2Allele-aware chromosome-level genome assembly of Artemisia annua reveals the correlation between ADS expansion and artemisinin yield显示文摘Artemisia annua is the major natural source of artemisinin,an anti-malarial medicine commonly used worldwide.Here,we present chromosome-level haploid maps for two A.annua strains with different artemisinin contents to explore the relationships between genomic organization and artemisinin production.High-fidelity sequencing,optical mapping,and chromatin conformation capture sequencing were used to assemble the heterogeneous and repetitive genome and resolve the haplotypes of A.annua.Approximately 5o,ooo genes were annotated for each haplotype genome,and a triplication event that occurred approximately 58.12million years ago was examined for the first time in this species.A total of 3,903,467-5,193,414 variants(SNPs,indels,and structural variants)were identified in the 1.5-Gb genome during pairwise comparison between haplotypes,consistent with the high heterozygosity of this species.Genomic analyses revealed a correlation between artemisinin concents and the copy number of amorpha-4,11-dienes ynthasegenes.This correlation was further confirmed by resequencing of 36A.annua samples with varied artemisinin contents.Circular consensus sequencing of transcripts facilitated the detection of paralog expression.Collectively,our study provides chromosome-level allele-aware genome assemblies for two A.annua strains and new insights into the biosynthesis of artemisinin and its regulation,which will contribute to conquering malaria worldwide.Baosheng Liao Xiaofeng Shen Li Xiang Shuai Guo Shiyu Chen Ying Meng Yu Liang Dandan Ding Junqi Bai Dong Zhang Tomasz Czechowski Yi Li Hui Yao Tingyu Mai Caroline Howard Chao Sun Haitao Liu Jiushi Liu Jin Pei Jihai Gao Jigang Wang Xiaohui Qiu Zhihai Huang Hongyi Li Ling Yuan Jianhe Wei lan Graham Jiang Xu Boli Zhang Shilin Chen 2022Molecular Plant2022,15,8:9
3Control Method for Steel Strip Roughness in Two-stand Temper Mill Rolling显示文摘How to control surface roughness of steel strip in a narrow range for a long time has become an important question because surface roughness would significantly influence the appearance of the products. However, there are few effective solutions to solve the problem currently. In this paper, considering both asperities of work roll pressing in and squeezing the steel strip, two asperity contact models including squeezing model and pressing in model in a two-stand temper mill rolling are established by using finite element method(FEM). The simulation investigates the influences of multiple process parameters, such as work roll surface roughness, roll radius and roll force on the surface roughness of steel strip. The simulation results indicate that work rolls surface roughness and roll force play important roles in the products; furthermore, the effect of roll force in the first stand is opposite to the second. According to the analysis, a control method for steel strip surface roughness in a narrow range for a long time is proposed, which applies higher work roll roughness in the first stand and lower roll roughness in the second to make the steel strip roughness in a required narrow range. In the later stage of the production, decreasing the roll force in the first stand and increasing the roll force in the second stand guarantee the steel strip roughness relatively stable in a long time. The following experimental measurements on the surface topography and roughness of the steel strips during the whole process are also conducted. The results validate the simulation conclusions and prove the effect of the control method. 5Geochemical characteristics of light hydrocarbons in natural gases from the Turpan-Hami Basin and identification of low-mature gas显示文摘Theoretical and practical knowledge regarding low-mature gasses is of significant importance to identifying potential natural gas resources.Light hydrocarbon parameters and C and H isotopes are useful tools to identify low-mature gas.Twenty gas samples were collected from the Turpan-Hami Basin for light hydrocarbon analyses.The results showed that the light hydrocarbon components of natural gases contain high methylcycloxane,high isoparaffin and low benzene.This implies that the gas-generating parent materials are of typical humus type and the paleoenvironment is a fresh water sedimentary environment.These features are consistent with the geological setting of the basin.Comparative studies of isoheptane,heptane,and the carbon isotopic compositions of methane in natural gases,and other maturity indices indicated that natural gases in the Turpan-Hami Basin are dominated by low-mature gas formed during the low evolution stage of Jurassic coal seams.The parent materials are of type III,and the maturation degree was in the low evolution stage.These are the fundamental characteristics of low-mature gas.Results of light hydrocarbon research provided further evidence to suggest that the Turpan-Hami Basin is a large-scale gas producer of low-mature gas in China.It is likely that this resource will play an important role in future exploration and development of low-mature gas in China.SHEN Ping WANG XiaoFeng WANG ZhiYong MENG QianXiang XU YongChang 2010Chinese Science Bulletin2010,55,29:8

8LncRNA DPP10-AS1 promotes malignant processes through epigenetically activating its cognate gene DPP10 and predicts poor prognosis in lung cancer patients显示文摘Objective:The purpose of this study was to explore the function and gene expression regulation of the newly identified lnc RNA DPP10-AS1 in lung cancer,and its potential value as a prognostic biomarker.Methods:q RT-PCR and Western blot were conducted to detect the expression of DDP10-AS1 and DPP10 in lung cancer cell lines and tissues.The effects of DDP10-AS1 on DPP10 expression,cell growth,invasion,apoptosis,and in vivo tumor growth were investigated in lung cancer cells by Western blot,rescue experiments,colony formation,flow cytometry,and xenograft animal experiments.Results:The novel antisense lnc RNA DPP10-AS1 was found to be highly expressed in cancer tissues(P<0.0001),and its upregulation predicted poor prognosis in patients with lung cancer(P=0.0025).Notably,DPP10-AS1 promoted lung cancer cell growth,colony formation,and cell cycle progression,and repressed apoptosis in lung cancer cells by upregulating DPP10 expression.Additionally,DPP10-AS1 facilitated lung tumor growth via upregulation of DPP10 protein in a xenograft mouse model.Importantly,DPP10-AS1 positively regulated DPP10 gene expression,and both were coordinately upregulated in lung cancer tissues.Mechanically,DPP10-AS1 was found to associate with DPP10 m RNA but did not enhance DPP10 m RNA stability.Hypomethylation of DPP10-AS1 and DPP10 contributed to their coordinate upregulation in lung cancer.Conclusions:These findings indicated that the upregulation of the antisense lnc RNA DPP10-AS1 promotes lung cancer malignant processes and facilitates tumorigenesis by epigenetically regulating its cognate sense gene DPP10.DPP10-AS1 may serve as a candidate prognostic biomarker and a potential therapeutic target in lung cancer.Haihua Tian Jinchang Pan Shuai Fang Chengwei Zhou Hui Tian Jinxian He Weiyu Shen Xiaodan Meng Xiaofeng Jin Zhaohui Gong 2021Cancer Biology & Medicine2021,18,3:5
9Regulatory network and genetic interactions established by OsMADS34 in rice inflorescence and spikelet morphogenesis显示文摘Grasses display highly diversified inflorescence architectures that differ in the arrangement of spikelets and flowers and determine cereal yields. However, the molecular basis underlying grass inflorescence morphogenesis remains largely unknown. Here we investigate the role of a functionally diversified SEPALLATA MADS-box transcription factor, OsMADS34, in regulating rice(Oryza sativa L.)inflorescence and spikelet development. Microarray analysis showed that, at the very early stages of inflorescence formation, dysfunction of OsMADS34 caused altered expression of 379 genes that are associated with protein modification and degradation, transcriptional regulation,signaling and metabolism activity. Genetic analysis revealed that OsMADS34 controls different aspects of inflorescence structure, branching and meristem activity synergistically with LAX PANICLE1(LAX1) and FLORAL ORGAN NUMBER4(FON4), as evidenced by the enhanced phenotypes of osmads34 lax1 and osmads34 fon4 compared with the single mutants. Additionally, double mutant between osmads34 and the sterile lemma defective mutant elongated empty glume(ele) displayed an enhanced phenotype, that is,longer and wider sterile lemmas that were converted into lemma/palea-like organs, suggesting that ELE and OsMADS34 synergistically control the sterile lemma development. OsMADS34 may act together with OsMADS15 in controlling sterile lemma development. 10Microwave propagation in air drilling显示文摘This paper examines the feasibility in air drilling of transmitting down-hole signals by using microwaves. Firstly the basic theory of microwave propagation in the drill-pipe or casing was studied, including power loss, cutoff wavelength, and dust scattering. Theoretical analysis indicates that the microwave propagation distance in a Ф214mm casing can easily reach 5,000 m. When the effect of dust particles is taken into account, the propagation distance decreases to 2,000 m. We conducted both laboratory experiments and field tests in casings commonly used in oilfields. The field tests show that the effective propagation distance of microwave in the casing is about 1,300 m. The experimental results do not match well with theoretical prediction, but are acceptable. 12Synthesis, characteristics of hierarchical EU-1 zeolite for xylene isomerization probe reaction显示文摘Introduced a method of synthesizing hierarchical EU-1 zeolite with organosilanes as additive, and studied the influences of following different kinds of organosilanes on the synthesis of hierarchical EU-1 zeolite: γ-glycidoxy propyl trimethoxy silane(GPTMS), N-β-(aminoethyl)-γ-aminopropyl methyl dimethoxyl silane(APAEDMS),and N-(β-aminoethyl)-γ-aminopropyl dimethoxyl(ethyoxyl) silane(TMPED). The hierarchical EU-1 samples were characterized by XRD, SEM, N_2 adsorption, FT-IR and NH_3-TPD to analyze the crystallinity, morphology, surface area, pore size distribution and acidity. The results showed that hierarchical EU-1 zeolites were successfully synthesized; organosilanes have great influence on crystal morphology of EU-1 zeolites; the exterior surface area of hierarchical EU-1 zeolite, which synthesized with organosilanes(APAEDMS) adding into synthesis system, increased by 62.1% and mesopore volume increased by 129.1% compared with conventional EU-1 zeolites, thus can reduce the diffusional restriction markedly in catalytic reaction. The catalytic performance of hierarchical EU-1zeolites were evaluated in m-xylene isomerization on fixed bed reactor. The catalytic data showed that the isomerization activity PX/X of the hierarchical EU-1 zeolites reached around 24.09% in theoretical thermodynamic equilibrium from 23.83%, and the selectivity of C_8 aromatic hydrocarbon increased from 75.16% to 84.87%. 15Fast computation of scattering from 3D complex structures by MLFMA显示文摘This paper introduces the research work on the extension of multilevel fast multipole algorithm (MLFMA) to 3D complex structures including coating object, thin dielectric sheet, composite dielectric and con-ductor, cavity. The impedance boundary condition is used for scattering from the object coated by thin lossy material. Instead of volume integral equation, surface integral equation is applied in case of thin dielectric sheet through resistive sheet boundary condition. To realize the fast computation of scattering from composite homoge-neous dielectric and conductor, the surface integral equation based on equivalence principle is used. Compared with the traditional volume integral equation, the surface integral equation reduces greatly the number of unknowns. To compute conducting cavity with electrically large aperture, an electric field integral equation is applied. 17Effect of solution cooling rate on the microstructure and creep deformation mechanism of a rhenium-free second-generation single crystal superalloy显示文摘Various cooling scenarios(water,oil,air and furnace)were employed to study the impacts of the solution cooling rate(SCR)on the microstructure and creep behavior of a novel single-crystal(SX)superalloy.The results showed that the cubic degree and size of theγphases were inversely proportional to the SCR.The creep life first increased and then dropped dramatically with a reduction in the SCR.The creep life of the sample cooled with air cooling(AC)was the highest,up to 144.90 h at 800℃/750 MPa and160.15 h at 1100℃/137 MPa.During creep at 800℃/750 MPa,the improved creep life of the AC sample was mainly attributed to the fine cubicγphases,which decreased the rate ofγ-phase coarsening and favoured plastic deformation by promoting the active movement of dislocations.The AC helped theγphases become rich in Al,Ti and Ta while depleted in Co and Cr,which enhanced its stacking fault energy,thus promoting the formation of dislocation locks.Meanwhile,the largest negative lattice misfit caused by AC induced denserγ/γinterface dislocation networks at 1100℃/137 MPa,which efficiently reduced the minimum creep rate.The calculated average dislocation spacing results indicated that the smallest density of excess dislocations corresponded to the AC sample,proving its greatest creep resistance.Interestingly,the size of the secondaryγphases first decreased and then increased sharply with decreasing SCR during creep at 1100℃/137 MPa,when fine secondaryγphases had a positive role in the blockage of dislocation movement in the matrix.Eventually,the comprehensive SCR effect was explored to provide more guidance in the design of Re-free SX superalloys.Xipeng Tao Yunling Du Xinguang Wang Jie Meng Yizhou Zhou Jinguo Li Xiaofeng Sun 2022Journal of Materials Science & Technology2022,,36:3

19Novel nanometer scaffolds regulate the biological behaviors of neural stem cells显示文摘Ideal tissue-engineered scaffold materials regulate proliferation, apoptosis and differentiation of cells seeded on them by regulating gene expression. In this study, aligned and randomly oriented collagen nanofiber scaffolds were prepared using electronic spinning technology. Their diameters and appearance reached the standards of tissue-engineered nanometer scaffolds. The nanofiber scaffolds were characterized by a high swelling ratio, high porosity and good mechanical properties. The proliferation of spinal cord-derived neural stem cells on novel nanofiber scaffolds was obviously enhanced. The proportions of cells in the S and G 2 /M phases noticeably increased. Moreover, the proliferation rate of neural stem cells on the aligned collagen nanofiber scaffolds was high. The expression levels of cyclin D1 and cyclin-dependent kinase 2 were increased. Bcl-2 expression was significantly increased, but Bax and caspase-3 gene expressions were obviously decreased. There was no significant difference in the differentiation of neural stem cells into neurons on aligned and randomly oriented collagen nanofiber scaffolds. These results indicate that novel nanofiber scaffolds could promote the proliferation of spinal cord-derived neural stem cells and inhibit apoptosis without inducing differentiation. 20Query Translation on the Fly in Deep Web Integration显示文摘To facilitate users to access the desired information,many researches have dedicated to the Deep Web(i.e. Web data-bases) integration. We focus on query translation which is an im-portant part of the Deep Web integration. Our aim is to construct automatically a set of constraints mapping rules so that the system can translate the query from the integrated interface to the Web database interfaces based on them. We construct a concept hierar-chy for the attributes of the query interfaces,especially,store the synonyms and the types(e.g. Number,Text,etc.) for every concept. At the same time,we construct the data hierarchies for some con-cepts if necessary. Then we present an algorithm to generate the constraint mapping rules based on these hierarchies. The approach is suitable for the scalability of such application and can be ex-tended easily from one domain to another for its domain inde-pendent feature.
