| 1 | Spatial cloud computing: how can the geospatial sciences use and help shape cloud computing?显示文摘The geospatial sciences face grand information technology(IT)challenges in the twenty-first century:data intensity,computing intensity,concurrent access intensity and spatiotemporal intensity.These challenges require the readiness of a computing infrastructure that can:(1)better support discovery,access and utilization of data and data processing so as to relieve scientists and engineers of IT tasks and focus on scientific discoveries;(2)provide real-time IT resources to enable real-time applications,such as emergency response;(3)deal with access spikes;and(4)provide more reliable and scalable service for massive numbers of concurrent users to advance public knowledge.The emergence of cloud computing provides a potential solution with an elastic,on-demand computing platform to integrateobservation systems,parameter extracting algorithms,phenomena simulations,analytical visualization and decision support,and to provide social impact and user feedbackthe essential elements of the geospatial sciences.We discuss the utilization of cloud computing to support the intensities of geospatial sciences by reporting from our investigations on how cloud computing could enable the geospatial sciences and how spatiotemporal principles,the kernel of the geospatial sciences,could be utilized to ensure the benefits of cloud computing.Four research examples are presented to analyze how to:(1)search,access and utilize geospatial data;(2)configure computing infrastructure to enable the computability of intensive simulation models;(3)disseminate and utilize research results for massive numbers of concurrent users;and(4)adopt spatiotemporal principles to support spatiotemporal intensive applications.The paper concludes with a discussion of opportunities and challenges for spatial cloud computing(SCC). | Chaowei Yang Michael Goodchild Qunying Huang Doug Nebert Robert Raskin Yan Xu Myra Bambacus Daniel Fay | 2011 | International Journal of Digital Earth2011,4,4: | 55 |
| 2 | Patterns and Dynamics of Rifting on Passive Continental Margin from Shelf to Slope of the Northern South China Sea:Evidence from 3D Analogue Modeling显示文摘Affected by thermal perturbation due to mantle uprising, the rheological structure of the lithosphere could be modified, which could lead to different rifting patterns from shelf to slope in a pas-sive continental margin. From the observed deformation style on the northern South China Sea and analogue modeling experiments, we find that the rift zone located on the shelf is characterized by half grabens or simple grabens controlled mainly by long faults with large vertical offset, supposed to be formed with normal lithosphere extension. On the slope, where the lithosphere is very hot due to mantle upwelling and heating, composite grabens composed of symmetric grabens developed. The boundary and inner faults are all short with small vertical offset. Between the zones with very hot and normal lithosphere, composite half grabens composed of half grabens or asymmetric grabens formed, whose boundary faults are long with large vertical offset, while the inner faults are relatively short. Along with the thickness decrease of the brittle upper crust due to high temperature, the deformation becomesmore sensitive to the shape of a pre-existing weakness zone and shows orientation variation along strike. When there was a bend in the pre-existing weakness zone, and the basal plate was pulled by a clockwise rotating stress, the strongest deformation always occurs along the middle segment and at the transition area from the middle to the eastern segments, which con-tributes to a hotter lithosphere in the middle segment, where the Baiyun (白云) sag formed. | 孙珍 周蒂 吴世敏 钟志洪 Myra Keep 姜建群 樊浩 | 2009 | Journal of China University of Geosciences2009,20,1: | 14 |
| 3 | Mechanisms of immune checkpoint inhibitormediated liver injury显示文摘The immune checkpoints,cytotoxic T-lymphocyte-associated antigen 4(CTLA-4) and programmed cell death protein-1/ligand-1(PD-1/PD-L1) are vital contributors to immune resulation and tolerance.Recently immune checkpoint inhibitors(ICIs) have revolutionized cancer therapy;however,they come with the cost of immune related adverse events involving multiple organs such as the liver.Due to its constant expo sure to foreign antigens,the liver has evolved a high capacity for immune tolerance,therefore,blockade of the immune checkpoints can result in aberrant immune activation affecting the liver in up to 20% of patients depending on the agent(s) used and underlying factors.This type of hepatotoxicity is termed immune mediated liver injury from checkpoint inhibitors(ILICI) and is more common when CTLA4 and PD-1/PDL1 are used in combination.The underlying mechanisms of this unique type of hepatotoxicity are not fully understood;however,the contribution of CD8^(+) cytotoxic T lymphocytes,various CD4^(+) T cells populations,cytokines,and the secondary activation of the innate immune system leading to liver injury have all been suggested.This review summarizes our current understanding of the underlying mechanisms of liver injury in immunotherapy using animal models of ILICI and available patient data from clinical studies. | Layla Shojaie Myra Ali Andrea Iorga Lily Dara | 2021 | Acta Pharmaceutica Sinica B2021,11,12: | 6 |
| 4 | Anticancer effect of linalool via cancer-specific hydroxyl radical generation in human colon cancer显示文摘AIM To investigate the anticancer mechanisms of the monoterpenoid alcohol linalool in human colon cancer cells.METHODS The cytotoxic effect of linalool on the human colon cancer cell lines and a human fibroblast cell line was examined using the WST-8 assay. The apoptosisinducing effect of linalool was measured using the terminal deoxynucleotidyl transferase d UTP nickend labeling assay and flow cytometry with Annexin V. Oxidative stress was investigated by staining for diphenyl-1-pyrenylphosphine, which is a cellular lipid peroxidation marker, and electron spin resonance spectroscopy. Sixteen SCID mice xenografted with human cancer cells were randomized into 3 groups for in vivo analysis: control and low-dose and high-dose linalool groups. The control group was administered tap water orally every 3 d. The linalool treatment groups were administered 100 or 200 μg/kg linalool solution orally for the same period. All mice were sacrificed under anesthesia 21 d after tumor inoculation, and tumors and organs were collected for immunohistochemistry using an anti-4-hydroxynonenal antibody. Tumor weights were measured and compared between groups. RESULTS Linalool induced apoptosis of cancer cells in vitro, following the cancer-specific induction of oxidative stress, which was measured based on spontaneous hydroxyl radical production and delayed lipid peroxidation. Mice in the high-dose linalool group exhibited a 55% reduction in mean xenograft tumor weight compared with mice in the control group(P < 0.05). | 5 | Split-dose bowel preparation improves adequacy of bowel preparation and gastroenterologists' adherence to National Colorectal Cancer Screening and Surveillance Guidelines显示文摘AIM To quantify the impact of split-dose regimen on endoscopists' compliance with guideline recommendations for timing of repeat colonoscopy in patients with normal colonoscopy or 1-2 small polyps(< 10 mm).METHODS A retrospective chart review of all endoscopy reports was undertaken in average-risk individuals > 50 years old with a normal screening colonoscopy and 1-2 small polyps. Data were abstracted from two time periods, pre and post-split-dose bowel preparation institution. Main outcome measurements were recommendation for timing of repeat colonoscopy and bowel preparation quality. Bivariate analysis by χ~2 tests and Student's t-tests were performed to assess differences between the two cohorts. Multivariable logistic regression was used with guideline consistent recommendations as the dependent variables and an indicator for 2011 cohort as the primary predictor. RESULTS Four thousand two hundred and twenty-five patients were included in the study; 47.0%(1987) prior to the institution of split dose bowel preparation, and 53.0%(2238) after the institution of split dose bowel preparation. Overall, 82.2%(n = 3472) of the colonoscopies were compliant with guideline recommendations, with a small but significantly increased compliance rate in year 2011(83.7%) compared to year 2009(80.4%, P = 0.005), corresponding to an unadjusted odds ratio of 1.25(95%CI: 1.07-1.47; P = 0.005). Colonoscopies with either 'Adequate' or 'Excellent' had increased from 30.6% in year 2009 to 39.6% in year 2011(P < 0.001). However, there was no significant difference in poor/inadequate category of bowel preparation as there was a mild increase from 4.6% in year 2009 to 5.1% in year 2011(P = 0.50). | 7 | A survey of ampullectomy practices显示文摘AIM:To investigate the endoscopic ampullectomy practices of expert biliary endoscopists.METHODS:An anonymous survey was mailed to 79 expert biliary endoscopists to assess ampullectomy practices.RESULTS:Forty six(58%) biliary endoscopists returned the questionnaire.Of these, 63% were in academia and in practice for an average of 16.4 years(± 8.6).Endoscopists performed an average of 1.1(± 0.8) ampullectomies per month.Prior to ampullectomy, endoscopic ultrasound was'always'utilized by 67% of respondents vs'sometimes'in 31% of respondents.Empiric biliary sphincterotomy was not utilized uniformly, only 26%'always'and 37%'sometimes'performed it prior to resection.Fifty three percent reported'never'performing empiric pancreatic sphincterotomy prior to ampullectomy.Practitioners with high endoscopic retrograde cholangiopancreatography volumes were the most likely to perform a pancreatic sphincterotomy(OR = 10.9;P = 0.09).Participants overwhelmingly favored'always'placing a prophylactic pancreatic stent, with 86% placing it after ampullectomy rather than prior to resection(23%).Argon plasma coagulation was the favored adjunct modality(83%) for removal of residual adenomatous tissue.Practitioners uniformly(100%) preferred follow-up examination to be within 6 mo post-ampullectomy.CONCLUSION:Among biliary experts, there is less variation in ampullectomy practices than is reflected in the literature. | Stacy B Menees Philip Schoenfeld Hyungjin Myra Kim Grace H Elta | 2009 | World Journal of Gastroenterology2009,15,28: | 2 |
