1MicroRNAs, development of Barrett’s esophagus, and progression to esophageal adenocarcinoma显示文摘Barrett's esophagus is a premalignant condition caused by gastroesophageal reflux. Once developed, it can progress through varying grades of dysplasia to esoph-ageal adenocarcinoma. Whilst it is well accepted that Barrett's esophagus is caused by gastroesophageal reflux, the molecular mechanisms of its pathogenesis and progression to cancer remain unclear. MicroRNAs (miRNAs) are short segments of RNA that have been shown to control the expression of many human genes. They have been implicated in most cellular processes, and the role of miRNAs in disease development is be-coming increasingly evident. 2Design of 16S rRNA gene primers for 454 pyrosequencing of the human foregut microbiome显示文摘AIM:To design and validate broad-range 16S rRNA primers for use in high throughput sequencing to classify bacteria isolated from the human foregut microbiome.METHODS:A foregut microbiome dataset was constructed using 16S rRNA gene sequences obtained from oral,esophageal,and gastric microbiomes produced by Sanger sequencing in previous studies represented by 219 bacterial species.Candidate primers evaluated were from the European rRNA database.To assess the effect of sequence length on accuracy of classification,16S rRNA genes of various lengths were created by trimming the full length sequences.Sequences spanning various hypervariable regions were selected to simulate the amplicons that would be obtained using possible primer pairs.The sequences were compared with full length 16S rRNA genes for accuracy in taxonomic classification using online software at the Ribosomal Database Project (RDP).The universality of the primer set was evaluated using the RDP 16S rRNA database which is comprised of 433 306 16S rRNA genes,represented by 36 phyla.RESULTS:Truncation to 100 nucleotides(nt)downstream from the position corresponding to base 28 in the Escherichia coli 16S rRNA gene caused misclassification of 87(39.7%)of the 219 sequences,compared with misclassification of only 29(13.2%)sequences with truncation to 350 nt.Among 350-nt sequence reads within various regions of the 16S rRNA gene,the reverse read of an amplicon generated using the 343F/798R primers had the least(8.2%)effect on classification.In comparison,truncation to 900 nt mimicking single pass Sanger reads misclassified 5.0%of the 219 sequences.The 343F/798R amplicon accurately assigned 91.8%of the 219 sequences at the species level.Weighted by abundance of the species in the esophageal dataset,the 343F/798R amplicon yielded similar classification accuracy without a significant loss in species coverage(92%).Modification of the 343F/798R primers to 347F/803R increased their universality among foregut species.Assuming that a typicalpolymerase chain reaction can tolerate 2 mismatches between a primer and a template,the modified 347F and 803R primers should be able to anneal 98%and 99.6%of all 16S rRNA genes in the RDP database.CONCLUSION:347F/803R is the most suitable pair of primers for classification of foregut 16S rRNA genes but also possess universality suitable for analyses of other complex microbiomes.Carlos W Nossa William E Oberdorf Jφrn A Aas Bruce J Paster Todd Z DeSantis Eoin L Brodie Daniel Malamud Michael A Poles Zhiheng Pei 2010World Journal of Gastroenterology2010,16,33:16
3miR-200 family expression is downregulated upon neoplastic progression of Barrett's esophagus显示文摘AIM: To investigate miR-200 family expression in Barrett's epithelium, gastric and duodenal epithelia, and esophageal adenocarcinoma. METHODS: Real-time reverse transcriptase-polymerase chain reaction was used to measure miR-200, ZEB1 and ZEB2 expression. Ingenuity Pathway Analysis of miR-200 targets was used to predict biological outcomes. RESULTS: Barrett's epithelium expressed lower levels of miR-141 and miR-200c than did gastric and duodenal epithelia (P < 0.001). In silico analysis indicated roles for the miR-200 family in molecular pathways that distinguish Barrett's epithelium from gastric and duodenalepithelia, and which control apoptosis and proliferation. All miR-200 members were downregulated in adenocarcinoma (P < 0.02), and miR-200c expression was also downregulated in non-invasive epithelium adjacent to adenocarcinoma (P < 0.02). The expression of all miR-200 members was lower in Barrett's epithelium derived high-grade dysplastic cell lines than in a cell line derived from benign Barrett's epithelium. We observed signif icant inverse correlations between miR-200 family expression and ZEB1 and ZEB2 expression in Barrett's epithelium and esophageal adenocarcinoma (P < 0.05). 5Intratidal and neap-spring variations of suspended sediment concentrations and sediment transport processes in the North Branch of the Changjiang Estuary显示文摘Profiles of tidal current and suspended sediment concentration(SSC) were measured in the North Branch of the Changjiang Estuary from neap tide to spring tide in April 2010. The measurement data were analyzed to determine the characteristics of intratidal and neap-spring variations of SSC and suspended sediment transport. Modulated by tidal range and current speed, the tidal mean SSC increased from 0.5 kg/m3 in neap tide to 3.5 kg/m3 in spring tide. The intratidal variation of the depth-mean SSC can be summarized into three types: V-shape variation in neap tide, M-shape and mixed M-V shape variation in medium and spring tides. The occurrence of these variation types is controlled by the relative intensity and interaction of resuspension, settling and impact of water exchange from the rise and fall of tide. In neap tide the V-shape variation is mainly due to the dominant effect of the water exchange from the rise and fall of tide. During medium and spring tides, resuspension and settling processes become dominant. The interactions of these processes, together with the sustained high ebb current and shorter duration of low-tide slack, are responsible for the M-shape and M-V shape SSC variation. Weakly consolidated mud and high current speed cause significant resuspension and remarkable flood and ebb SSC peaks. Settling occurs at the slack water periods to cause SSC troughs and formation of a thin fluff layer on the bed. 6MicroRNA signatures in chemotherapy resistant esophageal cancer cell lines显示文摘AIM:To investigate expression of microRNA(miRNA)and potential targets in chemotherapy resistant esoph-ageal cancer cell lines.METHODS:An in-vitro model of acquired chemotherapy resistance in esophageal adeno-(EAC)and squamous cell carcinoma(ESCC)cells was used,and microRNA expression profiles for cisplatin or 5-fluorouracil(5-FU)resistant variants vs chemotherapy sensitive controls were compared using microarray and quantitative real-time polymerase chain reaction(PCR).The expression of chemotherapy-relevant genes potentially targeted by the dysregulated microRNAs in the chemotherapy resistant variants was also evaluated.RESULTS:Chemotherapy resistant sublines were found to have specific miRNA signatures,and these miRNA signatures were different for the cisplatin vs 5-FU resistant cells from the same tumor cell line,and also for EAC vs ESCC cells with resistance to the same specific chemotherapy agent.Amongst others,miR-27b-3p,miR-193b-3p,miR-192-5p,miR-378 a-3p,miR-125a-5p and miR-18a-3p were dysregulated,consistent with negative posttranscriptional control of KRAS,TYMS,ABCC3,CBL-B and ERBB2 expression via these miRNAs.CONCLUSION:The current study supports the hypothesis that microRNA expression has an impact on chemotherapy resistance in esophageal cancer.Richard Hummel Corina Sie David I Watson Tingting Wang Alfiya Ansar Michael Z Michael Mark Van der Hoek Joerg Haier Damian J Hussey 2014World Journal of Gastroenterology2014,20,40:8
7Adenosquamous carcinoma of the pancreas: Molecular characterization of 23 patients along with a literature review显示文摘Adenosquamous carcinoma of the pancreas(ASCP)is a rare entity. Like adenocarcinoma of the pancreas,overall survival is poor. Characteristics of ASCP include central tumor necrosis, along with osteoclasts and hypercalcemia. Various theories exist as to why this histological subtype exists, as normal pancreas tissue has no benign squamous epithelium. Due to the rarity of this disease, limited molecular analysis has been performed, and those reports indicate unique molecular features of ASCP. In this paper, we characterize 23 patients diagnosed with ASCP through molecular profiling using immunohistochemistry staining, fluorescent in situ hybridization, chromogenic in situ hybridization, and gene sequencing, Additionally, we provide a comprehensive literature review of what is known to date of ASCP.Molecular characterization revealed overexpression in MRP1(80%), MGMT(79%), TOP2A(75), RRM1(42%),TOPO1(42%), PTEN(45%), CMET(40%), and C-KIT(10%) among others. One hundred percent of samples tested were positive for KRAS mutations. This analysis shows heretofore unsuspected leads to be considered for treatments of this rare type of exocrine pancreas cancer. Molecular profiling may be appropriate to provide maximum information regarding the patient's tumor. 8复合钙钛矿太阳能电池电荷传输层材料研究进展显示文摘有机无机复合钙钛矿太阳能电池因具有适合的载流子扩散长度而成为备受关注的有望获得高效率的光伏器件。复合钙钛矿材料本身不含贵金属元素,可以采用液相法或物理气相法低温制备,成本低廉,但目前应用最多的电子传输层材料TiO2需400~500℃煅烧,与柔性基底及低温制备技术适应性差;空穴传输层材料SpiroOMeTAD合成工艺复杂,价格高昂,限制了复合钙钛矿太阳能电池的开发应用。开发和研究导电性好、成本低、稳定性好的电子和空穴传输层材料是复合钙钛矿太阳能电池研究中的一个非常重要的方面。综述了复合钙钛矿太阳能电池中电荷传输层材料的研究进展及发展方向。电子传输层材料方面通过对TiO2的改性以及与石墨烯的复合,采用ZnO、石墨烯或PCBM作为电子传输层材料,以与柔性基底及低温制备技术相适应。空穴传输层材料方面,采用其它低成本、导电性高的有机p型半导体替代spiro-OMeTAD;采用无机空穴传输层材料以避免有机空穴传输层材料的老化问题,提高电池的长期稳定性;利用复合钙钛矿材料兼作吸收层与空穴传输层,制备无空穴传输层材料结构电池以降低成本,提高稳定性。陈建清 杨东辉 江静华 马爱斌 宋丹 Chaoying Ni Michael Z Hu 2015材料导报2015,29,5:4

11Resonance-enhanced multi-octave supercontinuum generation in antiresonant hollow-core fibers显示文摘Ultrafast supercontinuum generation in gas-filled waveguides is an enabling technology for many intriguing applications ranging from attosecond metrology towards biophotonics,with the amount of spectral broadening crucially depending on the pulse dispersion of the propagating mode.In this study,we show that structural resonances in a gas-filled antiresonant hollow core optical fiber provide an additional degree of freedom in dispersion engineering,which enables the generation of more than three octaves of broadband light that ranges from deep UV wavelengths to near infrared.Our observation relies on the introduction of a geometric-induced resonance in the spectral vicinity of the ultrafast pump laser,outperforming gas dispersion and yielding a unique dispersion profile independent of core size,which is highly relevant for scaling input powers.Using a krypton-filled fiber,we observe spectral broadening from 200 nm to 1.7μm at an output energy of B 23μJ within a single optical mode across the entire spectral bandwidth.Simulations show that the frequency generation results from an accelerated fission process of solitonlike waveforms in a non-adiabatic dispersion regime associated with the emission of multiple phase-matched Cherenkov radiations on both sides of the resonance.This effect,along with the dispersion tuning and scaling capabilities of the fiber geometry,enables coherent ultra-broadband and high-energy sources,which range from the UV to the mid‐infrared spectral range.Rudrakant Sollapur Daniil Kartashov Michael Zürch Andreas Hoffmann Teodora Grigorova Gregor Sauer Alexander Hartung Anka Schwuchow Joerg Bierlich Jens Kobelke Mario Chemnitz Markus A Schmidt Christian Spielmann 2017Light(Science & Applications)2017,6,1:2
