1Relationship between T-lymphocyte cytokine levels and sero-response to hepatitis B vaccines显示文摘AIM: To investigate the cellular defects by analyzing the (Th1/Th2) cytokine levels in vaccine responders and non-responders. METHODS: Peripheral blood mononuclear cell (PBMC) from responders and non-responders were stimulated with or with out recombinant HBsAg or PHA. Broad spectrum of cytokines viz (Th1) IFN-γ, IL-2, TNF-α, IL-12 and (Th2) IL-10, IL-4 were measured after in vitro stimulation with recombinant HBsAg and were compared with respective antibody titers. RESULTS: A significant decrease (P = 0.001) in Th1 and Th2 cytokines namely, IL-2, INF-γ, TNF-α and IL-10in non-responders was observed. The level of IL-4 was not significant between the three groups. Furthermore, despite a strong Th1 and Th2 cytokine response, the level of IL-12 was elevated in high-responders compared to other groups (P = 0.001) and demonstrated a positive correlation with anti-HBs titers and Th1 cytokine response. 2Gastric cancer risk in relation to tobacco use and alcohol drinking in Kerala, India-Karunagappally cohort study显示文摘AIM: To assess the risk of gastric cancer(GC) in relation to tobacco use and alcohol drinking in the Karunagappally cohort in Kerala, South India.METHODS: This study examined the association of tobacco use and alcohol drinking with GC incidence among 65553 men aged 30-84 in the Karunagappally cohort. During the period from 1990-2009, 116 GC cases in the cohort were identified as incident cancers. These cases were identified from the populationbased cancer registry. Information regarding risk factors such as socioeconomic factors and tobacco and alcohol habits of cohort members were collected from the database of the baseline survey conducted during 1990-1997. The relative risks(RRs) and the corresponding 95% confidence intervals(95%CIs) fortobacco use were obtained from Poisson regression analysis of grouped survival data, considering age, follow-up period, occupation and education.RESULTS: Bidi smoking was associated with GC risk(P = 0.042). The RR comparing current versus never smokers was 1.6(95%CI: 1.0-2.5). GC risk was associated with the number of bidis smoked daily(P = 0.012) and with the duration of bidi smoking(P = 0.036). Those who started bidi smoking at younger ages were at an elevated GC risk; the RRs for those starting bidi smoking under the age of 18 and ages 18-22 were 2.0(95%CI: 1.0-3.9) and 1.8(95%CI: 1.1-2.9), respectively, when their risks were compared with lifetime non-smokers of bidis. Bidi smoking increased the risk of GC among never cigarette smokers more evidently(RR = 2.2; 95%CI: 1.3-4.0). GC risk increased with the cumulative amount of bidi smoking, which was calculated as the number of bidis smoked per day x years of smoking(bidi-year; P = 0.017). Cigarette smoking, tobacco chewing or alcohol drinking was not significantly associated with GC risk. 3Antioxidant and antimutagenic activities of bark extract of Terminalia arjuna显示文摘Objective:The alcoholic extract of stem bark of Terminalia arjuna(ALTA) was screened for antioxidant and antimutagenic(anticlastogenic) activity.Methods:Antioxidant property was determined by l,l,Diphynyl,2-Picryl hydrazyl(DPPH) assay,super oxide radical scavenging activity,lipid peroxidation assay and total polyphenolic content was determined by Folin-Ciocalteau’s reagent.Antimutagenic activity was evaluated using micronucleus test in mice. Results:The ALTA has shown potent antioxidant activity with EC50 of 2.491±0.160,50.110±0.150 & 71.000±0.250 in DPPH assay,superoxide radical scavenging activity and lipid peroxidation assay, which is comparable with ascorbic acid with EC50 of 2.471±0.140,40.500±0.390 and 63.000±0.360 respectively.In micronucleus test,ALTA(100 & 200 mg/kg,p.o.) showed significant reduction in percentage of micronucleus in both polychromatic erythrocytes(PCE) and normochromatie erythrocytes(NCE) and also shown significant reduction in P/N ratio.Conclusions:These results suggested that ALTA possess significant antioxidant and antimutagenic activity.Gollapalle Lakshminarayana shastry Viswanatha Santosh Kumar Vaidya Ramesh C Nandakumar Krishnadas Srinath Rangappa 2010Asian Pacific Journal of Tropical Medicine2010,3,12:9

6Coinfection of hepatitis B and hepatitis C virus in HIV-infectedpatients in south India显示文摘AIM: To screen for the co-infection of hepatitis B (HBV) and hepatitis C virus (HCV) in human immunodeficiency virus (HIV) infected patients in southern India. METHODS: Five hundred consecutive HIV infected patients were screened for Hepatitis B Virus (HBsAg and HBV-DNA) and Hepatitis C virus (anti-HCV and HCV-RNA) using commercially available ELISA kits; HBsAg, HBeAg/ anti-HBe (Biorad laboratories, USA) and anti-HCV (Murex Diagnostics, UK). The HBV-DNA PCR was performed to detect the surface antigen region (pre S-S). HCV-RNA was detected by RT-PCR for the detection of the constant 5' putative non-coding region of HCV. RESULTS: HBV co-infection was detected in 45/500 (9%) patients and HCV co-infection in 11/500 (2.2%) subjects. Among the 45 co-infected patients only 40 patients could be studied, where the detection rates of HBe was 55% (22/40), antiHBe was 45% (18/40) and HBV-DNA was 56% (23/40). Among 11 HCV co-infected subjects, 6 (54.5%) were anti-HCV and HCV RNA positive, while 3 (27.2%) were positive for anti-HCV alone and 2 (18%) were positive for HCV RNA alone. CONCLUSION: Since the principal routes for HIV transmission are similar to that followed by the hepatotropic viruses, as a consequence, infections with HBV and HCV are expected in HIV infected patients. 7Comparison of three different recombinant hepatitis B vaccines:GeneVac-B,Engerix B and Shanvac B in high risk infants born to HBsAg positive mothers in India显示文摘AIM: To evaluate a low cost Indian recombinant hepatitis B vaccine GeneVac-B for its immunogenicity and safety in comparison to Engerix B and Shanvac B vaccine in high risk newborn infants born to hepatitis B surface antigen (HBsAg) positive mothers. METHODS: A total of 158 infants were enrolled in the study. Fifty eight infants were enrolled in the GeneVac-B group while 50 each were included for Engerix B and Shanvac B groups. A three-dose regimen of vaccination; at birth (within 24 h of birth), 1st mo and 6 mo. were adopted with 10 μg dosage administered uniformly in all the three groups. Clinical and immunological parameters were assessed for safety and immunogenicity of the vaccines, in all the enrolled infants. RESULTS: Successful follow up until seven months of age was achieved in 83/ (48/58) for GeneVac-B, 76/ (38/50) and 64/ (32/50) for Engerix B and Shanvac B groups respectively. 100/ seroconversion and seroprotection was achieved in all the three groups of infants. The geometric mean titers of anti-HBs one month after the completion of three dose of vaccination were 90.5, 80.9 and 72.5 mIU/mL in GeneVac-B, Engerix B and Shanvac B vaccine group respectively. Furthermore the level of anti-HBs increases with age ofbabies who were born to HBsAg positive mothers. The GMT values of anti-HBs were 226.7, 193.9 and 173.6 mIU/mL respectively in GeneVac-B, Engerix B and Shanvac B groups one year after the completion of the three doses of vaccine. No systemic reactions were reported in infants during the entire vaccination process of GeneVac-B and the other two vaccines. Clinical safety parameters remained within the normal limits throughout the study period.CONCLUSION: The study concludes that there is no significant difference between the three recombinant hepatitis B vaccines. 9Oxidative stress-induced mitochondrial dysfunction in a normal colon epithelial cell line显示文摘AIM To determine how a normal human colon cell line reacts to microbial challenge as a way to study oxidative stress-induced responses associated with inflammatory bowel disease.METHODS Normal human colon epithelial cells(ATCC?CRL.1790?)were stimulated with either heat killed E.coli or heat killed murine cecal contents(HKC)and examined for several relevant biomarkers associated with inflammation and oxidative stress including cytokine production,mitochondrial autophagy and oxidant status.TNFα,IL-1βand IL-8 protein concentrations were measured within the supernatants.Fluorescent microscopy was performed to quantify the production of reactive oxygen species(ROS)using an oxidation responsive fluorogenic probe.Mitochondrial morphology and mitochondrial membrane potential was assessed by dual staining using COXIV antibody and a dye concentrating in active mitochondria.Mitochondrial ROS scavenger was used to determine the source of ROS in stimulated cells.Autophagy was detected by staining for the presence of autophagic vesicles.Positive controls for autophagy and ROS/RNS experiments were treated with rapamycin and chloroquine.Mitochondrial morphology,ROS production and autophagy microscopy experiments were analyzed using a custom acquisition and analysis microscopy software(Image J).RESULTS Exposing CRL.1790 cells to microbial challenge stimulated cells to produce several relevant biomarkers associated with inflammation and oxidative stress.Heat killed cecal contents treatment induced a 10-12fold increase in IL-8 production by CRL.1790 cells compared to unstimulated controls at 6 and 12 h(P<0.001).Heat killed E.coli stimulation resulted in a4-5 fold increase in IL-8 compared to the unstimulated control cells at each time point(P<0.001).Both heat killed E.coli and HKC stimulated robust ROS production at 6(P<0.001),and 12 h(P<0.01).Mitochondrial morphologic abnormalities were detected at 6 and12 h based on reduced mitochondrial circularity and decreased mitochondrial membrane potential,P<0.01.Microbial stimulation also induced significant autophagy at 6 and 12 h,P<0.01.Lastly,blocking mitochondrial ROS generation using mitochondrial specific ROS scavenger reversed microbial challenge induced mitochondrial morphologic abnormalities and autophagy.CONCLUSION The findings from this study suggest that CRL.1790cells may be a useful alternative to other colon cancer cell lines in studying the mechanisms of oxidative stress events associated with intestinal inflammatory disorders.Nandakumar Packiriswamy Kari F Coulson Susan J Holcombe Lorraine M Sordillo 2017World Journal of Gastroenterology2017,23,19:3
10Small molecule SMU-CX24 targeting toll-like receptor 3 counteracts inflammation:A novel approach to atherosclerosis therapy显示文摘Toll-like receptor 3(TLR3),as an important pattern recognition receptor(PRR),dominates the innate and adaptive immunity regulating many acute and chronic inflammatory diseases.Atherosclerosis is proved as an inflammatory disease,and inflammatory events involved in the entire process of initiation and deterioration.However,the contribution of TLR3 to atherosclerosis remains unclear.Herein,we identified the clinical relevance of TLR3 upregulation and disease processes in human atherosclerosis.Besides,activation of TLR3 also directly led to significant expression of atherogenic chemokines and adhesion molecules.Conversely,silencing TLR3 inhibited the uptake of oxLDL by macrophages and significantly reduced foam cell formation.Given the aberrance in TLR3 functions on atherosclerosis progression,we hypothesized that TLR3 could serve as novel target for clinical atherosclerosis therapy.Therefore,we developed the novel ellipticine derivative SMU-CX24,which specifically inhibited TLR3(IC_(50)=18.87±2.21 nmol/L).In vivo,atherosclerotic burden was alleviated in Western diet fed ApoE^(−/−)mice in response to SMU-CX24 treatment,accompanying notable reductions in TLR3 expression and inflammation infiltration within atherosclerotic lesion.Thus,for the first time,we revealed that pharmacological downregulation of TLR3 with specific inhibitor regenerated inflammatory environment to counteract atherosclerosis progression,thereby proposing a new strategy and probe for atherosclerosis therapy.Xiaohong Cen Baoqu Wang Yuqing Liang Yanlin Chen Yu Xiao Shaohua Du Kutty Selva Nandakumar Hang Yin Shuwen Liu Kui Cheng 2022Acta Pharmaceutica Sinica B2022,12,9:2
11Graphene quantum dots rescue protein dysregulation of pancreatic β-cells exposed to human islet amyloid polypeptide显示文摘The amyloid aggregation of peptides and proteins is a hallmark of neurological disorders and type 2 diabetes.Human islet amyloid polypeptide(IAPP),co-secreted with insulin by pancreaticβ-cells,plays dual roles in both glycemic control and the pathology of type 2 diabetes.While IAPP can activate the NLRP3 inflammasome and modulate cellular autophagy,apoptosis and extracellular matrix metabolism,no data is available concerning intracellular protein expression upon exposure to the polypeptide.More surprisingly,how intracellular protein expression is modulated by nanoparticle inhibitors of protein aggregation remains entirely unknown.In this study,we first examined the changing proteomes ofβTC6,a pancreaticβ-cell line,upon exposure to monomeric,oligomeric and fibrillar IAPP,and detailed cellular protein expression rescued by graphene quantum dots(GQDs),an IAPP inhibitor.We found that 29 proteins were significantly dysregulated by the IAPP species,while majority of these proteins were nucleotide-binding proteins.Collectively,our liquid chromatography tandem-mass spectrometry,fluorescence quenching,helium ion microscopy,cytotoxicity and discreet molecular dynamics simulations data revealed a remarkable capacity of GQDs in regulating aberrant protein expression through H-bonding and hydrophobic interactions,pointing to nanomedicine as a new frontier against human amyloid diseases.Ava Faridi Yunxiang Sun Monika Mortimer Ritchlynn R.Aranha Aparna Nandakumar Yuhuan Li Ibrahim Javed Aleksandr Kakinen Qingqing Fan Anthony W.Purcell Thomas P.Davis Feng Ding Pouya Faridi Pu Chun Ke 2019Nano Research2019,12,11:2

13高强度炎症反应诱导关节软骨特异性瓜氨酸化抗原表达显示文摘目的探讨关节软骨蛋白瓜氨酸表位的表达情况,以及瓜氨酸化表位对抗瓜氨酸蛋白抗体(ACPAs)的免疫应答的影响。方法体外诱导实验:实验组加不同浓度梯度的脂多糖、热灭活细菌(大肠杆菌、金色葡萄球菌)或二型胶原(CII)特异性单克隆抗体等不同刺激物诱导体外培养关节软骨蛋白瓜氨酸化,等量PBS作为对照组,利用特异性结合瓜氨酸化表位的单克隆抗体ACC4(IgG1)进行免疫组织化学染色,ACC1(IgG2a)作为阳性对照,Hy2.15(IgG1)和L243(IgG2a)分别作为同型阴性对照,显微镜下观察瓜氨酸化蛋白表达;体内诱导实验:SD大鼠右膝关节内注射不同浓度脂多糖作为实验组,左膝关节内注射等量PBS为对照组,3 d后取膝关节组织,制成冰冻切片,利用上述抗体进行免疫组化染色,显微镜下观察瓜氨酸化蛋白表达;建立不同种系的胶原诱导性关节炎(CIA)小鼠模型,评价其疾病发生率和严重程度,同时收集小鼠血清与高滴度抗瓜氨酸化抗体的类风湿性关节炎患者血清,采用液相芯片技术检测小鼠及类风湿患者血清中抗瓜氨酸化蛋白抗体水平,并用免疫组织化学检测关节炎小鼠关节软骨瓜氨酸化蛋白的表达水平。结果特异性结合瓜氨酸化CII表位的ACC4抗体未能结合至经不同诱导的关节软骨组织;与健康对照组相比,ACC4抗体和含高滴度的抗瓜氨酸化蛋白抗体的类风湿性关节炎患者血清中的抗体均能结合至CIA小鼠关节炎症关节软骨周围的滑膜组织;成功构建不同种系小鼠类风湿性关节炎模型,其发病率和疾病严重程度无统计学差异(P>0.05);液相芯片技术分析结果显示,小鼠血清中的抗瓜氨酸化抗体低表达或几乎不表达,但抗CII三螺旋结构多肽的抗体反应性较强。结论轻度急性炎症反应不足以引起关节软骨蛋白瓜氨酸化,其特异性抗原表位的表达需要高强度的炎症反应。二型胶原诱导的小鼠关节炎模型中炎症关节软骨蛋白可表达瓜氨酸化抗原表位,但瓜氨酸化抗原表位的表达不足以引起小鼠体内抗瓜氨酸化抗体的免疫应答,其免疫应答需要更多的刺激信号来诱导。覃桂城 林晓吟 梁培彬 李艳鹏 周春 Kutty Selva Nandakumar Rikard Holmdahl 2020南方医科大学学报2020,40,8:2
