| 1 | The principle of measuring unusual change of underground mass by optical astrometric instrument显示文摘Optical observations from a ground-based astrometric instrument provide a unique set of data sensitive to variations of local plumb line, which represent the change of local gravitational field on the ground. The local gravitational change could be caused by density variation or movement of underground mass in addition to tidal action. Earthquake was found to be related with gravitational change. A seismic zone could have abnormal underground material density, causing the variation of plume line on the ground. In this frame, the astrometric telescope can measure the variation of local plumb line by observing the local astronomical longitudes and latitudes from stars in the Hipparcos catalog. In this study, we estimate the deflection angle of the plumb line on a ground site, and give a relation between the angle, abnormal mass and site distance (depth and horizontal distance). Then we derive the abnormality of underground material density using the plumb lines measured at different sites, and study the earthquake gestation, development and occurrence. Using the deflection angles of plumb lines observed at two sites, we give a method to calculate the mass and the center of gravity of underground materials. | 10 | Functional divergence of oligoadenylate synthetase 1(OAS1)proteins in Tetrapods显示文摘OASs play critical roles in immune response against virus infection by polymerizing ATP into 2-5 As,which initiate the classical OAS/RNase L pathway and induce degradation of viral RNA.OAS members are functionally diverged in four known innate immune pathways(OAS/RNase L,OASL/IRF7,OASL/RIG-I,and OASL/cGAS),but how they functionally diverged is unclear.Here,we focus on evolutionary patterns and explore the link between evolutionary processes and functional divergence of Tetrapod OAS1.We show that Palaeognathae and Primate OAS1 genes are conserved in genomic and protein structures but differ in function.The former(i.e.,ostrich)efficiently synthesized long 2-5 A and activated RNase L,while the latter(i.e.,human)synthesized short 2-5 A and did not activate RNase L.We predicted and verified that two in-frame indels and one positively selected site in the active site pocket contributed to the functional divergence of Palaeognathae and Primate OAS1.Moreover,we discovered and validated that an in-frame indel in the C-terminus of Palaeognathae OAS1 affected the binding affinity of ds RNA and enzymatic activity,and contributed to the functional divergence of Palaeognathae OAS1 proteins.Our findings unravel the molecular mechanism for functional divergence and give insights into the emergence of novel functions in Tetrapod OAS1. | Xiaoxue Wang Jiaxiang Hu Linfei Song Enguang Rong Chenghuai Yang Xiaoyun Chen Juan Pu Honglei Sun Chuze Gao David WBurt Jinhua Liu Ning Li Yinhua Huang | 2022 | Science China(Life Sciences)2022,65,7: | 1 |

| 13 | Aptamer-functionalized nanoplatforms overcoming temozolomide resistance in synergistic chemo/photothermal therapy through alleviating tumor hypoxia显示文摘Tumor hypoxia is intimately associated with gliomas,which represents a significant threat to human health and are resistant to the first-line chemotherapeutic drug temozolomide(TMZ)due to hypoxia.In this work,to overcome TMZ resistance in orthotopic gliomas,aptamer-functionalized liposomes are manufactured to encapsulate TMZ and photothermal agent IR780,and can cross the blood-brain barrier and actively target gliomas.It is possible to employ liposomes for both fluorescence and photoacoustic imaging simultaneously due to their stability and excellent photothermal conversion capabilities.This chemo/photothermal synergistic therapeutic effect of liposomes on gliomas is demonstrated by their abilities to target orthotopic gliomas,alleviate tumor hypoxia and consequently reverse resistance of glioma cells to TMZ,thereby extending the survival time of tumor-bearing mice,making the nanoplatforms and their synergistic chemo/photothermal therapy as a potential clinical treatment for gliomas. | Yun Zeng Linfei Zhao Ke Li Jingwen Ma Dan Chen Changhu Liu Wenhua Zhan Yonghua Zhan | 2023 | Nano Research2023,16,7: | 0 |
