1Baicalin induces ferroptosis in bladder cancer cells by downregulating FTH1显示文摘Ferroptosis is a non-apoptotic regulated cell death caused by iron accumulation and subsequent lipid peroxidation.Currently,the therapeutic role of ferroptosis on cancer is gaining increasing interest.Baicalin an active component in Scutellaria baicalensis Georgi with anticancer potential various cancer types;however,the effects of baicalein on bladder cancer and the underlying molecular mechanisms remain largely unknown.In the study,we investigated the effect of baicalin on bladder cancer cells5637 and KU-19-19.As a result,we show baicalin exerted its anticancer activity by inducing apoptosis and cell death in bladder cancer cells.Subsequently,we for the first time demonstrate baicalin-induced ferroptotic cell death in vitro and in vivo,accompanied by reactive oxygen species(ROS) accumulation and intracellular chelate iron enrichment.The ferroptosis inhibitor deferoxamine but not necrostatin-1,chloroquine(CQ),N-acetyl-L-cysteine,L-glutathione reduced,or carbobenzoxy-valyl-alanyl-aspartyl-[O-methyl]-fluoromethylketone(Z-VAD-FMK) rescued baicalin-induced cell death,indicating ferroptosis contributed to baicalin-induced cell death.Mechanistically,we show that ferritin heavy chain1(FTH1) was a key determinant for baicalin-induced ferroptosis.Overexpression of FTH1 abrogated the anticancer effects of baicalin in both 5637 and KU19-19 cells.Taken together,our data for the first time suggest that the natural product baicalin exerts its anticancer activity by inducing FTH1-dependent ferroptosis,which will hopefully provide a prospective compound for bladder cancer treatment.Na Kong Xiaying Chen Jiao Feng Ting Duan Shuiping Liu Xueni Sun Peng Chen Ting Pan Lili Yan Ting Jin Yu Xiang Quan Gao Chengyong Wen Weirui Ma Wencheng Liu Mingming Zhang Zuyi Yang Wengang Wang Ruonan Zhang Bi Chen Tian Xie Xinbing Sui Wei Tao 2021Acta Pharmaceutica Sinica B2021,11,12:23
2Curcumenol triggered ferroptosis in lung cancer cells via lncRNA H19/miR-19b-3p/FTH1 axis显示文摘Curcumenol,an effective ingredient of Wenyujin,has been reported that exerted its antitumor potential in a few cancer types.However,the effect and molecular mechanism of curcumenol in lung cancer are largely unknown.Here,we found that curcumenol induced cell death and suppressed cell proliferation in lung cancer cells.Next,we demonstrated that ferroptosis was the predominant method that contributed to curcumenol-induced cell death of lung cancer in vitro and vivo for the first time.Subsequently,using RNA sequencing,we found that the long non-coding RNA H19(lncRNA H19)was significantly downregulated in lung cancer cells treated with curcumenol,when compared to untreated controls.Overexpression of lncRNA H19 eliminated the anticancer effect of curcumenol,while lncRNA H19 knockdown promoted ferroptosis induced by curcumenol treatment.Mechanistically,we showed that lncRNA H19 functioned as a competing endogenous RNA to bind to miR-19b-3p,thereby enhanced the transcription activity of its endogenous target,ferritin heavy chain 1(FTH1),a marker of ferroptosis.In conclusion,our data show that the natural product curcumenol exerted its antitumor effects on lung cancer by triggering ferroptosis,and the lncRNA H19/miR-19b-3p/FTH1 axis plays an essential role in curcumenol-induced ferroptotic cell death.Therefore,our findings will hopefully provide a valuable drug for treating lung cancer patients.Ruonan Zhang Ting Pan Yu Xiang Mingming Zhang Han Xie Zimao Liang Bi Chen Cong Xu Jing Wang Xingxing Huang Qianru Zhu Ziming Zhao Quan Gao Chengyong Wen Wencheng Liu Weirui Ma Jiao Feng Xueni Sun Ting Duan Elaine Lai-Han Leung Tian Xie Qibiao Wu Xinbing Sui 2022Bioactive Materials2022,7,7:8
3Feature Selection Method Based on Class Discriminative Degree for Intelligent Medical Diagnosis显示文摘By using efficient and timely medical diagnostic decision making,clinicians can positively impact the quality and cost of medical care.However,the high similarity of clinical manifestations between diseases and the limitation of clinicians’knowledge both bring much difficulty to decision making in diagnosis.Therefore,building a decision support system that can assist medical staff in diagnosing and treating diseases has lately received growing attentions in the medical domain.In this paper,we employ a multi-label classification framework to classify the Chinese electronic medical records to establish corresponding relation between the medical records and disease categories,and compare this method with the traditional medical expert system to verify the performance.To select the best subset of patient features,we propose a feature selection method based on the composition and distribution of symptoms in electronic medical records and compare it with the traditional feature selection methods such as chi-square test.We evaluate the feature selection methods and diagnostic models from two aspects,false negative rate(FNR)and accuracy.Extensive experiments have conducted on a real-world Chinese electronic medical record database.The evaluation results demonstrate that our proposed feature selection method can improve the accuracy and reduce the FNR compare to the traditional feature selection methods,and the multi-label classification framework have better accuracy and lower FNR than the traditional expert system.Shengqun Fang Zhiping Cai Wencheng Sun Anfeng Liu Fang Liu Zhiyao Liang Guoyan Wang 2018Computers, Materials & Continua2018,,6:5

6Texture synthesis via the matching compatibility between patches显示文摘A new patch-based texture synthesis method is presented in this paper. By the method, a set of patches that can be matched with a sampled patch for growing textures effectively, called the matching compat- ibility between patches, is generated first for each patch, and the set is further optimized by culling the patches that may cause synthesis confficts. In this way, similarity measurement calculation for selecting suitable patches in texture synthesis can be greatly saved, and synthesis confficts between neighbouring patches are substantially reduced. Furthermore, retrace computation is integrated in the synthesis process to improve the texture quality. As a result, the new method can produce high quality textures as texture optimization, the best method to date for producing good textures, and run in a time complexity linear to the size of the output texture. 7A Stiffness Surface Method to Analyze the Cross-Sectional Mechanical Properties of Reinforced Thermoplastic Pipes Subjected to Axisymmetric Loads显示文摘Axial and hoop stiffness can describe the elastic responses of reinforced thermoplastic pipes(RTPs)subjected to axisymmetric loads,such as tension,compression,pressure,and crushing loads.However,an accurate analytical prediction cannot be provided because of the anisotropy of RTP laminates.In the present study,a stiffness surface method,in which the analytical expressions of the axial and hoop stiffness are derived as two concise formulas,is proposed.The axial stiffness formula is obtained by solving the equilibrium equations of RTPs under a uniaxial stress state based on the homogenization assumption,whereas the hoop stiffness formula is derived from the combination of the elastic stability theory,the classical lamination theory,and NASA SP-8007 formula.To verify the proposed method,three types of RTPs are modeled to conduct the quasi-static analyses of the tension and crushing cases.The consistency between numerical and analytical results verifies the effectiveness of the proposed method on the prediction of the axial and hoop stiffness of RTPs,which also proves the existence of stiffness surfaces.As the axial stiffness is proportional to the radii,the axial stiffness surface consists of a series of straight lines,which can be used to predict both thin-walled and thick-walled RTPs.Meanwhile,the hoop stiffness is more applicable for thin-walled RTPs because the proposed method ignores the proportional relationship between the homogenized hoop elastic moduli and the reciprocal radii in helical structures.LIU Wencheng WANG Shuqing 2021Journal of Ocean University of China2021,20,4:2
8Iron oxide nanoparticles promote vascular endothelial cells survival from oxidative stress by enhancement of autophagy显示文摘Dextran-coated superparamagnetic iron oxide nanoparticles(Dex-SPIONs)are excellent magnetic resonance imaging contrast agents for disease diagnosis and therapy.They can be delivered to target tissues mainly though vascular endothelium cells,which are major targets of oxidative stress.In cardiovascular cells,autophagy serves primarily on a pro-survival approach that protects the cells from oxidative stress even some autophagy inducers have been developed for adjuvant therapy of cardiovascular disorders.Our study demonstrated that the nanoparticles could be taken up by human umbilical vein endothelial cells(HUVECs)without causing obvious cytotoxicity but triggering autophagy.Furthermore,our results revealed that Dex-SPIONs could enhance HUVECs survival and reverse the reduction of nitric oxide secretion under the condition of H2O2 damage.However,these effects could be diminished by the autophagy inhibitor.In particular,we discovered that Dex-SPIONs evoked autophagy in HUVECs by reducing the phosphorylation of PRAS40,an upstream regulator of autophagy initiation.These results suggested that Dex-SPIONs functions as an autophagic-related antioxidant in HUVECs which may be utilized as an adjuvant therapy to cardiovascular disease associated with oxidative stress.Jimei Duan Jiuju Du Rongrong Jin Wencheng Zhu Li Liu Li Yang Mengye Li Qiyong Gong Bin Song James MAnderson and Hua Ai 2019Regenerative Biomaterials2019,6,4:2
9A natural allele of TAW1 contributes to high grain number and grain yield in rice显示文摘Grain number per panicle (GNP) is a complex trait controlled by quantitative trait loci (QTL),directly determining grain yield in rice.Identifying GNP-associated QTL is desirable for increasing rice yield.A rice chromosome segment substitution line (CSSL),F771,which showed increased panicle length and GNP,was identified in a set of CSSLs derived from a cross between two indica cultivars,R498 (recipient) and WY11327 (donor).Genetic analysis showed that the panicle traits in F771 were semidominant and controlled by multiple QTL.Six QTL were consistently identified by QTL-seq analysis.Among them,the major QTL q PLN10 for panicle length and GNP was localized to a 121-kb interval between markers N802 and N909 on chromosome 10.Based on quantitative real-time PCR and sequence analysis,TAWAWA1(TAW1),a known regulator of rice inflorescence architecture,was identified as the candidate gene for q PLN10.A near-isogenic line,NIL-TAW1,was developed to evaluate its effects.In comparison with the recurrent parent R498,NIL-TAW1 showed increased panicle length (14.0%),number of secondary branches (20.9%) and GNP (22.0%),and the final grain yield per plant of NIL-TAW1 was increased by18.6%.Transgenic experiments showed that an appropriate expression level of TAW1 was necessary for panicle development.Haplotype analysis suggested that the favorable F771-type (Hap 13) of TAW1was introduced from aus accessions and had great potential value in high-yield breeding both in indica and japonica varieties.Our results provide a promising genetic resource for rice grain yield improvement.Hua Yuan Zhengyan Xu Xueqin Tan Peng Gao Mengya Jin Wencheng Song Shiguang Wang Yunhai Kang Peixiong Liu Bin Tu Yuping Wang Peng Qin Shigui Li Bingtian Ma Weilan Chen 2021The Crop Journal2021,9,5:2

12SARS-CoV-2 impairs the disassembly of stress granules and promotes ALS-associated amyloid aggregation显示文摘The nucleocapsid(N)protein of SARS-CoV-2 has been reported to have a high ability of liquid-liquid phase separation,which enables its incorporation into stress granules(SGs)of host cells.However,whether SG invasion by N protein occurs in the scenario of SARS-CoV-2 infection is unknow,neither do we know its con-sequence.Here,we used SARS-CoV-2 to infect mam-malian cells and observed the incorporation of N protein into SGs,which resulted in markedly impaired self-dis-assembly but stimulated cell cellular clearance of SGs.NMR experiments further showed that N protein binds to the SG-related amyloid proteins via non-specific tran-sient interactions,which not only expedites the phase transition of these proteins to aberrant amyloid aggre-gation in vitro,but also promotes the aggregation of FUS with ALS-associated P525L mutation in cells.In addition,we found that ACE2 is not necessary for the infection of SARS-CoV-2 to mammalian cells.Our work indicates that SARS-CoV-2 infection can impair the dis-assembly of host SGs and promote the aggregation of SG-related amyloid proteins,which may lead to an increased risk of neurodegeneration.Yichen Li Shuaiyao Lu Jinge Gu Wencheng Xia Shengnan Zhang Shenqing Zhang Yan Wang Chong Zhang Yunpeng Sun Jian Lei Cong Liu Zhaoming Su Juntao Yang Xiaozhong Peng Dan Li 2022Protein & Cell2022,13,8:1

17Lactosylated N-Alkyl polyethylenimine coated iron oxide nanoparticles induced autophagy in mouse dendritic cells显示文摘Dendritic cell(DC)-based vaccines have shown promising therapeutic results in cancer and some immune disorders.It is critical to track in vivo migration behaviours of DCs and monitor the whole process dynamically and non-invasively.Superparamagnetic iron oxide(SPIO)nanoparticles are chosen for DC labelling under magnetic resonance imaging(MRI)because of their proven biosafety as contrast agents.However,when used for cell labelling,sensitive biological indicators such as cell autophagy may be helpful to better understand the process and improve the probe design.Here,lactosylated N-Alkyl polyethylenimine coated SPIO nanoparticles are used for DC labelling.This probe shows satisfactory cell labelling efficiency and low cytotoxicity.In this study,autophagy was used as a key factor to understand how DCs react to nanoparticles after labelling.Our results demonstrate that the nanoparticles can induce protective autophagy in DCs,as inhibition of the autophagy flux could lead to cell death.Meanwhile,the nanoparticles induced autophagy could promote DC maturation which is an essential process for its migration and antigen presentation.Autophagy induced DC maturation is known to enhance the vaccine functions of DCs,therefore,our results suggest that beyond the MRI tracking ability,this probe might enhance therapeutic immune activation as well.Taipeng Shen Wencheng Zhu Li Yang Li Liu Rongrong Jin Jimei Duan James M.Anderson Hua Ai 2018Regenerative Biomaterials2018,5,3:1
