| 1 | 内质网应激在大鼠心肌缺血再灌注损伤中的作用:与细胞自噬的关系显示文摘目的评价内质网应激在大鼠心肌缺血再灌注损伤中的作用及其与细胞自噬的关系。方法清洁级健康成年雄性SD大鼠36只,体重250~300 g,采用随机数字表法分为3组(n=12):假手术组(Sham组)、心肌缺血再灌注组(IR组)和内质网应激抑制剂4-PBA组(PBA组)。采用阻断冠状动脉左前降支(LAD)缺血30 min,再灌注120 min的方法制备大鼠心肌缺血再灌注损伤模型。Sham组仅行开胸处理但不阻断LAD;PBA组于造模前3 d连续灌胃给予内质网应激抑制剂4-PBA 500 mg·kg^(-1)·d^(-1),Sham组和IR组给予等量生理盐水。于再灌注120 min时采集髂静脉血样,采用ELISA法测定血浆CK-MB和cTnI浓度;随后处死大鼠取心肌组织,采用Western blot法检测心肌葡萄糖调节蛋白78(GRP78)、微管相关蛋白轻链3Ⅱ(LC3Ⅱ)和自噬相关蛋白5(ATG5)的表达。结果与Sham组比较,IR组和PBA组血浆CK-MB和cTnI浓度升高,IR组心肌组织GRP78、ATG5和LC3Ⅱ表达上调(P<0.05),病理学损伤加重;与IR组比较,PBA组血浆CK-MB和cTnI浓度降低,心肌组织GRP78、ATG5和LC3Ⅱ表达下调(P<0.05),病理学损伤减轻。结论内质网应激参与了大鼠心肌缺血再灌注损伤的过程,其机制可能与促进细胞自噬有关。 | 和凤 颜学滔 卢清 周慧敏 夏韵 Mohamed Bassirou MY 柯剑娟 王焱林 | 2021 | 中华麻醉学杂志2021,41,12: | 2 |

| 5 | Antioxidant,antimicrobial,and α-glucosidase inhibitory activities of saponin extracts from walnut(Juglans regia L.) leaves显示文摘Objective:To investigate the relationship between triterpenoid saponin content and antioxidant,antimicrobial,and α-glucosidase inhibitory activities of 70%ethanolic,butanolic,aqueous,supernate and precipitate extracts of Juglans regia leaves.Methods:Triterpenoid saponins of different Juglans regia leaf extracts were measured by the vanillin method.Antioxidant activity was evaluated against DPPH and ABTS free radicals.We also assessed α-glucosidase inhibitory and antimicrobial activities of the leaf extracts.Pearson’s correlation coefficient was evaluated to determine the correlation between the saponin content and biological activities.Results:The butanolic extract was most effective against DPPH with an IC50of 6.63μg/mL,while the aqueous extract showed the highest scavenging activity against ABTS free radical with an IC50of 42.27μg/mL.Pearson’s correlation analysis indicated a strong negative correlation (r=-0.956) between DPPH radical scavenging activity (IC50) and the saponin content in the samples examined.In addition,the aqueous extract showed the best α-glucosidase inhibitory activity compared with other extracts.All the extracts had fair antibacterial activity against Bacillus subtilis,Escherichia coli,and Klebsiella pneumoniae except for the aqueous extract.Conclusions:Juglans regia extracts show potent antioxidant,antimicrobial,and α-glucosidase inhibitory activities.There is a correlation between saponin levels in Juglans regia leaf extracts and the studied activities.However,additional research is required to establish these relationships by identifying the specific saponin molecules responsible for these activities and elucidating their mechanisms of action. | Youssef Elouafy Adil El Yadini Salma Mortada Mohamed Hnini Hicham Harhar Asaad Khalid Ashraf NAbdalla Abdelhakim Bouyahya Khang Wen Goh Long Chiau Ming My El Abbes Faouzi Mohamed Tabyaoui | 2023 | Asian Pacific Journal of Tropical Biomedicine2023,13,2: | 1 |

| 9 | 激活腺苷A2B受体对大鼠心肌缺血再灌注时自噬的影响:PI3K/Akt信号通路在其中的作用显示文摘目的:评价激活腺苷A2B受体对大鼠心肌缺血再灌注时自噬的影响及磷脂酰肌醇3-激酶/蛋白质丝氨酸苏氨酸激酶(PI3K/Akt)信号通路在其中的作用。方法:清洁级健康雄性SD大鼠48只,体重220~280 g,采用随机数字表法分为4组(n=12):假手术组(Sham组)、心肌缺血再灌注组(I/R组)、腺苷A2B受体激动剂BAY 60-6583组(BAY组)和BAY 60-6583+PI3K抑制剂LY 294002组(BAY+LY组)。采用结扎左冠状动脉前降支30 min、再灌注120 min的方法制备大鼠心肌缺血再灌注损伤模型。BAY组于再灌注前5 min时腹腔注射BAY60-65831 mg/kg,BAY+LY组于再灌注前10 min时腹腔注射LY 29400210 mg/kg,再灌注前5 min时腹腔注射BAY60-65831 mg/kg。于再灌注结束时,采用ELISA法检测血清LDH和CK-MB浓度,处死取心肌组织,采用伊文氏蓝、TTC双重染色法确定心肌梗死体积百分比,采用Western blot法检测微管相关蛋白1轻链3Ⅰ(LC3Ⅰ)、LC3Ⅱ、Beclin-1和磷酸化Akt(p-Akt)的表达水平。结果:与Sham组比较,I/R组血清LDH、CK-MB浓度和心肌梗死体积百分比升高,心肌p-Akt表达下调,Beclin-1和LC3Ⅱ表达上调,LC3Ⅱ/LC3Ⅰ比值升高(P<0.05)。与I/R组比较,BAY组血清LDH、CK-MB浓度和心肌梗死面积百分比降低,心肌p-Akt表达上调,Beclin-1和LC3Ⅱ表达下调,LC3Ⅱ/LC3Ⅰ比值降低(P<0.05),BAY+LY组上述指标差异无统计学意义(P>0.05)。与BAY组比较,BAY+LY组血清LDH、CK-MB浓度和心肌梗死面积百分比升高,心肌p-Akt表达下调,Beclin-1和LC3Ⅱ表达上调,LC3Ⅱ/LC3Ⅰ比值升高(P<0.05)。结论:激活腺苷A2B受体可降低大鼠心肌缺血再灌注时心肌细胞自噬水平,机制可能与激活PI3K/Akt信号通路有关。 | 周慧敏 Mohamed Bassirou MY 夏韵 和凤 柯剑娟 王焱林 | 2021 | 中华麻醉学杂志2021,41,5: | 1 |

| 12 | Imaging volumes during COVID-19:A Victorian health service experience显示文摘BACKGROUND The World Health Organisation declared the coronavirus disease 2019(COVID-19)a pandemic on March 11,2020.While globally,the relative caseload has been high,Australia’s has been relatively low.During the pandemic,radiology services have seen significant changes in workflow across modalities and a reduction in imaging volumes.AIM To investigate differences in modality imaging volumes during the COVID-19 pandemic across a large Victorian public health network.METHODS A retrospective analysis from January 2019 to December 2020 compared imaging volumes across two periods corresponding to the pandemic’s first and second waves.Weekly volumes across patient class,modality and mobile imaging were summed for periods:wave 1(weeks 11 to 16 for 2019;weeks 63 to 68 for 2020)and wave 2(weeks 28 to 43 for 2019;weeks 80 to 95 for 2020).Microsoft Power Business Intelligence linked to the radiology information system was used to mine all completed examinations.RESULTS Summed weekly data during the pandemic’s first wave showed the greatest decrease of 29.8%in adult outpatient imaging volumes and 46.3%in paediatric emergency department imaging volumes.Adult nuclear medicine demonstrated the greatest decrease of 37.1%for the same period.Paediatric nuclear medicine showed the greatest decrease of 47.8%,with angiography increasing by 50%.The pandemic’s second wave demonstrated the greatest decrease of 23.5%in adult outpatient imaging volumes,with an increase of 18.2%in inpatient imaging volumes.The greatest decrease was 28.5%in paediatric emergency department imaging volumes.Nuclear medicine showed the greatest decrease of 37.1%for the same period.Paediatric nuclear medicine showed the greatest decrease of 36.7%.Mobile imaging utilisation increased between 57.8%and 135.1%during the first and second waves.A strong correlation was observed between mobile and nonmobile imaging in the emergency setting(Spearman’s correlation coefficient=-0.743,P=0.000).No correlation was observed in the inpatient setting(Spearman’s correlation coefficient=-0.059,P=0.554).CONCLUSION Nuclear medicine was most impacted,while computed tomography and angiography were the least affected by the pandemic.The impact was less during the pandemic’s second wave.Mobile imaging shows continuous growth during both waves. | Jo-Anne Pinson My Linh Diep Vinay Krishnan Caroline Aird Cassie Cooper Christopher Leong Jeff Chen Nicholas Ardley Eldho Paul Mohamed Khaldoun Badawy | 2022 | World Journal of Radiology2022,14,8: | 0 |
| 13 | 大鼠RNAi-A2aR慢病毒载体的构建及鉴定显示文摘目的构建大鼠RNAi-腺苷A2a受体(A2aR)慢病毒载体并进行鉴定。方法首先构建3对短发夹RNA(shRNA)-A2aR序列(shRNA-A2aR 1、shRNA-A2aR 2、shRNA-A2aR 3),在shRNA慢病毒载体中分别插入3对双链shRNA oligo,shRNA慢病毒重组质粒构建成功后,将重组质粒、包装载体、穿梭载体共转染293T细胞,从而获得病毒液。实验Ⅰ采用随机数字表法将大鼠原代心肌细胞分为3组(n=6):空载组(V组)、shRNA-A2aR 1组和shRNA-A2aR 3组,各组分别转染MOI为10的相应病毒液,转染48 h时,Western blot法检测A2aR表达水平,确定干扰效率,以筛选最佳慢病毒载体。实验Ⅱ采用随机数字表法将大鼠原代心肌细胞分为5组(n=36):空载组(V组)、MOI5组、MOI10组、MOI15组和MOI20组,各组分别转染相应MOI的病毒液(最佳慢病毒载体),于转染24、48和72 h时,采用CCK-8法测定细胞存活率,荧光显微镜下观察细胞活力和细胞死亡情况,Western blot法检测A2aR表达水平,确定干扰效率。结果实验Ⅰ成功构建了2种shRNA-A2aR慢病毒载体(shRNA-A2aR 1、3)。shRNA-A2aR 3病毒液滴度为3.5×10^8 TU/ml。与V组和shRNA-A2aR 1组比较,shRNA-A2aR 3组心肌细胞A2aR表达下调(P<0.01),shRNA-A2aR 3慢病毒载体干扰效率73%。实验Ⅱ选择shRNA-A2aR 3慢病毒载体转染24 h时,各组细胞存活率均>85%;转染48 h时,MOI5组和MOI10组细胞存活率>80%;转染72 h各组细胞存活率<70%。倒置荧光显微镜下可见,MOI5组荧光密度稍低,MOI10组转染48 h及MOI20组转染24 h时,荧光密度较高且细胞状态较好;转染72 h时各组心肌细胞活力明显下降、死亡细胞增加。Western blot显示:MOI10组转染48 h、MOI15组转染48 h、MOI20组转染24和48 h时干扰效率均>70%。结论成功构建了大鼠shRNA-A2aR慢病毒载体,且MOI为10、转染48 h或MOI为20、转染24 h为最佳转染方案。 | 夏韵 Mohamed Bassirou MY 周慧敏 李婧凡 陈超 柯剑娟 王焱林 | 2020 | 中华麻醉学杂志2020,40,7: | 0 |
