Targeting the PI3K/Akt signaling pathway in gastric carcinoma: A reality for personalized medicine?显示文摘Frequent activation of phosphatidylinositol-3 kinases(PI3K)/Akt/m TOR signaling pathway in gastric cancer(GC) is gaining immense popularity with identification of mutations and/or amplifications of PIK3 CA gene or loss of function of PTEN,a tumor suppressor protein,to name a few; both playing a crucial role in regulating this pathway. These aberrations result in dysregulation of this pathway eventually leading to gastric oncogenesis, hence, there is a need for targeted therapy for more effective anticancer treatment. Several inhibitors are currently in either preclinical or clinical stages for treatment of solid tumors like GC. With so many inhibitors under development, further studies on predictive biomarkers are needed to measure the specificity of any therapeutic intervention. Herein,we review the common dysregulation of PI3K/Akt/m TOR pathway in GC and the various types of single or dual pathway inhibitors under development that might have a superior role in GC treatment. We also summarize the recent developments in identification of predictive biomarkers and propose use of predictive biomarkers to facilitate more personalized cancer therapy with effective PI3K/Akt/m TOR pathway inhibition. | Shikha Satendra Singh Wei Ney Yap Frank Arfuso Shreya Kar Chao Wang Wanpei Cai Arunasalam M Dharmarajan Gautam Sethi Alan Prem Kumar | 2015 | World Journal of Gastroenterology2015,21,43: | 36
Biochar increases soil microbial biomass with changes in extra-and intracellular enzyme activities:a global meta-analysis显示文摘Biochar application to soil has been proposed as a potential management strategy to enhance soil carbon(C)sequestration, reduce greenhouse gas emission, improve soil quality, and increase crop productivity. The effects of biochar on soil microbial and enzyme activities are integrally linked to the potential of biochar in achieving these benefits. We conducted a global meta-analysis to assess the effects of biochar on soil microbial biomass C and nitrogen(N)and the activities of 12 enzymes, and identify key factors affecting those soil microbial properties using 964 data points from 72 papers. We found that biochar effects on enzyme activities vary widely with soil type, biochar property and the type of enzyme studied.Biochar significantly increased microbial biomass C(MBC)and urease, alkaline phosphatase and dehydrogenase activities by 21.7%,23.1%,25.4%and 19.8%,respectively,with no significant negative effects on any of the enzymes analyzed in this study.Biochar application was more effective in increasing MBC and enzyme activities in soils with low pH(<6.5),TC(<20 g kg^(−1)),TN(<2 g kg^(−1)),and a fine texture(including clay, clay loam and silt clay).Biochars produced at pyrolysis temperature of 350-550℃ with a high pH(>10)and low C/N ratio(<50)increased MBC and urease and dehydrogenase activities.Biochar increased MBC and N-acquisition enzyme activities in the field but not in lab incubation experiments.Urease was increased in short-term studies(within 100 days of biochar application)while alkaline phosphatase was increased in long-term studies that span more than 1 year. The increase in MBC and activities of some soil enzymes in response to biochar application with no negative effects on any hydrolytic and oxidative enzymes illustrate its potential to enhance soil quality particularly in the degraded soils with low nutrient availability and fertility due to limited soil microbial and enzymatic activities. This study also shows that biochars can be designed to achieve specific properties for enhancing microbial and enzymatic activities for specific soils. | Prem Pokharel Zilong Ma Scott X.Chang | 2020 | Biochar2020,2,1: | 18

Efficacy of small-volume simethicone given at least 30 min before gastroscopy显示文摘AIM To evaluate the efficacy of 5 m L simethicone solution in decreasing gastric foam if given at least 30 min before gastroscopy.METHODS This was a randomized, placebo controlled, endoscopist blinded study performed at Changi General Hospital. Patients were at least 21 years old, had no prior sur-gical resection of the upper gastrointestinal tract, and scheduled for elective diagnostic gastroscopies. The primary outcome was the total mucosal visibility score(TMVS) which was evaluated using Mc Nally score. The sample size was calculated to be 24 per group(SD 2.4, 80% power, P < 0.05, 2-sample t test). RESULTS Fifty-four patients were randomised to receive either simethicone [1 m L liquid simethicone(100 mg) in 5 m L of water] or placebo(5 m L of water) at least 30 min before their gastroscopy. Six accredited consultants conductedthe gastroscopy, and the interobserver agreement of scoring TMVS was good with a Kappa statistic of 0.73. The simethicone group had significantly better mean TMVS compared to placebo(5.78 ± SD 1.65 vs 8.89 ± SD 1.97, P < 0.001). The improvement was statistically significant for the duodenum and the gastric antrum, angularis, body, and fundus. Percent 51.9 of patients in the simethicone group had a TMVS of 4(no bubbles at all) to 5(only 1 area with minimal bubbles), while in the placebo group 3.7% of patients had TMVS of 4 or 5. The number needed to treat was 2.1 to avoid a TMVS of 6 and more. The simethicone group also had a significantly shorter procedure time with less volume of additional flushes required during gastroscopy to clear away obscuring gastric foam.CONCLUSION With a premedication time of at least 30 min, 5 m L simethicone can significantly decrease gastric foam, decrease the volume of additional flushes, and shorten gastroscopy time. | Mingjun Song Andrew Boon Eu Kwek Ngai Moh Law Jeannie Peng Lan Ong Jessica Yi-Lyn Tan Prem Harichander Thurairajah Daphne Shih Wen Ang Tiing Leong Ang | 2016 | World Journal of Gastrointestinal Pharmacology and Therapeutics2016,7,4: | 10
Proximate Composition,Mineral Content and Fatty Acids Analyses of Aromatic and Non-Aromatic Indian Rice显示文摘Awareness on nutritive value and health benefits of rice is of vital importance in order to increase the consumption of rice in daily diet of the human beings. In this study, a total of six aromatic and two non-aromatic rice accessions grown in India were analysed for their nutritional quality attributes including proximate composition, mineral contents and fatty acids. Data with three replications were used to measure Pearson's simple correlation co-efficient in order to establish the relationship among various nutritional quality attributes. The result on proximate composition showed that Govind Bhog had the highest moisture(13.57%) and fat(0.92%) content, which signifies its tasty attribute. Badshah Bhog exhibited the highest fibre content(0.85%), carbohydrate content(82.70%) and food energy(365.23 k Cal per 100 g). Among the minerals, the higher Ca(98.75 mg/kg), Zn(17.00 mg/kg) and Fe(31.50 mg/kg) were in Gopal Bhog, whereas the highest Na(68.85 mg/kg) was in Badshah Bhog, the highest K(500.00 mg/kg) was in Swetganga, Khushboo and Sarbati. The highest contents of unsaturated fatty acids viz. oleic acid(49.14%), linoleic acid(46.99%) and linolenic acid(1.27%) were found in Sarbati, whereas the highest content of saturated fatty acids viz. myristic acid(4.60%) and palmitic acid(31.91%) were found in Govind Bhog and stearic acid(6.47%) in Todal. The identified aromatic rice accessions Gopal Bhog, Govind Bhog and Badshah Bhog and non-aromatic rice accession Sarbati were found nutritionally superior among all eight tested accessions. The nutritional quality oriented attributes in this study were competent with recognized prominent aromatic and non-aromatic rice accessions as an index of their nutritional worth and recommend to farmers and consumers which may be graded as export quality rice with good unique nutritional values in international market. | Deepak Kumar VERMA Prem Prakash SRIVASTAV | 2017 | Rice science2017,24,1: | 8
Prem N. | 7 | Recent advances in catalytic combustion of AP-based composite solid propellants显示文摘Composite solid propellants(CSPs) have widely been used as main energy source for propelling the rockets in both space and military applications. Internal ballistic parameters of rockets like characteristic exhaust velocity, specific impulse, thrust, burning rate etc., are measured to assess and control the performance of rocket motors. The burn rate of solid propellants has been considered as most vital parameter for design of solid rocket motors to meet specific mission requirements. The burning rate of solid propellants can be tailored by using different constituents, extent of oxidizer loading and its particle size and more commonly by incorporating suitable combustion catalysts. Various metal oxides(MOs),complexes, metal powders and metal alloys have shown positive catalytic behaviour during the combustion of CSPs. These are usually solid-state catalysts that play multiple roles in combustion of CSPs such as reduction in activation energy, enhancement of rate of reaction, modification of sequences in reaction-phase, influence on condensed-phase combustion and participation in combustion process in gas-phase reactions. The application of nanoscale catalysts in CSPs has increased considerably in recent past due to their superior catalytic properties as compared to their bulk-sized counterparts. A large surface-to-volume ratio and quantum size effect of nanocatalysts are considered to be plausible reasons for improving the combustion characteristics of propellants. Several efforts have been made to produce nanoscale combustion catalysts for advanced propellant formulations to improve their energetics. The work done so far is largely scattered. In this review, an effort has been made to introduce various combustion catalysts having at least a metallic entity. Recent developments of nanoscale combustion catalysts with their specific merits are discussed. The combustion chemistry of a typical CSP is briefly discussed for providing a better understanding on role of combustion catalysts in burning rate enhancement. Available information on different types of combustion nanocatalysts is also presented with critical comments. | Narendra Yadav Prem Kumar Srivastava Mohan Varma | 2021 | Defence Technology(防务技术)2021,17,3: | 8
Tuning the particle size and morphology of high energetic material nanocrystals显示文摘Morphology controlled synthesis of nanoparticles of powerful high energetic compounds(HECs) such as l,3,5-trinitro-l,3,5-triazinane(RDX) and 1,3,5,7-tetranitro-l,3,5,7-tetrazocane(HMX) were achieved by a simple solvent—antisolvent interaction(SAI) method at 70 ℃.The effects of different solvents on particle size and morphology of the prepared nano-HECs were studied systematically.Particle size and morphology of the nano-HECs was characterized using field emission scanning electron microscopy(FE-SEM) imaging.X-ray diffraction(XRD) and Fourier transform infrared(FTIR) spectroscopy studies revealed that RDX and HMX were precipitated in their most stable polymorphic forms,i.e.a and P,respectively.Thermogravimetric analysis coupled with differential scanning calorimetry(TGA-DSC) studies showed that the thermal response of the nanoparticles was similar to the respective raw-HECs.HEC nanoparticles with spherical and rod shaped morphology were observed under different solvent conditions. The mean particle size also varied considerably with the use of different solvents. | Raj KUMAR Prem Felix SIRIL Pramod SONI | 2015 | Defence Technology(防务技术)2015,11,4: | 7
A Comparative Study on the Role of Cytokinins in Caryopsis Development in the Maize miniature1 Seed Mutant and Its Wild Type显示文摘We report here on a comparative developmental profile of plant hormone cytokinins in relation to cell size, cell number and endoreduplication in developing maize caryopsis of a cell wall invertase-deficient miniature1(mn1) seed mutant and its wild type,Mn1,genotype.Both genotypes showed extremely high levels of total cytokinins during the very early stages of development, followed by a marked and genotype specific reduction. While the decrease of cytokinins in Mn1 was associated with their deactivation by 9-glucosylation,the absolute and the relative part of active cytokinin forms was higher in the mutant.During the exponential growth phase of endosperm between 6 d after pollination and 9 d after pollination, the mean cell doubling time, the absolute growth rate and the level of endoreduplication were similar in the two genotypes. However, the entire duration of growth was longer in Mn1 compared with mn1,resulting in a significantly higher cell number in the Mn1 endosperm. These data correlate with the previously reported peak levels of the Mn1-encoded cell wall invertase-2(INCW2) at 12 d after pollination in the Mn1 endosperm.A model showing possible crosstalk among cytokinins, cell cycle and cell wall invertase as causal to increased cell number and sink strength of the Mn1 developing endosperm is discussed. | Tomaz Rijavec Maja Kova Ale Kladnik Prem S.Chourey Marina Dermastia | 2009 | Journal of Integrative Plant Biology2009,51,9: | 7
A review on assessment and treatment of the trunk in stroke A need or luxury显示文摘Trunk function has been identified as an important early predictor of functional outcome after stroke and the same deteriorates on both contralateral and ipsilateral sides of the body following stroke. The primary contribution of the trunk muscles is to allow the body to remain upright, adjust weight shifts, and control movements against constant pull of gravity and is considered central key point of the body.Proximal stability of the trunk is a pre-requisite for distal limb mobility, balance, gait and functional activities and its positive correlation in hemiplegia has been demonstrated in a cross-sectional study. Both isokinetic and handheld dynamometer muscle strength testing demonstrated the weakness of bilateral trunk flexors, extensors and rotator muscles in both acute and chronic hemiplegic patients. This was confirmed by electromyography analysis which identified poor bilateral trunk muscles activity in patients with stroke.Trunk impairment scale is sensitive to evaluate the selective muscle control of upper and lower trunk, and it has been reported that lateral flexion of the trunk is easier than rotation of the trunk and the clinical observation concurs to the difficulty in lower trunk rotation of stroke patients. However, trunk exercises given early after stroke could produce enhanced balance performance post-stroke.This review attempts to report the evidence supporting the involvement of the trunk and its influence on balance and functional performance in post-stroke hemiplegia. | Suruliraj Karthikbabu Mahabala Chakrapani Sailakshmi Ganeshan Kedambadi C Rakshith Syed Nafeez Venkatesan Prem | 2012 | Neural Regeneration Research2012,7,25: | 5
Assessment of tumor markers CA 19-9,CEA,CA 125,and CA 242 for the early diagnosis and prognosis prediction of gallbladder cancer显示文摘BACKGROUND Gallbladder cancer(GBC)is one of the leading and aggressive cancers in this region of India. It is very difficult to diagnose in the early stage,as it lacks typical early signs and symptoms;thus, the diagnosis is often in the advanced stage, which ultimately leads to a poor 5-year survival outcome.Tumor markers including carbohydrate antigen 19-9(CA 19-9),carcinoembryonic antigen(CEA),CA 125,CA 242,and alpha fetoprotein are used as indicators in the diagnosis and prognosis of GBC.AIM To compare tumor marker levels between GBC and benign GB diseases(GBDs)and to assess the combined use of tumor markers to increase the diagnostic accuracy for GBC.METHODS Patients of either sex aged≥18 years, with suspected GBC(GB polyp, irregular thick GB wall,GB mass, porcelain GB)on the basis of radiological imaging were included in this study.GB wall thickness using ultrasonography and tumor markers CEA,CA 125,CA 19-9,and CA 242 in all patients were recorded. All cases after surgical intervention were divided into two groups,GBC and benign GBD,according to histopathological examination findings. The cases were followed up and clinical findings, radiological findings, and levels of tumor markers were assessed.RESULTS A total of 200 patients were included in this study,of whom 80 patients had GBC and 120 patients had benign GBD.The median(interquartile range)age was 52.0(41.0-60.0)years and the majority of patients(132,66.0%)were women.Tumor markers including CA 19-9,CA 125,CEA,and CA 242 were significantly elevated in patients with GBC(P<0.001).There was a significant reduction in tumor markers at 3 and 6 mo from baseline(P<0.001).The mean survival of patients with normal and elevated levels of tumor markers CA 125,CA 19-9,and CEA was comparable;however lymph node metastasis and CA 242 expression level were independent prognostic factors.CONCLUSION Serum levels of tumor markers including CA 19-9,CA 125,CEA,and CA 242 were significantly associated with GBC.However,no significant association was observed between the presence of elevated levels of any tumor marker with respect to survival.Tumor marker assessment during follow-up may represent a treatment response. | Seema Rani Sinha Prem Prakash Rakesh Kumar Singh Dinesh Kumar Sinha | 2022 | World Journal of Gastrointestinal Surgery2022,14,11: | 5
Garcinol sensitizes human head and neck carcinoma to cisplatin in a xenograft mouse model despite downregulation of proliferative biomarkers显示文摘OBJECTIVE Platinum compounds such as cisplatin and carboplatin are frequently used as the first-line chemotherapy for the treatment of the head and neck squamous cell carcinoma(HNSCC).In the present study,we investigated whether garcinol, apolyisoprenylated benzophenone can chemosensitize HNSCC to cisplatin.METHODS The effect of garcinol and cisplatin on HNSCC was assessed by MTT,Western blotting, real time PCR,FACS,immunohistochemistry,DNA binding assay and xenograft mouse model.RESULTS We found that garcinol inhibited the viability of a panel of diverse HNSCC cell lines, enhanced the apoptotic effect of cisplatin, suppressed constitutive as well as cisplatin-induced NF-κB activation, and downregulated the expression of various oncogenic gene products(cyclin D1,Bcl-2,survivin and VEGF).In vivo study showed that administration of garcinol alone(0.5 mg·kg-1,ip five times/week)significantly suppressed the growth of the tumor, and this effect was further increased by cisplatin. Both the markers of proliferation index(Ki-67)and microvessel density(CD31)were downregulated in tumor tissues by the combination of cisplatin and garcinol. The pharmacokinetic results of garcinol indicated that good systemic exposure was achievable after ip administration of garcinol at 0.5and 2mg·kg-1 with mean peak concentration(cmax)of 1825.4 and 6635.7nmol·L-1 in the mouse serum, respectively.CONCLUSION Overall, our results suggest that garcinol can indeed potentiate the effects of cisplatin by negative regulation of various inflammatory and proliferative biomarkers. | Feng Li Muthu K. | 18 | Influence of A-site cation disorder on structural and magnetocaloric properties of Nd_(0.7–x)La_xSr_(0.3)MnO_3(x=0.0, 0.1, 0.2 & 0.3)显示文摘In the present research, the influence of A-site ionic size disorder on structural, magnetic and magnetocaloric properties of Nd0.7–xLaxSr0.3MnO3(for x=0.0, 0.1, 0.2 & 0.3) compounds were investigated. These compounds were synthesized by solid state reaction route and crystallized in orthorhombic structure with Pnma space group. An average ionic radius increased with La-substitution which influenced the physical properties such as the magnetic entropy varying from 4.78 J/kg·K(for x=0.0) to 3.55 J/kg·K(for x=0.3) and the maximum magnetization at 5 K changing from 3.92 μB(for x=0.0) to 3.59 μB(for x=0.3). The change in maximum magnetic entropy decreased with increase of A-site average ionic radius. The decrease in magnetic entropy indicated the suppression of lattice disorder that influenced the spin-lattice coupling. The ferromagnetic transition temperature shifted toward room temperature with La substitution. As increased, the phase transition changed from first order(for x=0.0) to second order(for x≥ 0.1) and the refrigeration capacity increased to a significant level. The modest change in magnetic entropy over a wide range of transition temperature might be useful for future active magnetic refrigeration technology. | Sudharshan Vadnala Prem Pal Saket Asthana | 2015 | Journal of Rare Earths2015,33,10: | 3

Modeling Spatial–Temporal Dynamics of Urban Residential Fire Risk Using a Markov Chain Technique显示文摘This article applies a Markov chain method to compute the probability of residential fire occurrence based on past fire history. Fitted with the fire incidence data gathered over a period of 10 years in Melbourne, Australia, the spatially-integrated fire risk model predicts the likely occurrence of fire incidents using space and time as key model parameters. The mapped probabilities of fire occurrence across Melbourne show a city-centric spatial pattern where inner-city areas are relatively more vulnerable to a fire than outer suburbia. Fire risk reduces in a neighborhood when there is at least one fire in the last1 month. The results show that the time threshold of reduced fire risk after the fire occurrence is about 2 months.Fire risk increases when there is no fire in the last 1 month within the third-order neighborhood(within 5 km). A fire that occurs within this distance range, however, has no significant effect on reducing fire risk level within the neighborhood. The spatial–temporal dependencies of fire risk provide new empirical evidence useful for fire agencies to effectively plan and implement geo-targeted fire risk interventions and education programs to mitigate potential fire risk in areas where and when they are most needed. | Rifan Ardianto Prem Chhetri | 2019 | International Journal of Disaster Risk Science2019,10,1: | 2
