| 1 | Purification and Characterization of a New Thermostable κ-CarrageenasefromtheMarineBacterium Pseudoalteromonas sp. QY203显示文摘A new extracellular κ-carrageenase, namely CgkP, 34.0 kDa in molecular weight, was purified from Pseudoalteromonas sp. QY203. CgkP showed relatively high activity at acidities ranging from pH6.0 to pH9.0 and temperatures ranging from 30℃ to 50℃ with the highest activity at 45℃ and pH7.2. Sodium chloride increased its activity markedly, and KCl increased its activity slightly. The divalent and trivalent metal ions including Cu2+ , Ni2+ , Zn2+ , Mn2+ , Al3+ and Fe3+ significantly inhibited its activity, while Mg2+ did not. CgkP remained 70% of original activity after being incubated at 40℃ for 48 h, and remained 80% of the activity after being incubated at 45℃ for 1 h. It exhibited endo-κ-carrageenase activity, mainly depolymerizing the κ-carrageenan into disaccharide and tetrasaccharide. | 2 | Increased leptin by hypoxic-preconditioning promotes autophagy of mesenchymal stem cells and protects them from apoptosis显示文摘Autophagy is the basic catabolic progress involved in cell degradation of unnecessary or dysfunctional cellular components.It has been proven that autophagy could be utilized for cell survival under stresses.Hypoxic-preconditioning(HPC)could reduce apoptosis induced by ischemia and hypoxia/serum deprivation(H/SD)in bone marrow-derived mesenchymal stem cells(BMSCs).Previous studies have shown that both leptin signaling and autophagy activation were involved in the protection against apoptosis induced by various stress,including ischemia-reperfusion.However,it has never been fully understood how leptin was involved in the protective effects conferred by autophagy.In the present study,we demonstrated that HPC can induce autophagy in BMSCs by increased LC3-II/LC3-I ratio and autophagosome formation.Interestingly,similar effects were also observed when BMSCs were pretreated with rapamycin.The beneficial effects offered by HPC were absent when BMSCs were incubated with autophagy inhibitor,3-methyladenine(3-MA).In addition,down-regulated leptin expression by leptin-shRNA also attenuated HPC-induced autophagy in BMSCs,which in turn was associated with increased apoptosis after exposed to sustained H/SD.Furthermore,increased AMP-activated protein kinase phosphorylation and decreased mammalian target of rapamycin phosphorylation that were observed in HPC-treated BMSCs can also be attenuated by down-regulation of leptin expression.Our data suggests that leptin has impact on HPC-induced autophagy in BMSCs which confers protection against apoptosis under H/SD,possibly through modulating both AMPK and mTOR pathway. | WANG LiHan HU XinYang ZHU Wei JIANG Zhi ZHOU Yu CHEN PanPan WANG JianAn | 2014 | Science China(Life Sciences)2014,57,2: | 8 |
| 3 | Preferential adsorption behaviour of CH_4 and CO_2 on high-rank coal from Qinshui Basin,China显示文摘In order to better understand the prevailing mechanism of CO2 storage in coal and estimate CO2 sequestration capacity of a coal seam and enhanced coalbed methane recovery(ECBM) with CO2 injection into coal, we investigated the preferential adsorption of CH4 and CO2on coals. Adsorption of pure CO2,CH4 and their binary mixtures on high-rank coals from Qinshui Basin in China were employed to study the preferential adsorption behaviour. Multiple regression equations were presented to predict CH4 equilibrium concentration from equilibrium pressure and its initial-composition in feed gas. The results show that preferential adsorption of CO2 on coals over the entire pressure range under competitive sorption conditions was observed, however, preferential adsorption of CH4 over CO2on low-volatile bituminous coal from higher CH4-compostion in source gas was found at up to 10 MPa pressure. Preferential adsorption of CO2 increases with increase of CH4 concentration in source gas, and decreases with increasing pressure. Although there was no systematic investigation of the effect of coal rank on preferential adsorption, there are obvious differences in preferential adsorption of gas between low-volatile bituminous coal and anthracite. | 4 | Purification of starch and phosphorus wastewater using core-shell magnetic seeds prepared by sulfated roasting显示文摘Core-shell magnetic seeds with certain adsorption capacity that were prepared by sulfated roasting, served as the core of a magnetic separation technology for purification of starch wastewater. XRD and SEM results indicate that magnetite's surface transformed to be porous α-Fe_2O_3 structure. Compared with magnetite particles, the specific surface area was significantly improved to be 8.361 from 2.591 m^2/g, with little decrease in specific susceptibility. Zeta potential, FT-IR and XPS experiments indicate that both phosphate and starch adsorbed on the surface of the core-shell magnetic seeds by chemical adsorption, which fits well with the Langmuir adsorption model. | 7 | Inhibitor of DNA binding 2 accelerates nerve regeneration after sciatic nerve injury in mice显示文摘Inhibitor of DNA binding 2(Id2)can promote axonal regeneration after injury of the central nervous system.However,whether Id2 can promote axonal regeneration and functional recovery after peripheral nerve injury is currently unknown.In this study,we established a mouse model of bilateral sciatic nerve crush injury.Two weeks before injury,AAV9-Id2-3×Flag-GFP was injected stereotaxically into the bilateral ventral horn of lumbar spinal cord.Our results showed that Id2 was successfully delivered into spinal cord motor neurons projecting to the sciatic nerve,and the number of regenerated motor axons in the sciatic nerve distal to the crush site was increased at 2 weeks after injury,arriving at the tibial nerve and reinnervating a few endplates in the gastrocnemius muscle.By 1 month after injury,extensive neuromuscular reinnervation occurred.In addition,the amplitude of compound muscle action potentials of the gastrocnemius muscle was markedly recovered,and their latency was shortened.These findings suggest that Id2 can accelerate axonal regeneration,promote neuromuscular reinnervation,and enhance functional improvement following sciatic nerve injury.Therefore,elevating the level of Id2 in adult neurons may present a promising strategy for peripheral nerve repair following injury.The study was approved by the Experimental Animal Ethics Committee of Jinan University(approval No.20160302003)on March 2,2016. | Zhong-Hai Huang Ai-Ying Feng Jing Liu Libing Zhou Bing Zhou Panpan Yu | 2021 | Neural Regeneration Research2021,16,12: | 2 |

| 11 | Effect of chondroitin sulfate proteoglycans on neuronal cell adhesion, spreading and neurite growth in culture显示文摘As one major component of extracellular matrix(ECM) in the central nervous system, chondroitin sulfate proteoglycans(CSPGs) have long been known as inhibitors enriched in the glial scar that prevent axon regeneration after injury. Although many studies have shown that CSPGs inhibited neurite outgrowth in vitro using different types of neurons, the mechanism by which CSPGs inhibit axonal growth remains poorly understood. Using cerebellar granule neuron(CGN) culture, in this study, we evaluated the effects of different concentrations of both immobilized and soluble CSPGs on neuronal growth, including cell adhesion, spreading and neurite growth. Neurite length decreased while CSPGs concentration arised, meanwhile, a decrease in cell density accompanied by an increase in cell aggregates formation was observed. Soluble CSPGs also showed an inhibition on neurite outgrowth, but it required a higher concentration to induce cell aggregates formation than coated CSPGs. We also found that growth cone size was significantly reduced on CSPGs and neuronal cell spreading was restrained by CSPGs, attributing to an inhibition on lamellipodial extension. The effect of CSPGs on neuron adhesion was further evidenced by interference reflection microscopy(IRM) which directly demonstrated that both CGNs and cerebral cortical neurons were more loosely adherent to a CSPG substrate. | 14 | Multi-objective and multi-scheme research on water and sediment regulation potential of reservoirs in the upper Yellow River显示文摘The problems of the deteriorating relation between water and sediment,and the escalating conflict between water supply and demand in the upper Yellow River(YR),need to be addressed.Reservoirs of Longyangxia,Liujiaxia,and Heishanxia(Long-Liu-Hei)in the upper YR are taken as the research object,and a multi-objective,water-sediment optimal operation model for cascade reservoirs Long-Liu-Hei has been developed.This operation model considers the comprehensive requirements of water supply,power generation,flood control,and especially,water and sediment regulation.The improved Non-dominated Sorting Genetic Algorithms(NSGA-II)method is used to demonstrate the water and sediment regulation potential of three scenario schemes under long-term conditions,and the adjustable water allocation of four schemes under typical year conditions are calculated.It is recommended to use the mode of generating hydropower depending on the available water amount as the best operating mode under the conditions of a long time series and for typical years.This recommended operation mode has verified the rationality and accuracy of the reservoir operations model.At the same time,compared with the calculation results for water and sediment regulation after the construction of the Heishanxia,the current research indicates:(1)Under the recommended model,the potential for water and sediment regulation under the long-term series and typical years is once every four years.(2)Inter-basin water transfer greatly improves the cascade power generation.When the adjustable water volume is converted into the controlled volume for water and sediment regulation,the intensity frequency of water and sediment regulation of cascade reservoirs are improved.(3)With the intensification of the contradiction between the supply of and demand for water resources in the upper YR in the future,the amount of transferable water exhibits a decreasing trend,and the regulation of water and sediment in the upper YR is facing huge challenges.(4)Through joint operation with the Longyangxia and Liujiaxia reservoirs(Long-Liu),Heishanxia will play an important role in the regulation and control of power generation,water supply,flood control,and ice prevention in the upper YR.The current research results can provide a theoretical basis and technical support for water and sediment regulation in the YR basin. | Tao Bai Jia Yu Wenting Jin Jiaquan Wan Shaojie Gou Xu Ma Panpan Ma | 2023 | International Journal of Sediment Research2023,38,2: | 1 |

| 16 | A computational model of topological and geometric recovery for visual curve completion显示文摘Visual curve completion is a fundamental problem in understanding the principles of the human visual system. This problem is usually divided into two problems: a grouping problem and a shape problem.On one hand, though perception of the visually completed curve is clearly a global task(for example,a human perceives the Kanizsa triangle only when seeing all three black objects), conventional methods for solving the grouping problem are generally based on local Gestalt laws. On the other hand, the shape of the visually completed curve is usually recovered by minimizing shape energy in existing methods.However, not only do these methods lack mechanisms to adjust the shape of the recovered visual curve using perceptual, psychophysical, and neurophysiological knowledge, but it is also difficult to calculate an explicit representation of the visually completed curve. In this paper, we present a systematic computational model for generating a visually completed curve. Firstly, based on recent studies of perception, psychophysics, and neurophysiology, we formulate a grouping procedure based on the human visual system by seeking a minimum Hamiltonian cycle in a graph, solving the grouping problem in a global manner. Secondly, we employ a B′ezier curve-based model to represent the visually completed curve. Not only is an explicit representation deduced, but we also present a means to integrate knowledge from related areas, such as perception, psychophysics, and neurophysiology, and so on.
