1Phospholipase C Delta 3 inhibits apoptosis and promotes proliferation,migration,and invasion of thyroid cancer cells via Hippo pathway显示文摘In recent decades,the incidence of thyroid cancer(TC)has rapidly increased,leading us to explore the complex underlying mechanisms.We identified the gene Phospholipase C Delta 3(PLCD3)as a potential oncogene in TC by conducting the whole transcriptome sequencing.Our study is to understand the oncogenic role of PLCD3 in TC.We verified the overexpression of PLCD3 in TC from The Cancer Genome Atlas,Gene Expression Omnibus databases,and a locally validated cohort.Clinical correlation analysis showed that PLCD3 expression was related to histological type,T stage,lymph node metastasis(LNM),and disease stage.The high expression of PLCD3 could be a distinguishing factor for TC and its LNM.The biological function was examined using small interfering RNA-transfected TC cell lines.Silenced PLCD3 could inhibit colony formation,migration,and invasion ability and promote apoptosis of TC cell lines.PLCD3 silencing reversed the epithelial-mesenchymal transition but induced the apoptotic progress.Further exploration revealed that PLCD3 might be associated with critical genes of the Hippo pathway.The expressions of RHOA,YAP1/TAZ,and their downstream targets were decreased significantly when PLCD3 was down-regulated.YAP1 overexpression rescued the tumor-suppressive effect caused by PLCD3 silencing.This study demonstrates that PLCD3 is an oncogene that supports tumorigenesis and progression in TC,and PLCD3 may be a potential target gene for TC treatment.Lizhi Lin Jialiang Wen Bangyi Lin Hao Chen Adheesh Bhandari Yufeng Qi Danni Zheng Ouchen Wang 2021Acta Biochimica et Biophysica Sinica2021,53,4:4
2Downregulating integrin subunit alpha 7(ITGA7)promotes proliferation,invasion,and migration of papillary thyroid carcinoma cells through regulating epithelial-to-mesenchymal transition显示文摘Thyroid cancer is one of the common malignancies of the endocrine system and the number of thyroid cancer cases is increasing constantly.Significant work has focused on the molecular mechanisms of thyroid cancer,but many mechanisms remain undiscovered.In this study,we employed a comprehensive analysis of whole-transcriptome resequencing derived from paired papillary thyroid cancer(PTC)and normal thyroid tissues.We performed a massive parallel whole-transcriptome resequencing of matched PTC and normal thyroid tissues in 19 patients and found that integrin subunit alpha 7(ITGA7)was downregulated in thyroid tumor tissues,but the function of ITGA7 in this cancer is still unclear.We also discovered that ITGA7 gene in thyroid cancer tissues was downregulated compared to paired adjacent non-tumor tissues by real-time quantitative polymerase chain reaction.After transfection with small interfering RNA to knock down ITGA7 the abilities of colony formation,proliferation,migration,and invasion were enhanced in PTC cell lines(TPC1 and KTC-1).Meanwhile,ITGA7 knockdown decreased apoptotic cell death in thyroid cells but promoted the expressions of N-cadherin and vimentin and decreased E-cadherin expression by epithelial-to-mesenchymal transition,which may induce invasion and migration.In conclusion,these results indicated that ITGA7 is involved in the progress of PTC and might act as a tumor suppressor gene.Yaoyao Guan Adheesh Bhandari Erjie Xia Lingguo Kong Xiaohua Zhang Ouchen Wang 2020Acta Biochimica et Biophysica Sinica2020,52,2:2

12AB073.Correlations between control of saccadic eye movements and performance in other cognitive tasks in younger adults,older adults and patients with Parkinson’s disease显示文摘Background:Cognitive control is defined as the ability to act flexibly in the environment by either behaving automatically or inhibiting said automatic behaviour and it can be measured using an interleaved pro/anti-saccade task.Decline in cognitive control has been attributed to normal aging and neurological illnesses such as Parkinson’s disease(PD)as well as decline in other cognitive abilities.This parallel might highlight the role played by cognitive control in information processing and working memory.However,little is known about the relationship between cognitive control and other cognitive processes such as visual memory,decision making,and visual search.We thus propose to correlate the incidence of impaired cognitive control with deficits in visual memory,decision making and visual search in three groups:younger adults,older adults and patients with idiopathic PD.Methods:Seventy-one participants,namely 34 adults(M=22.75,SD=3.8),22 older adults(M=67.4,SD=8.3),and 20 PD patients(M=65.59,SD=8.2)performed four tasks:interleaved pro/anti-saccade,visual memory,decision making,and serial and pop-out visual search.Results:Results show that within each group,anti-saccade error rate(ER)were significantly and negatively correlated with visual memory ER(ryounger=−0.378,P=0.036;rolder=−0.440,Polder=0.046;rPD=−0.609,P=0.016).On the other hand,correct decision-making reaction times(RT)were significantly correlated with anti-saccade ER,and RTs only in older adults(rER=0.529,P=0.014;rRT=0.512,P=0.018)and PD patients(rER=0.727,P=0.012;rRT=0.769,P=0.001).For visual search,PD patients showed a significant relationship between RTs for correct pro-saccades and pop-out(r=0.665,P=0.007),and serial(r=0.641,P=0.010)search RTs.Furthermore,there was a significant correlation between MoCA scores and anti-saccade RTs(r=−0.559,P=0.030)and ER(r=−0.562,P=0.029)in PD patients.Taken together,these results support the hypothesis of PD patients’reliance on bottom-up processes as top-down processes decline.For younger adults,there was a significant correlation between serial search performance and both anti-saccade ER(r=0.488,P=0.005),and correct pro-saccade ER(r=0.413,P=0.021).In older adults,this relationship was absent,but anti-saccade ER significantly correlated with pop-out search times(r=0.473,P=0.030).Conclusions:We found significant relationships between cognitive tasks and cognitive control as measured through the interleaved pro/anti-saccade task across and within participant groups,providing evidence of the appropriateness of the use of the interleaved pro/anti-saccade task as a measure of overall cognitive control.Julie Ouerfelli-Ethier Basma Elsaeid Julie Des Groseilliers Lydia Ouchene Douglas P.Munoz Gunnar Blohm Aarlenne Z.Khan 2018Annals of Eye Science2018,,1:0
13Phosphorus transformation under the influence of aluminum,organic carbon,and dissolved oxygen at the water-sediment interface:A simulative study显示文摘The effects of sediment aluminum(Al),organic carbon(OC),and dissolved oxygen(DO)on phosphorus(P)transformation,at the water-sediment interface of a eutrophic constructed lake,were investigated via a series of simulative experiments.The above three factors had various influences on dissolved P concentration,water pH,water and surface sediment appearance,and P fractions.Additions of Al had the greatest effect on suppressing P release,and the water p H remained alkaline in the water-sediment system under various OC and DO conditions.No dissolution of the added Al was detected.31P-N M R characterization suggested that OC addition did not promote biological P uptake to polyphosphates under oxic conditions.The simulation result on the added phytate indicated the absence of phytate in the original lake sediment.As compared to the reported natural lakes and wetland,the water-sediment system of the constructed lake responded differently to some simulative conditions.Since Al,OC,and DO can be controlled with engineering methods,the results of this study provide insights for the practical site restorations.Ouchen Cai Yuanxiao Xiong Haijun Yang Jinyong Liu Hui Wang 2020Frontiers of Environmental Science & Engineering2020,14,3:0
