5Three-dimensional Culture of Human Airway Epithelium in Matrigel for Evaluation of Human Rhinovirus C and Bocavirus Infections显示文摘Objective Newly identified human rhinovirus C(HRV-C) and human bocavirus(HBoV) cannot propagate in vitro in traditional cell culture models; thus obtaining knowledge about these viruses and developing related vaccines are difficult. Therefore, it is necessary to develop a novel platform for the propagation of these types of viruses. Methods A platform for culturing human airway epithelia in a three-dimensional(3 D) pattern using Matrigel as scaffold was developed. The features of 3 D culture were identified by immunochemical staining and transmission electron microscopy. Nucleic acid levels of HRV-C and HBoV in 3 D cells at designated time points were quantitated by real-time polymerase chain reaction(PCR). Levels of cytokines, whose secretion was induced by the viruses, were measured by ELISA. Results Properties of bronchial-like tissues, such as the expression of biomarkers CK5, ZO-1, and PCK, and the development of cilium-like protuberances indicative of the human respiration tract, were observed in 3 D-cultured human airway epithelial(HAE) cultures, but not in monolayer-cultured cells. Nucleic acid levels of HRV-C and HBoV and levels of virus-induced cytokines were also measured using the 3 D culture system. 6Taxonomy notes on Vandeae(Orchidaceae)from China:Five new species and two new records显示文摘Five new species(Gastrochilus yei,Gastrochilus minimus,Luisia simaoensis,Taeniophyllum xizangense,Tuberolabium subulatum)and two newly recorded species(Cleisostoma tricornutum,Luisia inconspicua)of Vandeae(Orchidaceae)from China are described and illustrated.Gastrochilus yei is similar to G.affinis and G.nepalensis,but differs from them by having an epichile not lobed,the apex of the hypochile not bilobed,and a tine on the apex of the leaf.Gastrochilus minimus is similar to G.acinacifolius,but can be distinguished from the latter by having a flabellate epichile that is densely hirsute on the adaxial surface and an inconspicuous central cushion;in addition,the hypochile of G.minimus has a keel that extends to the apex of the epichile.Taeniophyllum xizangense is similar to T.stella and T.radiatum,but it is distinguished from them by having much bigger flowers,inflorescences densely covered with short-bristly hairs,papillae on the external surface of sepals,and bigger triangular-ovate viscidium.Luisia simaoensis is similar to L magniflora and L ramosii,but can be easily distinguished from them by having lateral sepals longer than dorsal sepals and petals,lip with irregular and waved margins,and lip with bilobed apex.Luisia inconspicua is moved from Gastrochilus to Luisia based on phylogenetic analyses of plastid matK sequence data.Tuberolabium subulatum is similar to T.camosum,but it can be easily distinguished from the latter by having an inflorescence much shorter than the leaves,yellow sepals and petals,and many small papillae outside the lip lobes.Jian-Wu Li Ji-Dong Ya De-Ping Ye Cheng Liu Qiang Liu Rui Pan Zai-Xing He Bo Pan Jie Cai Dong-Liang Lin Xiao-Hua Jin 2021Plant Diversity2021,43,5:5
7Thioredoxin-Interacting Protein(TXNIP) Regulates Parkin/PINK1-mediated Mitophagy in Dopaminergic Neurons Under High-glucose Conditions: Implications for Molecular Links Between Parkinson’s Disease and Diabetes显示文摘Patients with diabetes mellitus have a higher risk of developing Parkinson’s disease(PD). However, the molecular links between PD and diabetes remain unclear.In this study, we investigated the roles of thioredoxininteracting protein(TXNIP) in Parkin/PINK1-mediated mitophagy in dopaminergic(DA) cells under high-glucose(HG) conditions. In streptozotocin-induced diabetic mice,TXNIP was upregulated and autophagy was inhibited in the midbrain, while the loss of DA neurons was accelerated by hyperglycemia. In cultured PC12 cells under HG, TXNIP expression was upregulated and the intracellular reactive oxygen species(ROS) levels increased, leading to cell death. Autophagic flux was further blocked and PINK1 expression was decreased under HG conditions. Parkin expression in the mitochondrial fraction and carbonyl cyanide 3-chlorophenylhydrazone(CCCP)-induced co-localization of COX IV(marker for mitochondria) and LAMP1(marker for lysosomes) were also significantly decreased by HG. Overexpression of TXNIP was sufficient to decrease the expression of both PINK1 and Parkin in PC12 cells, while knockdown of the expression of TXNIP by si RNA decreased intracellular ROS and attenuated cellular injury under HG. Moreover, inhibition of TXNIP improved the CCCP-induced co-localization of COX IV and LAMP1 in PC12 cells under HG. 10Radix Achyranthis Bidentatae improves learning and memory capabilities in ovariectomized rats显示文摘Kidney-tonifying recipe can reduce the accumulation of advanced glycation end products, prevent neuronal degeneration and improve cognitive functions in ovariectomized rats. Radix Achyranthis Bidentatae alcohol extracts may dose-dependently inhibit non-enzymatic saccharification in vitro. This study aimed to examine the effect of Radix Achyranthis Bidentatae on advanced glycation end products and on learning and memory capabilities in ovariectomized rats. Ovariectomized rats were treated with Radix Achyranthis Bidentatae alcohol extracts (containing 1.5 g/kg crude drug) or 0.1% aminoguanidine for 12 weeks and behavioral testing was performed with the Y-electrical maze. This test revealed that Radix Achyranthis Bidentatae and aminoguanidine could improve the learning and memory capabilities of ovariectomized rats. Results of competitive enzyme-linked immunosorbent assay showed that treatment with Radix Achyranthis Bidentatae or aminoguanidine reduced the accumulation of advanced glycation end products in the frontal cortex of ovariectomized rats, while increasing content in the blood and urine. Biochemical tests showed that treatment with Radix Achyranthis Bidentatae or aminoguanidine decreased superoxide dismutase activity in the serum and frontal cortex, and increased serum levels of glutathione peroxidase in ovariectomized rats. In addition, there was no apparent effect on malondialdehyde levels. These experimental findings indicate that Radix Achyranthis Bidentatae inhibits production of advanced glycation end products and its accumulation in brain tissue, and improves learning and memory capabilities in ovariectomized rats. 11Non-dormant Axillary Bud 1 regulates axillary bud outgrowth in sorghum显示文摘Tillering contributes to grain yield and plant architecture and therefore is an agronomically important trait in sorghum(Sorghum bicolor).Here,we identified and functionally characterized a mutant of the Nondormant Axillary Bud 1(NAB1)gene from an ethyl methanesulfonate-mutagenized sorghum population.The nab1 mutants have increased tillering and reduced plant height.Map-based cloning revealed that NAB1encodes a carotenoid-cleavage dioxygenase 7(CCD7)orthologous to rice(Oryza sativa)HIGH-TILLERING DWARF1/DWARF17 and Arabidopsis thaliana MORE AXILLARY BRANCHING 3.NAB1 is primarily expressed in axillary nodes and tiller bases and NAB1 localizes to chloroplasts.The nab1 mutation causes outgrowth of basal axillary buds;removing these non-dormant basal axillary buds restored the wild-type phenotype.The tillering of nab1 plants was completely suppressed by exogenous application of the synthetic strigolactone analog GR24.Moreover,the nab1plants had no detectable strigolactones and displayed stronger polar auxin transport than wild-type plants.Finally,RNA-seq showed that the expression of genes involved in multiple processes,including auxin-related genes,was significantly altered in nab1.These results suggest that NAB1 functions in strigolactone biosynthesis and the regulation of shoot branching via an interaction with auxin transport.Jun Chen Limin Zhang Mengjiao Zhu Lijie Han Ya Lv Yishan Liu Pan Li Haichun Jing Hongwei Cai 2018Journal of Integrative Plant Biology2018,60,10:3

13A new gain-of-function OsGS2/GRF4 allele generated by CRISPR/Cas9 genome editing increases rice grain size and yield显示文摘Grain size is one of the most important factors affecting rice grain quality and yield,and attracts great attention from molecular biologists and breeders.In this study,we engineered a CRISPR/Cas9 system targeting the miR396 recognition site of the rice GS2 gene,which encodes growth-regulating factor 4(OsGRF4)and regulates multiple agronomic traits including grain size,grain quality,nitrogen use efficiency,abiotic stress response,and seed shattering.In contrast to most previous genome editing efforts in which indel mutations were chosen to obtain null mutants,a mutant named GS2^(E) carrying an in-frame 6-bp deletion and 1-bp substitution within the miR396-targeted sequence was identified.GS2^(E) plants showed increased expression of GS2 in consistent with impaired repression by miR396.As expected,the gain-of-function GS2^(E) mutant exhibited multiple beneficial traits including increased grain size and yield and bigger grain length/width ratio.Thousand grain weight and grain yield per plant of GS2^(E) plants were increased by 23.5%and 10.4%,respectively.These improved traits were passed to hybrids in a semidominant way,suggesting that the new GS2^(E) allele has great potential in rice improvement.Taken together,we report new GS2 germplasm and describe a novel gene-editing strategy that can be widely employed to improve grain size and yield in rice.This trait-improvement strategy could be applied to other genes containing miRNA target sites,in particular the conserved miR396-GRF/GIF module that governs plant growth,development and environmental response.Wenshu Wang Weipeng Wang Yanlin Pan Chao Tan Hongjing Li Ya Chen Xingdan Liu Jing Wei Nian Xu Yu Han Han Gu Rongjian Ye Qi Ding Chonglie Ma 2022The Crop Journal2022,10,4:3

17Association of Catalase Genotype with Oxidative Stress in the Predication of Colorectal Cancer:Modification by Epidemiological Factors显示文摘Objective This paper aims to assess the interaction between common variations in catalase(CAT) polymorphic gene and environmental factors for antioxidant defense enzyme in modulating individual susceptibility to colorectal cancer(CRC).Methods A case-control study with 880 colorectal cancer cases and 848 controls was conducted to investigate whether variations in the catalase(CAT) gene,one of the genes involved in scavenging oxidative stress,influenced susceptibility to CRC.Results The interaction between life style and genotypes as well as with their effects on colorectal cancer was deduced from the present study.Significant difference(P=0.01) was identified in the distribution of CAT genotype between the colorectal cancer cases and the controls.The CRC cases had significantly lower mean activity than the controls(P<0.01).Correlation analyses revealed statistically significant correlations between CAT activity and CAT genotype(P<0.01).Conclusion The risk of CRC was associated with smoking,low vegetable consumption,high pork and poultry consumptions,and low or high BMI.This is the first study reporting an association of polymorphism CAT-21A>T with colorectal cancer.Low CAT activity was associated with an increased risk of CRC;however,no evidence was found to support an association between CAT-21A>T polymorphism and CRC risk.CHANG Dong HU Zhang Liang ZHANG Lin ZHAO Ya Shuang MENG Qing Hui GUAN Qing Bai ZHOU Jin PAN Hong Zhi 2012Biomedical and Environmental Sciences2012,25,2:2

19Metal-organic frameworks as functional materials for implantable flexible biochemical sensors显示文摘Metal-organic frameworks(MOFs)exhibit attractive properties such as highly accessible surface area,large porosity,tunable pore size,and built-in redox-active metal sites.They may serve as excellent candidates to construct implantable flexible devices for biochemical sensing due to their high thermal and solution stability.However,MOFs-based sensors have only been mostly reported for in-vitro chemical sensing,their use in implantable chemical sensing and combination with flexible electronics to achieve excellent mechanical compatibility with tissues and organs has rarely been summarized.This paper systematically reviews the biochemical sensors based on MOFs and discusses the feasibility to achieve implantable biochemical sensing through MOFs-based flexible electronics.The properties of MOFs and underlying mechanisms have been intrcxluced,followed by a summarization of different biochemical sensing applications.Strategies to integrate MOFs with flexible devices have been supplied from the standpoints of matching mechanics and compatible fabrication processes.Issues that should be addressed in developing flexible MOFs sensors and potential solutions have also been provided,followed by the perspective for future applications of flexible MOFs sensors.This paper may serve as a reference to offer potential guidelines for the development of flexible MOFs-based biochemical sensors that may benefit future applications in personal healthcare,disease diagnosis and treatment,and fundamental study of various biological processes.Ya Li Wei Ling Xinyu Liu Xue Shang Pan Zhou Zhaorun Chen Hang Xu Xian Huang 2021Nano Research2021,14,9:2
