1A novel vector of topological and structural information for amino acids and its QSAR applications for peptides and analogues显示文摘A new descriptor, called vector of topological and structural information for coded and noncoded amino acids (VTSA), was derived by principal component analysis (PCA) from a matrix of 66 topological and structural variables of 134 amino acids. The VTSA vector was then applied into two sets of peptide quantitative structure-activity relationships or quantitative sequence-activity modelings (QSARs/ QSAMs). Molded by genetic partial least squares (GPLS), support vector machine (SVM), and immune neural network (INN), good results were obtained. 2Effects of fire and nitrogen addition on photosynthesis and growth of three dominant understory plant species in a temperate forest显示文摘Aims Fire and atmospheric nitrogen(N)deposition have the potential to influence growth and productivity of forest canopy.However,their impacts on photosynthesis and growth traits of understory plants in forests remain largely unexplored.This study was conducted to examine the effects of burning and N addition on foliar N content,net photosynthesis and growth traits of three dominant shrub species(Vitex negundo,Lindera glauca and Symplocos chinensis)in a temperate forest in Central China.Methods The experiment used a pair-nested design,with four treatments(control,burning,N addition and burning plus N addition)and five replicates.Leaf mass area(LMA),area-based concentrations of foliar N and chlorophyll(N_(area) and Chl_(area)),net photosynthesis(A_(n)),stomatal conductance(g_(s)),maximum photosynthetic rate(A_(max))and maximal carboxylation rate(V_(cmax)),basal diameter,height and branch length(BL)of the three species were measured.Important Findings Across the three species,burning stimulated LMA,N_(area),Chl_(area),A_(n),g_(s),A_(max) and V_(cmax),and consequently enhanced basal diam-eter,height and BL.Nitrogen addition increased A_(n) and gs but did not affect LMA,N_(area),Chl_(area),A_(max),V_(cmax),basal diameter,height or BL.However,N addition strengthened the positive effects of burning on g_(s),V_(cmax),A_(n) and BL.The findings indicate the primary role of light resources in determining plant photosynthesis and growth of understory shrub species after fire and highlight that understory plants should be considered in projection of biomass accumulation and productivity of forests under environmental perturbations.Mengjun Hu Shiqiang Wan 2019Journal of Plant Ecology2019,12,4:2
3Discovery of an orally effective double-stapled peptide for reducing ovariectomy-induced bone loss in mice显示文摘Stapled peptides with significantly enhanced pharmacological profiles have emerged as promising therapeutic molecules due to their remarkable resistance to proteolysis and performance to penetrate cells.The all-hydrocarbon peptide stapling technique has already widely adopted with great success,yielding numerous potent peptide-based molecules.Based on our prior efforts,we conceived and prepared a double-stapled peptide in this study,termed FRNC-1,which effectively attenuated the bone resorption capacity of mature osteoclasts in vitro through specific inhibition of phosphorylated GSK-3β.The double-stapled peptide FRNC-1 displayed notably improved helical contents and resistance to proteolysis than its linear form.Additionally,FRNC-1 effectively prevented osteoclast activation and improved bone density for ovariectomized(OVX)mice after intravenous injection and importantly,after oral(intragastric)administration.The double-stapled peptide FRNC-1 is the first orally effective peptide that has been validated to date as a therapeutic candidate for postmenopausal osteoporosis(PMOP).Wei Cong Huaxing Shen Xiufei Liao Mengjun Zheng Xianglong Kong Zhe Wang Si Chen Yulei Li Honggang Hu Xiang Li 2023Acta Pharmaceutica Sinica B2023,13,9:0
4Establishment of an in vitro reporter system for screening HBx-targeting molecules显示文摘Chronic hepatitis B virus (HBV) infection remains a global public health problem. HBV-encoded X protein (HBx) is a multifunctional regulator that is required to initiate and maintain productive HBV infection, and is involved in HBV-related hepatocellular carci noma (HCC). Inhibitors that in terfere with the functions of HBx could be useful not only for the inhibition of HBV replication but also for the prevention or treatment of HBV-related HCC. To screen molecules that target HBx on a large scale remains a tech nical challenge due to a lack of sensitive and high-throughput system .In this work, we established an in vitro bioluminescent reporter system for screening HBx-targeting molecules. The system is based on a secretory fusion protein that combines HBx and NanoLuc (HBx-Nluc). The measured activity of NanoLuc in the culture supernatant of HBx-Nluc-expressing cells directly reflects the level of secreted HBx-Nluc. HBx protein-targeting intracellular anti-HBx single-chain variable fragment and RNA-targeting shRNA significantly reduced the secreted NanoLuc activity in HBx-Nluc-expressing cells. 5Study on dynamic changes of land desertification in the circum-lake zone of the Qinghai Lake in the past 30 years supported by Remote Sensing and Geographical Information System显示文摘The circum-lake zone of the Qinghai Lake is one of the most serious regions of desertification on the Qinghai-Tibetan Plateau. Based on Remote Sensing(RS) and Geographical Information System(GIS) techniques, the four sets of remote sensing images acquired in 1976, 1987, 1995, and 2006 were selected to conduct thematic information extraction in 14,300 km2 of land in the circum-lake zone using the Earth Resource Data Analysis System(ERDAS) image and information processing function. In the meantime the off-site interpretation and filed check-correction methods were used to reconstruct the temporal-spatial dynamic changes and differentiation patterns of land desertification in the circum-lake zone in the past 30 years, and based on this, the developmental trend of land desertification in the next 20 years was predicted. The results of the study show that in the past 30 years land desertification in the region experienced a continuous fluctuation between rapid- and slow-development processes in time, while in space it exhibited a few small patchy distributions or sporadic distributions in many places while at the same time, four large sand-dominated distributions stood out and evolved into a circum-lake belt differentiation pattern with a lakeside dike as the connecting line. 6Effects of Polygonatum odoratum Polysaccharide on Reproductive Hormones in Male Rats Fed a High-fat Diet显示文摘[Objectives]This study was conducted to investigate the effects of Polygonatum odoratum polysaccharide(POP)on organ relative weights and reproductive hormone levels in male rats fed a high-fat diet.[Methods]Thirty healthy male Sprague-Dawley(SD)rats were randomly divided into two groups according to their body weight:10 in normal control group(Group NC,n=10)and 20 in experimental group(n=20).The rats in experimental group were fed a high-fat diet for eight weeks before they were further randomly divided into two groups:high fat group(Group HF)and high fat+400 mg/(kg·d)POP group(Group HF+POP).In Group HF+POP,the rats were administered with POP for another six weeks,before their blood plasma was collected,and the relative weights of their testis and epididymis were calculated.The plasma levels of testosterone(T),estrogen(E2),follicle-stimulating hormone(FSH),cortisol(C)and luteinizing hormone(LH)were measured by radioimmunoassay,and the plasma levels of sex hormone-binding globulin(SHBG)and insulin-like growth factor-1(IGF-1)were determined by enzyme-linked immunosorbent assay.[Results]Compared with Group HF,POP could effectively inhibit rat obesity caused by high-fat diets,increase the relative weights of their testis and epididymis,plasma levels of LH,E2,FSH,T,SHBG and IGF-1,and reduce the plasma level of E2.[Conclusions]Polygonatum odoratum polysaccharide(POP)is able to effectively regulate the level of reproductive hormones in high-fat diet fed rats,and helps to protect their reproductive function.Yixin YAN Gang ZHANG Lingli BAI Xin TANG Mengjun YUAN Yihong HU Yan WANG Chenzhong JIN 2023Agricultural Biotechnology2023,12,2:0

8Chromosome-scale assembly reveals asymmetric paleo-subgenome evolution and targets for the acceleration of fungal resistance breeding in the nut crop, pecan显示文摘Pecan(Carya illinoinensis)is a tree nut crop of worldwide economic importance that is rich in healthpromoting factors.However,pecan production and nut quality are greatly challenged by environmental stresses such as the outbreak of severe fungal diseases.Here,we report a high-quality,chromosome-scale genome assembly of the controlled-cross pecan cultivar‘Pawnee’constructed by integrating Nanopore sequencing and Hi-C technologies.Phylogenetic and evolutionary analyses reveal two whole-genome duplication(WGD)events and two paleo-subgenomes in pecan and walnut.Time estimates suggest that the recent WGD event and considerable genome rearrangements in pecan and walnut account for expansions in genome size and chromosome number after the divergence from bayberry.The two paleo-subgenomes differ in size and protein-coding gene sets.They exhibit uneven ancient gene loss,asymmetrical distribution of transposable elements(especially LTR/Copia and LTR/Gypsy),and expansions in transcription factor families(such as the extreme pecan-specific expansion in the far-red impaired response 1 family),which are likely to reflect the long evolutionary history of species in the Juglandaceae.A whole-genome scan of resequencing data from 86 pecan scab-associated core accessions identified 47 chromosome regions containing 185 putative candidate genes.Significant changes were detected in the expression of candidate genes associated with the chitin response pathway under chitin treatment in the scab-resistant and scabsusceptible cultivars‘Excell’and‘Pawnee’.These findings enable us to identify key genes that may be important susceptibility factors for fungal diseases in pecan.The high-quality sequences are valuable resources for pecan breeders and will provide a foundation for the production and quality improvement of tree nut crops.Lihong Xiao Mengjun Yu Ying Zhang Jie Hu Rui Zhang Jianhua Wang Haobing Guo He Zhang Xinyu Guo Tianquan Deng Saibin Lv Xuan Li Jianqin Huang Guangyi Fan 2021Plant Communications2021,2,6:0
9Recognition for avian influenza virus proteins based on support vector machine and linear discriminant analysis显示文摘Total 200 properties related to structural characteristics were employed to represent structures of 400 HA coded proteins of influenza virus as training samples. Some recognition models for HA proteins of avian influenza virus (AIV) were developed using support vector machine (SVM) and linear discriminant analysis (LDA). The results obtained from LDA are as follows: the identification accuracy (Ria) for training samples is 99.8% and Ria by leave one out cross validation is 99.5%. Both Ria of 99.8% for training samples and Ria of 99.3% by leave one out cross validation are obtained using SVM model, respectively. External 200 HA proteins of influenza virus were used to validate the external predictive power of the resulting model.
