3FAT1, a direct transcriptional target of E2F1, suppresses cell proliferation, migration and invasion in esophageal squamous cell carcinoma显示文摘Objective: Growing evidence indicates that FAT atypical cadherin 1(FAT1) has aberrant genetic alterations and exhibits potential tumor suppressive function in esophageal squamous cell carcinoma(ESCC). However, the role of FAT1 in ESCC tumorigenesis remains not well elucidated. The aim of this study was to further investigate genetic alterations and biological functions of FAT1, as well as to explore its transcriptional regulation and downstream targets in ESCC.Methods: The mutations of FAT1 in ESCC were achieved by analyzing a combined study from seven published genomic data, while the copy number variants of FAT1 were obtained from an analysis of our previous data as well as of The Cancer Genome Atlas(TCGA) and Cancer Cell Line Encyclopedia(CCLE) databases using the cBioPortal. The transcriptional regulation of FAT1 expression was investigated by chromatin immunoprecipitation(ChIP) and the luciferase reporter assays. In-cell western, Western blot and reverse transcription-quantitative polymerase chain reaction(RT-qPCR) were used to assess the indicated gene expression. In addition, colony formation and Transwell migration/invasion assays were employed to test cell proliferation, migration and invasion.Finally, RNA sequencing was used to study the transcriptomes.Results: FAT1 was frequently mutated in ESCC and was deleted in multiple cancers. Furthermore, the transcription factor E2 F1 occupied the promoter region of FAT1, and depletion of E2 F1 led to a decrease in transcription activity and mRNA levels of FAT1. Moreover, we found that knockdown of FAT1 promoted KYSE30 and KYSE150 cell proliferation, migration and invasion;while overexpression of FAT1 inhibited KYSE30 and KYSE410 cell proliferation, migration and invasion. 6PTBP1 promotes IRES-mediated translation of cyclin B1 in cancer显示文摘Cyclin B1 is an essential cyclin-dependent protein that involves in the G2/M transition.Multiple studies report that cyclin B1 is upregulated in cancers and promotes cancer progression.However,the mechanism of cyclin B1 upregulation remains unclear.Here we report that the 5′UTR of cyclin B1 mRNA contains an internal ribosome entry site(IRES)by using a bicistronic fluorescent reporter.We show that IRES can initiate the translation of cyclin B1,and the IRES-mediated translation is further activated under cell stress.Interacting trans-acting factors(ITAFs)are required by most IRES to initiate the translation.We find that PTBP1 promotes the IRES-mediated translation of cyclin B1 by binding to the 5′UTR of cyclin B1.On top of that,PTBP1 promotes the malignancy of ESCC cells.Our data suggest that the IRES-mediated translation of cyclin B1 plays an essential role in the cyclin B1 upregulation in cancers.Xinyi Fan Zitong Zhao Liying Ma Xuanlin Huang Qimin Zhan Yongmei Song 2022Acta Biochimica et Biophysica Sinica2022,54,5:1
7Estimation of the Maximum Allowable Loading Amount of COD in Luoyuan Bay by a 3-D COD Transport and Transformation Model显示文摘The rapid economic and social developments in the Luoyuan and Lianjiang counties of Fujian Province, China, raise certain environment and ecosystem issues. The unusual phytoplankton bloom and eutrophication, for example, have increased in severity in Luoyuan Bay(LB). The constant increase of nutrient loads has largely caused the environmental degradation in LB. Several countermeasures have been implemented to solve these environmental problems. The most effective of these strategies is the reduction of pollutant loadings into the sea in accordance with total pollutant load control(TPLC) plans. A combined three-dimensional hydrodynamic transport-transformation model was constructed to estimate the marine environmental capacity of chemical oxygen demand(COD). The allowed maximum loadings for each discharge unit in LB were calculated with applicable simulation results. The simulation results indicated that the environmental capacity of COD is approximately 11×104 t year-1 when the water quality complies with the marine functional zoning standards for LB. A pollutant reduction scheme to diminish the present levels of mariculture- and domestic-based COD loadings is based on the estimated marine COD environmental capacity. The obtained values imply that the LB waters could comply with the targeted water quality criteria. 8Effect of rare-earth doping on adsorption of carbon atom on ferrum surface and in ferrum subsurface:A first-principles study显示文摘The adsorption of carbon atom on Fe surface and in Fe subsurface with and without rare earth(La and Ce)substitution in the surface layer and subsurface layer was studied by first-principles calculations.The carbon atom is predicted to adsorb at hollow and long bridge site on Fe(100)and Fe(110),respectively.However,the carbon atom shifts to occupy preferentially hollow site on both Fe(100)and Fe(110)with rare earth atom doping at surface layer.The lower adsorption energies involved with stronger adsorption abilities were obtained for carbon atoms on Fe surface with rare earth doping at surface layer,which was determined by the electronic structure of the surface atoms.The La atom was pulled out the surface after carbon adsorption due to strong interaction of La-C,which is consistent with the more charge transfer.In the subsurface region,the carbon atom prefers to occupy at octahedral site with rare earth doping at surface layer in Fe slab.These strong adsorption energies of the carbon atoms on Fe surface and in Fe subsurface with rare earth pose relevant insights into the interaction between carbon and rare earth,which helps to understanding the influence mechanism of rare earth in carburizing.Yang Yang Xiang Zhou Faqing Pan Zuju Ma Rongjian Sa Jun Zheng Qimin Wang 2021Journal of Rare Earths2021,39,9:1

14The Biomarker Changes of a Heavy Fuel Oil After Different Weathering Times显示文摘This paper presents the experimental results of composition changes of heavy fuel oil by simulating weathering in static seawater under natural environmental conditions. The results indicate: n-C10 to n-C15 were lost gradually in 24 weeks and the relative abundance of alkanes with long chains (>n-C19) increased markedly. The aromatic compounds with less than two rings (except C4N) were completely lost in 24 weeks and CnP and CnD became the main aromatics in the heavy fuel oil after 24 weeks. The ratios of n- C17/ Pristane (Pr) and n-C18/ Phytane (Ph) were suitable for identifying lightly weathered (3 weeks) heavy fuel oil. 15Membrane-spanning domain of bovine foamy virus transmembrane protein having cytotoxicity显示文摘Foamy viruses(FVs)have broad cellular tropism infecting vertebrates from fish to human being,which indicates that Env protein has a high capability for membrane fusion.Conservative features in all FV transmembrane(TM)proteins include a region of hydrophobic domain called membrane-spanning domain(MSD),which contains several stretches of hydrophobic amino acids.To investigate whether these features were associated with the cytotoxicity effect of TM on Escherichia coli,a series of mutants were constructed and expressed in the E.coli BL21(DE3)using pET-32a(+)as expressing vector.The results showed that only TM3 without MSD was expressed in E.coli,whereas the other two containing full or part of the MSD(TM1 and TM2)could not be expressed.Furthermore,the bacterial amount and living bacteria analysis revealed that the cytotoxicity of TM was dependent on its MSD,especially on the stretches of hydrophobic amino acids.Western blotting analysis showed that TM3 protein was purified with affinity purification.MA Yonggang YU Hong WANG Jinzhong CHEN Qimin GENG Yunqi 2006Frontiers in Biology2006,1,4:0
16Nip promotes autophagy through facilitating the interaction of Rab7 and FYCO1显示文摘Autophagy is the main degradation pathway to eliminate long-lived and aggregated proteins,aged or malfunctioning organelles,which is essential for the intracellular homeostasis and prevention of malignant transformation.Although the processes of autophagosome biogenesis have been well illuminated,the mechanism of autophagosome transport remains largely unclear.In this study,we demonstrated that the ninein-like protein(Nip),a well-characterized centrosomal associated protein,was able to modulate autophagosome transport and facilitate autophagy.During autophagy,Nip colocalized with autophagosomes and physically interacted with autophagosome marker LC3,autophagosome sorting protein Rab7 and its downstream effector FYCOl.Interestingly,Nip enhanced the interaction between Rab7 and FYC01,thus accelerated autophagic flux and the formation of autophagolysosomes.Furthermore,compared to the wild-type mice,NLP deficient mice treated with chemical agent DMBA were prone to increased incidence of hepatomegaly and liver cancer,which were tight associated with the hepatic autophagic defect.Taken together,our findings provide a new insight for the first time that the well-known centrosomal protein Nip is also a new regulator of autophagy,which promotes the interaction of Rab7 and FYC01 and facilitates the formation of autophagolysosome.Wenchang Xiao Danna Yeerken Jia Li Zhangfu Li Lanfang Jiang Dan Li Ming Fu Liying Ma Yongmei Song Weimin Zhang Qimin Zhan 2021Signal Transduction and Targeted Therapy2021,6,5:0

18Changes in the Chemical Components of Light Crude Oil During Simulated Short Term Weathering显示文摘To unambiguously identify spilled oils and to link them to the known sources are extremely important in settling ques-tions of environmental impact and legal liability. The fate and behavior of spilled oils in the environment depend on a number of physicochemical and biological factors. This paper presents the results regarding changes in chemical composition of light crude oil during simulated short-term weathering based on natural environmental conditions. The results show that the saturated hydrocarbons of the light crude oil mainly distribute between n-C8 and n-C23 and the most abundant n-alkanes are found in the n-C10 to n-C16. The main chemical components of the light crude oil are n-alkanes and isoprenoids. The aromatic compounds are subordinate chemical components. Under the conditions of the weathering simulation experiment,n-alkanes less than n-C12,toluene and 1,3-dimethyl ben-zene are lost after 1 d weathering,the n-C13,n-C14,naphthalene and 2-methyl-naphthalene are lost on the fifth day of weathering,and n-C15 alkane components show certain weatherproof capability. The ratios n-C17/pristane and n-C18/phytane are unaltered and can be used to identify the source of the light crude oil during the first 8 d of weathering. After 21 d,the ratio pristine/phytane can not pro-vide much information on the source of the spilled light crude oil. 19Targeting lncRNA16 by GalNAc-siRNA conjugates facilitates chemotherapeutic sensibilization via the HBB/NDUFAF5/ROS pathway显示文摘Chemoresistance is a significant barrier to effective cancer treatment.Potential mechanisms for chemoresistance include reactive oxygen species(ROS)accumulation and expression of chemoresistance-promoting genes.Here,we report a novel function of lncRNA16 in the inhibition of ROS generation and the progression of chemoresistance.By analyzing the serum levels of lncRNA16 in a cohort of 35 patients with non-small cell lung cancer(NSCLC)and paired serum samples pre-and post-treatment from 10 NSCLC patients receiving neoadjuvant platinum-based chemotherapy,performing immunohistochemistry(IHC)assays on 188 NSCLC tumor samples,using comprehensive identification of RNA-binding proteins by mass spectrometry(ChIRP-MS)assays,as well as RNA immunoprecipitation(RIP)and RNA pull-down analyses,we discovered that patients with increased serum levels of lncRNA16 exhibited a poor response to platinum-based chemotherapy.The expression of hemoglobin subunit beta(HBB)and NDUFAF5 significantly increases with the development of chemoresistance.LncRNA16 binds to HBB and promotes HBB accumulation by inhibiting autophagy.LncRNA16 can also inhibit ROS generation via the HBB/NDUFAF5 axis and function as a scaffold to facilitate the colocalization of HBB and NDUFAF5 in the mitochondria.Importantly,preclinical studies in mouse models of chemo-resistant NSCLC have suggested that lncRNA16 targeting by trivalent N-acetylgalactosamine(GalNAc)-conjugated siRNA restores chemosensitivity and results in tumor growth inhibition with no detectable toxicity in vivo.Overall,lncRNA16 is a promising therapeutic target for overcoming chemoresistance,and the combination of first-line platinum-based chemotherapy with lncRNA16 intervention can substantially enhance anti-tumor efficacy.Yanfang Liu Yan Wang Bing Liu Wenzhong Liu Yuanyuan Ma Yiren Cao Shi Yan Panpan Zhang Lixin Zhou Qimin Zhan Nan Wu 2024Science China(Life Sciences)2024,67,4:0
