| 1 | Focal adhesion protein Kindlin-2 regulates bone homeostasis in mice显示文摘Our recent studies demonstrate that the focal adhesion protein Kindlin-2 is critical for chondrogenesis and early skeletal development. Here, we show that deleting Kindlin-2 from osteoblasts using the 2.3-kb mouse Col1 a1-Cre transgene minimally impacts bone mass in mice, but deleting Kindlin-2 using the 10-kb mouse Dmp1-Cre transgene, which targets osteocytes and mature osteoblasts, results in striking osteopenia in mice. Kindlin-2 loss reduces the osteoblastic population but increases the osteoclastic and adipocytic populations in the bone microenvironment. Kindlin-2 loss upregulates sclerostin in osteocytes,downregulates β-catenin in osteoblasts, and inhibits osteoblast formation and differentiation in vitro and in vivo. Upregulation ofβ-catenin in the mutant cells reverses the osteopenia induced by Kindlin-2 deficiency. Kindlin-2 loss additionally increases the expression of RANKL in osteocytes and increases osteoclast formation and bone resorption. Kindlin-2 deletion in osteocytes promotes osteoclast formation in osteocyte/bone marrow monocyte cocultures, which is significantly blocked by an anti-RANKLneutralizing antibody. | 2 | Potassium Uptake and Transport in Apple Roots Under Drought Stress显示文摘As one of the most important mineral nutrient elements, potassium(K^+) plays an important role in many plant physiological processes and determines both the yield and quality of crops. There are two typical gene families that regulate K^+transport in higher plants, including K^+channels and K^+transporters. However, little is known about how these channels and transporters divide their work in response to drought stress. In this study, the hydroponic experiment was conducted on Malus hupehensis. The K^+content was found to decrease in response to drought stress in M. hupehensis, the aboveground decreased by 34.15% and the underground decreased by 3.97%. Meanwhile, the root morphology change was detected by scanning the root system. Under conditions of drought, the genes encoding K^+transporters were upregulated including MdCHX1.3, MdCHX4.11, MdCHX4.8, MdCHX4.9, Md HKT1, and MdHAK3.2. The net influx of K^+was inhibited by 19.47% with the action of K^+channel inhibitors(CsCl), however a significant decrease(80.99%; P < 0.05) was found in roots exposed to a PM H+-ATPase(orthovanadate) inhibitor by utilizing a non-invasive micro-test technique. The trend of H+efflux was similar to that of K^+. The data suggested that the positive influx of K^+through the transporter accounted for the main K^+uptake under drought stress. These results suggest that we can improve the uptake of K^+by purposely up-regulating specific K^+transporters under drought stress. | 4 | The interaction between polyphyllin I and SQLE protein induces hepatotoxicity through SREBP-2/HMGCR/SQLE/LSS pathway显示文摘Polyphyllin I(PPI)and polyphyllin II(PII)are the main active substances in the Paris polyphylla.However,liver toxicity of these compounds has impeded their clinical application and the potential hepatotoxicity mechanisms remain to be elucidated.In this work,we found that PPI and PII exposure could induce significant hepatotoxicity in human liver cell line L-02 and zebrafish in a dose-dependent manner.The results of the proteomic analysis in L-02 cells and transcriptome in zebrafish indicated that the hepatotoxicity of PPI and PII was associated with the cholesterol biosynthetic pathway disorders,which were alleviated by the cholesterol biosynthesis inhibitor lovastatin.Additionally,3-hydroxy-3-methy-lglutaryl CoA reductase(HMGCR)and squalene epoxidase(SQLE),the two rate-limiting enzymes in the cholesterol synthesis,selected as the potential targets,were confirmed by the molecular docking,the overexpression,and knockdown of HMGCR or SQLE with siRNA.Finally,the pull-down and surface plasmon resonance technology revealed that PPI could directly bind with SQLE but not with HMGCR.Collectively,these data demonstrated that PPI-induced hepatotoxicity resulted from the direct binding with SQLE protein and impaired the sterol-regulatory element binding protein 2/HMGCR/SQLE/lanosterol synthase pathways,thus disturbing the cholesterol biosynthesis pathway.The findings of this research can contribute to a better understanding of the key role of SQLE as a potential target in drug-induced hepatotoxicity and provide a therapeutic strategy for the prevention of drug toxic effects with similar structures in the future. | Zhiqi Li Qiqi Fan Meilin Chen Ying Dong Farong Li Mingshuang Wang Yulin Gu Simin Guo Xianwen Ye Jiarui Wu Shengyun Dai Ruichao Lin Chongjun Zhao | 2023 | Journal of Pharmaceutical Analysis2023,13,1: | 2 |

| 6 | The Synthesis of Sub-Nano-Thick Pd Nanobelt-Based Materials for Enhanced Hydrogen Evolution Reaction Activity显示文摘Tailoring atomic structures of noblemetal nanomaterials with size close to single-unit cell range is essential in both fundamental researchand applications,including their development into high catalytic performance materials in renewable,green energy conversions,devices for energy storage,and as biosensors for environmental pollutants.However,several strategies used in fabricating these materials still impose enormous challenges,arising from lack of even size distribution,shape uniformity,and controlled composition,which are critical in determining their specific activities and efficiencies.Herein,we report a facile approach for preparing sub-nano-thick palladium nanobelt-based(PdNB)materials.Then we rationalized the formation mechanism of such highly anisotropic structures by morphology-related thermodynamic and kinetic analysis.Moreover,we investigated if electrocatalysis performance of these NB-basedmaterialswere enhanced.Thepalladium(Pd)NBs featured a thickness of∼0.9-1.2 nm and width of 5-18nmwith length extending to severalmicrometers[denoted as Pd(0.9)],or a thickness of∼0.7-0.9 nm and width of 2.5-6 nmwith length of several hundreds of nanometers[denoted as Pd(0.7)].According to our theoretical analysis,one-dimensional(1D)growth encountered almost no energy barrier at optimal reaction conditions,whereas the growth of Pd nanostructures with other dimensions confronted high barriers,indicating that it was plausible to prepare 1D structures with sizes close to single-unit cells.Also,platinum(Pt)could be successfully doped into the Pd(0.9)NBs through a galvanic epitaxial growth,forming edge-Pt-enriched Pd NBs(eePtPd NBs).Further,electron transfer from Pd to Pt imparted the eePtPd NBs with high hydrogen evolution reaction(HER)activity.The eePtPd NBs showed a 3.5 and 1.8 times higher in exchange current density and mass activity(at−0.1 V),respectively,compared to those of Pt catalysts in perchloric acid(HClO_(4))solutions.Finally,the NBs all showed high activity toward ethanol and formic acid oxidation reactions.Our current work aids in gaining insights into tailoring Pd nanostructures at an atomic level and provides Pd sub-nanometric 1D structures for further research.Moreover,our morphology-related thermodynamic and kinetic analysis extend our understanding of the control of nanostructure morphology and might shed light on the precision of designing specific morphologies of noble metal nanocrystal structures. | Bing Ni Qinghua Zhang Chen Ouyang Simin Zhang Biao Yu Jing Zhuang Lin Gu Xun Wang | 2020 | CCS Chemistry2020,2,1: | 1 |

| 10 | Tectonic features,genetic mechanisms and basin evolution of the eastern Doseo Basin,Chad显示文摘The features of the unconformity,fault and tectonic inversion in the eastern Doseo Basin,Chad,were analyzed,and the genetic mechanisms and basin evolution were discussed using seismic and drilling data.The following results are obtained.First,four stratigraphic unconformities,i.e.basement(Tg),Mangara Group(T10),lower Upper Cretaceous(T5)and Cretaceous(T4),four faulting stages,i.e.Barremian extensional faults,Aptian–Coniacian strike-slip faults,Campanian strike-slip faults,and Eocene strike-slip faults,and two tectonic inversions,i.e.Santonian and end of Cretaceous,were developed in the Doseo Basin.Second,the Doseo Basin was an early failed intracontinental passive rift basin transformed by the strike-slip movement and tectonic inversion.The initial rifting between the African and South American plates induced the nearly N-S stretching of the Doseo Basin,giving rise to the formation of the embryonic Doseo rift basin.The nearly E-W strike-slip movement of Borogop(F1)in the western section of the Central African Shear Zone resulted in the gradual cease of the near north-south rifting and long-term strike-slip transformation,forming a dextral transtension fault system with inherited activity but gradually weakened in intensity(interrupted by two tectonic inversions).This fault system was composed of the main shear(F1),R-type shear(F2-F3)and P-type shear(F4-F5)faults,with the strike-slip associated faults as branches.The strike-slip movements of F1 in Cretaceous and Eocene were controlled by the dextral shear opening of the equatorial south Atlantic and rapid expanding of the Indian Ocean,respectively.The combined function of the strike-slip movement of F1 and the convergence between Africa and Eurasia made the Doseo Basin underwent the Santonian dextral transpressional inversion characterized by intensive folding deformation leading to the echelon NE-SW and NNE-SSW nose-shaped uplifts and unconformity(T5)on high parts of the uplifts.The convergence between Africa and Eurasia caused the intensive tectonic inversion of Doseo Basin at the end of Cretaceous manifesting as intensive uplift,denudation and folding deformation,forming the regional unconformity(T4)and superposing a nearly E-W structural configuration on the Santonian structures.Third,the Doseo Basin experienced four evolutional stages with the features of short rifting and long depression,i.e.Barremian rifting,Aptian rifting–depression transition,Albian–Late Cretaceous depression,and Cenozoic extinction,under the control of the tectonic movements between Africa and its peripheral plates. | GAO Huahua DU Yebo WANG Lin GAO Simin HU Jie BAI Jianfeng MA Hong WANG Yuhua ZHANG Xinshun LIU Hao | 2023 | Petroleum Exploration and Development2023,50,5: | 0 |
| 11 | Detection and classification of breast lesions using multiple information on contrast-enhanced mammography by a multiprocess deep-learning system: A multicenter study显示文摘Objective: Accurate detection and classification of breast lesions in early stage is crucial to timely formulate effective treatments for patients. We aim to develop a fully automatic system to detect and classify breast lesions using multiple contrast-enhanced mammography(CEM) images.Methods: In this study, a total of 1,903 females who underwent CEM examination from three hospitals were enrolled as the training set, internal testing set, pooled external testing set and prospective testing set. Here we developed a CEM-based multiprocess detection and classification system(MDCS) to perform the task of detection and classification of breast lesions. In this system, we introduced an innovative auxiliary feature fusion(AFF)algorithm that could intelligently incorporates multiple types of information from CEM images. The average freeresponse receiver operating characteristic score(AFROC-Score) was presented to validate system’s detection performance, and the performance of classification was evaluated by area under the receiver operating characteristic curve(AUC). Furthermore, we assessed the diagnostic value of MDCS through visual analysis of disputed cases,comparing its performance and efficiency with that of radiologists and exploring whether it could augment radiologists’ performance.Results: On the pooled external and prospective testing sets, MDCS always maintained a high standalone performance, with AFROC-Scores of 0.953 and 0.963 for detection task, and AUCs for classification were 0.909[95% confidence interval(95% CI): 0.822-0.996] and 0.912(95% CI: 0.840-0.985), respectively. It also achieved higher sensitivity than all senior radiologists and higher specificity than all junior radiologists on pooled external and prospective testing sets. Moreover, MDCS performed superior diagnostic efficiency with an average reading time of 5 seconds, compared to the radiologists’ average reading time of 3.2 min. | 12 | Orthogonal carbazole-perylene bisimide pentad:a photoconversion-tunable photosensitizer with diversified excitation and excited-state relaxation pathways显示文摘Integrating multiple photosensitive properties into an“all-in-one”photosensitizer(PS)shows great promise for the treatment of cancers owing to synergistic effect among them.However,the development of such PSs,especially those that need a single laser source,remains a challenge.Herein,we report an orchestration of electron donors and acceptors in a propeller-like pentad,PBI-4Cz,where four carbazole(Cz)units are covalently linked to the ortho-positions of the perylene bisimide(PBI)core.Strong intramolecular donor-acceptor interaction significantly quenches the luminescence and largely extends the absorption spectra to near-infrared region.Excited-state dynamics investigated via femto-and nano-second transient absorption spectroscopy revealed exclusive charge separation of the PBI-4Cz within initial 0.5 ps when photoexcited regardless of which intermediate is involved.Energy dissipation of the resulting charge-separated state(PBI^(•−)-4Cz^(•−))is subjected to the toggle between intersystem-crossing toward excited triplet states and charge recombination toward ground states.Relative importance of the two pathways can be tuned by micro-environmental polarity,which endows PBI-4Cz remarkable performances of singlet-oxygen generation(>90.0%)in toluene and photothermal conversion(∼28.6%)in DMSO.Harnessing intrinsic photostability and excited-state processes of heavy-atom-free PBI derivatives not only holds a promise for multifunctional phototheranostics,but also provides a prototype for designing high-performance PSs with tunable photoconversion pathways. | Zhaolong Wang Yue Sun Simin Lin Gang Wang Xingmao Chang Xinyu Gou Taihong Liu Shengye Jin Gang He Yu-Chen Wei Pi-Tai Chou Yu Fang | 2021 | Science China Chemistry2021,64,12: | 0 |
| 13 | Mechanisms of high-frequency dinoflagellate blooms of Scrippsiella trochoidea in Daya Bay,South China Sea显示文摘Harmful algal blooms(HABs),such as dinoflagellate blooms,have adverse eff ects on coastal water environments,causing seafood contamination and aquaculture economic losses.Based on historical HAB records data(1983-2017)and phytoplankton data(1982-2008),field investigation and laboratory experiments on frequently bloomed dinoflagellate species,we analyzed the possible mechanisms of high frequency dinoflagellate blooms in Daya Bay,South China Sea.The HAB records data show that the proportion of dinoflagellate blooms increased from 15% before 1994 to 77.27% after 2011.The bloom occurred mostly in summer(57.58%)in Yaling Bay(76.92%).The species inducing dinoflagellate blooms tended to be singular over time,with high proportion of Scrippsiella trochoidea(42.86%-88.88%).Laboratory experiment and field investigation of S.trochoidea bloom showed that this species grew rapidly at 30℃ and 32.1℃(r=0.47,P<0.05),respectively.Results indicate that temperature rise in Yaling Bay was aff ected by nearby nuclear plant effluent,which favored the S.trochoidea bloom.Coincidently,nearly 70% of S.trochoidea blooms occurred in about 7 days after local spring tide and strong wind(wind velocity>7 m/s),which disturbed the resting cysts in the sediment and bring them into surface layer.Taking into account of environmental parameters,algae source,and atmospheric conditions,the probable mechanism of S.trochoidea bloom could be explained.The cysts were resuspended by the spring tide or strong wind,germinated at the bottom layer,reproduced rapidly at higher temperature,and then aggregated in the inner bay(Yaling Bay),resulting the frequent S.trochoidea bloom.Considering the influences of global warming and human activities,it should be taken more attention that dinoflagellate blooms might increase along the coastal waters. | Yuan TIAN Simin HU Xianzhi LIN Hui HUANG Xingyu SONG Yan YAN Xuedong XIE Tao LI Sheng LIU | 2021 | Journal of Oceanology and Limnology2021,39,4: | 0 |

| 15 | A machine learning-assisted multifunctional tactile sensor for smart prosthetics显示文摘The absence of tactile perception limits the dexterity of a prosthetic hand and its acceptance by amputees.Recreating the sensing properties of the skin using a flexible tactile sensor could have profound implications for prosthetics,whereas existing tactile sensors often have limited functionality with cross-interference.In this study,we propose a machine-learning-assisted multifunctional tactile sensor for smart prosthetics,providing a human-like tactile sensing approach for amputations.This flexible sensor is based on a poly(3,4-ethylenedioxythiophene):poly(styrene sulfonate)(PEDOT:PSS)-melamine sponge,which enables the detection of force and temperature with low cross-coupling owing to two separate sensing mechanisms:the open-circuit voltage of the sensor as a force-insensitive intrinsic variable to measure the absolute temperature and the resistance as a temperature-insensitive extrinsic variable to measure force.Furthermore,by analyzing the unsteady heat conduction and characterizing it using real-time thermal imaging,we demonstrated that the process of open-circuit voltage variation resulting from the unsteady heat conduction is closely correlated with the heatconducting capabilities of materials,which can be utilized to discriminate between substances.Assisted by the decision tree algorithm,the device is endowed with thermal conductivity sensing ability,which allows it to identify 10 types of substances with an accuracy of 94.7%.Furthermore,an individual wearing an advanced myoelectric prosthesis equipped with the above sensor can sense pressure,temperature,and recognize different materials.We demonstrated that our multifunctional tactile sensor provides a new strategy to help amputees feel force,temperature and identify the material of objects without the aid of vision. | Yue Li Lin Yang Shihao Deng Hong Huang Yingyi Wang Zuoping Xiong Simin Feng Shuqi Wang Tie Li Ting Zhang | 2023 | InfoMat2023,5,9: | 0 |
| 16 | Genetic dissection and validation of a major QTL for grain weight on chromosome 3B in bread wheat(Triticum aestivum L.)显示文摘Grain weight is one of the key components of wheat(Triticum aestivum L.)yield.Genetic manipulation of grain weight is an efficient approach for improving yield potential in breeding programs.A recombinant inbred line(RIL)population derived from a cross between W7268 and Chuanyu 12(CY12)was employed to detect quantitative trait loci(QTLs)for thousand-grain weight(TGW),grain length(GL),grain width(GW),and the ratio of grain length to width(GLW)in six environments.Seven major QTLs,QGl.cib-2D,QGw.cib-2D,QGw.cib-3B,QGw.cib-4B.1,QGlw.cib-2D.1,QTgw.cib-2D.1 and QTgw.cib-3B.1,were consistently identified in at least four environments and the best linear unbiased estimation(BLUE)datasets,and they explained 2.61 to 34.85%of the phenotypic variance.Significant interactions were detected between the two major TGW QTLs and three major GW loci.In addition,QTgw.cib-3B.1 and QGw.cib-3B were co-located,and the improved TGW at this locus was contributed by GW.Unlike other loci,QTgw.cib-3B.1/QGw.cib-3B had no effect on grain number per spike(GNS).They were further validated in advanced lines using Kompetitive Allele Specific PCR(KASP)markers,and a comparison analysis indicated that QTgw.cib-3B.1/QGw.cib-3B is likely a novel locus.Six haplotypes were identified in the region of this QTL and their distribution frequencies varied between the landraces and cultivars.According to gene annotation,spatial expression patterns,ortholog analysis and sequence variation,the candidate gene of QTgw.cib-3B.1/QGw.cib-3B was predicted.Collectively,the major QTLs and KASP markers reported here provide valuable information for elucidating the genetic architecture of grain weight and for molecular marker-assisted breeding in grain yield improvement. | Simin Liao Zhibin Xu Xiaoli Fan Qiang Zhou Xiaofeng Liu Cheng Jiang Liangen Chen Dian Lin Bo Feng Tao Wang | 2024 | Journal of Integrative Agriculture2024,23,1: | 0 |
| 17 | 3D Joint inversion of gravity and gravity tensor data显示文摘The processing and interpretation of gravity and gradient data plays an important role in geophysics.The cross gradient joint inversion is usually used for achieving structure coupling of multiple geophysical models. In order to realize the coupling of gravity and gravity tensor data,the authors analyzed each component.The results show that different types of data contain different direction information,and derived the joint inversion based on cross gradient function and applied it to model data. | 18 | Biological Tissue-Inspired Ultrasoft,Ultrathin,and Mechanically Enhanced Microfiber Composite Hydrogel for Flexible Bioelectronics显示文摘Hydrogels offer tissue-like softness,stretchability,fracture toughness,ionic conductivity,and compatibility with biological tissues,which make them promising candidates for fabricating flexible bioelectronics.A soft hydrogel film offers an ideal interface to directly bridge thin-film electronics with the soft tissues.However,it remains difficult to fabricate a soft hydrogel film with an ultrathin configuration and excellent mechanical strength.Here we report a biological tissue-inspired ultrasoft microfiber composite ultrathin(<5μm)hydrogel film,which is currently the thinnest hydrogel film as far as we know.The embedded microfibers endow the composite hydrogel with prominent mechanical strength(tensile stress~6 MPa)and anti-tearing property.Moreover,our microfiber composite hydrogel offers the capability of tunable mechanical properties in a broad range,allowing for matching the modulus of most biological tissues and organs.The incorporation of glycerol and salt ions imparts the microfiber composite hydrogel with high ionic conductivity and prominent anti-dehydration behavior.Such microfiber composite hydrogels are promising for constructing attaching-type flexible bioelectronics to monitor biosignals. | Qiang Gao Fuqin Sun Yue Li Lianhui Li Mengyuan Liu Shuqi Wang Yongfeng Wang Tie Li Lin Liu Simin Feng Xiaowei Wang Seema Agarwal Ting Zhang | 2023 | Nano-Micro Letters2023,15,9: | 0 |
