2Intranasal delivery of nanostructured lipid carriers,solid lipid nanoparticles and nanoemulsions:A current overview of in vivo studies显示文摘The management of the central nervous system(CNS)disorders is challenging,due to the need of drugs to cross the blood-brain barrier(BBB)and reach the brain.Among the various strategies that have been studied to circumvent this challenge,the use of the intranasal route to transport drugs from the nose directly to the brain has been showing promising results.In addition,the encapsulation of the drugs in lipid-based nanocarriers,such as solid lipid nanoparticles(SLNs),nanostructured lipid carriers(NLCs)or nanoemulsions(NEs),can improve nose-to-brain transport by increasing the bioavailability and site-specifc delivery.This review provides the state-of-the-art of in vivo studies with lipid-based nanocarriers(SLNs,NLCs and NEs)for nose-to-brain delivery.Based on the literature available from the past two years,we present an insight into the different mechanisms that drugs can follow to reach the brain after intranasal administration.The results of pharmacokinetic and pharmacodynamics studies are reported and a critical analysis of the differences between the anatomy of the nasal cavity of the different animal species used in in vivo studies is carried out.Although the exact mechanism of drug transport from the nose to the brain is not fully understood and its effectiveness in humans is unclear,it appears that the intranasal route together with the use of NLCs,SLNs or NEs is advantageous for targeting drugs to the brain.These systems have been shown to be more effective for nose-to-brain delivery than other routes or formulations with non-encapsulated drugs,so they are expected to be approved by regulatory authorities in the coming years.Cláudia Pina Costa Joao Nuno Moreira JoséManuel Sousa Lobo Ana Catarina Silva 2021Acta Pharmaceutica Sinica B2021,11,4:5
3Prevalence and dietetic management of mild gastrointestinal disorders in milk-fed infants显示文摘AIM: To assess the prevalence of mild gastrointestinal disorders in milk-fed infants in paediatric practice, and to evaluate the effectiveness and satisfaction with dietetic treatment.METHODS: A cross-sectional epidemiological study was first carried out. A total of 285 paediatricians included 3487 children seen during a period of one week. In a second phase an observational, prospective and multicentre study was conducted and 2069 milk-fed infants with mild gastrointestinal disorders (colic, constipation, regurgitation and diarrhoea) were included. There was a baseline visit (start of treatment) and a final visit four weeks later. The effectiveness of the various Novalac formulas, as well as the satisfaction of the parents/tutors and paediatricians with the dietetic treatment were assessed at the final visit.RESULTS: The prevalence of mild gastrointestinal disorders was 27.8% of all paediatrician consultations (9.2%, 7.8%, 6.1% and 4.6% in relation to colic, constipation, regurgitation and diarrhoea, respectively). The several Novalac adapted milk formulas resolved 88.4% of the mild gastrointestinal disorders. Depending on the type of disorder, differences in response rate were observed. The highest effectiveness was recorded with respect to diarrhoea (92.6%), followed by constipation (91.6%), colic (87.6%) and regurgitation (81%). Overall, 91% of the paediatricians and 88.8% of the parents/tutors were satisfied or very satisfied with the Novalac adapted milk formulas.CONCLUSION: Mild gastrointestinal disorders show a high prevalence in paediatric practice. 5Inventory and action plan for PFOS and related substances in Suriname as basis for Stockholm Convention implementation显示文摘Within the framework of the Stockholm Convention implementation in Suriname,the first inventory of perfluorooctane sulfonic acid(PFOS)has been developed.The major stocks of PFOS are firefighting foams with minor stocks in pesticides(ant baits)and possibly carpets.The inventory of firefighting foams assessed the import,current stocks and major use sites and recent releases.Considerable amount of PFOS and potentially PFOS containing firefighting foams are stored at the military base,the oil company and small firefighting departments throughout the country.The total amount of stored PFAS firefighting foam in Suriname is estimated to be at least 275.6 tonnes,where the major stockpile of 206 tonnes is at the oil storage facilities.For some of these foams PFOS content is confirmed and for others it is not clear if the foam contains PFOS or other per-or polyfluorinated alkylated substances(PFAS).The upper estimate of PFOS content is 4133 kg in these foams.Additionally,PFOS or other PFAS has also been discovered in fire extinguishers in institutions.The total amount of PFOS related substance EtFOSA imported with the pesticide sulfluramid was 75 kg.The imports were banned in 2014 and is controlled.A considerable amount of synthetic carpets are in stores even produced before 2003 where main use of PFOS from 3 M took place possibly containing PFOS related substances.A compilation of potentially PFOS contaminated sites has been made.Potentially contaminated sites result from firefighting foam use,dumpsites and sulfluramid use.The airport where firefighting foam was applied is above a major drinking water reservoir,where an urgent assessment is needed.Due to the lack of analytical capacity for PFOS and precursors,drinking water,firefighting foam or impacted carpets,could not been analysed.An action plan for the control of PFOS and other PFAS has been developed considering the synergy of the Stockholm Convention and the Strategic Approach to International Chemicals Management(SAICM)addressing this emerging pollution issue for developing countries.Victorine Pinas Carmen Van Dijk Roland Weber 2020Emerging Contaminants2020,6,1:3

7Platelet aggregation but not activation and degranulation during the acute post-ischemic reperfusion phase in livers with no underlying disease显示文摘Background:Platelets and P-selectin(CD62P)play an unequivocal role in the pathology of hepatic ischemia/reperfusion(I/R)injury.Inhibition or knock-out of P-selectin or immunodepletion of platelets results in amelioration of post-ischemic inflammation,reduced hepatocellular damage,and improved survival.However,P-selectin expression on platelets and endothelial cells,which concurs with platelet activation,has never been clearly demonstrated in I/R-subjected livers.Aims:To determine whether platelets become activated and degranulate in the acute phase of liver I/R and whether the platelets interact with neutrophils.Methods:Hepatic I/R was induced in male C57BL/6J mice(N=12)using 37.5-min ischemia time.Platelets,endothelial cells,and neutrophils were fluorescently labeled by systemic administration of non-blocking antibodies.Cell kinetics were monitored by intravital spinning disk confocal microscopy during 90 min of reperfusion.Image analysis and quantification was performed with dedicated software.Results:Platelets adhered to sinusoids more extensively in post-ischemic livers compared to livers not subjected to I/R and formed aggregates,which occurred directly after ischemia.Platelets and endothelial cells did not express P-selectin in post-ischemic livers.There was no interaction between platelets and neutrophils.Conclusions:Platelets aggregate but do not become activated and do not degranulate in post-ischemic livers.There is no platelet-neutrophil interplay during the early reperfusion phase in a moderate model of hepatic I/R injury.The mechanisms underlying the biological effects of platelets and P-selectin in this setting warrant further investigation.Relevance for patients:I/R in surgical liver patients may compromise outcome due to post-ischemic oxidative stress and sterile inflammation.Both processes are mediated in part by platelets.Understanding platelet function during I/R is key to developing effective interventions for I/R injury and improving clinical outcomes.Rowan F.van Golen Katarzyna M.Stevens Pina Colarusso Hartmut Jaeschke Michal Heger 2015Journal of Clinical & Translational Research2015,1,2:2

10Indirect printing of hierarchical patient-specific scaffolds for meniscus tissue engineering显示文摘The complex meniscus tissue plays a critical role in the knee. The high susceptibility to injury has led to an intense pursuit for better tissue engineering regenerative strategies, where scaffolds play a major role. In this study, indirect printed hierarchical multilayered sca ffolds composed by a silk fibroin (SF) upper layer and an 80/20 (w/w) ratio of SF/ionic-doped β-tricalcium phosphate (TCP) bottom layer were developed. Furthermore, a comparative analysis between two types of sca ffolds pro- duced using di fferent SF concentrations, i.e., 8% (w/v) (Hi8) and 16% (w/v) (Hi16) was performed. In terms of architecture and morphology, the produced sca ffolds presented homogeneous porosity in both layers and no di fferences were observed when comparing both sca ffolds. A decrease in terms of mechanical performance of the sca ffolds was observed when SF concentration decreased from 16 to 8% (w/v). Hi16 revealed a static compressive modulus of 0.66 ± 0.05 MPa and dynamical mechanical properties ranging from 2.17 ± 0.25 to 3.19 ± 0.38 MPa. By its turn, Hi8 presented a compressive modulus of 0.27 ± 0.08 MPa and dynamical mechanical properties ranging from 1.03 ± 0.08 MPa to 1.56 ± 0.13 MPa. In vitro bioactivity studies showed formation of apatite crystals onto the surface of Hi8 and Hi16 bottom layers. Human meniscus cells (hMCs) and human primary osteoblasts were cultured separately onto the top layer (SF8 and SF16) and bottom layer (SF8/TCP and SF16/TCP) of the hierarchical sca ffolds Hi8 and Hi16, respectively. Both cell types showed good adhesion and proliferation as denoted by the live/dead staining, Alamar Blue assay and DNA quanti fication analysis. Subcutaneous implantation in mice revealed weak in flammation and sca ffold’s integrity.
