uponRTK activation could be specifically inhibited by Src family kinase inhibitors and by depletion of endogenous c-Src withRNAi, and could be mimicked by over-expressed c-Src. Moreover, blockage of BACE internalization by a dominantnegative form of Rab5 also abolished the enhancement of BACE activity and Aβ production, indicating the requirementof BACE internalization for the enhanced activity. 7Cranial irradiation impairs intrinsic excitability and synaptic plasticity of hippocampal CA1 pyramidal neurons with implications for cognitive function显示文摘Radiation therapy is a standard treatment for head and neck tumors.However,patients often exhibit cognitive impairments following radiation therapy.Previous studies have revealed that hippocampal dysfunction,specifically abnormal hippocampal neurogenesis or neuroinflammation,plays a key role in radiation-induced cognitive impairment.However,the long-term effects of radiation with respect to the electrophysiological adaptation of hippocampal neurons remain poorly characterized.We found that mice exhibited cognitive impairment 3 months after undergoing 10 minutes of cranial irradiation at a dose rate of 3 Gy/min.Furthermore,we observed a remarkable reduction in spike firing and excitatory synaptic input,as well as greatly enhanced inhibitory inputs,in hippocampal CA1 pyramidal neurons.Corresponding to the electrophysiological adaptation,we found reduced expression of synaptic plasticity marker VGLUT1 and increased expression of VGAT.Furthermore,in irradiated mice,long-term potentiation in the hippocampus was weakened and GluR1 expression was inhibited.These findings suggest that radiation can impair intrinsic excitability and synaptic plasticity in hippocampal CA1 pyramidal neurons.Min-Yi Wu Wen-Jun Zou Pei Yu Yuhua Yang Shao-Jian Li Qiang Liu Jiatian Xie Si-Qi Chen Wei-Jye Lin Yamei Tang 2022Neural Regeneration Research2022,17,10:5

9Responsiveness and minimal clinically important difference of the Chinese version of the Low Vision Quality of Life Questionnaire after cataract surgery显示文摘AIM: To investigate the Chinese version of the Low Vision Quality of Life Questionnaire(CLVQOL) as an instrument for obtaining clinically important changes after cataract surgery.METHODS: Patients underwent cataract surgery in Shanghai General Hospital, Shanghai Jiao Tong University, who fit the inclusion criteria were recruited. Two CLVQOLs were administered, including a preoperative CLVQOL and a CLVQOL at the end of the 3 mo follow-up period, and were completed using face-to-face interviews or phone interviews conducted by trained investigators. The minimal clinically important difference(MCID) was calculated using an anchor-based method and a distribution method. In addition, the responsiveness of the questionnaire was measured.RESULTS: A total of 155 residents were enrolled. The average visual acuity(VA) preoperatively was 0.08(SD=0.05), and it increased to 0.47(SD=0.28) at the end of followup. Statistically significant positive changes in the CLVQOL scores indicated significant improvement of vision related quality of life after cataract surgery. With the larger value between the two results as the final value, the MCID values of the CLVQOL(scores of the four scales as well as the total score) were 8.94, 2.61, 4.34, 3.10 and 17.63, respectively. The CLVQOL has both good internal and external responsiveness.CONCLUSION: CLVQOL scores are appropriate instruments for obtaining clinically important changes after cataract surgery. 10Simulation study of energy resolution with changing pixel size for radon monitor based on Topmetal-Ⅱ^- TPC显示文摘In this paper, we study how pixel size influences energy resolution for a proposed pixelated detector—a high sensitivity, low cost, and real-time radon monitor based on a Topmetal-Ⅱ^- time projection chamber(TPC). This monitor was designed to improve spatial resolution for detecting radon alpha particles using Topmetal-Ⅱ^- sensors assembled by a 0.35 lm CMOS integrated circuit process.Owing to concerns that small pixel size might have the side effect of worsening energy resolution due to lower signalto-noise ratio, a Geant4-based simulation was used to investigate the dependence of energy resolution on pixel sizes ranging from 60 to 600 lm. A non-monotonic trend in this region shows the combined effect of pixel size and threshold on pixels, analyzed by introducing an empirical expression. Pixel noise contributes 50 keV full-width at half-maximum energy resolution for 400 lm pixel size at 1–4σ threshold that is comparable to the energy resolution caused by energy fluctuations in the TPC ionization process( ~20 keV). The total energy resolution after combining both factors is estimated to be 54 keV for a pixel size of 400 lm at 1–4σ threshold. 11Prevalence and risk factors of diabetic retinopathy in patients with type 2 diabetes in Shanghai显示文摘AIM:To investigate the prevalence of diabetic retinopathy(DR)in residents of Shanghai and analyze the risk factors of DR.METHODS:This study involved 7233 patients with diabetes in 2016.The demographic data of the participants were collected using a questionnaire survey.Physical examination,laboratory tests,and ophthalmological examinations were conducted.Two professional ophthalmologists diagnosed and graded DR by fundus examination and then combined the results with fundus images.The unconditional multivariate Logistic regression analysis was used to determine the risk factors.RESULTS:In total,6978 patients with type 2 diabetes in Shanghai with a mean age of 68.33±8.40 y were recruited,including 2975 males(42.6%)and 4003 females(57.4%).Overall,1184 patients were diagnosed with DR,with a prevalence rate of 16.97%.Regression analysis showed that duration of diabetes(OR 1.061,95%CI 1.049-1.073),high systolic blood pressure(SBP;OR 1.071,95%CI 1.037-1.106),increased glycosylated hemoglobin level(OR 1.234,95%CI 1.162-1.311),high blood glucose level(OR 1.061,95%CI 1.023-1.099),increased neutrophil-to-lymphocyte ratio(NLR;OR 1.132,95%CI 1.053-1.217)and mean platelet volume(MPV;OR 1.077,95%CI 1.016-1.142)were risk factors of DR.Conversely,hematocrit(HCT;OR 0.971,95%CI 0.954-0.988)and mean corpuscular volume(MCV;OR 0.980,95%CI 0.965-0.994)were protective factors.CONCLUSION:The prevalence rate of DR in Shanghai is 16.97%.The duration of diabetes,high SBP,increased glycosylated hemoglobin,NLR,and MPV were determined as risk factors of DR.Pei Zhang Wen-Wen Xue Xiao-Bo Huang Yi Xu Li-Na Lu Kai-Rong Zheng Hai-Dong Zou 2021International Journal of Ophthalmology(English edition)2021,14,7:2
12Experimental study of freak waves due to three-dimensional island terrain in random wave显示文摘An experimental study is carried out for waves passing an isolated reef terrain in a wave tank. A three-dimensional model of a representative and isolated reef terrain in the West Pacific is built. Random wave trains with various periods and wave heights are generated by a wave maker using the improved JONSWAP spectrum. It is observed that there are different kinds of generation processes and waveforms of freak waves. The freak wave factor Hm/Hs (where Hm is the maximum wave height of wave series, and Hs is significant wave height) is analyzed in detail, in terms of the skewness, kurtosis and water depth, as well as the relationship between freak wave height H& and skewness. The freak wave factor Hm/Hs is found to be in positive correlation with the kurtosis, while larger H[x tends to be related with bigger skewness. 19Raman and UV-Vis Spectroscopy Applied to the Analysis of Liver Tissues from Rats with Myocardial Ischemia Induced by Isoproterenol显示文摘The application of the laser Raman spectroscopic(LRS) technique for the analysis of liver tissues from rats with myocardial ischemia induced by isoproterenol(ISO) was described.Animal model of myocardial ischemia was established for rats induced by ISO.Rats were randomly divided into four groups as normal group and myocardial ischemia groups.We observed the successful myocardial ischemia model via serum enzymes levels and hematoxylin-eosin(HE) staining,and detected the liver tissue of the rats from normal group and liver tissue of the rats from myocardial ischemia groups via UV-Vis spectroscopy(UV-Vis) and LRS,and the changes of the absorbance spectra were compared in the above four different groups.The results show that ISO can induce rat myocardial ischemia successfully.The spectrum of normal liver tissue supernatant exhibits a strong absorption band at 968 nm,but no absorption band appears in the spectra of liver tissue supernatant solutions from the rats with myocardial ischemia induction after 2,12 and 72 h presented at 968 nm.LRS results show that Raman intensities of the precipitates suffered from ISO-treatment after 2,12 and 72 h were obviously increased compared with that of the precipitate of the liver tissue of the normal rats suffered from 0.9 g/L normal saline(NS) treatment.These results indicate that LRS and UV-Vis can be harmless,nondestructive,rapid and effective methods for analyzing different pathological specimens of liver tissue from myocardial ischemia rats.GAO Hai-cheng ZOU Ying-gang HUANG Yu-xin GAO Hai-mei CHEN Lei PEI Jin 2011Chemical Research in Chinese Universities2011,27,4:1
