5Laboratory Diagnosis and Monitoring the Viral Shedding of SARS-CoV-2 Infection显示文摘The worldwide epidemic of coronavirus disease 2019(COVID-19)is ongoing.Rapid and accurate detection of the causative virus SARSCoV-2 is vital for the treatment and control of COVID-19.In this study,the comparative sensitivity of different respiratory specimen types were retrospectively analyzed using 3,552 clinical samples from 410 COVID-19 patients confirmed by Guangdong CDC(Center for Disease Control and Prevention).Except for bronchoalveolar lavage fluid(BALF),the sputum possessed the highest positive rate(73.4%–87.5%),followed by nasal swabs(53.1%–85.3%)for both severe and mild cases during the first 14 days after illness onset(d.a.o.).Viral RNA could be detected in all BALF samples collected from the severe group within 14 d.a.o.and lasted up to 46 d.a.o.Moreover,although viral RNA was negative in the upper respiratory samples,it was also positive in BALF samples in most cases from the severe group during treatment.Notably,no viral RNA was detected in BALF samples from the mild group.Despite typical ground-glass opacity observed via computed tomographic scans,no viral RNA was detected in the first three or all upper respiratory tract specimens from some COVID-19 patients.In conclusion,sputum is most sensitive for routine laboratory diagnosis of COVID-19,followed by nasal swabs.Detection of viral RNA in BALF improves diagnostic accuracy in severe COVID-19 patients.Yang Yang Minghui Yang Jing Yuan Fuxiang Wang Zhaoqin Wang Jinxiu Li Mingxia Zhang Jinli Wei Ling Peng Gary Wong Haixia Zheng Weibo Wu Chenguang Shen Mingfeng Liao Kai Feng Jianming Li Qianting Yang Juanjuan Zhao Lei Liu Yingxia Liu 2020The Innovation2020,1,3:1

7Taxi origin and destination demand prediction based on deep learning:a review显示文摘Taxi demand prediction is a crucial component of intelligent transportation system research.Compared to region-based demand prediction,origin-destination(OD)demand prediction has a wide range of potential applications,including real-time matching,idle vehicle allocation,ridesharing services,and dynamic pricing,among others.However,because OD demand involves complex spatiotemporal dependence,research in this area has been limited thus far.In this paper,we first review existing research from four perspectives:topology construction,temporal and spatial feature processing,and other relevant factors.We then elaborate on the advantages and limitations of OD prediction methods based on deep learning architecture theory.Next,we discuss ongoing challenges in OD prediction,such as dynamics,spatiotemporal dependence,semantic differentiation,time window selection,and data sparsity problems,and summarize and compare potential solutions to each challenge.These findings offer valuable insights for model selection in OD demand prediction.Finally,we provide public datasets and open-source code,along with suggestions for future research directions.Dan Peng Mingxia Huang Zhibo Xing 2023Digital Transportation and Safety2023,2,3:0
8MANF brakes TLR4 signaling by competitively binding S100A8 with S100A9 to regulate macrophage phenotypes in hepatic fibrosis显示文摘The mesencephalic astrocyte-derived neurotrophic factor(MANF)has been recently identified as a neurotrophic factor,but its role in hepatic fibrosis is unknown.Here,we found that MANF was upregulated in the fibrotic liver tissues of the patients with chronic liver diseases and of mice treated with CCl4.MANF deficiency in either hepatocytes or hepatic mono-macrophages,particularly in hepatic mono-macrophages,clearly exacerbated hepatic fibrosis.Myeloid-specific MANF knockout increased the population of hepatic Ly6C^(high)macrophages and promoted HSCs activation.Furthermore,MANF-sufficient macrophages(from WT mice)transfusion ameliorated CCl4-induced hepatic fibrosis in myeloid cells-specific MANF knockout(MKO)mice.Mechanistically,MANF interacted with S100A8 to competitively block S100A8/A9 heterodimer formation and inhibited S100A8/A9-mediated TLR4-NF-κB signal activation.Pharmacologically,systemic administration of recombinant human MANF significantly alleviated CCl_(4)-induced hepatic fibrosis in both WT and hepatocytes-specific MANF knockout(HKO)mice.This study reveals a mechanism by which MANF targets S100A8/A9-TLR4 as a“brake”on the upstream of NF-κB pathway,which exerts an impact on macrophage differentiation and shed light on hepatic fibrosis treatment.Chao Hou Dong Wang Mingxia Zhao Petek Ballar Xinru Zhang Qiong Mei Wei Wang Xiang Li Qiang Sheng Jun Liu Chuansheng Wei Yujun Shen Yi Yang Peng Wang Juntang Shao Sa Xu Fuyan Wang Yang Sun Yuxian Shen 2023Acta Pharmaceutica Sinica B2023,13,10:0
9Sorting nexin 3 exacerbates doxorubicin-induced cardiomyopathy via regulation of TFRC-dependent ferroptosis显示文摘The clinical utilization of doxorubicin(Dox)in various malignancies is restrained by its major adverse effect:irreversible cardiomyopathy.Extensive studies have been done to explore the prevention of Dox cardiomyopathy.Currently,ferroptosis has been shown to participate in the incidence and development of Dox cardiomyopathy.Sorting Nexin 3(SNX3),the retromer-associated cargo binding protein with important physiological functions,was identified as a potent therapeutic target for cardiac hypertrophy in our previous study.However,few study has shown whether SNX3 plays a critical role in Dox-induced cardiomyopathy.In this study,a decreased level of SNX3 in Dox-induced cardiomyopathy was observed.Cardiac-specific Snx3 knockout(Snx3-cKO)significantly alleviated cardiomyopathy by downregulating Dox-induced ferroptosis significantly.SNX3 was further demonstrated to exacerbate Dox-induced cardiomyopathy via induction of ferroptosis in vivo and in vitro,and cardiac-specific Snx3 transgenic(Snx3-cTg)mice were more susceptible to Dox-induced feroptosis and cardiomyopathy.Mechanistically,SNX3 facilitated the recycling of transferrin 1 receptor(TFRC)via direct interaction,disrupting iron homeostasis,increasing the accumulation of iron,triggering ferroptosis,and eventually exacerbating Dox-induced cardiomyopathy.Overall,these findings established a direct SNX3-TFRC-ferroptosis positive regulatory axis in Dox-induced cardiomyopathy and suggested that targeting SNX3 provided a new effective therapeutic strategy for Dox-induced cardiomyopathy through TFRCdependentferroptosis.Wenjing Yu Yuehuai Hu Zhiping Liu Kaiteng Guo Dinghu Ma Mingxia Peng Yuemei Wang Jing Zhang Xiaolei Zhang Panxia Wang Jiguo Zhang Peiqing Liu Jing Lu 2023Acta Pharmaceutica Sinica B2023,13,12:0
10Aqueous synthesis of bright near-infrared-emitting Zn-Cu-In-Se quantum dots for multiplexed detection of tumor markers显示文摘Aqueous synthesis is a green but challenging pathway to produce cadmium-free,bright,and near-infrared(NIR)emitting quantum dots(QDs).Herein,we develop a straightforward aqueous phase approach to prepare Zn-Cu-In-Se QDs with tunable emission from 630 to 800 nm through varying the molar ratio of Zn and Cu.The ultrasmall nanoparticle size remains nearly fixed(~2.6 nm)when the QD composition is varied,allowing for the effects of composition on the optical properties of QDs to be decoupled from the impact of particle size.By leveraging the off-stoichiometric effect on photoluminescence(PL),Zn-Cu-In-Se QDs with a Zn:Cu molar ratio of 1.0 show a long PL decay lifetime of 211 ns and an absolute photoluminescence quantum yield(PLQY)of 14.2%.The PLQY is further enhanced by overcoating a ZnS shell,reaching up to 25.8%.Based on the bright,biocompatible,and emission-tunable QDs,nanoprobes with targeting capability for multiple tumor markers are further constructed and employed to simultaneously ascertain targets in the cytoplasm and cell membrane.This study provides a green and effective strategy for achieving bright and biocompatible NIR quantum dots for multiplexed biodetection applications.Hui Liu Peng Cai Kevin J.McHugh Collin F.Perkinson Laishuo Li Shuna Wang Wei Wang Mingxia Jiao Xiliang Luo Lihong Jing 2022Nano Research2022,15,9:0
11Developing allometric equations to estimate forest biomass for tree species categories based on phylogenetic relationships显示文摘The development of allometric biomass models is important process in biomass estimation because the reliability of forest biomass and carbon estimations largely depends on the accuracy and precision of such models.National Forest Inventories(NFI)are detailed assessments of forest resources at national and regional levels that provide valuable data for forest biomass estimation.However,the lack of biomass allometric equations for each tree species in the NFI currently hampers the estimation of national-scale forest biomass.The main objective of this study was to develop allometric biomass regression equations for each tree species in the NFI of China based on limited biomass observations.These equations optimally grouped NFI and biomass observation species according to their phylogenetic relationships.Significant phylogenetic signals demonstrated phylogenetic conservation of the crown-to-stem biomass ratio.Based on phylogenetic relationships,we grouped and matched NFI and biomass observation species into 22 categories.Allometric biomass regression models were developed for each of these 22 species categories,and the models performed successfully(R^(2)=0.97,root mean square error(RMSE)=12.9​t·ha^(–1),relative RMSE=11.5%).Furthermore,we found that phylogeny-based models performed more effectively than wood density-based models.The results suggest that grouping species based on their phylogenetic relationships is a reliable approach for the development and selection of accurate allometric equations.Mingxia Yang Xiaolu Zhou Changhui Peng Tong Li Kexin Chen Zelin Liu Peng Li Cicheng Zhang Jiayi Tang Ziying Zou 2023Forest Ecosystems2023,10,4:0
