1OSBPL2-disrupted pigs recapitulate dual features of human hearing loss and hypercholesterolaemia显示文摘Oxysterol binding protein like 2(OSBPL2), an important regulator in cellular lipid metabolism and transport, was identified as a novel deafness-causal gene in our previous work. To resemble the phenotypic features of OSBPL2 mutation in animal models and elucidate the potential genotypephenotype associations, the OSBPL2-disrupted Bama miniature(BM) pig model was constructed using CRISPR/Cas9-mediated gene editing, somatic cell nuclear transfer(SCNT) and embryo transplantation approaches, and then subjected to phenotypic characterization of auditory function and serum lipid profiles. The OSBPL2-disrupted pigs displayed progressive hearing loss(HL) with degeneration/apoptosis of cochlea hair cells(HCs) and morphological abnormalities in HC stereocilia, as well as hypercholesterolaemia. High-fat diet(HFD) feeding aggravated the development of HL and led to more severe hypercholesterolaemia. 2Design of task priority model and algorithm for imaging observation problem显示文摘In the imaging observation system, imaging task scheduling is an important topic. Most scholars study the imaging task scheduling from the perspective of static priority, and only a few from the perspective of dynamic priority. However,the priority of the imaging task is dynamic in actual engineering. To supplement the research on imaging observation, this paper proposes the task priority model, dynamic scheduling strategy and Heuristic algorithm. At first, this paper analyzes the relevant theoretical basis of imaging observation, decomposes the task priority into four parts, including target priority, imaging task priority, track, telemetry & control(TT&C)requirement priority and data transmission requirement priority, summarizes the attribute factors that affect the above four types of priority in detail, and designs the corresponding priority model. Then, this paper takes the emergency tasks scheduling problem as the background, proposes the dynamic scheduling strategy and heuristic algorithm. 16Inactivity of YGL082W in vitro due to impairment of conformational change in the catalytic center loop显示文摘MINDY-1 is a recently discovered new family of deubiquitinating enzymes(DUB),but one of its yeast homologs,YGL082 W,does not show any DUB activity in vitro.Sequence alignment shows that YGL082 W possesses the correct catalytic triad,and yet did not catalyze either the hydrolysis of di-ubiquitin,crosslinking with C-terminally propargylated ubiquitin,or hydrolysis of ubiquitin-7-amino-4-methylcoumarin.After obtaining a crystal structure of the catalytic domain of YGL082 W,we identified an interesting difference between the catalytic center loop of YGL082 W and that of its human homolog MINDY-1.Because the conformation of the catalytic center loop was previously reported to be important for the deubiquitination activity of MINDY-1,we hypothesized that Glu27(instead of the corresponding Pro136 in MINDY-1) of the catalytic center loop of YGL082 W may impair the conformational change and account for the lack of activity.This hypothesis was supported by homology modeling and molecular dynamics simulations,which showed that the Pro-to-Glu mutation(P136 E mutation for MINDY-1) creates a hydrogen bond that inhibits the conformation change of the catalytic center loop of MINDY-1.Further experiments through site-directed mutation validated this hypothesis,showing that the P27 E mutation caused MIY1(a homologous active DUB from yeast) to lose activity.Lining Lu Yu Guo Tian Wang Lujun Liang Suwen Zhao Feng Wang Lei Liu 2020Science China Chemistry2020,63,2:1

20Zinc homeostasis in the metabolic syndrome and diabetes显示文摘Zinc(Zn)is an essential mineral that is required for various cellular functions.Zn dyshomeostasis always is related to certain disorders such as metabolic syndrome,diabetes and diabetic complications.The associations of Zn with metabolic syndrome,diabetes and diabetic complications,thus,stem from the multiple roles of Zn:(1)a constructive component of many important enzymes or proteins,(2)a requirement for insulin storage and secretion,(3)a direct or indirect antioxidant action,and(4)an insulin-like action.However,whether there is a clear cause-and-effect relationship of Zn with metabolic syndrome,diabetes,or diabetic complications remains unclear.In fact,it is known that Zn deficiency is a common phenomenon in diabetic patients.Chronic low intake of Zn was associated with the increased risk of diabetes and diabetes also impairs Zn metabolism.Theoretically Zn supplementation should prevent the metabolic syndrome,diabetes,and diabetic complications;however,limited available data are not always supportive of the above notion.Therefore,this review has tried to summarize these pieces of available information,possible mechanisms by which Zn prevents the metabolic syndrome,diabetes,and diabetic complications.In the final part,what are the current issues for Zn supplementation were also discussed.Xiao Miao Weixia Sun Yaowen Fu Lining Miao Lu Cai 2013Frontiers of Medicine2013,7,1:1
