3CRISPR/Cas9: Development and Application in Rice Breeding显示文摘Rice(Oryza sativa L.)is an important staple food crop worldwide due to its adaptability to different environmental conditions.Because of its great economic and social importance,there is a constant requirement for new varieties with improved agronomic characteristics,such as tolerance to different biotic(such as bacterium,fungus,insect and virus)and abiotic stresses(such as salinity,drought and temperature),higher yield and better organoleptic and nutritional value.Among the new genome editing technologies,the clustered regularly interspaced short palindromic repeats(CRISPR)/CRISPR-associated protein(Cas)(CRISPR/Cas)system allows precise and specific edition in a targeted genome region.It is one of the most frequently used techniques for the study of the function of new genes and for the development of mutant lines with enhanced tolerance to biotic and abiotic stresses,herbicide resistance or improved yield.The wide varieties of applications for this technology include simple non-homologous end joining,homologous recombination,gene replacement,and base editing.In this review,we analyzed how some of these applications have been used in rice cultivars to obtain rice varieties better adapted to current environmental conditions and market requirements.Fernando Matías ROMERO Andrés GATICA-ARIAS 2019Rice science2019,26,5:6

14Mapping aboveground biomass and its prediction uncertainty using LiDAR and field data, accounting for tree-level allometric and LiDAR model errors显示文摘Background: The increasing availability of remotely sensed data has recently challenged the traditional way of performing forest inventories, and induced an interest in model-based inference. Like traditional design-based inference, model-based inference allows for regional estimates of totals and means, but in addition for wall-to-wall mapping of forest characteristics. Recently Light Detection and Ranging(LiDAR)-based maps of forest attributes have been developed in many countries and been well received by users due to their accurate spatial representation of forest resources. However, the correspondence between such mapping and model-based inference is seldom appreciated. In this study we applied hierarchical model-based inference to produce aboveground biomass maps as well as maps of the corresponding prediction uncertainties with the same spatial resolution. Further, an estimator of mean biomass at regional level, and its uncertainty, was developed to demonstrate how mapping and regional level assessment can be combined within the framework of model-based inference.Results: Through a new version of hierarchical model-based estimation, allowing models to be nonlinear, we accounted for uncertainties in both the individual tree-level biomass models and the models linking plot level biomass predictions with LiDAR metrics. In a 5005 km2 large study area in south-central Sweden the predicted aboveground biomass at the level of 18 m×18 m map units was found to range between 9 and 447 Mg·ha^-1. The corresponding root mean square errors ranged between 10 and 162 Mg·ha^-1. For the entire study region, the mean aboveground biomass was 55 Mg·ha^-1 and the corresponding relative root mean square error 8%. At this level 75%of the mean square error was due to the uncertainty associated with tree-level models.Conclusions: Through the proposed method it is possible to link mapping and estimation within the framework of model-based inference. Uncertainties in both tree-level biomass models and models linking plot level biomass with LiDAR data are accounted for, both for the uncertainty maps and the overall estimates. 16Porous titanium granules in critical size defects of rabbit tibia with or without membranes显示文摘Recently,porous titanium granules(PTGs)have been indicated for the preservation of the dimensions of post-extraction sockets,as a filler in sinus lift procedures and for the treatment of peri-implant and periodontal defects,based on the osteoconductivity and dimensional stability of the titanium granules.However,there is a lack of information regarding the use of this material in larger defects and in conjunction with membranes.The objective of this study is to test the behavior of PTGs used to fill critical size defects in rabbit tibiae,with and without membranes.Critical defects were created in both tibiae of rabbits,divided randomly into three groups:Group A(defect filled with PTG),Group B(defect filled with PTG1collagen membrane)and a control group(empty defect).After six weeks,histomorphometric analysis was performed.The results showed more defect closures at the cortical area(87.37%62.2%)and more bone formation at the marrow area(57.6%61.3%)in Group B,in comparison with the other groups(P,0.05);the use of membranes improved the material stability expressed as more percentages of the original material when membranes were used(P,0.05).Finally,inflammatory reactions were observed when the granules were not protected by membranes.In spite of the limitations of this animal study,it may be concluded that PTG particles are osteoconductive and allow bone growth.The PTG particles must be covered by a membrane,especially when grafting larger defects,in order to control particle migration,promote clot stabilization and separate the PTG graft from undesired soft tissue cells.Rafael Arcesio Delgado-Ruiz Jose Luis Calvo-Guirado Marcus Abboud Maria Piedad Ramirez-Ferna'ndez Jose Eduardo Maté-Snchez Bruno Negri Alex Won Georgios Romanos 2014International Journal of Oral Science2014,6,2:1
