1Hepatic steatosis is associated with an increased risk of carotid atherosclerosis显示文摘AIM: Although an association between helatic steatosis and vascular risk factors has been described, direct relationships between fatty liver and atherosclerosis have not yet been investigated. The aim of the present study has been to investigate those relationships.METHODS: The Study of Health in Pomerania examined a random population sample aged between 20 and 79 years.A study population of 4 222 subjects without hepatitis B and C infections and without liver cirrhosis was available for the present analysis. Hepatic steatosis was defined sonographically and intima-media thickness (IMT) as well as plaque prevalence were estimated by carotid ultrasound.RESULTS: The prevalence rate of hepatic steatosis was 29.9%. Among subjects aged ≥45 years, an association between hepatic steatosis and IMT of the carotid arteries was found in bivariate analysis, but not after adjustment for atherosclerotic risk factors. Individuals with fatty liver had more often carotid plaques than persons without fatty liver (plaque prevalence rate 76.8% vs 66.6%; P<0.001).This association persisted after adjustment for confounding factors and was predominantly present in subjects with no to mild alcohol consumption.CONCLUSION: There is an independent association between hepatic steatosis and carotid atherosclerotic plaques. 2Bioengineering bacterial encapsulin nanocompartments as targeted drug delivery system显示文摘The development of Drug Delivery Systems(DDS)has led to increasingly efficient therapies for the treatment and detection of various diseases.DDS use a range of nanoscale delivery platforms produced from polymeric of inorganic materials,such as micelles,and metal and polymeric nanoparticles,but their variant chemical composition make alterations to their size,shape,or structures inherently complex.Genetically encoded protein nanocages are highly promising DDS candidates because of their modular composition,ease of recombinant production in a range of hosts,control over assembly and loading of cargo molecules and biodegradability.One example of naturally occurring nanocompartments are encapsulins,recently discovered bacterial organelles that have been shown to be reprogrammable as nanobioreactors and vaccine candidates.Here we report the design and application of a targeted DDS platform based on the Thermotoga maritima encapsulin reprogrammed to display an antibody mimic protein called Designed Ankyrin repeat protein(DARPin)on the outer surface and to encapsulate a cytotoxic payload.The DARPin9.29 chosen in this study specifically binds to human epidermal growth factor receptor 2(HER2)on breast cancer cells,as demonstrated in an in vitro cell culture model.The encapsulin-based DDS is assembled in one step in vivo by co-expressing the encapsulin-DARPin9.29 fusion protein with an engineered flavin-binding protein mini-singlet oxygen generator(MiniSOG),from a single plasmid in Escherichia coli.Purified encapsulin-DARPin_miniSOG nanocompartments bind specifically to HER2 positive breast cancer cells and trigger apoptosis,indicating that the system is functional and specific.The DDS is modular and has the potential to form the basis of a multi-receptor targeted system by utilising the DARPin screening libraries,allowing use of new DARPins of known specificities,and through the proven flexibility of the encapsulin cargo loading mechanism,allowing selection of cargo proteins of choice.Alexander Van de Steen Rana Khalife Noelle Colant Hasan Mustafa Khan Matas Deveikis Saverio Charalambous Clare M.Robinson Rupali Dabas Sofia Esteban Serna Diana A.Catana Konstantin Pildish Vladimir Kalinovskiy Kenth Gustafsson Stefanie Frank 2021Synthetic and Systems Biotechnology2021,6,3:2

It illustrates how developments associated with globalisation and growing urban markets are re-shaping rural areas. The interplay between global and local is investigated in two different contexts. A Chinese example reflects transformations affecting the countryside following national economic reforms. Focusing on the hinterland of Xi'an, capital of Shaanxi Province, it examines farmers' responses to the changing socio-political context and the rising size and wealth of the local market. Individual initiatives backed by government support have spawned localised concentrations of cherry growing and increased horticultural production.Farm-based tourism is creating new relationships between farmers and consumers, with farms becoming more diversified and multifunctional. The second example is the Adelaide Hills, South Australia, where cherry growing is increasingly combined with direct sales to consumers and gastronomic tourism. The paper addresses contrasts and similarities between the two examples in the interplay between global and local, and a ‘multifunctional transition' in farming. Concluding remarks include reference to new economic links forged between China and Australia through relaxations on cherry imports to China and new patterns of Chinese foreign direct investment into Australian cherry production. 10Exploring the tectonostratigraphy of the Pakistan Himalaya with new detrital zircon geochronology and neodymium isotope data from the Kaghan Valley显示文摘In the central Himalaya,the Greater and Lesser Himalayan tectonostratigraphic zones can be distinguished from each other by combining detrital zircon U-Pb ages and whole-rock Nd isotopic data.In the Hazara-Kashmir syntaxis of northern Pakistan,rock units in the Kaghan valley have been variously assigned to the Tethyan,Greater,and Lesser Himalayan zones.To determine which tectonostratigraphic zones are represented,we sampled across the Batal thrust in the northern Kaghan valley because this structure is identified in some research as the Main Central thrust.Samples from the footwall yield Paleo-to Mesoproterozoic maximum depositional ages(-1.8-1.1 Ga)and an averageεNd(0)value of-14.3.Samples from the hanging wall yield Neoproterozoic maximum depositional ages(~1000-600 Ma)andεNd(0)values of-17.3.The contrasting detrital zircon ages show that the Batal thrust in the Kaghan valley is a major structural division.However,these data lack the characteristic detrital zircon age spectra andεNd(0)values of Greater and Lesser Himalayan rocks north and south of the Main Central thrust in the central Himalaya,respectively.Therefore,either the Batal thrust is not the Main Central thrust,or the Main Central thrust in the Kaghan valley cuts a different structural level and carries rock units not seen outside the Hazara-Kashmir syntaxis.W.Joel Schiffer Delores M.Robinson Shah Faisal Harold H.Stowell 2023Geoscience Frontiers2023,14,6:0
11Geochemistry and in-situ U-Th/Pb Geochronology of the Jambil Meta-Carbonatites, Northern Pakistan: Implications on Petrogenesis and Tectonic Evolution显示文摘The putative Jambil meta-carbonatites of Swat,northern Pakistan,occur as discrete intrusions into the Proterozoic Manglaur Formation,which are difficult to be distinguished from nearby calc-silicate marble because both rock types experienced regional metamorphism during Himalayan orogenesis that resulted in similar mosaic textures and mineral assemblages.Carbonatites are often significant repositories of economic mineral resources and,therefore,are important to be distinguished from calc-silicate marble.We present new geochemical and geochronology data to distinguish between the two rock types and interpret the petrogenesis and tectonic evolution of the Jambil metacarbonatites.Whole rock chemical data from the Jambil meta-carbonatites show characteristically high rare earth element(REE),Sr contents and lack of negative Eu anomaly,consistent with average calcio-carbonatite values worldwide and an igneous origin.More than 0.5 wt.% SrO in the metacarbonatites and SrO> 0.15 wt.% in constituent rock forming calcite are discriminating signatures of the Jambil meta-carbonatites.Chemically,the Jambil meta-carbonatites are relatively depleted in Rb,Nb,Ta,Ti,Zr and Hf,relatively enriched in Ba,Th,Sr,and have a high LREE/HREE ratio when normalized to primitive mantle.Their carbon and oxygen isotope compositions vary from-3.5‰ to-4.3‰and from 9.7‰ to 12.3‰,respectively.These geochemical characteristics indicate generation of the carbonatites through small degree of partial melting from a carbonated eclogitic source.In-situ,U/Pb analysis of titanite indicates that the Jambil meta-carbonatites were emplacement at 438 ±3 Ma.When combined with regional geological observations,we interpret the emplacement of the Jambil metacarbonatites to have taken place during the Silurian back arc extension within greater Gondwana and mark a transition from a compressional tectonic regime,brought about by collision of microcontinental blocks along the northern margin of Gondwana,to post-orogenic extension in the waning stages of the pre-Himalayan Ordovician orogeny.Finally,in-situ ^(208)Pb/^(232)Th monazite dates(40.3-27.6Ma) extracted from the meta-carbonatites are consistent with the Cenozoic metamorphism of the area.Asad Khan Shah Faisal Kyle P.Larson Delores M.Robinson Huan Li Zaheen Ullah Mark Button Javed Nawab Muhammad Farhan Liaqat Ali Muhammad Ali 2023Journal of Earth Science2023,34,1:0

14Dynamics of Submersible Mussel Rafts in Waves and Current显示文摘To investigate the dynamics of submersible mussel rafts, the finite element program Aqua-FETM, developed by the University of New Hampshire(UNH), was applied to rafts moored at the surface and submerged. The submerged configuration is used to reduce wave forcing and to avoid contact with floating ice during winters in northern waters. Each raft consists of three pontoons connected by a grid framework. Rafts are intended to support densely spaced mussel ropes hung from the framework. When submerged, the pontoons are flooded, and the raft is held vertically by floats attached by lines. The computer models were developed in Aqua-FE? to simulate the effects of waves and current. They were validated by comparison with wave tank results by use of a 1/10 scale raft physical model. Comparisons showed good agreement for the important heave(vertical) and pitch(rotational) motions, though there was a tendency towards conservative results for wave and current drag. Full-scale simulations of surface and submerged single raft and two rafts connected in tandem were performed. Submerged raft wave response was found to be reduced relative to that at the surface for both the single and two-raft configurations. In particular, the vertical motion of mussel rope connection points was significantly reduced by submergence, resulting in reduced potential for mussel drop-off.
