2Effect of Dy^(3+) on osteogenic and adipogenic differentiation of mouse primary bone marrow stromal cells and adipocytic trans-differentiation of mouse primary osteoblasts显示文摘A series of experimental methods including 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bro-mide (MTT) test,alkaline phosphatase (ALP) activity measurement,mineralized function,Oil Red O stain and measurement were employed to assess the effect of Dy3+ on the osteogenic and adipogenic differentiation of mouse primary bone marrow stromal cells (BMSCs) and the adipogenic trans-differ-entiation of mouse primary osteoblasts (OBs). The results showed that Dy3+ had no effect on BMSC proliferation at concentrations of 1×10-8 and 1×10-5 mol/L,but inhibited BMSC proliferation at other concentrations. Dy3+ had no effect on OB proliferation at concentrations of 1×10-10 and 1×10-9 mol/L,but inhibited OB proliferation at other concentrations. Dy3+ had no effect on the osteogenic differentia-tion of BMSCs at concentrations of 1×10-9 and 1×10-7 mol/L,and promoted osteogenic differentiation of BMSCs at other concentrations at the 7th day. The osteogenic differentiation of BMSCs was inhibited by Dy3+ at concentration of 1×10-5 mol/L at the 14th day,but promoted osteogenic differentiation of BMSCs at concentrations of 1×10-9,1×10-8,1×10-7 and 1×10-6 mol/L with the maximal effect at concen-tration of 10-6 mol/L. Dy3+ promoted mineralized function of BMSCs at any concentration. Dy3+ had no effect on adipogenic differentiation of BMSCs at concentration of 1×10-7 mol/L,but inhibited adipogenic differentiation of BMSCs at other concentrations. Dy3+ inhibited adipocytic trans-differentiation of OBs at any concentration,suggesting that Dy3+ had protective effect on bone and the protective effect on bone may be mediated by modulating differentiation of BMSCs away from the adipocyte and inhibiting adipocytic trans-differentiation of OBs which may promote differentiation and mineralization of OBs. 3Effects of genistein, daidzein and glycitein on the osteogenic and adipogenic differentiation of bone marrow stromal cells and on the adipogenic trans-differentiation of osteoblasts显示文摘The 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyl tetrazolium bromide (MTT), alkaline phosphatase (ALP) activity and oil red O assays were used to examine the effects of genistein, daidzein and glycitein on the osteogenic and adipogenic differentiation of primary mouse bone marrow stromal cells (MSCs) and the adipogenic trans-differentiation of primary mouse osteoblasts. The results indicated that daidzein, genistein and glycitein at concentrations from 1×10 -8 mol/L to 1×10 -5 mol/L promoted the proliferation of MSCs and osteoblasts; genistein, daidzein and glycitein promoted osteogenic differentiation and inhibited adipogenic differentiation of MSCs, and inhibited adipocytic transdifferentiation of osteoblasts at appropriate concentrations as 17β-estradiol. It suggests that genistein, daidzein and glycitein regulate a dual differentiational process of MSCs into the osteogenic and adipogenic lineages, and trans-differentiational process of primary osteoblasts into the adipocyte lineages, causing a lineage shift toward osteoblast. Protective effects of them on bone may be mediated by a reversal of adipogenesis which may promote the proliferation, differentiation and mineralization of osteoblasts, and make adipocytes secrete less cytokines which may promote osteoclast formation and activation. 4Numerical manifold method modeling of coupled processes in fractured geological media at multiple scales显示文摘The greatest challenges of rigorously modeling coupled hydro-mechanical(HM)processes in fractured geological media at different scales are associated with computational geometry.These challenges include dynamic shearing and opening of intersecting fractures at discrete fracture scales as a result of coupled processes,and contact alteration along rough fracture surfaces that triggers structural and physical changes of fractures at micro-asperity scale.In this paper,these challenges are tackled by developing a comprehensive modeling approach for coupled processes in fractured geological media based on numerical manifold method(NMM)at multiple scales.Based on their distinct geometric features,fractures are categorized into three different scales:dominant fracture,discrete fracture,and discontinuum asperity scales.Here the scale is relative,that of the fracture relative to that of the research interest or domain.Different geometric representations of fractures at different scales are used,and different governing equations and constitutive relationships are applied.For dominant fractures,a finite thickness zone model is developed to treat a fracture as a porous nonlinear domain.Nonlinear fracture mechanical behavior is accurately modeled with an implicit approach based on strain energy.For discrete fractures,a zero-dimensional model was developed for analyzing fluid flow and mechanics in fractures that are geometrically treated as boundaries of the rock matrix.With the zero-dimensional model,these fractures can be modeled with arbitrary orientations and intersections.They can be fluid conduits or seals,and can be open,bonded or sliding.For the discontinuum asperity scale,the geometry of rough fracture surfaces is explicitly represented and contacts involving dynamic alteration of contacts among asperities are rigorously calculated.Using this approach,fracture alteration caused by deformation,re-arrangement and sliding of rough surfaces can be captured.Our comprehensive model is able to handle the computational challenges with accurate representation of intersections and shearing of fractures at the discrete fracture scale and rigorously treats contacts along rough fracture surfaces at the discontinuum asperity scale.With future development of three-dimensional(3D)geometric representation of discrete fracture networks in porous rock and contacts among multi-body systems,this model is promising as a basis of 3D fully coupled analysis of fractures at multiple scales,for advancing understanding and optimizing energy recovery and storage in fractured geological media.Mengsu Hu Jonny Rutqvist 2020Journal of Rock Mechanics and Geotechnical Engineering2020,12,4:4

6The influence of ^(18)F-fluorodeoxyglucose positron emission tomography/computed tomography on the N-and M-staging and subsequent clinical management of intrahepatic cholangiocarcinoma显示文摘Background:Intrahepatic cholangiocarcinoma(ICC)is a highly metastatic cancer.^(18)F-fluorodeoxyglucose positron emission tomography/computed tomography(^(18)F-FDG PET/CT)enables sensitive tumor and metastasis detection.Our aim is to evaluate the influence of pre-treatment PET/CT on the N-and M-staging and subsequent clinical management in ICC patients.Methods:Between August 2010 and August 2018,660 consecutive ICC patients,without prior anti-tumor treatments nor other malignancies,were enrolled.The diagnostic performance of PET/CT on the N-and M-staging was compared with conventional imaging,and the preoperative staging accuracy and treatment re-allocation by PET/CT were retrospectively calculated.Survival difference was compared between patients receiving PET/CT or not after propensity score matching.Results:Patients were divided into group A(n=291)and group B(n=369)according to whether PET/CT was performed.Among 291 patients with both PET/CT and conventional imaging for staging in group A,PET/CT showed significantly higher sensitivity(83.0%vs.70.5%,P=0.001),specificity(88.3%vs.74.9%,P<0.001)and accuracy(86.3%vs.73.2%,P<0.001)than conventional imaging in diagnosing regional lymph node metastasis,as well as higher sensitivity(87.8%vs.67.6%,P<0.001)and accuracy(93.5%vs.89.3%,P=0.023)in diagnosing distant metastasis.Overall,PET/CT improved the accuracy of preoperative staging from 60.1%to 71.8%(P<0.001),and modified clinical treatment strategy in 5.8%(17/291)of ICC patients,with unique roles in different tumor-node-metastasis(TNM)stages.High tumor-to-non-tumor ratio(TNR)predicted poor overall survival[hazard ratio(HR)=2.17;95%confidence interval(CI):1.49-3.15;P<0.001].Furthermore,patients performing PET/CT had longer overall survival compared with those without PET/CT(HR=0.74;95%CI:0.58-0.93;P=0.011)after propensity score matching.Conclusions:PET/CT was valuable for diagnosing regional lymph node metastasis and distant metastasis in ICC patients,and facilitated accurate tumor staging and optimal treatment allocation.Youpei Lin Huanhuan Chong Guohe Song Chenhao Zhang Liangqing Dong Ling Aye Fei Liang Shuaixi Yang Mengsu Zeng Guangyu Ding Shu Zhang Jieyi Shi Aiwu Ke Xiaoying Wang Jian Zhou Jia Fan Qiang Gao 2022Hepatobiliary Surgery and Nutrition2022,11,5:3
7The effects of gold nanoparticles on the proliferation, differentiation, and mineralization function of MC3T3-E1 cells in vitro显示文摘This study has investigated the effects of gold nanoparticles (Au NPs) on the proliferation, differentiation, and mineralization of a murine preosteoblast cell line MC3T3-E1 in vitro. The results show that Au NPs with diameters of both 20 and 40 nm promoted the proliferation, differentiation, and mineralization of MC3T3-E1 cells in a time- and dose-dependent manner at the concentra-tions of 1.5×10-5, 3.0×10-5, and 1.5×10-4 μmol/L. The reverse transcriptase polymerase chain reaction (RT-PCR) indicates that the expressions of runt-related transcription factor 2 (Runx2), bone morphogenetic protein 2 (BMP-2), alkaline phosphatase (ALP), and osteocalcin (OCN) genes increased after the 20 and 40 nm Au NP treatments, and the expression levels were higher than those of the NaF group. The above results suggest that Au NPs have the potential to promote the osteogenic differentiation and miner- alization of MC3T3-E1 cells and the particle size plays a significant role in the process. 20Synthesis and antitumor activity of iodo-bridged binuclear platinum complex显示文摘Iodo-bridged binuclear platinum(II) com- plex[Pt( -NH2)I2]2(BPA) has been synthesized and characterized by elemental analysis, conductivity, differential thermal analysis, IR, UV and 1HNMR spectra techniques. The cytotoxicity of the complex was tested by MTT and SRB assays. The results show that complex BPA demonstrates better cyto-toxicity than that of the clinically established cisplatin against EJ, HCT-8, BGC-823, HL-60 and MCF-7 cell lines. The complex BPA at concentrations of 1.00 and 2.00 μmol/L induces G1 cell cycle arrest in HL-60 cells. The level of total platinum bound to DNA in HL-60 cells is significantly higher than that of cisplatin under the same experimental conditions. Acute tox-icity experimental results indiacte that LD50 of BPA is 815.3 mg/kg by intraperitoneal administration. BPA at dose of 12 mg/kg significantly inhibits the growth of nude mice implanted by human A2780 and HCT-116 carcinomas, and inhibition rate is similar to that of cisplatin at dose of 4 mg/kg by intraperitoneal admini-stration.
