| 1 | Tissue differential expression of lycopene β-cyclase gene in papaya显示文摘Carotene pigments in flowers and fruits are distinct features related to fitness advantages such as attracting insects forpollination and birds for seed dispersal.In papaya,the flesh color of the fruit is considered a quality trait that correlateswith nutritional value and is linked to shelf-life of the fruit.To elucidate the carotenoid biosynthesis pathway in papaya,we took a candidate gene approach to clone the lycopene β-cyclase gene,LCY-B.A papaya LCY-B ortholog,cpLCY-B,was successfully identified from both cDNA and bacterial artificial chromosome(BAC)libraries and complete genomicsequence was obtained from the positive BAC including the promoter region.This cpLCY-B shared 80% amino acididentity with citrus LCY-B.However,full genomic sequences from both yellow- and red-fleshed papaya were identical.Quantitative real-time PCR(qPCR)revealed similar levels of expression at six different maturing stages of fruits forboth yellow-and red-fleshed genotypes.Further expression analyses of cpLCY-B showed that its expression levels wereseven- and three-fold higher in leaves and,respectively,flowers than in fruits,suggesting that cpLCY-B is down-regulatedduring the fruit ripening process. | Rachel L Skelton Qingyi Yu Rajeswari Srinivasan Richard Manshardt Paul H Moore Ray Ming | 2006 | Cell Research2006,16,8: | 8 |

| 3 | Mid-infrared dual-comb spectroscopy with electro-optic modulators显示文摘Absorption spectroscopy of fundamental ro-vibrational transitions in the mid-infrared region provides a powerful tool for studying the structure and dynamics of molecules in the gas phase and for sensitive and quantitative gas sensing.Laser frequency combs permit novel approaches to perform broadband molecular spectroscopy.Multiplex dual-comb spectroscopy without moving parts can achieve particularly high speed,sensitivity and resolution.However,achieving Doppler-limited resolution in the mid-infrared still requires overcoming instrumental challenges.Here we demonstrate a new approach based on difference-frequency generation of frequency-agile near-infrared frequency combs that are produced using electro-optic modulators.The combs have a remarkably flat intensity distribution,and their positions and line spacings can be freely selected by simply dialing a knob.Using the proposed technique,we record,in the 3-μm region,Doppler-limited absorption spectra with resolved comb lines within milliseconds,and precise molecular line parameters are retrieved.Our technique holds promise for fast and sensitive time-resolved studies of,for example,trace gases. | Ming Yan Pei-Ling Luo Kana Iwakuni Guy Millot Theodor W Hänsch Nathalie Picqué | 2017 | Light(Science & Applications)2017,6,1: | 5 |
| 4 | Reprogramming of a defense signaling pathway in rough lemon and sweet orange is a critical element of the early response to‘Candidatus Liberibacter asiaticus’显示文摘Huanglongbing(HLB)in citrus infected by Candidatus Liberibacter asiaticus(CLas)has caused tremendous losses to the citrus industry.No resistant genotypes have been identified in citrus species or close relatives.Among citrus varieties,rough lemon(Citrus jambhiri)has been considered tolerant due to its ability to produce a healthy flush of new growth after infection.The difference between tolerance and susceptibility is often defined by the speed and intensity of a plant’s response to a pathogen,especially early defense responses.RNA-seq data were collected from three biological replicates of CLas-and mock-inoculated rough lemon and sweet orange at week 0 and 7 following infection.Functional analysis of the differentially expressed genes(DEGs)indicated that genes involved in the mitogen activated protein kinase(MAPK)signaling pathway were highly upregulated in rough lemon.MAPK induces the transcription of WRKY and other transcription factors which potentially turn on multiple defense-related genes.A Subnetwork Enrichment Analysis further revealed different patterns of regulation of several functional categories,suggesting DEGs with different functions were subjected to reprogramming.In general,the amplitude of the expression of defense-related genes is much greater in rough lemon than in sweet orange.A quantitative disease resistance response may contribute to the durable tolerance level to HLB observed in rough lemon. | Qibin Yu Chunxian Chen Dongliang Du Ming Huang Jiqiang Yao Fahong Yu Ronald H Brlansky Frederick G.Gmitter Jr | 2017 | Horticulture Research2017,4,1: | 5 |
| 5 | Screening Helicobacter pylori genes induced during infection of mouse stomachs显示文摘AIM:To investigate the effect of in vivo environment on gene expression in Helicobacter pylori(H.pylori) as it relates to its survival in the host.METHODS:In vivo expression technology(IVET) systems are used to identify microbial virulence genes.We modified the IVET-transcriptional fusion vector,pIVET8,which uses antibiotic resistance as the basis for selection of candidate genes in host tissues to develop two unique IVET-promoter-screening vectors,pIVET11 and pIVET12.Our novel IVET systems were developed by the fusion of random Sau3A DNA fragments of H.pylori and a tandem-reporter system of chloramphenicol acetyltransferase and beta-galactosidase.Additionally,each vector contains a kanamycin resistance gene.We used a mouse macrophage cell line,RAW 264.7 and mice,as selective media to identify specific genes that H.pylori expresses in vivo.Gene expression studies were conducted by infecting RAW 264.7 cells with H.pylori.This was followed by real time polymerase chain reaction(PCR) analysis to determine the relative expression levels of in vivo induced genes.RESULTS:In this study,we have identified 31 in vivo induced(ivi) genes in the initial screens.These 31 genes belong to several functional gene families,including several well-known virulence factors that are expressed by the bacterium in infected mouse stomachs.Virulence factors,vacA and cagA,were found in this screen and are known to play important roles in H.pylori infection,colonization and pathogenesis.Their detection validates the efficacy of these screening systems.Some of the identified ivi genes have already been implicated to play an important role in the pathogenesis of H.pylori and other bacterial pathogens such as Escherichia coli and Vibrio cholerae.Transcription profiles of allivi genes were confirmed by real time PCR analysis of H.pylori RNA isolated from H.pylori infected RAW 264.7 macrophages.We compared the expression profile of H.pylori and RAW 264.7 coculture with that of H.pylori only.Some genes such as cag A,vac A,lpx C,mur I,tlp C,trx B,sod B,tnp B,pgi,rbf A and inf B showed a 2-20 fold upregulation.Statistically significant upregulation was obtained for all the above mentioned genes(P < 0.05).tlp C,cag A,vac A,sod B,rbf A,inf B,tnp B,lpx C and mur I were also significantly upregulated(P < 0.01).These data suggest a strong correlation between results obtained in vitro in the macrophage cell line and in the intact animal.CONCLUSION:The positive identification of these genes demonstrates that our IVET systems are powerful tools for studying H.pylori gene expression in the host environment. | Aparna Singh Nathaniel Hodgson Ming Yan Jungsoo Joo Lei Gu Hong Sang Emmalena Gregory-Bryson William G Wood Yisheng Ni Kimberly Smith Sharon H Jackson William G Coleman | 2012 | World Journal of Gastroenterology2012,18,32: | 5 |
