1Comparison of the toxicity of silver nanoparticles and silver ions on the growth of terrestrial plant model Arabidopsis thaliana显示文摘Silver nanoparticles (AgNPs) are one of the most widely used nanomaterials, but the mechanism of AgNP toxicity in terrestrial plants is still unclear. We compared the toxic effects of AgNPs and Ag+ on Arabidopsis thaliana at the physiological, ultrastructural and molecular levels. AgNPs did not affect seed germination; however, they showed stronger inhibitory effect on root elongation than Ag+ . The results of transmission electron microscopy and metal content analysis showed that AgNPs could be accumulated in leaves. These absorbed AgNPs disrupted the thylakoid membrane structure and decreased chlorophyll content, which can inhibit plant growth. By comparison, a small amount of Ag+ was absorbed by seedlings, and it did not pronouncedly affect chloroplast structure and other metal ion absorption as AgNPs did. Compared with Ag+ , AgNPs could alter the transcription of antioxidant and aquaporin genes, indicating that AgNPs changed the balance between the oxidant and antioxidant systems, and also affected the homeostasis of water and other small molecules within the plant body. 2Gold nanorods together with HSP inhibitor-VER-155008 micelles for colon cancer mild-temperature photothermal therapy显示文摘Enhancing the heat-sensitivity of tumor cells provides an alternative solution to maintaining the therapeutic outcome of photothermal therapy(PTT). In this study, we constructed a therapeutic system, which was composed of methoxy-polyethylene-glycol-coated-gold-nanorods(MPEG-AuN R) and VER-155008-micelles, to evaluate the effect of VER-155008 on the sensitivity of tumor cells to heat, and further investigate the therapeutic outcome of MPEG-AuN R mediated PTT combined with VER-155008-micelles. VER-155008-micelles downregulate the expression of heat shock proteins and attenuate the heat-resistance of tumor cell. The survival of HCT116 cells treated with VER-155008-micelles under 45 ℃ is equal to that treated with high temperature hyperthermia(55 ℃) in vitro. Furthermore, we proved either the MPEG-AuN R or VER-155008-micelles can be accumulate in the tumor site by photoacoustic imaging and fluorescent imaging. In vivo anti-cancer evaluation showed that tumor size remarkably decreased(smaller than 100 mm^3 or vanished) when treated with combing 45℃ mild PTT system, which contrasted to the tumor size when treated with individual 45℃ mild PTT(around 500nm^3) or normal saline as control(larger than 2000 nm^3). 4IL-17 sustains the plasma cell response via p38-mediated Bcl-xL RNA stability in lupus pathogenesis显示文摘Recent studies have demonstrated a central role for plasma cells in the development of autoimmune diseases,such as systemic lupus erythematosus(SLE).Currently,both the phenotypic features and functional regulation of autoreactive plasma cells during SLE pathogenesis remain largely unclear.In this study,we first found that a major subset of IL-17 receptor-expressing plasma cells potently produced anti-dsDNA IgG upon IL-17A(IL-17)stimulation in SLE patients and lupus mice.Using a humanized lupus mouse model,we showed that the transfer of Th17 cell-depleted PBMCs from lupus patients resulted in a significantly reduced plasma cell response and attenuated renal damage in recipient mice compared to the transfer of total SLE PBMCs.Moreover,long-term BrdU incorporation in lupus mice detected highly enriched long-lived BrdU+subsets among IL-17 receptor-expressing plasma cells.Lupus mice deficient in IL-17 or IL-17 receptor C(IL-17RC)exhibited a diminished plasma cell response and reduced autoantibody production with attenuated renal damage,while the adoptive transfer of Th17 cells triggered the plasma cell response and renal damage in IL-17-deficient lupus mice.In reconstituted chimeric mice,IL-17RC deficiency resulted in severely impaired plasma cell generation but showed no obvious effect on germinal center B cells.Further mechanistic studies revealed that IL-17 significantly promoted plasma cell survival via p38-mediated Bcl-xL transcript stabilization.Together,our findings identified a novel function of IL-17 in enhancing plasma cell survival for autoantibody production in lupus pathogenesis,which may provide new therapeutic strategies for the treatment of SLE.Kongyang Ma Wenhan Du Fan Xiao Man Han Enyu Huang Na Peng Yuan Tang Chong Deng Lixiong Liu Yulan Chen Jingjing Li Shiwen Yuan Qin Huang Xiaoping Hong Dajun Hu Xiaoyan Cai Quan Jiang Dongzhou Liu Liwei Lu 2021Cellular & Molecular Immunology2021,18,7:6

6Characteristics on microstructure and mechanical performances of 6111Al influenced by Ce-containing precipitates显示文摘In this work,the effects of Ce addition(0,0.1 wt%,0.3 wt%,0.5 wt%and 0.7 wt%)on the evolution of microstructure and mechanical properties of 6111 Al alloy and strengthening mechanism of 6111 Al-Ce alloy were systematically investigated by a polarizing microscope,a scanning electronic microscope,an energy dispersive spectroscope and a high-resolution transmission electron microscope.The results indicate that with 0.3 wt%Ce addition,theα-Al grains show the equiaxed crystal morphology with the average size decreasing from 137 to 57μm and numerous small AlCeSi phases with lump-like or platelike morphology are distributed closely along the grain boundary.The peak yield strength,ultimate tensile strength and elongation of 6111 Al-Ce alloy reach to 279 MPa,316 MPa and 12.1%,respectively,which is attributed to the grain refinement strengthening and the formation of nanosized Al11Ce3 precipitates.Eventually,this investigation gives us instructive suggestion to prepare the new kind of aluminum alloy with high strength and high ductility.Xu Gao Yutao Zhao Xizhou Kai Wei Qian Liwei Jin Chuang Guan Peng Sheng 2022Journal of Rare Earths2022,40,1:3
7A polyimide cathode with superior stability and rate capability for lithium-ion batteries显示文摘Organic-based electrode materials for lithium-ion batteries (LIBs) are promising due to their high theoretical capacity,structure versatility and environmental benignity.However,the poor intrinsic electric conductivity of most polymers results in slow reaction kinetics and hinders their application as electrode materials for LIBs.A binder-free self-supporting organic electrode with excellent redox kinetics is herein demonstrated via in situ polymerization of a uniform thin polyimide (PI) layer on a porous and highly conductive carbonized nanofiber (CNF) framework.The PI active material in the porous PI@CNF film has large physical contact area with both the CNF and the electrolyte thus obtains superior electronic and ionic conduction.As a result,the PI@CNF cathode exhibits a discharge capacity of 170 mAh·g^-1 at 1 C (175 mA·g^-1),remarkable rate-performance (70.5% of 0.5 C capacity can be obtained at a 100 C discharge rate),and superior cycling stability with 81.3% capacity retention after 1,000 cycles at 1 C.Last but not least,a four-electron transfer redox process of the PI polymer was realized for the first time thanks to the excellent redox kinetics of the PI@CNF electrode,showing a discharge capacity exceeding 300 mAh·g^-1 at a current of 175 mA·g^-1.Jianghui Zhao Tuo Kang Yanli Chu Peng Chen Feng Jin Yanbin Shen Liwei Chen 2019Nano Research2019,12,6:3
8Diffuse Intrinsic Pontine Gliomas Exhibit Cell Biological and Molecular Signatures of Fetal Hindbrain-Derived Neural Progenitor Cells显示文摘Diffuse intrinsic pontine glioma(DIPG) is the main cause of brain tumor-related death among children.Until now, there is still a lack of effective therapy with prolonged overall survival for this disease. A typical strategy for preclinical cancer research is to find out the molecular differences between tumor tissue and para-tumor normal tissue, in order to identify potential therapeutic targets. Unfortunately, it is impossible to obtain normal tissue for DIPG because of the vital functions of the pons.Here we report the human fetal hindbrain-derived neural progenitor cells(pontine progenitor cells, PPCs) as normal control cells for DIPG. The PPCs not only harbored similar cell biological and molecular signatures as DIPG glioma stem cells, but also had the potential to be immortalized by the DIPG-specific mutation H3 K27 M in vitro. 9Spatial expansion effects on urban ecosystem services supply-demand mismatching in Guanzhong Plain Urban Agglomeration of China显示文摘Global urbanization has led to drastic land use change,interfering the ecosystem services(ES)supply-demand balance,in turn threatening the well-being of humans.However,existing studies mainly stranded at the historical and current analysis,and the effects of urban spatial expansion on the relationship between ES supply and demand in the future are less clear,in particular at an urban agglomeration scale.This study was constructed with a framework of assessing the effects of urban spatial expansion on ES supply-demand mismatching under different future scenarios in the Guanzhong Plain Urban Agglomeration(GPUA)by using the Future Land Use Simulation(FLUS)model and expert-based Land-Use and Land-Cover Change(LUCC)matrix.The results showed that:(1)Urban expansion is significant in the natural development(ND)scenario,mainly manifesting the great transfer of dry land to construction land.(2)The gap between total ES supply and demand is narrowed from 2000 to 2030 and the mismatch between ES supply and demand is mainly reflected in the spatial distribution pattern in the GPUA.The ES budgets were in high surplus in Northern Qinling Mountains and northeast mountain areas,while they were in severe deficit in urban center areas.The budgets deficit under the ND scenario in 2030 is the most severe.(3)The gradient differences of ES budgets of the GPUA between urban centers and suburbs increase from 2000 to 2030 under two scenarios.The deficit region expands largest under ND scenario.The findings revealed that ES declining and supply-demand mismatching were triggered by the drastic land-use change driven by rapid urban expansion.The expansion has brought about an increasing material demand and growing industries,threatening the sustainability of ecosystems.Scenarios setting could contribute to coordinating the relationship between future urban development and ecological protection,and the policy strategies proposed in the study could inform ecological management and urban planning in the regions facing the similar urbanization situation.PENG Lixian ZHANG Liwei LI Xupu WANG Zhuangzhuang WANG Hao JIAO Lei 2022Journal of Geographical Sciences2022,32,5:2

11Early Age of Onset, Multiple Primary Malignancies and Poor Prognosis Are Indicative of an Inherited Predisposition to Esophageal Squamous Cell Carcinoma in Familial Rather Than Sporadic Disease- An Update Based on 14- to 23-year Follow-up显示文摘<正> OBJECTIVE To demonstrate the effects of an inheritedpredisposition to familial esophageal squamous cellcarcinoma (ESCC) through the comparison and analysis of theclinicopathologic differences between familial and sporadic ESCCcases.METHODS Differences in age of onset, prevalence rates ofdouble primary ESCC, and survival rates between familial ESCC (n= 476) and sporadic ESCC cases (n = 1226) were analyzed.RESULTS Overall, familial ESCC cases showed a significantlyyounger age of onset (51.9 ± 8.2 vs. 53.4 ± 8.0, P_(t-test) = 0.00), asignificantly higher prevalence rate for double ESCC (2.73 % vs.1.22%, adjusted with TNM:χ_(MH)~2= 4.029, P = 0.045), and a lowersurvival rate than in sporadic cases (P_(wald) = 0.04). The familial casesshowed both a younger age of onset and poorer survival in mostsubgroups, and the differences were more marked in early-stagerather than in the late-stage disease groups.CONCLUSION Theses findings confirm the existence of familialas opposed to sporadic ESCC. 12Single-cell transcriptome analysis reveals the regulatory effects of artesunate on splenic immune cells in polymicrobial sepsis显示文摘Sepsis is characterized by a severe and life-threatening host immune response to polymicrobial infection accompanied by organ dysfunction.Studies on the therapeutic effect and mechanism of immunomodulatory drugs on the sepsis-induced hyperinflammatory or immunosuppression states of various immune cells remain limited.This study aimed to investigate the protective effects and underlying mechanism of artesunate(ART)on the splenic microenvironment of cecal ligation and puncture-induced sepsis model mice using single-cell RNA sequencing(scRNA-seq)and experimental validations.The scRNA-seq analysis revealed that ART inhibited the activation of pro-inflammatory macrophages recruited during sepsis.ART could restore neutrophils’chemotaxis and immune function in the septic spleen.It inhibited the activation of T regulatory cells but promoted the cytotoxic function of natural killer cells during sepsis.ART also promoted the differentiation and activity of splenic B cells in mice with sepsis.These results indicated that ART could alleviate the inflammatory and/or immunosuppressive states of various immune cells involved in sepsis to balance the immune homeostasis within the host.Overall,this study provided a comprehensive investigation of the regulatory effect of ART on the splenic microenvironment in sepsis,thus contributing to the application of ART as adjunctive therapy for the clinical treatment of sepsis.Jiayun Chen Xueling He Yunmeng Bai Jing Liu Yin Kwan Wong Lulin Xie Qian Zhang Piao Luo Peng Gao Liwei Gu Qiuyan Guo Guangqing Cheng Chen Wang Jigang Wang 2023Journal of Pharmaceutical Analysis2023,13,7:1

17Tetrahedral framework nucleic acids promote the biological functions and related mechanism of synovium-derived mesenchymal stem cells and show improved articular cartilage regeneration activity in situ显示文摘Many recent studies have shown that joint-resident mesenchymal stem cells(MSCs)play a vital role in articular cartilage(AC)in situ regeneration.Specifically,synovium-derived MSCs(SMSCs),which have strong chondrogenic differentiation potential,may be the main driver of cartilage repair.However,both the insufficient number of MSCs and the lack of an ideal regenerative microenvironment in the defect area will seriously affect the regeneration of AC.Tetrahedral framework nucleic acids(tFNAs),notable novel nanomaterials,are considered prospective biological regulators in biomedical engineering.Here,we aimed to explore whether tFNAs have positive effects on AC in situ regeneration and to investigate the related mechanism.The results of in vitro experiments showed that the proliferation and migration of SMSCs were significantly enhanced by tFNAs.In addition,tFNAs,which were added to chondrogenic induction medium,were shown to promote the chondrogenic capacity of SMSCs by increasing the phosphorylation of Smad2/3.In animal models,the injection of tFNAs improved the therapeutic outcome of cartilage defects compared with that of the control treatments without tFNAs.In conclusion,this is the first report to demonstrate that tFNAs can promote the chondrogenic differentiation of SMSCs in vitro and enhance AC regeneration in vivo,indicating that tFNAs may become a promising therapeutic for AC regeneration.Liwei Fu Pinxue Li Junyao Zhu Zhiyao Liao Cangjian Gao Hao Li Zhen Yang Tianyuan Zhao Wei Chen Yu Peng Fuyang Cao Chao Ning Xiang Sui Quanyi Guo Yunfeng Lin Shuyun Liu 2022Bioactive Materials2022,7,3:1
