2CD5 expression promotes IL-10 production through activation of the MAPK/Erk pathway and upregulation of TRPC1 channels in B lymphocytes显示文摘CD5 is constitutively expressed on T cells and a subset of mature normal and leukemic B cells in patients with chronic lymphocytic leukemia(CLL).Important functional properties are associated with CD5 expression in B cells,including signal transducer and activator of transcription 3 activation,IL-10 production and the promotion of B-lymphocyte survival and transformation.However,the pathway(s)by which CD5 influences the biology of B cells and its dependence on B-cell receptor(BCR)co-signaling remain unknown.In this study,we show that CD5 expression activates a number of important signaling pathways,including Erk1/2,leading to IL-10 production through a novel pathway independent of BCR engagement.This pathway is dependent on extracellular calcium(Ca2+)entry facilitated by upregulation of the transient receptor potential channel 1(TRPC1)protein.We also show that Erk1/2 activation in a subgroup of CLL patients is associated with TRPC1 overexpression.In this subgroup of CLL patients,small inhibitory RNA(siRNA)for CD5 reduces TRPC1 expression.Furthermore,siRNAs for CD5 or for TRPC1 inhibit IL-10 production.These findings provide new insights into the role of CD5 in B-cell biology in health and disease and could pave the way for new treatment strategies for patients with B-CLL.Soizic Garaud Taher E Taher Marjolaine Debant Miguel Burgos Sarra Melayah Christian Berthou Kaushal Parikh Jacques-Olivier Pers Damien Luque-Paz Gilles Chiocchia Maikel Peppelenbosch David A Isenberg Pierre Youinou Olivier Mignen Yves Renaudineau Rizgar A Mageed 2018Cellular & Molecular Immunology2018,15,2:2
3Rapid phenotypic evolution with shallow genomic differentiation during early stages of high elevation adaptation in Eurasian Tree Sparrows显示文摘Known as the‘third polar region’,the Qinghai-Tibet Plateau represents one of the harshest highland environments in the world and yet a number of organisms thrive there.Previous studies of birds,animals and humans have focused on well-differentiated populations in later stages of phenotypic divergence.The adaptive processes during the initial phase of highland adaptation remain poorly understood.We studied a human commensal,the Eurasian Tree Sparrow,which has followed human agriculture to the Qinghai-Tibet Plateau.Despite strong phenotypic differentiation at multiple levels,in particular in muscle-related phenotypes,highland and lowland populations show shallow genomic divergence and the colonization event occurred within the past few thousand years.In a one-month acclimation experiment investigating phenotypic plasticity,we exposed adult lowland tree sparrows to a hypoxic environment and did not observe muscle changes.Through population genetic analyses,we identified a signature of polygenic adaptation,whereby shifts in allele frequencies are spread across multiple loci,many of which are associated with muscle-related processes.Our results reveal a case of positive selection in which polygenic adaptation appears to drive rapid phenotypic evolution,shedding light on early stages of adaptive evolution to a novel environment.Yanhua Qu Chunhai Chen Ying Xiong Huishang She Yong E Zhang Yalin Cheng Shane DuBay Dongming Li Per G P Ericson Yan Hao Hongyuan Wang Hongfeng Zhao Gang Song Hailin Zhang Ting Yang Chi Zhang Liping Liang Tianyu Wu Jinyang Zhao Qiang Gao Weiwei Zhai Fumin Lei 2020National Science Review2020,7,1:2
4Pit-and trench-forming osteoclasts:a distinction that matters显示文摘Osteoclasts(OCs)seeded on bone slices either drill round pits or dig long trenches.Whereas pits correspond to intermittent resorption,trenches correspond to continuous and faster resorption and require a distinct assembly of the resorption apparatus.It is unknown whether the distinction between pits and trenches has any biological relevance.Using OCs prepared from different blood donors,we found that female OCs achieved increased resorption mainly through pit formation,whereas male OCs did so through trench formation.Trench formation went along with high collagenolytic activity and high cathepsin K(CatK)expression,thereby allowing deeper demineralization.A specific CatK inhibitor abrogated the generation of trenches,while still allowing the generation of pits.OCs obtained from bone marrow were more prone to generate trenches than those obtained from blood.Scanning electron microscopy of bone surfaces eroded in vivo showed trenches and pits of similar size as those made by OCs in culture.We conclude that the distinction between trench-and pit-forming OCs is relevant to the differences among OCs from different skeletal sites,different individuals,including gender,and results from differences in colIagenolytic power.This indicates a biological relevance and highlights the importance of discriminating between pits and trenches when assessing resorption.Ditte MH Merrild Dinisha C Pirapaharan Christina M Andreasen Per Kjxrsgaard-Andersen Anas MJ Mùller Ming Ding Jean-Marie Delaissé Kent Sùe 2015Bone Research2015,3,4:2
