1Comparison of sentinel lymph node detection performances using blue dye in conjunction with indocyanine green or radioisotope in breast cancer patients: a prospective singlecenter randomized study显示文摘Objective:This randomized study aimed to compare the clinical efficacy between the novel dual tracer composed of indocyanine green(ICG)and blue dye(BD)and the conventional dual tracer composed of radioisotope and BD for sentinel lymph node(SLN)mapping in patients with breast cancer.Methods:This study enrolled 471 clinically lymph node-negative patients with primary breast cancer.All patients underwent mastectomy,and those undergoing sentinel lymph node biopsy(SLNB)were randomized to receive blue dye plus radioisotope(RB group)or BD plus ICG(IB group).The detection performances on SLN identification rate,positive SLN counts,detection sensitivity,and false-negative rate were compared between the two groups.Results:In the IB group,97%(194/200)of the patients who underwent the ICG and BD dual tracer injection showed fluorescentpositive lymphatic vessels within 2–5 min.The identification rate of SLNs was comparable between the IB group(99.0%,198/200)and the RB group(99.6%,270/271)(P=0.79).No significant differences were observed in the identification rate of metastatic SLNs(22.5%vs.22.9%,P>0.05,RB group vs.IB group,the same below),positive SLN counts(3.72±2.28 vs.3.91±2.13,P>0.05),positive metastatic SLN counts(0.38±0.84 vs.0.34±0.78,P>0.05),SLNB detection sensitivity(94.4%vs.92.5%,P>0.05),or false-negative rate(5.6%vs.7.5%,P>0.05)between the two groups.Conclusions:ICG can be used as a promising alternative tracer for radioisotope in SLN mapping,and when it is combined with BD in lymphangiography,it offers comparable detection sensitivity compared to the conventional lymphatic mapping strategies that are widely used in clinical practice.Long Yuan Xiaowei Qi Yi Zhang Xinhua Yang Fan Zhang Linjun Fan Li Chen Kongyong Zhang Ling Zhong Yanling Li Sijie Gan Wenying Fu Jun Jiang 2018Cancer Biology & Medicine2018,15,4:9
2CO2 capture over molecular basket sorbents:Effects of SiO2 supports and PEG additive显示文摘The objective of this work is to study the influences of silica supports and PEG additive on the sorption performance of molecular basket sorbent(MBS) for CO_2 capture consisting of polyethylenimine and one of the following supports: SBA-15(2-D structure), TUD-1(3-D sponge-like structure) and fumed silica HS-5(3-D disordered structure). Effects of the supports regarding pore structures and pore properties, the PEI loading amount as well as the sorption temperature were examined. Furthermore, polyethylene glycol(PEG) was introduced as an additive into the sorbents and its effect was investigated at different PEI loadings and sorption temperatures. The results suggest that the pore properties of MBS(after PEI loading) play a more important role in the CO_2 sorption capacity, rather than those of the supports alone.MBS with 3D pore structure exhibits higher CO_2 sorption capacity and amine efficiency than those with 2D-structured support. Among the sorbents studied, fumed silica(HS-5) based MBS showed the highest CO_2 sorption capacity in the temperature range of 30-95 °C, probably due to its unique interstitial pores formed by the aggregation of polymer-loaded SiO_2 particles. It was found that the temperature dependence is directly related to the PEI surface coverage layers. The more PEI surface coverage layers, the higher diffusion barrier for CO_2 and the stronger temperature dependence of CO_2 capacity. 3D MBS exceeds 2D MBS at the same PEI coverage layers due to lower diffusion barrier. 4Injectable self-healing hydrogel with siRNA delivery property for sustained STING silencing and enhanced therapy of intervertebral disc degeneration显示文摘Inflammatory responses of nucleus pulposus(NP)can induce imbalanced anabolism and catabolism of extracellular matrix,and the cytosolic dsDNA accumulation and STING-NF-κB pathway activation found in NP inflammation are considered as fairly important cause of intervertebral disc(IVD)degeneration.Herein,we constructed a siSTING delivery hydrogel of aldehyde hyaluronic acid(HA-CHO)and poly(amidoamine)PAMAM/siRNA complex to intervene the abnormal STING signal for IVD degeneration treatment,where the formation of dynamic Schiff base bonds in the system(siSTING@HPgel)was able to overcome the shortcomings such as low cellular uptake,short half-life,and rapid degradation of siRNA-based strategy.PAMAM not only formed complexes with siRNA to promote siRNA transfection,but also served as dynamic crosslinker to construct hydrogel,and the injectable and self-healing hydrogel efficiently and steadily silenced STING expression in NP cells.Finally,the siSTING@HPgel significantly eased IVD inflammation and slowed IVD degeneration by prolonging STING knockdown in puncture-induced IVD degeneration rat model,revealing that STING pathway was a therapeutic target for IVD degeneration and such novel hydrogel had great potential for being applied to many other diseases for gene delivery.Jiaxin Chen Haifeng Zhu Yutao Zhu Chenchen Zhao Shengyu Wang Yixin Zheng Ziang Xie Yang Jin Honghai Song Linjun Yang Jin Zhang Jiayong Dai Zhijun Hu Huaiyu Wang 2022Bioactive Materials2022,7,3:2
5Fractal Uniform Circular Arrays Based Multi-Orbital-Angular-Momentum-Mode Multiplexing Vortex Radio MIMO显示文摘A radio wave driven by Orbital angular momentum(OAM) is called a vortex radio and has a helical wavefront. The differential helical wavefronts of several vortex radios are closely related to their topological charges or mode numbers. In physics, two or more radio waves with different mode numbers are orthogonal to their azimuth angles. With the development of radio communication technologies, some researchers have been exploring the OAM-based multi-mode multiplexing(multi-OAM-mode multiplexing) technologies in order to enhance the channel spectrum efficiency(SE) of a radio communication system by using the orthogonal properties of vortex radios. After reviewing the reported researches of OAM-based radio communication, we find that some breakthroughs have been made in the combination of OAM and traditional Multi-Input-Multi-Output(MIMO). However, the existing technology is not sufficient to support OAM-based MIMO system to achieve maximum the channel SE. To maximize the spectrum efficiency of OAM-based MIMO system, we present a reused multi-OAM-mode multiplexing vortex radio(RMMVR) MIMO system, which is based on fractal uniform cir-cular arrays(UCAs). The scheme described in this study can effectively combine multiOAM-mode multiplexing with MIMO spatial multiplexing. First, we present the generation of RMMVR MIMO signals. Second, under line-of-sight(LOS) propagation conditions, we derive the channels of the RMMVR MIMO system. Third, we separate the RMMVR MIMO signals using an orthogonal separation method based on full azimuth sampling. Finally, we introduce the method for calculating the channel capacity of the RMMVR MIMO system. Theoretical analysis shows that the scheme proposed in this study is feasible. Moreover, the simulation results show that spatial and mode diversity are obtained by exploiting fractal UCAs. However, to enhance the channel SE of RMMVR MIMO system, an interference cancellation method needs to be introduced for zero-mode vortex radios, and some methods of multi-OAM-mode beams convergence and mode power optimization strategy should be introduced in the future.Linjun Zhao Hailin Zhang Wenchi Cheng 2018China Communications2018,15,9:2
6Emerging phagocytosis checkpoints in cancer immunotherapy显示文摘Cancer immunotherapy,mainly including immune checkpoints-targeted therapy and the adoptive transfer of engineered immune cells,has revolutionized the oncology landscape as it utilizes patients’own immune systems in combating the cancer cells.Cancer cells escape immune surveillance by hijacking the corresponding inhibitory pathways via overexpressing checkpoint genes.Phagocytosis checkpoints,such as CD47,CD24,MHC-I,PD-L1,STC-1 and GD2,have emerged as essential checkpoints for cancer immunotherapy by functioning as“don’t eat me”signals or interacting with“eat me”signals to suppress immune responses.Phagocytosis checkpoints link innate immunity and adaptive immunity in cancer immunotherapy.Genetic ablation of these phagocytosis checkpoints,as well as blockade of their signaling pathways,robustly augments phagocytosis and reduces tumor size.Among all phagocytosis checkpoints,CD47 is the most thoroughly studied and has emerged as a rising star among targets for cancer treatment.CD47-targeting antibodies and inhibitors have been investigated in various preclinical and clinical trials.However,anemia and thrombocytopenia appear to be formidable challenges since CD47 is ubiquitously expressed on erythrocytes.Here,we review the reported phagocytosis checkpoints by discussing their mechanisms and functions in cancer immunotherapy,highlight clinical progress in targeting these checkpoints and discuss challenges and potential solutions to smooth the way for combination immunotherapeutic strategies that involve both innate and adaptive immune responses.Yu’e Liu Yanjin Wang Yanrong Yang Linjun Weng Qi Wu Jin Zhang Pengcheng Zhao Lan Fang Yufeng Shi Ping Wang 2023Signal Transduction and Targeted Therapy2023,8,4:1

12FGF8-mediated signaling regulates tooth developmental pace during odontogenesis显示文摘The developing human and mouse teeth constitute an ideal model system to study the regulatory mechanism underlying organ growth control since their teeth share highly conserved and well-characterized developmental processes, and their developmental tempo varies notably. In the current study, we manipulated heterogenous recombination between human and mouse dental tissues and demonstrated that the dental mesenchyme dominates the tooth developmental tempo and FGF8 could be a critical player during this developmental process. Forced activation of FGF8 signaling in the dental mesenchyme of mice promoted cell proliferation, prevented cell apoptosis via p38 and perhaps PI3 K-Akt intracellular signaling,and impelled the transition of the cell cycle from G1-to S-phase in the tooth germ, resulting in the slowdown of the tooth developmental pace.
