5Anti-infection effects of heparin on SARS-CoV-2 in a diabetic mouse model显示文摘Severe acute respiratory syndrome coronavirus 2(SARSCo V-2)infection can result in more severe syndromes and poorer outcomes in patients with diabetes and obesity.However,the precise mechanisms responsible for the combined impact of coronavirus disease 2019(COVID-19)and diabetes have not yet been elucidated,and effective treatment options for SARS-Co V-2-infected diabetic patients remain limited.To investigate the disease pathogenesis,K18-h ACE2 transgenic(h ACE2^(Tg))mice with a leptin receptor deficiency(h ACE2-Lepr^(-/-))and high-fat diet(h ACE2-HFD)background were generated.The two mouse models were intranasally infected with a 5×10^(5) median tissue culture infectious dose(TCID_(50))of SARSCo V-2,with serum and lung tissue samples collected at 3days post-infection.The h ACE2-Lepr^(-/-)mice were then administered a combination of low-molecular-weight heparin(LMWH)(1 mg/kg or 5 mg/kg)and insulin via subcutaneous injection prior to intranasal infection with1×10^(4) TCID_(50)of SARS-Co V-2.Daily drug administration continued until the euthanasia of the mice.Analyses of viral RNA loads,histopathological changes in lung tissue,and inflammation factors were conducted.Results demonstrated similar SARS-Co V-2 susceptibility in h ACE2^(Tg)mice under both lean(chow diet)and obese(HFD)conditions.However,compared to the h ACE2-Lepr^(+/+)mice,h ACE2-Lepr^(-/-)mice exhibited more severe lung injury,enhanced expression of inflammatory cytokines and hypoxia-inducible factor-1α(HIF-1α),and increased apoptosis.Moreover,combined LMWH and insulin treatment effectively reduced disease progression and severity,attenuated lung pathological changes,and mitigated inflammatory responses.In conclusion,preexisting diabetes can lead to more severe lung damage upon SARS-Co V-2 infection,and LMWH may be a valuable therapeutic approach for managing COVID-19patients with diabetes.Zhongyun Zhang Ning Zhang Xuancheng Lu Min Zhou Xiaoxiang Yan Weiqiong Gu Jingru Yang Qin Zhang Cheng Zhang Yuhuan Gong Mingjun Jia Xiaoyu Zhang Peng Ning Mei Liu Xiaoyan Li Xiaomeng Shi Wenjun Liu George FGao Guang Ning Jiqiu Wang Yuhai Bi 2023Zoological Research2023,44,6:1
6Aldosterone may induce visfatin expression via the GR-ERK1/2 signal pathway in 3T3-L1 adipocytes显示文摘The main function of aldosterone is to regulate blood pressure and electrolytic balance;however,recent studies have also focused on the effect of aldosterone on glucose metabolism especially in adipose tissue.Aldosterone was found to inhibit insulin-induced glucose uptake in 3T3-L1 adipocytes [1] and aldosterone deficiency was found to prevent high-fat-feeding-induced hyperglycemia and adipocyte dysfunction in mice [2].Visfatin,an adipokine discovered by the team of Fukuhara,is mainly released by adipose tissue [3].Treatment of 3T3-L1 adipocytes and L6 myocytes with visfatin increased basal glucose uptake,and glucose release was suppressed in H4IIEC3 hepatocytes after visfatin treatment [3].It has also been suggested that visfatin may act as a proinflammatory cytokine [4] and produce chronic inflammation in adipocytes [5].Eyileten et al.[6] reported that treatment with angiotensin-converting enzyme inhibitor,which can reduce plasma aldosterone concentration,decreased circulating visfatin level in diabetic nephropathy patients.These findings suggest a possible interaction between the renin–angiotensin–aldosterone system(RAAS)and visfatin.Considering the important roles of aldosterone and visfatin and the association between RAAS and visfatin,we hypothesize that there may be a direct regulatory interaction between aldosterone and visfatin.To test this hypothesis,3T3-L1 adipocytes were used as the cell model and the expression of visfatin was measured by qRT-PCR and western blot analysis.Junhua Ma Xuemei Zhao Jichen Zhang Fei Sun Jiancan Lu Hongling Zhu Mingjun Gu Jingnan Wang Xiangqi Li 2019Acta Biochimica et Biophysica Sinica2019,51,3:1

11A bionic controllable strain membrane for cell stretching at air–liquid interface inspired by papercutting显示文摘Lung diseases associated with alveoli,such as acute respiratory distress syndrome,have posed a long-term threat to human health.However,an in vitro model capable of simulating different deformations of the alveoli and a suitable material for mimicking basement membrane are currently lacking.Here,we present an innovative biomimetic controllable strain membrane(BCSM)at an air–liquid interface(ALI)to reconstruct alveolar respiration.The BCSM consists of a high-precision three-dimensional printing melt-electrowritten polycaprolactone(PCL)mesh,coated with a hydrogel substrate—to simulate the important functions(such as stiffness,porosity,wettability,and ALI)of alveolar microenvironments,and seeded pulmonary epithelial cells and vascular endothelial cells on either side,respectively.Inspired by papercutting,the BCSM was fabricated in the plane while it operated in three dimensions.A series of the topological structure of the BCSM was designed to control various local-area strain,mimicking alveolar varied deformation.Lopinavir/ritonavir could reduce Lamin A expression under over-stretch condition,which might be effective in preventing ventilator-induced lung injury.The biomimetic lung-unit model with BCSM has broader application prospects in alveoli-related research in the future,such as in drug toxicology and metabolism.Yuanrong Li Mingjun Xie Shang Lv Yuan Sun Zhuang Li Zeming Gu Yong He 2023International Journal of Extreme Manufacturing2023,5,4:0

13A simple and rapid model of vascular calcification显示文摘Vascular calcification is the aggregation and deposition of hydroxyapatite crystals within arterial wall cells,which can lead to increased vascular wall hardness,decreased compliance,thrombosis,and plaque rupture,and is an important factor for the high incidence and mortality of vascular diseases[1].Arterial calcification was discovered 5000 years ago in the Iceman[2].Nowadays,vascular calcification has become a very common adverse phenotype of vascular lesions,a common phenotypic feature of diabetes,atherosclerosis,chronic kidney disease,hypertension,and other diseases,as well as an independent risk factor for its occurrence,development,and death[3].Vascular calcification is also an independent risk factor and a sign of disease worsening.For example,in patients with diabetes and chronic kidney disease,accelerated calcification occurs in the vascular intima,media,and heart valves[4].The incidence of vascular calcification increases with age,however even young people have a significant proportion of vascular calcification[5].However,there is no effective clinical response to this,especially no specific drugs to treat it.Therefore,the study of vascular calcification will not only improve our understanding of the pathogenesis of diseases and the aging mechanism but also help to develop new treatment strategies and drugs for these diseases.Mingjun Gu Lianyong Liu Chaobao Zhang Juan Chen Xiangqi Li 2021Acta Biochimica et Biophysica Sinica2021,53,5:0
14New values of a type 2 taste receptor TAS2R14 in thyroid cancer显示文摘Recognition of bitter substances is mediated by type 2 taste receptors(TAS2Rs)family which consists of over 20 members.Traditionally,TAS2Rs are thought to be specifically expressed in the tongue.Recent studies on TAS2R member TAS2R14 in non-gustatory tissues supported its new functions beyond the perception of bitter taste.TAS2R14 regulates resveratrol transport across the human blood-cerebrospinal fluid barrier[1],which renders hope for resveratrol to be used in the treatment of brain diseases.TAS2R14 also mediates the relaxation of smooth muscle[2],indicating its value for the development of effective tocolytics for preterm birth management.TAS2R14 also has an anti-inflammatory property.TAS2R14 can inhibit IgE-induced mast cell degranulation with anti-asthma activity when activated directly by Saikosaponin b[3],and prevent LPS-induced cytokine production in human lung macrophages[4].Of note,TAS2R14 also has endocrine regulatory effect.It stimulates glucagon-like peptide 1 secretion in rat gastrointestinal tract[5],and may also have appetite-suppressant effects by inducing cholecystokinin secretion[6].Of greater interest is that we found that TAS2R14 might be a receptor for bitter medicines to fight COVID-19[7,8].Meanwhile,TAS2R14 has recently been linked to oncology.Agonists of TAS2R14,including quercetin and naringenin,exhibit anti-cancer activities in numerous cancer types[9].Noscapine stimulation of TAS2R14 increases apoptosis in ovarian cancer cells[10].However,the involvement of TAS2R14 in the risk and development of cancers including thyroid cancer(THCA)has yet to be clarified.Lianyong Liu Qingyun Zhu Hong Du Chao Shi Mingjun Gu Xiangqi Li 2022Acta Biochimica et Biophysica Sinica2022,54,4:0
