1Kinsenoside attenuates osteoarthritis by repolarizing macrophages through inactivating NF-κB/MAPK signaling and protecting chondrocytes显示文摘The objective was to investigate the effect of kinsenoside(Kin) treatments on macrophage polarity and evaluate the resulting protection of chondrocytes to attenuate osteoarthritis(OA) progression.RAW264.7 macrophages were polarized to M1/M2 subtypes then administered with different concentrations of Kin. The polarization transitions were evaluated with quantitative real-time polymerase chain reaction(q RT-PCR), confocal observation and flow cytometry analysis. The mechanism of Kin repolarizing M1 macrophages was evaluated by Western blot. Further, macrophage conditioned medium(CM) and IL-1β were administered to chondrocytes. Micro-CT scanning and histological observations were conducted in vivo on anterior cruciate ligament transection(ACLT) mice with or without Kin treatment. We found that Kin repolarized M1 macrophages to the M2 phenotype. Mechanistically, Kin inhibited the phosphorylation of IκBα, which further reduced the downstream phosphorylation of P65 in nuclear factor-κB(NF-κB) signaling. Moreover, Kin inhibited mitogen-activated protein kinases(MAPK) signaling molecules p-JNK, p-ERK and p-P38. Additionally, Kin attenuated macrophage CM and IL-1β-induced chondrocyte damage. In vivo, Kin reduced the infiltration of M1 macrophages,promoted M2 macrophages in the synovium, inhibited subchondral bone destruction and reduced articular cartilage damage induced by ACLT. 2Pathobiological behavior and molecular mechanism of signet ring cell carcinoma and mucinous adenocarcinoma of the stomach:A comparative study显示文摘AIM:To elucidate the distinctive pathobiological behavior between signet ring cell carcinoma (SRC) and mucinous adenocarcinoma of the stomach.METHODS: Based on the histological growth patterns and cell-functional differentiation classifications of stomach carcinoma, we conducted a series of comparative studies.All paraffin-embedded and frozen blocks were collected from the files of Cancer Institute of China Medical University. On the basis of histopathological observation, we applied enzymatic and mucous histochemistry, immunohistochemistry,flow cytometry (FCM) and molecular biology to compare these two categories of gastric cancers in terms of the DNA ploidy, proliferative kinetics, the expression of gastric carcinoma associated gene product and instabilities of mitochondrial DNA (mtDNA).RESULTS:Gastric SRC was commonly seen in females below 45 years, mostly presenting diffuse growth and ovary or uterine cervix metastasis. The majority of SRC were absorptive and mucus-producing functional differentiation type (AMPFDT), which growth relied on estrogen. Meanwhile,stomach mucinous adenocarcinomas were mostly observed in males over 50 years, prone to massive growth or nest growth and extensive peritoneal infiltration, showing two categories of cell-functional differentiation types: AMPFDT and mucus-secreting functional differentiation type (MSFDT).Expressions of ER, enzyme c-PDE and 67kDaLN-R in SRC were evidently higher than that in mucinous adenocarcinoma,while expressions of LN, CN-Ⅳ, CD44v6, and PTEN protein were obviously lower in SRC than that in mucinous adenocarcinoma (P<0.05).There was no statistic significance in VEGF, ECD and instabilities of mtDNA (P>0.05) between the above two gastric carcinomas.CONCLUSION: Though SRC and mucinous adenocarcinoma were both characterized by abundant mucus-secretion, they were quite different in morphology, ultrastructure, cell-functional differentiation and protein expression, indicating different mechanisms of carcinogenesis. 3Sleep and Cognitive Abnormalities in Acute Minor Thalamic Infarction显示文摘In order to characterize sleep and the cognitive patterns in patients with acute minor thalamic infarction(AMTI), we enrolled 27 patients with AMTI and 12 matched healthy individuals. Questionnaires about sleep and cognition as well as polysomnography(PSG) were performed on days 14 and 90 post-stroke. Compared to healthy controls, in patients with AMTI, hyposomnia was more prevalent; sleep architecture was disrupted as indicated by decreased sleep efficiency, increased sleep latency, and decreased non-rapid eye movement sleep stages 2 and 3;more sleep-related breathing disorders occurred; and cognitive functions were worse, especially memory. While sleep apnea and long-delay memory recovered to a large extent in the patients, other sleep and cognitive function deficit often persisted. Patients with AMTI are at an increased risk for hyposomnia, sleep structure disturbance,sleep apnea, and memory deficits. 5Six-year follow-up of pancreatic β cell function in adults with latent autoimmune diabetes显示文摘AIM: To investigate the characteristics of the progression of islet β cell function in Chinese latent autoimmune diabetes in adult (LADA) patients with glutamic acid decarboxylase antibody (GAD-Ab) positivity, and to explore the prognostic factors for β cell function. METHODS: Forty-five LADA patients with GAD-Ab positivity screened from phenotypic type 2 diabetic (T2DM) patients and 45 T2DM patients without GAD-Ab matched as controls were followed-up every 6 mo. Sixteen patients in LADA1 and T2DM1 groups respectively have been followed-up for 6 years, while 29 patients in LADA2 and T2DM2 groups respectively for only 1.5 years. GAD-Ab was determined by radioligand assay, and C-peptides (CP) by radioimmune assay.RESULTS: The percentage of patients whose fasting CP(FCP) decreased more than 50% compared with thebaseline reached to 25.0% at 1.5th year in LADA1 group, and FCP level decreased (395.8±71.5 vs 572.8±72.3 pmol/L, P<0.05) at 2.5th year and continuously went down to the end of follow-up. No significant changes of the above parameters were found in T2DM1 group. The average decreased percentages of FCP per year in LADA and T2DM patients were 15.8% (4.0-91.0%) and 5.2% (-3.5 to 35.5%, P= 0.000) respectively. The index of GAD-Ab was negatively correlated with the FCP in LADA patients (rs= -0.483, P = 0.000). The decreased percentage of FCP per year in LADA patients were correlated with GAD-Ab index, body mass index (BMI) and age at onset (rs = 0.408, -0.301 and -0.523 respectively, P<0.05). Moreover, GAD-Ab wasthe only risk factor for predicting βcell failure in LADA patients (B = 1.455, EXP (B) = 4.283, P = 0.023). CONCLUSION: The decreasing rate of islet β cell function in LADA, being highly heterogeneous, is three times that of T2DM patients. 6Prediction of the coupled heat radiation and conduction parameters and boundary condition using the unscented Kalman filter显示文摘The boundary emissivity, thermal conductivity, and boundary time-varying heat flux in the participating media are retrieved in this work. In the forward model, the coupled radiation-conduction heat transfer in the participating medium is resolved by the finite volume method combined with discrete ordinates method. The inverse model utilizes the temperature signals at the appropriate position of the medium as the known information and uses the unscented Kalman filter(UKF) as an optimization tool to reconstruct the boundary emissivity, thermal conductivity, and boundary time-varying heat flux. It is found that when the emissivity, thermal conductivity, and boundary time-varying heat flux are reconstructed simultaneously, only the temperature information of two locations is required. The influence of the measurement noise, sampling interval, absorption coefficient,process noise covariance, measurement noise covariance on the accuracy and stability of the retrieval results is investigated in detail. 8Longitudinal Measurement of Hemodynamic Changes within the Posterior Optic Nerve Head in Rodent Nonarteritic Anterior Ischemic Optic Neuropathy显示文摘ObjectiveTo assess the in vivo dynamic blood flow features of posterior optic nerve head (ONH) in rat model of nonarteritic anterior ischemic optic neuropathy (rNAION). MethodsrNAION was established with Rose Bengal and argon green laser in Sprague-Dawley rats. Fundus photography and fundus fluorescein angiography (FFA) were performed to assess the dynamic changes of optic disc in morphology in 90 days and in blood perfusion in 3 hours after the induction of disease. Histological examinations were performed to evaluate the success of modeling. Thedynamic blood flow kinetics of posterior ONH in rNAION were measured by Laser Doppler Flowmetry (LDF) on the day 3, 7, 14, 21, and 40 after the disease induction. One-way ANOVA, Student'st-test and Bonferroni adjustment were used for multiple comparisons of kinetic measurements of blood flow. ResultsOptic disc edema and subsequent resolution associated with the development of optic disc pallor were observed in rNAION. FFA showed that the optic disc was hypofluorescence in the early phase and hyperfluorescence in the late phase. Histological studies suggested edema and loosened tissues of ONH, loss of retinal ganglion cells (RGCs), optic nerve substance and gliosis. Compared to the naive rats, the blood flow kinetics of posterior ONH in rNAION significant reduced at each time point after modeling (F=175.06,P<0.0001). The reductions were specifically remarkable in 14 days after the disease induction (AllP<0.01). Conclusions Continuous blood perfusion reduction was found in rNAION, with significant alteration in 14 days after disease induction. 10A Liquid Metal Reaction System for Advanced Material Manufacturing显示文摘CONSPECTUS:Liquid metal exhibits the unique advantages of both the liquid and metal,including excellent deformability,high thermal conductivity,and electrical conductivity.The exploration of liquid metal is a new opportunity and revolution for the application of metal materials,including the flexible devices,catalysis,microfluidic,and drug delivery.In addition to the above applications,the characteristics of excellent inclusive ability with the majority of the elements and abundant vacancies in the bulk make it a new type of reaction medium different from traditional aqueous and organic solutions,exhibiting great potential in the precise construction of materials.To date,the research of using liquid metal as a reaction system to synthesize materials is still in its infancy.When acting as a reaction system,the vacancies inside and the smooth layering surfaces without grain boundaries allow liquid metals to encapsulate heterogeneous atoms,confine the precursors in atomically thick layers and realize the self-limiting growth of 2D material.Besides,the good rheological property makes it possible to construct 2D arrays on its surface.Except for the properties mentioned above,as a kind of metal,its excellent electrical conductivity and ductility provide a new idea for the preparation of composite materials in the energy field.Indeed,liquid metals provide attractive prospects in manufacturing advanced materials including 2D materials and functional composite materials.Thus,this Account aims to focus on the controllable fabrication of 2D materials and functional composite materials by liquid metals.Based on the characteristics of the surface layering and solidification and of excellent fluidity,the self-limited growth and ordered arrangement of 2D materials on liquid metal surfaces can be achieved,which enriches the material structures and leads to new properties.By constructing an in situ synthesis and observation system,the growth and assembly behavior of 2D materials on the liquid metal can be observed directly.Combining the electrical property,deformability,ductility,and high inclusive ability with other materials,the liquid metal reaction system can also realize the preparation of new functional composite materials toward various applications,such as the energy field.Except for the 2D materials and functional composite materials mentioned here,liquid metals also provide more possibilities for fabricating other promising materials,like wafer-scale semiconductors,magic-angle graphene,flexible functional materials,biomedical materials,and so on.The research concerning the manufacturing of advanced materials on liquid metals is still in its infancy.We believe that the development of related technologies offering in-depth investigation and theoretical understanding on liquid metals will lay a solid foundation for the basic research and practical application of more advanced materials.Mengqi Zeng Linyang Li Xiaohui Zhu Lei Fu 2021Accounts of Materials Research2021,2,8:1

15Catalytic conversion of asphaltenes to BTXN using metal-loaded modified HZSM-5显示文摘HZSM-5 zeolites with Si/Al ratios of 20,35,50 and 65 were prepared by the directing crystallization process of silicalite-1 seeds.The influence of Si/Al ratios on the production of benzene,toluene,xylene and naphthalene(BTXN)originated from asphaltenes catalytic pyrolysis was explored by adopting Py-GC/MS.Modified Z5-50 zeolites were prepared by various metal ions(Ni^(2+),Mo^(6+),Fe^(3+),and Co^(2+))with different loading rates(3%(mass),5%(mass),7%(mass),and 9%(mass))and the physical and chemical properties of these zeolites were characterized by XRD,SEM,ICP-OES,XPS,NH_(3)-TPD,FTIR,Py-IR and N_(2)adsorption-desorption isotherm.In addition,they were employed to catalyze the conversion of asphaltenes pyrolysis production to BTXN using Py-GC/MS.Results show that the highest relative content of aromatics has been obtained over HZSM-5 with Si/Al ratio of 50(Z5-50),reaching 61.87%.Besides,the loading of Ni,Mo,Fe,and Co on Z5-50 leads to an increase of acid strength and provides new active sites.The relative content of BTXN increases by 3.17%over 3Ni-Z5,which may be ascribed to that Ni promoted the conversion of polycyclic aromatic hydrocarbons(PAHs)to monocyclic aromatics due to the cracking of aliphatic side chains of PAHs and the decrease of phenolic activation energy.While under the catalysis of 5Mo-Z5,the relative content of aromatics and BTXN augmented by 5.75%and 4.02%,respectively.In addition,the highest relative content of aromatics reaches 70.09%when the loading rate of Fe was 7%(mass),and the relative content of BTXN increases from 25.87%to 29.42%.The results demonstrate that the active sites provided by different metal species expressed diverse effects on BTXN.Although the Bronsted/Lewis acid ratios of HZSM-5 modified by metal decreased,the acid strength and the relative content of BTXN both increased,which illustrated that there is a synergistic catalysis with the Bronsted acid sites and Lewis acid sites provided by metal species.In general,the performance of the catalyst is affected by the pore structure,acidity and metal active sites.Moreover,the possible formation mechanism of BTXN derived from asphaltenes catalytic pyrolysis was proposed on the basis of structural features and catalytic performances of a series of zeolites.Linyang Wang Qiang Wang Yongqi Liu Qiuxiang Yao Ming Sun Xiaoxun Ma 2022Chinese Journal of Chemical Engineering2022,35,9:1
16Review of PPP-RTK:achievements,challenges,and opportunities显示文摘The PPP–RTK method,which combines the concepts of Precise of Point Positioning(PPP)and Real-Time Kinematic(RTK),is proposed to provide a centimeter-accuracy positioning service for an unlimited number of users.Recently,the PPP–RTK technique is becoming a promising tool for emerging applications such as autonomous vehicles and unmanned logistics as it has several advantages including high precision,full flexibility,and good privacy.This paper gives a detailed review of PPP–RTK focusing on its implementation methods,recent achievements as well as challenges and opportunities.Firstly,the fundamental approach to implement PPP–RTK is described and an overview of the research on key techniques,such as Uncalibrated Phase Delay(UPD)estimation,precise atmospheric correction retrieval and modeling,and fast PPP ambiguity resolution,is given.Then,the recent efforts and progress are addressed,such as improving the performance of PPP–RTK by combining multi-GNSS and multi-frequency observations,single-frequency PPP–RTK for low-cost devices,and PPP–RTK for vehicle navigation.Also,the system construction and applications based on the PPP–RTK method are summarized.Moreover,the main issues that impact PPP–RTK performance are highlighted,including signal occlusion in complex urban areas and atmosphere modeling in extreme weather events.The new opportunities brought by the rapid development of low-cost markets,multiple sensors,and new-generation Low Earth Orbit(LEO)navigation constellation are also discussed.Finally,the paper concludes with some comments and the prospects for future research.Xingxing Li Jiaxin Huang Xin Li Zhiheng Shen Junjie Han Linyang Li Bo Wang 2022Satellite Navigation2022,3,3:1
